Abstract
生体適合性および生分解性を有し、石灰化なしに自己組織化および成長を許容する、生体由来の再生医療用スキャフォールドの作製方法を提供する。この方法は生体軟組織をグルタルアルデヒドで部分的に架橋固定するステップ、および部分的に架橋固定化した組織をエラスターゼと共にインキュベートするステップを含む。
Description
生物由来スキヤフォールドの作製方法
技術分野
[0001] 本発明は、再生医療技術の分野、詳しくは哺乳動物の軟組織力 生物由来のスキ ャフォールドの作製方法に関する。
背景技術
[0002] 患者の欠損した器官を再生するため、人工デバイスによる置換、人工または生物由 来のスキヤフォールドを移植して自己組織ィ匕等により、失われた機能を回復させる医 療が行われている。例えば、現在我国では、年間 1万件を超える心臓弁置換術が行 われている力 その約 7割は機械弁による置換である。残りの 3割はゥシ、ブタ等の組 織をダルタルアルデヒドで固定ィ匕した異種生体弁の移植である。機械弁置換後は継 続して抗血栓剤の投与が不可欠であり、患者のクオーリティ ·ォブ ·ライフを損ねる問 題がある。移植した異種生体弁は石灰化などによって 15年程度の耐久性しかなぐ 若年者では再度の移植を必要とする。
[0003] ダルタルアルデヒドで固定化した異種生体血管による血管置換術では、心臓弁置 換術と同様に石灰化などによる耐久性の問題に加えて、生体組織の本来の機械的 性質 (強度および弾力性)が失われる問題がある。生分解性ポリマー、例えばポリ乳 酸を用いた人工血管の移植は小児患者の静脈系で臨床応用されているが、狭窄等 の問題も報告されており、また生分解は単純な加水分解によるため動脈系では破断 のおそれがある。
[0004] 生体組織力ゝら酵素や界面活性剤を用いて細胞を取り去り、細胞外マトリックスを残し 、移植用スキヤフォールドを作製する方法も知られている。このスキヤフォールドは、 そのままでは例えば比較的高圧力に耐える必要のある左心系の大動脈弁では破断 に耐えられる強度が不足しており、ダルタルアルデヒド架橋等の安定化処理が不可 欠である。しかしながらダルタルアルデヒドによる固定化はコラーゲンを主体とするマ トリックスの生分解性を消失させ、移植後体内で分解されずに残るので細胞の浸潤が 殆んど見られず、自己組織の再生を目的とする再生医療には適さない。
[0005] 特公昭 54— 43833号公報には、ブタ血管をキモトリプリンで消化処理し、次いで得 られる消化血管をダルタルアルデヒドなどで固定することよりなる異種代用血管の製 法が開示されている。しかしながらこの代用血管は血液透析を受けている患者の血 管へのアクセスを提供する動脈一静脈シャントとして移植することを意図しているので 、細胞の浸潤による自己組織ィ匕は必須条件ではない。
[0006] 従って、宿主に拒絶反応を誘発しないことおよび抗血栓性であることを含む生体適 合性を有し、終局的には体内で分解されるが自己組織化されるまでの耐久性を有し 、細胞の浸潤を妨げる石灰化なしに自己組織化および成長を許容する、生物由来の スキヤフォールドの提供が望まれる。
発明の開示
[0007] 上記の課題は、
(a)生態軟組織をダルタルアルデヒドで部分的に架橋固定ィ匕するステップ、および
(b)部分的に架橋固定ィ匕した組織をエラスターゼと共にインキュベートし、選択的に エラスチンを除去するステップを含む、本発明の移植用スキヤフォールドの作製方法 によって達成される。

詳細な説明
[0009] 本発明は、生体組織の構造タンパク成分であるエラスチンが石灰化に関与している との研究報告を基礎にして 、る。し力しながら組織力もエラスチンを除去しただけで は他の要件特に満足な機械的強度を保っためには不充分であるため、部分的なグ
ルタルアルデヒド固定ィ匕を併用する。
[0010] 一般にダルタルアルデヒドの固定化は、組織がそれ以上ダルタルアルデヒド
と反応しなくなるまでダルタルアルデヒド水溶液中に浸漬して行われる。このようにグ ルタルアルデヒドと完全に反応させた組織内の構造タンパクは不溶性に変性し、生 分解性でもなくタンパク分解酵素の基質にもならない。しかしながら構造タンパクに含 まれるダルタルアルデヒドと反応し得る官能基の多くを未反応のまま残して反応を停 止することにより、組織を部分的に架橋することができる。
[0011] エラスチンは高等動物の結合組織、大動脈外皮などを構成する硬タンパクであり、 それを構成するアミノ酸の大部分は電荷を持たな 、非芳香族アミノ酸である。グルタ ルアルデヒドは主としてリジン残基と反応し、分子間および分子内でタンパク分子を 架橋するので、リジン残基を持たな 、エラスチンにはダルタルアルデヒドが作用しな
V、。そのため酵素の基質特異性を利用してダルタルアルデヒドによって部分架橋され た組織から、エラスターゼを作用させてエラスチンを選択的に除去することができる。
[0012] ダルタルアルデヒドによる架橋の程度は温度が一定の場合ダルタルアルデヒドの濃 度と反応時間の関数である。哺乳動物組織力もスキヤフォールドを作製する方法に 関する米国特許第 4, 378, 224号は、最初約 0. 05%ないし約 0. 4wt%、好ましく は約 0. 15wt%のダルタルアルデヒドで約 12時間ないし約 64時間処理し、次にジァ ミンとカルポジイミドによりカルボキシル基間の架橋を行うことを開示して 、る。本発明 者らの研究によれば、ステップ (a)のダルタルアルデヒドによる部分架橋は 0. 1%グ ルタルアルデヒド水溶液中に約 1時間常温で浸漬するのが最適であることがわ力つた 。し力しながら上で述べたように、架橋の程度はダルタルアルデヒドと浸漬時間の関 数であるから、 0. 1%より高濃度のダルタルアルデヒド水溶液を使用し、 1時間より短 時間浸漬するか、または 0. 1%より低濃度で 1時間より長い時間浸漬することによつ ても匹敵する部分架橋を達成することが可能であろう。
[0013] 部分架橋した組織力ものエラスターゼによるエラスチンの選択的除去は、緩衝液中 エラスターゼの至適 pHにおいて実施される。我々は pH8. 0のトリスバッファ一中 37 °Cでインキュベーションを行って満足な結果を得た。勿論トリス以外の緩衝液を用い て特定のエラスターゼ供給者の推奨する pH範囲例えば pH7. 5ないし 8. 5の範囲で
実施することも本発明の範囲内である。酵素濃度は一般に 300ないし 1000UZL、 好ましくは 600UZLが適当であり、インキュベーション時間は 37°Cにおいて 1日ない し 1週間、好ましくは 2ないし 7日である。
[0014] エラスターゼ処理が終った組織は、純水、生理食塩水、緩衝液などを用いてリンス し、移植に使用されるまで PBSその他の保存液中で保存される。
実施例
[0015] 以下の実験は、(株)ジャパンファーム力 購入したブタ大動脈を用い、条件を変え てダルタルアルデヒド(GAと略す。)で処理し、処理後の組織をエラスターゼ(3. 85 UZmg、フナコシ) 150mgZL(577. 5UZUを添加した 0. 01M卜リスノ ッファー( PH8. 0)中で 37°C4日間の一定条件でインキュベートした。なお%は重量基準によ る。
[0016] 実施例 1
GA未処理、 0. 01%GA, 0. 1%GA, 1%GAおよび 25%GAに常温でそれぞれ 1時間浸漬し、上の一定条件でエラスチンを分解処理した。図 1の下列は輪切したブ タ大動脈の外観を示し、上列はへマトキシリンーェォジン染色した後の組織の拡大 光学顕微鏡写真である。見られるように、 GA未処理および GA0. 01%処理のもの はエラスチンの酵素分解除去により組織が過度に弛緩し、原形を保っていないが、 1 %および 25%GA処理したものは GAがエラスチンにも作用し、酵素分解後も組織内 にエラスチンが残留している様子が観察された。 0. 1%GAでは組織の原形が保た れたまま、エラスチンがほぼ完全に除去され、ドナー由来細胞の脱核も観察される。
[0017] 実施例 2
実施例 1と同様に、 GA無処理, 0. 1%GA, 1%GAおよび 10%GA処理後エラス ターゼ処理したブタ大動脈のへマトキシリンーェォジン染色した組織を異なる倍率で 光学顕微鏡写真撮影した。結果を図 2に示す。見られるように GA未処理のものは過 度の弛緩が認められ、 GA1%および 10%の処理では暗色のヒモ状部分として見え るエラスチンおよびドナー由来細胞核の残留が多く認められる。
[0018] 実施例 3
実施例 2と同じ条件で処理したブタ大動脈組織をエラスチカ'ワンギーソン染色法
により染色し、拡大光学顕微鏡写真撮影した。結果を図 3に示す。この染色法はエラ スチン線維を黒紫色に、コラーゼン線維を赤色に染め分ける染色法である。モノクロ ム写真ではエラスチン線維がより濃暗色に写るので、見られるように GA濃度が高くな るにつれ、エラスチン線維が多く残留することが見られる。
[0019] GAによる部分固定化、次いでエラスターゼによるエラスチンの酵素分解処理を受 けた組織が生体内で分解可能であることを確認するため、実施例 2の 0. 1%GA処 理後エラスターゼ処理した組織をコラゲナーゼ(228U/mg,和光純薬)のトリスバッ ファー溶液に浸漬処理し、分解溶液を分光学的に分析したところ、 280nm付近に芳 香族アミノ酸に起因する吸収が観察され、本発明方法によって作製されたスキヤフォ 一ルドは生分解性であることが推察された。
[0020] 以上の実験の結果、 GAによる適度の部分固定化、次 、でエラスターゼによるエラ スチンの酵素分解は、処理すべき生体組織の物理的形状を保持しつつ柔軟性を有 し、細胞の浸潤を助ける多孔質構造の再生医療用スキヤフォールドの作製に利用で きることが確かめられた。
Claims
[1] (a)生体軟組織をダルタルアルデヒドで部分的に架橋固定するステップ、および
(b)部分的に架橋固定ィ匕した組織をエラスターゼと共にインキュベートするステップ を含む、移植用スキヤフォールドの作製方法。
[2] ステップ(a)は、組織を 0. 1%またはそれ未満のダルタルアルデヒド水溶液中に 5 時間以下の期間浸漬することによって実施される請求項 1の方法。
[3] ダルタルアルデヒド濃度が 0. 1%であり、浸漬時間が常温において 1時間である請 求項 2の方法。
[4] ステップ(b)は、緩衝液中エラスターゼの至適 pHにお!/、て行われる請求項 1な!、し 3のいずれかの方法。
[5] 緩衝液が 7. 5ないし 8. 5の pHを有するトリス緩衝液である請求項 4の方法。
[6] ステップ (b)の後、組織を洗浄するステップをさらに含んで 、る請求項 1な 、し 5の いずれかの方法。
