T. Ema, D. Tanida, T. Sakai, Versatile and practical chiral shift reagent with hydrogen-bond donor/acceptor sites in a macrocyclic cavity. Org. Lett., 8(17), 3773-3775 (2006)に従って、酸クロリド(R)-3を合成した。酸クロリド(R)-3(785mg、1.79mmol)の乾燥テトラヒドロフラン溶液(180mL)と化合物2(654mg、1.44mmol)ならびにトリエチルアミン(0.4mL、2.9mmol)の乾燥テトラヒドロフラン溶液(180mL)を乾燥テトラヒドロフラン(120mL)に室温下でそれぞれ同じ速度で3時間半かけて滴下した。得られた混合液をさらに10時間攪拌し、前記混合液からエバポレーターによって揮発性物質を取り除いた。得られた固体残渣を塩化メチレンに溶かし、この溶液を飽和重曹水(40mL)で洗浄し、硫酸ナトリウムで乾燥後、減圧濃縮し、固体の生成物を得た。生成物はシリカゲルカラムクロマトグラフィー(塩化メチレン/テトラヒドロフラン(塩化メチレン:テトラヒドロフラン(体積比)=20:1))で精製した。得られた精製物を塩化メチレンから再結晶することにより、化合物(R)-4(753mg、収率64%)を白色結晶として得た。得られた化合物(R)-4のスペクトルデータを以下に示す。

化合物(R)-6(1.04g、1.23mmol)をトリフルオロ酢酸(1.9mL、25mmol)と乾燥塩化メチレン(0.6mL)の混合溶液に溶かして室温、窒素雰囲気下で18時間攪拌した。得られた反応液から溶媒を留去後、真空乾燥して、化合物(R)-6のt-ブチル基が水素に置き換わった化合物(R)-6のカルボン酸を得た。110℃で4時間半真空乾燥した後に窒素下で放冷しておいたアミノプロピルシリカゲル(4.11g)へ、化合物(R)-6のカルボン酸(1.56g、1.97mmol)を溶かした乾燥ジメチルホルムアミド(12mL)と乾燥テトラヒドロフラン(5.4mL)の混合溶液、1,3-ジシクロヘキシルカルボジイミド(DCC、621mg、3.01mmol) ならびに1-ヒドロキシベンゾトリアゾール(HOBT、411mg、3.04mmol)を加えた。得られたスラリーをメカニカルスターラーで室温下48時間攪拌後、DCC(619mg、3.00mmol)、HOBT(411mg、3.04mmol)、酢酸(0.35mL、6.1mmol)を前記スラリーに加えてさらに48時間攪拌した。得られたスラリーを濾過して得られるシリカゲルをテトラヒドロフラン、ジメチルホルムアミド、エタノール、熱エタノールの順序で洗浄し、真空乾燥することにより、CSP-1を白色粉末として得た。
以下に示す合成経路でキラル固定相CSP-2を調製した。

Mater., 15(13), 2534-2542 (2003)に従って、5-tert-ブチルオキシカルボニルイソフタル酸を合成した。5-tert-ブチルオキシカルボニルイソフタル酸(1.60g、6.01mmol)の乾燥塩化メチレンの懸濁液(60mL)に塩化オキサリル(2.0mL、23mmol)、ジメチルホルムアミド(1滴)を加えて室温下5時間攪拌した。得られた混合液からエバポレーターによって揮発性物質を取り除き、得られた固体を3時間真空乾燥した。得られた5-tert-ブチルオキシカルボニルイソフタル酸の酸クロリドはこれ以上精製せずに使用した。

化合物(R)-8(542mg、0.665mmol)をトリフルオロ酢酸(1.1 mL、14mmol)と乾燥塩化メチレン(0.5mL)の混合溶液に溶かして室温、窒素雰囲気下で4時間半攪拌した。得られた混合液から溶媒を留去後、得られた固体の生成物を真空乾燥して、化合物(R)-8のt-ブチル基が水素に置き換わった化合物(R)-8のカルボン酸を得た。150℃で4時間真空乾燥した後に窒素下で放冷しておいたアミノプロピルシリカゲル(2.99g)へ、化合物(R)-8のカルボン酸(1.11g、1.46mmol)を溶かした乾燥ジメチルホルムアミド(3.5mL)と乾燥テトラヒドロフラン(9.5mL)の混合溶液、DCC(459mg、2.23mmol)ならびにHOBT(301mg、2.23mmol)を加えた。得られたスラリーをメカニカルスターラーで室温下48時間攪拌した後、DCC(463mg、2.25mmol)、HOBT(302mg、2.23mmol)、酢酸(0.23mL、4.0mmol)を前記スラリーに加えてさらに24時間攪拌した。得られたスラリーを濾過して得られるシリカゲルをテトラヒドロフラン、エタノール、熱エタノールの順序で洗浄し、真空乾燥することにより、CSP-2を白色粉末として得た。
[CSP-1を用いたHPLC用カラムの作製]
CSP-1(3.5g)をメタノールに分散し、得られたスラリーを用いて、φ0.46cm×25cmLのステンレス製カラムにスラリー方式のカラム充填を行い、CSP-1が充填されてなるカラムを作製した。

下記条件でHPLCにて7種のサンプル(化合物番号:1、2、3、4、5、6、7)用いて不斉識別能力の評価を実施した。評価結果を表1に示す。また各評価サンプルのそれぞれのクロマトグラムを図1~7に示す。なお、表中、t0はデッドタイム(min)、t1はより弱く保持される対掌体の溶出時間(min)t2はより強く保持される対掌体の溶出時間(min)、k1’はより弱く保持される対掌体の保持係数、k2’はより強く保持される対掌体の保持係数を表す。

[超臨界流体クロマトグラフィー(SFC)による光学異性体の分離]
実施例3により得られたカラムを用いて、以下の超臨界もしくは亜臨界評価条件下、CSP-1の不斉識別能力の評価を行った。評価用サンプルには、先述の化合物3及び6を用いた。以下の評価結果における保持係数(k’)、分離係数(α)はそれぞれ先述と同じく定義づけられる。デッドタイムも先述と同じくトリターシャリブチルベンゼンの溶出時間である。
下記4条件でSFCにて2種サンプル(化合物番号:3、6)を用いて不斉識別能力の評価を実施した。サンプル3の評価結果を表9に、サンプル6の評価結果を表10に示す。また各評価サンプル、条件でのクロマトグラムを図48~図55に示す。なお、表中、t0はデッドタイム(min.)、t1はより弱く保持される対掌体の溶出時間(min.)、t2はより強く保持される対掌体の溶出時間(min.)、k1'はより弱く保持される対掌体の保持係数、k2'はより強く保持される対掌体の保持係数を表す。また下記表中、CO2は二酸化炭素、IPAは2-プロパノール、MeCNはアセトニトリル、EtOHはエタノールを表す。

以下に示す合成経路でキラル固定相CSP-3を調製した。
T. Ema, D. Tanida, K. Hamada, T. Sakai, Tuning the chiral cavity of macrocyclic receptor for chiral recognition and discrimination. J. Org. Chem., 73(22), 9129-9132 (2008) に従って、酸クロリド(R)-9を合成した。酸クロリド(R)-9(594mg、1.13mmol)の乾燥テトラヒドロフラン溶液(120mL)と化合物7(408mg、0.190mmol)ならびにトリエチルアミン(0.26mL、1.9mmol)の乾燥テトラヒドロフラン溶液(120mL)とを、乾燥テトラヒドロフラン(70mL)に室温下でそれぞれ同じ速度で3時間半かけて滴下した。得られた混合物をさらに12時間攪拌し、前記混合物からエバポレーターによって揮発性物質を取り除いた。得られた固体残渣を塩化メチレンに溶解し、この溶液を飽和重曹水(40mL)で洗浄し、硫酸ナトリウムで乾燥後、減圧濃縮し、固体の生成物を得た。得られた生成物をシリカゲルカラムクロマトグラフィー(塩化メチレン/テトラヒドロフラン(塩化メチレン:テトラヒドロフラン(体積比)(20:1))で精製することにより、化合物(R)-10(654mg、収率77%)を白色固体として得た。得られた化合物(R)-10のスペクトルデータを以下に示す。

化合物(R)-10(1.15g、1.24mmol)のトリフルオロ酢酸溶液(3.1mL、40mmol)を室温、窒素雰囲気下で7時間半攪拌した。得られた混合液から溶媒を留去後、得られた固体の生成物を真空乾燥して、化合物(R)-10のt-ブチル基が水素に置き換わった、化合物(R)-10のカルボン酸を得た。150 ℃で4時間真空乾燥した後に窒素下で放冷しておいた3-アミノプロピルシリカゲル(2.47g)へ、化合物(R)-10のカルボン酸(1.08g、1.24mmol)を溶かした乾燥塩化メチレン(5mL)と乾燥テトラヒドロフラン(6mL)の混合溶液、DCC(405mg、1.96mmol)ならびにHOBT(257mg、1.90mmol)を加えた。得られたスラリーをメカニカルスターラーで室温下48時間攪拌後、DCC(407mg、1.97mmol)、HOBT(256mg、1.89mmol)、酢酸(0.20mL、3.5mmol)を前記スラリーに加えてさらに24時間攪拌した。得られたスラリーを濾過して得られるシリカゲルをテトラヒドロフラン、塩化メチレン、エタノール、熱エタノールの順序で洗浄し、真空乾燥することにより、CSP-3を白色粉末として得た。
以下に示す合成経路でキラル固定相CSP-4を調製した。
(R)-3,3'-ジブロモ-1,1'-ビ-2-ナフトール(433mg、0.974mmol)とブロモ酢酸t-ブチルエステル(0.35mL、2.4mmol)ならびに炭酸カリウム(337mg、2.44mmol)を入れたアセトン混合溶液(15mL)を12時間加熱還流した。得られた反応混合物を濾過し、得られた濾液から溶媒を留去し、固体の生成物を得た。得られた生成物をアルミナカラムクロマトグラフィー(ヘキサン/酢酸エチル(ヘキサン:酢酸エチル(体積比)=5:1))で精製することにより、化合物(R)-11(628mg、収率96%) を白色固体として得た。得られた化合物(R)-11のスペクトルデータを以下に示す。

化合物(R)-13(1.51g、1.56mmol)のトリフルオロ酢酸溶液(3.5mL、45mmol)を室温、窒素雰囲気下で15時間攪拌した。得られた混合液から溶媒を留去後、得られた固体の生成物を真空乾燥して、化合物 (R)-13のt-ブチル基が水素に置き換わった、化合物(R)-13のカルボン酸を得た。150℃で4時間真空乾燥した後に窒素下で放冷しておいた3-アミノプロピルシリカゲル(2.96g)へ、化合物(R)-13のカルボン酸 (1.36g、1.48mmol)を溶かした乾燥テトラヒドロフラン(10mL)と乾燥塩化メチレン(10mL)の混合溶液、DCC(463mg、2.24mmol)ならびにHOBT(303mg、2.24mmol)を加えた。得られたスラリーをメカニカルスターラーで室温、窒素雰囲気下で12時間攪拌後、DCC(448mg、2.17mmol)、HOBT(118mg、0.871mmol)を前記スラリーに加えてさらに25時間半攪拌した。得られたスラリーを濾過して得られるシリカゲルをテトラヒドロフラン、塩化メチレン、エタノール、熱エタノールの順序で洗浄し、真空乾燥することにより修飾シリカゲルを白色粉末として得た。得られたシリカゲル(3.30g)の乾燥トルエンの懸濁液(20mL)に1,1,1,3,3,3-ヘキサメチルジシラザン(HMDS)(1.4mL、6.7mmol)を加えて、メカニカルスターラーで室温、窒素雰囲気下で2時間攪拌した。得られたスラリーを濾過して得られるシリカゲルをテトラヒドロフランで洗浄し、真空乾燥することにより、CSP-4を白色粉末として得た。
以下に示す合成経路でキラル固定相CSP-5を調製した。
室温で真空乾燥しておいた3-グリシジルオキシプロピルシリカゲル(2.95g)の乾燥テトラヒドロフランの懸濁液(20mL)に化合物(R)-5(1.46g、2.00mmol)、トリエチルアミン(0.28mL、2.0mmol)を加えてメカニカルスターラーで窒素雰囲気下、43時間半加熱還流した。得たれたスラリーを濾過して得られるシリカゲルをテトラヒドロフラン、アセトン、エタノール、ジエチルエーテルの順序で洗浄し、真空乾燥することにより、修飾シリカゲルを白色粉末として得た。得られたシリカゲル(3.27g)の乾燥トルエンの懸濁液(20mL)にHMDS(1.8mL、8.5mmol)を加えて、メカニカルスターラーで室温下2時間攪拌した。得られたスラリーを濾過して得られるシリカゲルをトルエン、テトラヒドロフラン、ジエチルエーテルの順序で洗浄し、真空乾燥することにより、CSP-5を白色粉末として得た。
より得た3-グリシジルオキシプロピルシリカゲルを用いた。
以下に示す合成経路でキラル固定相CSP-6を調製した。
化合物(R)-5(2.19g、3.00mmol)と1,6-ジブロモヘキサン(4.5mL、30mmol)ならびに炭酸カリウム(0.620g、4.49mmol)を入れた乾燥ジメチルホルムアミド混合溶液(10mL)を窒素雰囲気下、70℃で47時間攪拌した。得られた反応混合物を濾過し、得られた濾液から溶媒を留去し、固体の生成物を得た。得られた生成物をシリカゲルカラムクロマトグラフィー(塩化メチレン/テトラヒドロフラン(塩化メチレン:テトラヒドロフラン(体積比)(30:1))で精製することにより、化合物(R)-14(1.23g、収率46%)を白色固体として得た。得られた化合物(R)-14のスペクトルデータを以下に示す。

以下に示す合成経路でキラル固定相CSP-7を調製した。
化合物(R)-5(1.80g、2.46mmol)と臭化アリル(0.26mL、3.0mmol)ならびに炭酸カリウム(0.510g、3.69mmol)を入れた乾燥テトラヒドロフラン混合溶液(25mL)を窒素雰囲気下、2時間加熱還流した後、臭化アリル(0.26mL、3.0mmol)と乾燥テトラヒドロフラン(5mL)を前記反応混合物に加えてさらに16時間半加熱還流した。得られた反応混合物を濾過し、得られた濾液から溶媒を留去し、固体の生成物を得た。得られた生成物をシリカゲルカラムクロマトグラフィー(塩化メチレン/テトラヒドロフラン(塩化メチレン:テトラヒドロフラン(体積比)(20:1))で精製することにより、化合物(R)-15(0.791g、収率42%)を白色固体として得た。得られた化合物(R)-15のスペクトルデータを以下に示す。

室温で真空乾燥しておいた3-メルカプトプロピルシリカゲル(3.00g)の乾燥クロロホルムの懸濁液(20mL)に化合物(R)-15(947mg、1.23mmol)、アゾビスイソブチロニトリル(71mg、0.43mmol)を加えてメカニカルスターラーで窒素雰囲気下、24時間加熱還流した。得られたスラリーを濾過して得られるシリカゲルをクロロホルム、メタノール、ジエチルエーテルの順序で洗浄し、真空乾燥することにより、修飾シリカゲルを白色粉末として得た。得られたシリカゲル(3.29g)の乾燥テトラヒドロフランの懸濁液(20mL)にHMDS(1.8mL、8.5mmol)を加えて、メカニカルスターラーで室温下3時間攪拌した。得られたスラリーを濾過して得られるシリカゲルをテトラヒドロフラン、ジエチルエーテルの順序で洗浄し、真空乾燥することにより、CSP-7を白色粉末として得た。
[CSP-3]を用いたHPLC用カラムの作製
CSP-3(3.5g)をメタノールに分散し、得られたスラリーを用いて、φ0.46cm×25cmLのステンレス製カラムにスラリー方式のカラム充填を行い、CSP-3が充填されてなるカラムを作製した。
下記条件でHPLCにて4種の評価サンプル(化合物番号:3、4、5、6)を用いて不斉識別能力の評価を実施した。評価結果を表11に示す。また各評価サンプルのそれぞれのクロマトグラムを図56~59に示す。

- 前記水素結合受容性を有する二価の基が炭素数3~10の複素環基であることを特徴とする請求項1~3のいずれか一項に記載の分離剤。
- 一般式(1)におけるZが酸素、窒素、及び硫黄の少なくとも一つを含む二価の有機基であることを特徴とする請求項1~5のいずれか一項に記載の分離剤。

一般式(II)、(III)及び(V)~(VII)中、Ar1は独立して炭素数3~10の芳香族基又は複素環基を表し、
一般式(IV)~(VII)中、Rは炭素数2~30の不斉構造を有する有機基を表し、Z’は独立して単結合又は二価の基を表し、
一般式(VI)中、Y’は担体と化学的に結合している基を表し、
一般式(I)及び(IV)中、Acはカルボキシル基又は-COClを表す。) - 前記一般式(II)で表されるジアミン(II)の2当量と前記一般式(I)で表される酸化合物(I)の1当量とを反応させて、前記一般式(III)で表されるジアミン(III)を得る工程と、
ジアミン(III)と酸化合物(IV)とを同当量で反応させて、前記一般式(V)で表される環状アミド化合物(V)を得る工程と、
環状アミド化合物(V)のYと担体とを、直接又は架橋基を介して結合させて、前記一般式(VI)で表されるクロマトグラフィー用光学異性体分離剤を得る工程とを含む、請求項11に記載の分離剤の製造方法。 - 前記Z’が-O-、-O-CH2-、-CH2-、又は-S-CH2-であることを特徴とする請求項11~14のいずれか一項に記載の分離剤の製造方法。
- 前記担体がシリカゲルであることを特徴とする請求項11~15のいずれか一項に記載の分離剤の製造方法。
- 前記シリカゲルが前記架橋基となる基で表面修飾されていることを特徴とする請求項16に記載の分離剤の製造方法。
