本発明は内臓脂肪をエネルギー源として代謝を活性化することを目的に、 L一アルギニン、 — 3多価不飽和脂肪酸、 リポ核酸、 ミネラル、 ビタミンを適宜配合することで加齢、 内臓肥 満などの結果として代謝能が低下することにより発症する、 メ夕ポリックシンドロームなどの 生活習慣病 (糖尿病、 高脂血症、 および脂肪肝などの疾患) の予防および改善を可能とした健 康食品素材、 医薬に関するものである。 特に内臓脂肪の脂肪分解を促進するとともに、 それに 続く有用生理活性蛋白の発現増加による代謝能の活性化によりアディポネクチンをはじめと するアディポサイト力イン類の発現を正常化あるいは改善することで、 糖尿病、 高脂血症およ び脂肪肝などの生活習慣病のみならず、 メタボリックシンドロームなどの症候群の改善効果に も関する。 背景技術
近年、 生活習慣病と呼ばれている肥満、 糖尿病、 高脂血症や高血圧などの疾患が様々な重篤 な疾患 (動脈硬化、 脳血管障害、 虚血性心疾患、 など) の発症もしくは悪化因子となり社会問 題化している。 これらの疾患が重度な場合は対症療法に基づく有効な治療薬が存在するが、 日 常生活において血糖値、 肝機能、 血圧、 血中脂質などの値が正常値を低から中程度に逸脱した 場合は、 前記の洽療薬を投薬されることは少ない。 このように、 正常ではない未病あるいは境 界領域の病気の状態でこれらの数値の逸脱が複数重なる疾患をメタボリックシンドロームと 称している。 生活習慣病の中でもメ夕ポリックシンドロームは先に述べたように、 放置するこ とで非常に重篤な疾患を惹起する確率が高くなることが疫学的な研究から明らかになつてい る。 メ夕ポリックシンドロームは一般的には中年の男女で内臓脂肪が蓄積することが特徴であ り、 カロリー摂取過剩、 運動不足も原因となり、 肥満、 髙血圧、 髙脂血症あるいは糖尿病が複 合することでメタポリックシンドロームと呼ばれる症候群と認定される。 肥満とは、 脂肪細胞 に異常に脂肪が蓄積して細胞が肥大した状態である。脂肪細胞として皮下に蓄積する皮下脂肪
および内臓に蓄積する内臓脂肪があり、近年、内臓の脂肪細胞がレブチン、アディポネクチン、 P A I— 1、 TN F— αなどのアディポサイト力インの分泌にかかわり、 生体で最も活発な内 分泌臓器であることが分かってきた。特に肥大した脂肪細胞ではィンスリン抵钪性惹起分子と いわれる TN F— α、 動脈硬化を惹起するといわれる P A I— 1などを過剰分泌することが明 らかにされている。 一方で、 肥大した脂肪細胞では食欲、 糖代謝、 血管の炎症の修復などを正 常にコントロールするレブチンやアディポネクチンの分泌が抑制されることが明らかにされ ている。 このことがインスリン抵抗性の原因となり、 糖尿病、 高脂血症、 高血圧、 などの生活 習慣病の発症につながると考えられている。 即ち、 内臓脂肪の蓄積が生活習慣病の発症と密接 に閧係している。 : また、 若年から青年期は一般に代謝能が高く、 肥満傾向であってもメタポリックシンドロ一 ムの発症は起こりづらいことから生活習慣病の発症は加齢に伴う代謝能の低下と、 栄養過剰へ の適応力の低下による内臓脂肪の蓄積からアディポサイトカインの異常分泌が大きな原因と 考えられる。 発明の開示
発明が解決しょうとする課題
メタボリックシンドロームなどの生活習慣病の治療の一環として行われる食事療法では 体重減少の効果は見られるものの、 メタボリックシンドローム、 糖尿病、 髙脂血症や脂肪肝 などの生活習慣病に対して十分な予防および改善効果が得られないという問題があつた。本 発明では近年生活習慣病、 特にメタポリックシンドローム、 の罹患率が非常に高いにもかか わらず適切な対処法が確立されていなレ ^現状をふまえ、医科学的代謝理論と細胞生物学的知 見のもとに内臓脂肪の減少と代謝を促進する新たな栄養組成物の創出を試みた。
本発明は、 内臓脂肪の減少と代謝を促進する作用を有しながら、 脂質代謝異-常、 糖代謝異 常患者における脂質および血糖値コントロールおよび脂肪肝患者の肝機能改善する有効な 栄養組成物を提供することを課題とする。 より具体的には、 高脂血症の状況を反映するトリ グリセリド (T G) 、 および糖尿病の状況を反映する空腹時血糖、 グリコヘモグロビン (H
bAl c) 値の改善、 更に肝機能としてァー GTP、 AST, ALT値の改善、 アディポサ イト力イン (アディポネクチン、 PAI— 1など) 値の改善により、 生活習慣病、 なかでも 糖尿病、 高脂血症および脂肪肝の予防'改善、 更にはこれらの生活習慣病の複合疾患である メタポリックシンドロームおよびその予備軍の予防 ·改善を目的とする栄養組成物を提供す ることを課題とする。
尚、本発明における栄饕組成物とは、糖尿病、高脂血症または脂肪肝をはじめとし、肥満、 高血圧を含む生活習慣病およぴメ夕ポリックシンドロームの予防及び改善をもたらす経口 栄養剤、 改善剤、 在宅用予防食、 あるいは保健機能食品、 体重の減少、 肥満の改善、 姿の スリム化を含む各種美容的効果を有する機能性食品、 スポーツァスリ一卜の体質改善剤、 生 活習慣病が引き起こす様々な循環器系や代謝調節系又は神経系の障害に起因する各種疾患 の発症および進展の予防 '改善が可能となる経口栄養剤、 改善剤、 在宅用予防食、 あるいは 保健機能食品をいう。 課題を解決するための手段
糖尿病、 高脂血症や脂肪肝などの生活習慣病の予防 '改善において食事療法と運動療法は 必須の治療法であり、 その目的としては患者の代謝異常をできる限り正常化すること、 およ び肥満を解消することにある。 生活習慣病の中でも、 特に糖尿病、 髙脂血症、 および脂肪肝 は肥満を誘因として発症するとされている。肥満者の多くにィンスリンの過剰分泌が見られ るため、 一定以上の肥満になるとインスリン分泌力多くなり過ぎて、 肥満を悪化させている 可能性がある 〔Food S t y 1 e 21, PP. 46, 2002. 5 (Vo l. 6 No. 5) 〕 。 また、 近年、 ω— 3および ω— 6多価不飽和脂肪酸類に富んだ混合物を含有してい る食事補助品について開示されている。 例えば、 米国特許 4, 820, 731号 (文献 1) に 記載された ω— 3および ω— 6多価不飽和脂肪酸類を含有し、手術後の患者への感染症の危 険性を最少にする食事補助品に閧する発明。 また、 米国特許 4, 871, 768号 (文献 2) に記載された ω— 3油類および中鎖トリグリセリド類を含有している食事補助品に関する 発明。 さらに、 米国特許 5, 053, 387号 (文献 3) に記載された魚油およびリノ'ール酸
の ω— 3脂肪酸類などを含む外傷治療用の組成物に関する発明などが開示されており、食事 補助品に関する特許は数多く出願されている。 しかしながら、 糖尿病、 髙脂血症および脂肪 肝などの生活習慣病全般への予防および治療に関するものはなく、 1 ) P C T/ C A 9 7 / 0 0 1 7 6号(文献 4 )に記載された免疫栄養素( O— 3多価不飽和脂肪酸、短鎖脂肪酸、 グルタミン、 L—アルギニン、 酸化防止剤、 リポ核酸又はヌクレオチド) と胆汁酸封鎖剤を 組み合わせた肝脂肪予防ため組成物に関する発明、 2 ) 特開平 1 1一 1 8 7 2 5号 (文献 5 ) に記載された、 タンパク質、 脂質および糖質のエネルギー比率を特定し粘調な可溶性食 物繊維およびィヌリンまたはその加水分解物を加えた糖尿病患者用栄饕組成物に関する発 明、 などがみられる程度である。 文献 4の肝脂肪予防のためには胆汁酸封鎖剤の配合が必要 であり、文献 5の糖尿病患者用栄養組成物はエネルギーコントロールと炭水化物および糖質 の吸収阻害を目的としており、 代謝活性化を目論んだものではなく、 加齢により発症する各 種生活習慣病を同時に改善できるものではない。
L一アルギニンやオル二チンに代表されるアミノ酸成分、及び ω— 3多価不飽和脂肪酸の 生体に対する作用は既に多数報告されており、 これらのアミノ酸成分が NO Sの活性化およ び成長ホルモン分泌による免疫活性増強作用を保持することが知られている、 また、 to— 3 多価不飽和脂肪酸は免疫能の向上、 脂質代謝改善作用、 抗動脈硬化作用などの生理作用を保 持することが知られている。 以上の知見から、 L一アルギニンや ω— 3多価不飽和脂肪酸は 免疫栄養素として炎症性疾患や回復期の食事補助品中の栄養組成物の用途で用いられてい る (文献 1—文献 4 ) 。 また、 中程度の肥満 '髙血圧患者の体重減少に有効であることも既 に知られている。 しかしながら、 一般に上記のような栄養組成物を伴った食事療法と運動療 法による減量では若千の血糖値、 中性脂肪値、 血圧などが低下することは知られているが、 1〜3ヶ月の期間で血糖値、 中性脂肪値、 肝機能、 血圧などの異常値を正常値あるいはその 近傍まで低下することはまれである。 つまり、 通常の食事療法や運動療法による体重減少だ けでは、 糖尿病、 髙脂血症や脂肪肝などの疾患に対して十分な治療効果は期待できない。
発明の効果
本発明者は脂質代謝改善作用により産生されたエネルギーを細胞核酸蛋白生合成や細胞 組織生理活性の促進のために利用することで生体内での代謝能を活性化し、ひいては各種疾 病の予防あるいは改善させることを目論んだ。特に、生体内の代謝能を活性化させるために、 L一アルギニン、 co— 3多価不飽和脂肪酸に加え、 リポ核酸 (R NA) と合成された蛋白を 活性化するために必要となる亜鉛、 セレニウムなどを含む各種ミネラル、 およびビタミン B 群および K群などを含む各種ビタミンを添加した栄養組成物を考案した。生活習慣病として 発症する糖尿病、髙脂血症や脂肪肝は一般には若年層では起こらず、 3 0歳代以降に見られ、 単純に栄養過剰による肥満が発症の原因ではなく、加齢に伴う代謝能の低下のためであるこ とが知られている。 これらの代謝能減弱は蛋白の合成および活性化の低下を引き起こすこと から、 R NAを加えることで脂質代謝改善作用により得られた脂肪分解エネルギ一により積 極的に蛋白の合成を推し進め、各種ミネラルおよびビタミンを加えることで蛋白の活性化を 促進させることとした。
本発明で考案した栄養組成物を被験者に 3ヶ月間投与したところ、被験者全員の血糖値は 正常値あるいは正常値近傍まで低下した。 また、 T G値が正常範囲を逸脱した髙脂血症の被 験者に 3ヶ月間投与したところ、被験者全員の T G値は正常値あるいは正常値近傍まで低下 した。 更には、 ァ一 G T P、 AL Tなどの肝機能が異常値を示す脂肪肝の被験者に同様に栄 義組成物を 3ヶ月間飲用させた結果、被験者の肝機能に関わる数値が正常値あるいは正常値 近傍まで低下した。 驚くべき事に、 これらの被験者の血液生化学値を測定したところ、 血清 中のアルブミン値および総夕ンパク量が有意に上昇し蛋白合成能が改善されていることが 明らかになった。 また、 生活習慣病の発症 ·進展に深く関わるアディポサイト力イン類のう ちアディポネクチン値が有意に上昇、 P A I— 1力有意に減少しており、 脂肪細胞の代謝が 活性化し、脂肪細胞内の脂肪の減少とこれに続くアディポサイトカインの分泌が正常化ある いは改善し、複合疾患であるメタボリックシンドロームおよびその予備軍が正常化あるいは 大幅に改善していることが明らかになつた。
本発明で考案した栄 II組成物は、本来 L一アルギニン、 ω- 3多価不飽和脂肪酸が薬効用量
を示す量の 1 Z 1 0程度の含有量にもかかわらず、 また R N Aおよび適切な量のビタミン、 ミネラルが加えられることで各種生活習慣病が劇的に改善'チ防されるというこれらの知見 は、 予期しない且つ驚くべきことであり、 本発明の栄蓥組成物が単なる肥満改善効果ではな く、 糖尿病、 高脂血症や脂肪肝などの代謝能が低下することで発症する生活習慣病全般の予 防および改善、 またその他の生活習慣病に起因するその他の疾病の予防を可能とすることが 証明された。 発明を実施するための最良の形態
本発明の栄養組成物は、糖尿病患者の血糖コントロール、 高脂血症の T G値の改善および 脂肪肝患者の肝機能改善ならびに肥満予防などに有効である。 具体的には 2型糖尿病、 耐糖 能異常者、 術後耐糖能障害、 グルコース不耐症の患者などの予防および改善に有用であり、 血糖値コントロールに用いることができる。 さらに、 糖尿病患者の食事療法における補助剤 としても有用である。 また、 糖尿病誘発のリスクファクターにもなる肥満の予防にも有効で ある。 髙脂血症および脂肪肝患者の T G値や肝機能異常の改善による治療に有用である。 さ らに、 糖尿病、 高脂血症および脂肪肝患者だけでなく、 未病の人においても血糖コントロー ルに配慮した食事は糖尿病食事療法の根本であると同時に、上記の生活習慣病予防の上にも 重要である。 本発明の泶蘧組成物は、 このような観点から、 経口 ·経管栄養剤、 改善食、 在 宅用糖尿病食としての用途の他、 肥満予防食、 保健機能食品として用いることもできる。 患者への栄饕組成物の投与は、 患者の状態、 患者の体重、 患者の年令、 等により異なり、 その投与量および投与期間は、 患者の担当医により決定される。栄 組成物が他の食品の補 助剤あるいは代替食として広く一般に使用される場合には 1日に投与される栄養組成物は 一般に市販されている加工食品と同様の各極形態で提供可能であるが、その場合においても 必ずしも aが限定される必要はなく、 効果に応じて増 itあるいは減量することができる。 本発明の栄 組成物は水溶液として摂取することができるばかりでなく、本発明の水溶液 状栄卷組成物に寒天を加えるか、 または粉末状栄 $組成物に水と寒天を加え、 熱処理後に冷 却して固形化した栄蓰組成物として摂取することもできる。固形化については寒天に限られ
るものではなく、一般的に食品の固形化に用いられるゼラチンなどの素材を用いることで同 様な固形栄蘧組成物として有用である。固形化した栄蘧組成物は摂取後の満腹感が得られる ため、 通常の固形食の代替えとしてより好ましい。
以下に実施形態を詳述するが、 本発明は、 リボ核酸 (RNA) および ω— 3多価不飽和脂 肪酸と、 L一アルギニン、 を含み、 細胞および酵素などの生理活性蛋白を活性化するために 必要なミネラル類およびビ夕ミン類を含有する栄蘧組成物である。
第一に数多くのアミノ酸のなかでも L—アルギニンは、カティォニックでユニークな存在 であり、 ことに細胞内へ移行しやすい。 また、 L一アルギニンをいくつか連結したポリアミ ンは、単独では細胞内へ進入できない外来性分子でさえ細胞内へ運搬できるビークルとなる。 また、 L一アルギニンは成長ホルモンゃグルカゴンの分泌を促し、 その結果として内因性リ パーゼ活性を高めることで脂肪分解の活性化作用も呈する。 第二に、 単なる L一アルギニン 摂取では、 リパーゼにより一旦グリセロールと脂肪酸に分離された中性脂肪も、 再度もとの 脂肪に復元しやすい。 ここで、 ω— 3多価不飽和脂肪酸、 ω— 6多価不飽和脂肪酸、 あるい は外来性短中鎖脂肪酸が存在すれば、 中性脂肪は原形に復元できず内因性脂肪酸は燃焼代謝 経路に進入せざるをえない。 また、 モノグリセリド、 もしくはジグリセリ ドが存在すれば、 さらに効果的に体脂肪をエネルギーに変換できることとなる。第三の重要な要素としてリポ 核酸がある。 リポ核酸は上記のようにして産生された燃焼エネルギーをより効果的に細胞の 代謝活性化に結びつけるために必要であり、 リポ核酸の供給が充分であれば燃焼エネルギー を物質合成系に投入できるので糖類や脂肪に再転換されることも防止できる。
具体的にはリポ核酸、 L—アルギニン、 ω— 3多価不飽和脂肪酸を特異的な機能成分とす る食事を、 糖尿病、 高脂血症または脂肪肝をはじめとする、 肥満、 高血圧などを含む生活習 慣病およびメタボリックシンドロームおよびその予備軍の予防及び改善をもたらす栄狭-組 成物として供することを発案した。
この理論的根拠として L一アルギニンは成長ホルモンの分泌を促し新陳代謝を活発化す るのみならず、 その結果内因性リパーゼ活性を高めて脂肪分解をもたらすとともに、 R NA やポリアミンの合成に必須のアミノ酸である。一方、 ω— 3多価不飽和脂肪酸はエイコサぺ
ンタエン酸がインスリン作用を改善し、 ドコサへキサェン酸は低比重リポ蛋白を低下させる とともに脳神経の活動を高める。 また、 — 3多価不飽和脂肪酸は中性脂肪から自由となつ たグリセりンに結合して内因性脂肪酸の再結合を防止して内因性脂質の分解を促進する。さ らに、 リポ核酸を含む核酸類は主として R N A組み立てから蛋白合成に至るプロセスにおけ る細胞内代謝に要求されるエネルギー消費を増大する。例えば、 培養細胞における蛋白合成 が培義液中の熱量を消費するがごとく、ダリセリンへの再結合を阻害された内因性脂'肪酸の 燃焼によって発生するエネルギーは、 R NAアッセンブリーからアミノ酸のリクルートを経 てペプチド、 蛋白合成に至るプロセスで有効に消費される。
上記の成分の好ましい組み合わせは、余剰の脂肪組織を処理して体重減少に効果を発揮す るのみならず、燃焼エネルギーとリポ核酸により引き起こされる生理活性蛋白の増大が細胞 活性化をもたらすことで、 結果として生体全体の代謝レベルを改善することとなり、 生活習 慣病の予防および改善を可能とした。
本発明は、 一食あたりの摂取量としてリボ核酸 (R NA) が 0 . l g以上で且つ 1 . 0 g 以下であり、 ω— 3多価不飽和脂肪酸が 0 . l g以上で且つ 1 . 0 g以下であり、 L—アル ギニンが 0 . 5 g以上で且つ 5 . 0 g以下である栄義組成物である。
本発明の態様のひとつは、 ジァシルグリセロール、 中短鎖脂肪酸及び植物ステロールのう ち少なくとも 1つと、 リポ核酸、 L一アルギニン、 ω— 3多価不飽和脂肪酸を含むことを特 徴とする栄養組成物である。
本発明は、上記 L一アルギニンに加えて同様な活性を持つ L一オル二チンを含むことを特 徴とする栄義組成物とすることも可能である。
本発明は、上記 ω— 3多価不飽和脂肪酸に加えて ω - 6多価不飽和脂肪酸を含むことを特 徵とする栄義組成物とすることも可能である。
また、本発明で用いられる ω— 3多価不飽和脂肪酸と ω— 6多価不飽和脂肪酸は、 少なく とも部分的には、 ともに魚油や植物性油、 例えばしそ油、 として含むことを特徴とする泶¾| 組成物であることが好ましい。
本発明で用いられるリポ核酸類は、 ヌクレオシド、 ヌクレオチド、 ドヌクレオチドが重合

したポリヌクレオチドのうち少なくとも 1つであることが好ましく、核酸基質源が天然ヌク レオチド類の場合には、 天然ヌクレオシド類の燐酸エステル類、 例えばモノ燐酸エステル類 であるアデ二レート (AM P) 、 グァニレート (GM P ) 、 ゥリジレ一ト (UM P) 、 シチ ジレート (CM P ) 、 並びに天然ヌクレオシド類のジ燐酸エステル類およびトリ燐酸エステ ル類、 例えば AD Pおよび A T Pなどが含まれる。
また、本発明で用いられるリボ核酸類としては、 ビ一ルまたはパン酵母 R NAが好適であ るが、 他の全ての生物素材もまた出発源として利用できる。
本発明は、 リポ核酸、 L一アルギニン、 c — 3多価不飽和脂肪酸に加えミネラル類及びビ 夕ミン類を含有すること特徴とする栄養組成物である。 ミネラル類及びビタミン類は、細胞および生理活性蛋白の活性化に必須であり、特に肥満 を伴う生活習慣病患者はこれらのミネラル類およびビ夕ミン類が不足していることが多く、 本発明で提供される上記栄養組成物をより効果的にするためにミネラル類及びビタミン類 が含有されることが好ましく、細胞代謝の高まりおよび内因性脂肪の分解と脂質代謝改善お よび燃焼との相乗作用がもたらされ、 糖尿病、 高脂血症および脂肪肝などの生活習慣病およ びメタボリックシンドロ一ムおよびその予備軍の予防および改善のための効果が一層発揮 される。
本発明の組成物に包含させるのに好適なミネラル元素および微量元素の例は、薬理学的に 許容可能な形のナトリウム、 カリウム、 カルシウム、 リン、 マグネシウム、 マンガン、 銅、 亜鉛、 鉄、 セレン、 クロムおよびモリブデンを含む。
さらに、本発明の栄養組成物に包含させるのに好適なビタミン類の例は、薬理学的に許容 可能な形のビタミン A、 ビタミン E、 ビタミン C、 ビタミン D、 ビタミン K:、 のほか葉酸、 チアミン、 リボフラビン、 ビタミン B 6、 ビタミン B 1 2、 ナイァシン、 ピオチンおよびパ ントテン酸を含む。
また、 本発明の効果をより高めるためには以下の成分を加えることが好ましい。
本発明において、 ジァシルグリセロール及び中短鎖脂肪酸は、 肥満防止作用、 体重増加抑 制作用等を有する。 一方、 植物ステロールはコレステロールが胆汁酸に溶解して吸収される
過程でコレステロールの吸収阻害機能を有する。これらのいずれか又は両者を上記栄蘧組成 物とともに摂取するとき、細胞代謝の高まりおよび内因性脂肪の分解と脂質代謝改善および 燃焼との相乗作用がもたらされて、利用エネルギー増大に基づきエネルギー収支が臨界点を 超えて負に傾くことにより、 予防および改善効果が発現される。
本発明において、 上記栄養組成物は、 マルトデキストリン、 クラスターデキストリン、 難 消化デキストリンのうち少なくとも 1つを含むこととすることもできる。澱粉を酵素などで 処理して得られるマルトデキストリン、 クラス夕一デキストリン、 難消化デキストリンはダ イエット用炭水化物としてエネルギー量、 吸収様式、 味覚等を理想的に設定するために適切 な特性を有する。 単にグルコース、 砂糖などを摂取した場合には、 急激に血糖が上昇してィ ンスリン分泌を促し、 プロテインキナーゼ〇、 P 1 3キナーゼ、 N O産生など様々な代謝経 路が活性化されて、 中長期的には血管障害、 肥満や生活習慣病体質の増悪に帰結する。 ここ で、 特徴的な消化、 吸収、 また代謝様式を呈するマルトデキストリン、 クラスターデキスト リン、 難消化デキストリンは、 緩やかな血糖上昇をもたらし炭水化物としての人体中への移 行と利用を計画できる糖類グループである。 したがって、 栄養組成物へのマルトデキストリ ン、 クラスターデキストリン、 難消化デキストリンの配合比率を定めることにより、 より効 果的に糖尿病の予防および改善が実現できる。
本発明において、上記栄養組成物は、 ムコ多糖類あるいはゥロン酸類を含むこととするこ ともできる。 ぺクチン、 カラギーナン、 アルギン酸などゥロン酸含有多糖類は水分を引きつ け、食品として腹部充足感を与えると同時に低力口リ一かつ糖分の吸収を抑制できる優れた 性能を発揮する高分子である。 また、 ヒアルロン酸は皮膚や滑膜組織に保護と滑らかさを与 える。 従って、 本発明に係る栄養組成物にゥロン酸類を配合することにより、 これらの効果 を高めるための有力な手段となる。 リンゴゃ海草、 山芋などゥロン酸含有多糖類やムコ多糖 額を多く含有する天然素材を処理配合してもよい。
本発明において、 上記栄¾組成物は、糖 Sまたは脂笾の吸収抑制作用を有する成分を含む こととすることもできる。脂肪の分解を阻害するョヒンビンや吸収阻害のある天然に存在す る物質、さらに炭水化物吸収阻害作用のある 0;—ダルコシダーゼ阻害作用のある天然に存在
する物質、 ゥコギ科のタラの芽、 ギムネマエキス、 キチンキトサン、 ガルシニア等の従来型 の脂質や糖類の吸収代謝抑制作用のある多数の天然の物質を併用することは本発明に係る 栄養組成物の効能を強化する意味から有用である。 また、 カテキンなどのポリフエノール類 のように抗酸化作用を有する成分を添加することも一層好ましい。
本発明において、 上記栄養組成物は、 吸水性繊維成分を含むごととすることもできる。 吸 水性繊維質は消化管内で膨張して満腹感をもたらすことが知られる。 よって、 ダイエット食 にマンナン、 グァガム、 キタンサンガム、 ィヌリンなどを組みあわせて用いるときには、 胃 腸管内での水和膨張により物理神経学的な満腹感が達成されて過大な食欲を抑制できるの で、 予防および治療食に対する継続的な認容性がもたらされる。 また、 これらの物質には脂 質、 炭水化物などの食事成分の吸収低減作用も期待される。 セルロース、 へミセルロース、 リグニン、 寒天などの不溶性繊維質も食感の修飾などの目的で使用してもよい。 本発明にお いて、 上記栄養組成物は、 天然物由来の食欲抑制成分、 循環刺激成分、 脂肪分解消費促進成 分、 抗酸化成分のうち少なくとも 1つを含むこととすることもできる。 カブサイシン、 ラズ ベリーケトン、 カルニチン、 リコピン、 タウリン、 サポニン、 キサンチン類は、 体循環を刺 激し、 体脂肪を分解燃焼させる脂肪分解消費促進作用を有する。 また、 ポリフエノール類、 セサミンなど抗酸化成分には動脈硬化防止作用がある。
本発明は、糖尿病、 髙脂血症あるいは脂肪肝などの生活習慣病患者またはメ夕ポリツクシ ンドロームおよびその予備軍の患者における健康維持または体質改善用とすることができ る。
さらに、 本発明は、 肥満、 糖尿病、 高脂血症あるいは脂肪肝などの生活習慣病の家畜、 家 禽または愛玩動物における健康維持または体質改善用とすることができる。
本発明の食品を飲用に供する場合の機能成分の濃度は、 次の栄養組成物が良好であ?)。
(A) L一アルギニンを 0 . 2〜2重量%、 (B) ω— 3多価不飽和脂肪酸を 0 . 0 4〜 0 . 4重量%、 及び (C) リポ核酸類を 0 . 0 4〜0 . 4重量%含む。
さらにより効果的な液状栄養組成物は、 (A) L—アルギニンを 0 . 2〜2重量%、 (Β ) ω— 3多価不飽和脂肪酸を 0 . 0 4〜0 . 4重量%、 (C): リボ核酸類を 0 . 0 4〜0 .
4重量%、 (D) ミネラル類を 0 . 0 0 1〜0 . 0 1を重量%、 (F ) ビタミン類を 0 . 0 1〜0 . 1を重量% (G) 蛋白又はペプチドを 1 ~ 1 0を重量% (H) 脂質を 0 . 1〜: I重 量%を含むものとする。
本発明では、 上記 L—アルギニンは、 成長ホルモンの分泌を促し、 新陳代謝を活発化する のみならず、その結果として脂肪分解酵素である内因性リパーゼの活性を高める作用を有す る。 成長ホルモンの分泌によって活性が高められた内因性リパーゼは、 血液中や体内の脂肪 組織に蓄積されているトリグリセリドを効率よく加水分解し、脂肪酸とダリセリンを遊離さ せる。 一方、 L一オル二チンは、 下垂体による成長ホルモンめ合成と放出に関わつ いる。 従って、 L一オル二チンの摂取により、 成長ホルモンの合成及び放出が活性化され、 体細胞 の代謝が高まり、 物質合成が促進され体脂肪の分解 ·消費を促す。
さらに、上記 ω— 3多価不飽和脂肪酸であるエイコサペン夕ェン酸やドコサへキサェン酸 はリノレン酸からごく小量合成されるのみであり、必須の ω - 6多価不飽和脂肪酸であるァ ラキドン酸も体内合成は不充分である。上記 ω— 3多価不飽和脂肪酸や ω— 6多価不飽和脂 肪酸は、 摂取者の体内において、 体内の中性脂肪から遊離するグリセリンと結合する。 これ により、 ω— 3多価不飽和脂肪酸あるいは ω— 6多価不飽和脂肪酸は、 グリセリンと内因性 脂肪酸が再びトリァシルグリセロールや V L D Lとなるのを抑制し、血漿中の V L D L夕一 ンオーバ一を促進する。 この結果、 脂肪細胞、 動脈硬化巣へ V L D Lが移行蓄積されるのを 阻害する。 従って、 L—アルギニンにより、 分解へと導かれたトリグリセリドの引き受け手 として最適である。 また、 エイコサペンタエン酸はトロンポキサン A 3となり血栓性疾患に 対して防止的に作用し、 ドコサへキサェン酸は神経系の機能を高め、 両者とも生活習慣病素 因の発現に抑制的に機能する。上記栄養組成物に含まれる ω— 3多価不飽和脂肪酸や; ω— 6 多価不飽和脂肪酸は、 魚油中に比較的髙ぃ割合で含まれている。 魚油は比較的安価、 且つ容 易に入手が可能であり、 本発明の栄 組成物への添加に適している。 本発明において、 上記 栄 ¾組成物に含まれる魚油が力プセル化など低無臭化処理されていることにより、魚油特有 の臭気を封止することができるとともに、 魚油の酸化による劣化を防止することもできる。 本発明においては、 リポ核酸類は、主として R N Α組み立てから蛋白合成に至るプロセス
における細胞内代謝に要求されるエネルギー消費を増大させる作用を有する。従って、 ダリ セリンとの再結合を阻害された遊離脂肪酸の燃焼によって発生するエネルギーは、 R N Aァ ッセンプリ一からアミノ酸のリクルートを経てペプチド、蛋白合成に至るプロセスにおいて 有効に消費される。
これらをまとめると、 本発明における L一アルギニン、 ω— 3多価不飽和脂肪酸、 リポ核 酸類は三位一体となって、 単なるカロリー制瞌ゃ吸収阻害によるダイエットと異なり、 エネ ルギ一産生、 細胞活性、 R N Α合成から蛋白合成を高められることにより、 糖尿病、 高脂血 症あるいは脂肪肝および肥満、高血圧などの生活習慣病素因保有者およびメタポリックシン ドローム患者およびその予備軍の患者のための理想的な栄養組成物成分を構成する。
さらに、 ミネラル類及びビタミン類は、 細胞および生理活性蛋白の活性化に必須であり、 本発明で提供される上記栄養組成物をより効果的に作用させるためにミネラル類及びビ夕 ミン類力 S含有されることが好ましい。その結果として、 一層細胞代謝が高まり内因性脂肪の 分解と脂質代謝改善および燃焼との相乗作用がもたらされることにより、 糖尿病、 高脂血症 および脂肪肝などの生活習慣病およびメタポリックシンドロ一ムおよびその予備軍の予防 および改善効果が一層発揮される。
本発明において、上記栄養組成物にさらに包含させるのに好適な微量元素の例は、薬理学 的に許容可能な形のナトリウム、 カリウム、 カルシウム、 リン、 マグネシウム、 マンガン、 銅、 亜鉛、 鉄、 セレン、 クロムおよびモリブデンなどのミネラル元素である。 これらのミネ ラル元素は疾患を引き起こした患者で不足している場合が多く、 このような状態では細胞代 謝あるいは蛋白の活性化能が減弱してしまう。 これらのミネラル元素を加えることで、 代謝 能の改善効果を補助するとともに、治療期間中の必須栄璲素の補填によって摂取者の健康を 維持するために役立つ。
さらに、本発明において、上記栄養組成物にさらに包含させるのに好適な微量元素の例は、 I理学的に許容可能な形のビタミン A、ビタミン E、 ビタミン C、ビタミン D、ビタミン 、 のほか葉酸、 チアミン、 リボフラビン、 ビタミン B 6、 ビタミン B 1 2、 ナイァシン、 ピオ チンおよびパントテン酸などのビタミン類である。これらのビタミン類は上記栄養組成物の
予防および改善効果を補助するとともに、飲用期間中の必須栄養素の補填によって摂取者の 健康を維持するために役立つ。
本発明では、 ω— 3多価不飽和脂肪酸や ω— 6多価不飽和脂肪酸の一部に加えてあるいは 入れ替えて、 ジァシルグリセロール、 中短鎖脂肪酸や植物ステロールを使用した場合、 体脂 肪の再合成をさらに抑制することができる。 即ち、 グリセリン 1分子に脂肪酸が 2分子結合 したジァシルグリセ口一ルは、 体内に吸収された後も体脂肪に再合成されにくレ^ また、 炭 素数が 8〜1 0個の中短鎖脂肪酸は、 通常の長短鎖脂肪酸に比べて水溶性が高く、 細胞内で 代謝されやすい。 さらに、 植物ステロールは、 胆汁酸に溶け込む性質を有するため、 コレス テロ一ルの吸収量を減少させるように作用する。
L—アルギニンまたはオル二チンの摂取によれば、成長ホルモンの分泌とそれに続くリパ —ゼの活性化が誘導され、 中性脂肪が分解される。 さらに、 食事性の中性脂肪は脂肪^とグ リセリンに分解後吸収され内因性のグリセリンゃ脂肪酸と混在化した状態となる。
しかしながら、この分解過程は可逆的であることから上記脂肪酸類はグリセリンとともに 中性脂肪を再構成しやすく燃焼消費されづらい難点がある。 ここで食事性のジァシルグリセ ロールが組み込まれたときには中性脂肪は再構成されにくく、内因性脂肪酸を燃焼させる効 果を発揮する。 よって、 ジァシルグリセロールの存在により L—アルギニンによる細胞刺激 と成長ホルモン分泌促進によるリパーゼ活性化が有効に脂肪の燃焼と細胞代謝の上昇に連 結される。 このときに、 外来性には ω— 3多価不飽和脂肪酸、 または ω— 6多価不飽和脂肪 酸の脂肪酸を含めて調和のとれた脂肪酸配合を供給することが好ましい。
また、コレステロールや中性脂肪に分類される脂質群は血液中ではアポ蛋白と共役して大 きなミセルを形成するので、 中性脂肪の減少分にはコレステロールが補充されやすい。 ここ で、植物ステロールは体内コレステロールとの競合下において胆汁酸と複合体を構成して腸 肝循環に入る一方、植物ステロールは体内へ吸収されないのでコレステロールの吸収を抑制 でき、 ひいてはアポ蛋白の発現レベルとパターンをも調整できる。 したがって、 上記栄義組 成物と組み合わせてジァシルグリセロールや植物ステロールを摂取することは内因性脂肪 の再構成やアポ蛋白発現レベルの変化を介して肥満や生活習慣病の改善に寄与する。 一方、
ジァシルグリセロールや植物ステロールの摂取で内因性脂質を事前調整させた場合には、 L —アルギニン作用における細胞機能活発化による細胞組織ひいては身体機能向上に έ、要と されるエネルギー供給が準備されたこととなり、様々な機能物質形成プロセスが円滑に進行 し、 細胞組織の利用エネルギー増大を介する基礎代謝量増大に寄与する。
本発明において、上記栄養組成物に添加されるマルトデキストリン、 クラス夕一デキスト リンゃ難消化デキストリンは、 栄養組成物のエネルギーを理想的な形で補うことができる。 即ち、 上記各デキストリンは、 ブドウ糖、 ショ糖や果糖などに比べて、 摂取した場合の血糖 値の上昇が穏やかである。 そのため、 血中へのインスリン分泌も抑制されるため、 代謝経路 を過度に活性化することもなく、 高ィンスリンによる動脈硬化や脂質代謝増悪、 肥満刺激を 抑制できる。 従つて、 栄養組成物の摂取者は急激な空腹感ゃ疲労感におそわれることからも 防御される。
本発明において、上記栄養組成物に含まれるムコ多糖類としてコンドロイチン硫酸、 ヒア ルロン酸、 へパリン、 ケラタン硫酸、 デルマタン酸などがあげられ、 ヒト成長期には充分に 体内で作られる物質であるが、 年齢とともにデノポ合成が低下していくため、 積極的に添加 する意義がある。 これらムコ多糖類は、 ネバネバした粘度の高い物質で関節、 眼球、 粘膜、 血管壁などに多く含まれ組織の柔軟性や円滑性を担っている。 したがって、 上記ムコ多糖の 適切な補給は血管や皮膚をしなやかに保ち動脈硬化など生活習慣病体質の進展を抑制する ために有用である。
栄養組成物に含まれる糖質及び脂質の吸収抑制作用を有する成分として、例えば、脂肪の 分解を阻害するョヒンビンや吸収阻害のある天然に存在する物質、 さらに炭水化物吸収阻害 作用のある α—ダルコシダーゼ阻害作用のある天然に存在する物質、 ゥコギ科のタラの芽、 ギムネマエキス、 キチンキトサン、 ガルシニア等の従来型の脂質や糖類の吸収代謝抑制作用 のある多数の天然の物質を併用することは栄 51組成物の効能を強化する意味から有用であ る。 また、 ポリフエノール類は抗酸化作用も併せ持ち、 フランス人が欧州のなかで一番心臓 病の死亡率が低いのは赤ワイン中のポリフエノールを多く摂取するためであり、 緑茶には、 タンニン、 カテキン、 フラポノイドなどのポリフエノールが含まれる。 本発明においてはこ
れらの各種天然素材抽出物として吸収阻害機能成分を添加することが好ましい。
本発明において、 上記栄養組成物に添加される吸水性繊維成分は、 摂取された後、 消化管 内で膨張して満腹感を与えることができる。 また、 吸水性繊維成分は、 脂質や糖質の吸収低 減作用をも発揮する。
本発明において、栄養組成物に添加される食欲抑制成分として中枢性に食欲を減退させる アルカイド類やその誘導体なども配合可能である。
本発明において、栄養組成物の各有効成分は、安全性の高い天然物由来の物質であること が好ましい。
本発明は、 糖尿病、 高脂血症あるいは脂肪肝または肥満、 高血圧などの生活習慣病患者も しくはメ夕ポリックシンドロームおよびその予備軍の患者における健康維持または体質改 善に使用されて、 顕著な効果を得ることができる。 また、 本発明に係る栄養組成物は、 牛、 豚、 鶏などの家畜および家禽類、 及び犬、 猫などの愛玩動物に使用されて、 人間が摂取した 場合と同様にそれらの動物の健康維持や体質改善においても効果を発揮することができる。 本発明を以下の実施例によってさらに説明する。 実施例 1
A S T, A L T, ァー G P Tなどの肝機能の異常値を示すの 7名の被験者(脂肪肝) が通 常の朝昼夕食のうちいずれかの 1食に代えて、 (表 1 ) に示す主要成分を含有する本発明の 栄養組成物を 3ヶ月間摂取させた。 本実施例において、 上記泶薆組成物は液体状であり、 1 回当たりの摂取量は 2 5 0 m lとした。 その結果を (表 2 ) および (図 1 ) に示した。 異常 値を示していた G〇T、 G T P、 ァ— G P Tは 3ヶ月後に有意に改善または正常化した。 ま た全ての被験者において、 血清アルブミン値および総蛋白値が増加するとともに、 アディポ ネクチンが有意に増加し、 P A I— 1が有意に減少した。 '

実施例 2
中性脂肪 (TG) 異常値および HDLコレステロール低値の 7名の被験者(高脂血症) が 通常の朝昼夕食のうちいずれかの 1食に代えて、表 1に示す成分を含有する本発明の栄薆組 成物 (250nil) を 3ヶ月摂取した。 その結果を (表 3) および (図 2) に示した。 異常 値を示していた TG (正常値: <15 OmgZd 1) は 3ヶ月後有意に低下し、 正常値ある いは正常値の近傍まで推移した。 また HDLコレステロール (正常値:〉 40mgZd 1 ) は 3ヶ月後有意に増加した。 また全ての被験者において、 血清アルブミン値および総蛋白値 が増加するとともに、 アディポネクチンが有意に増加、 PA I— 1が有意に減少した。

実施例 3
空腹時血糖 · H b A 1 c高値の 6名の被験者(糖尿病) が通常の朝昼夕食のうちいずれか の 1食に代えて、 表 1に示す成分を有する本発明の栄養組成物 (250ml) を 3ヶ月摂取 した。 尚、 他の食生活および運動などの指導および制限は全く加えなかった。 その結果を ( 表 4) および (図 3) に示した。 異常値を示していた空腹時血糖 (正常値:く 11 OmgZ d 1) および HbAl c (正常値: <5. 8%) は 3ヶ月後有意に低下し、 正常値あるいは 正常値の近傍まで推移した。 また全ての被験者において、 血清アルブミン値および総蛋白値 が増加するとともに、 アディポネクチンが有意に増加、 PA I— 1が有意に減少した。

70歳の糖尿病以外の既往症のない男性 (身長 175 cm、 体重 75 Kg) に対して、 糖 尿病の入院ィンスリン治療の終了後、 退院後通常の朝食に代えて、 (表 1 ) に示す成分を有 する本発明の栄養組成物を任意摂取した。 同時にインスリン投与は中止した。 この結果を ( 図 4) に示した。 入院時異常値を示していた被験者の血糖はインスリン治療を終了後も本発
明の栄養組成物の服用のみで服用開始後 30日過ぎた時点においても 13 Omg/d 1前後 に維持され、 ィンスリン投与と同等の効果が得られることが示唆された。 実施例 5
メ夕ボリックシンドロームおよびその予備軍の 18名の被験者(上記実施例 1一 3の被験 者を含め) が通常の朝昼夕食のうちいずれかの 1食に代えて、 表 1に示す成分を有する本発 明の栄養組成物 (250ml) を 3ヶ月摂取した。 尚、 他の食生活および運動などの指導お よび制限は全く加えなかった。 被験者全体の平均値で体重 (3ヶ月平均 5. 9Kg±2. 7 7) 、 体脂肪 ( 3ヶ月平均 4. 0%±1. 67) が減少した。 収縮時血圧 ( 3ヶ月平均 1 8. 7mmHg±9. 1) 、 拡張時血圧 ( 3ヶ月平均 12. 6mmHg±5. 44) 、 空腹 時血糖 ( 3ヶ月平均 18. 6mgZd l ±13. l) 、 HbAl c ( 3ヶ月平均 0· 41 %±0. 19) 、 中性脂肪 ( 3ヶ月平均 129. 2mgZd l ±75. 2) 、 肝機能 ( A ST : 3ヶ月平均 21. 8 I U/m 1 ± 16. 3、 ALT: 3ヶ月平均 35. 7 I U/m 1 ±28. 7、 ァ一 GTP: 3ヶ月平均 44. 2 I U/m 1 ± 36. 6) の全て項目での値が 有意に改善低下した。 アルブミン値についても被験者全員で増加し、 3ヶ月平均 0. 25g /d 1 ±0. 14の増加であった。 同様に内臓脂肪のマーカーとなりうるアディポネクチン は被験者全員で 3ヶ月平均 2. 32;tig/ml ±0. 83増加し、 PAI— 1も被験者全員 で 3ヶ月平均 6. AOngZml iA. 80減少し、 被験者全員の内臓脂肪が減少している ことが示唆された。その結果、 本発明の栄養組成物の服用でメ夕ポリックシンドロームおよ びその予備軍が大幅改善および正常化することが明らかになり、代謝能の低下に伴い発症す る肥満、 糖尿病、 高脂血症、 高血圧症、 および脂肪肝などの生活習慣病に広く効果があるこ とが示された。 産業上の利用可能性
以上説明したように、 本発明における栄養組成物は、 脂肪肝などの肝疾患および糖尿病、 高脂血症、メ夕ポリックシンドロームなどをはじめとする生活習慣病の予防および改善に有

Claims
1. リポ核酸 (RNA) 類、 c — 3多価不飽和脂肪酸類、 L一アルギニンを含むことを特徴とす る栄養組成物。
2. 上記リポ核酸類は、 ヌクレオシド、 ヌクレオチド、 ヌクレオチドが重合したポリヌクレオチ ドのうち少なくとも 1つを含むことを特徴とする請求項 1に記載の栄養組成物。
3. 上記 ω— 3多価不飽和脂肪酸類の一部もしくは全部が魚油、 植物油のうち少なくとも 1つか ら由来することを特徴とする請求項 1に記載の栄養組成物。
4. 一食あたりの摂取量としてリポ核酸 (RNA) 力 0. l g以上で且つ 1. 0 g以下であり、 ω— 3多価不飽和脂肪酸が 0. l g以上で且つ 1. 0g以下であり、 L一アルギニンが 0. 5 g以上で且つ 5. 0 g以下であることを特徴とする請求項 1に記載の栄養組成物。
5. 銅、 亜鉛、 マンガン、 セレニウム、 クロム、 モリブデン、 ヨウ素などの生体に必須のミネラ ル類のうち少なくとも 1つを 1日の必要摂取量の 1 / 3以上且つ 1日必要摂取量以下の量で 含むことを特徴とする請求項 1記載の栄養組成物。
6. ビタミン A、 ビタミン B類、 ビタミン C、 ビタミン D類、 ビタミン E、 ビタミン K類、 など 生体に必須のビタミン類のうち少なくとも 1つを 1日の必要摂取量の 1/3以上且つ 1日必 要摂取量以下の量で含むことを特徴とする請求項 1または請求項 5に記載の栄養組成物。
7. ジァシルグリセロール、 中短鎖脂肪酸、 ω— 6多価不飽和脂肪酸及び乳蛋白植物ステロール のうち少なくとも 1つを含むことを特徴とする請求項 1、請求項 5または請求項 6に記載の 栄養組成物。
8. マルトデキストリン、 クラスタ一デキス卜リン、 難消化デキストリン、 オリゴ糖のうち少な くとも 1つを含むことを特徴とする請求項 1、請求項 5または請求項 6に記載の栄 31組成物。
9. ムコ多糖類、 ゥロン酸含有多糖類のうち少なくとも 1つを含むことを特徴とする請求項 1、 請求項 5または請求項 6に記載の栄蘧組成物。
10. 糖質または脂質の吸収抑制作用を有する成分を含むことを特徴とする請求項 1、 請求項 5
または請求項 6に記載の栄養組成物。
1 . 有効成分が天然物由来の物質であることを特徴とする請求項 1、 請求項 5または請求項 6 に記載の栄戔組成物。
2 . 糖尿病、 高脂血症、 脂肪肝、 およびメ夕ポリックシンドロームなどの生活習慣病の予防お よび改善に用いることを特徴とする請求項 1、請求項 5または請求項 6に記載の栄養組成物。3 . 家畜、 家禽、 または愛玩動物における糖尿病、 高脂血症、 脂肪肝、 およびメ夕ボリツクシ ンドロームなどの生活習慣病の予防および改善に用いることを特徴とする請求項 1、請求項 5または請求項 6に記載の栄養組成物。
4. 請求項 1、 請求項 5または請求項 6に記載の栄養組成物で水を溶媒とすることを特徴とす る液状栄養組成物。
5 . 請求項 1、 請求項 5または請求項 6に記載の栄養組成物で水を溶媒とした溶液に寒天、 ゼ ラチンのうち少なくとも 1つを加え固化することを特徴とする固相栄養組成物。 .
