Abstract
【課題】 臓器機能の低下した患者に対しても、副作用が問題とならず、更に高い抗菌効果を奏することが出来る新たなペプチドを提供すること。
【解決手段】 以下のアミノ酸配列から成るペプチド:
(1)配列番号1: Lys-Arg-Leu-Phe-Arg-Arg-Trp-Gln-Trp-Arg-Tyr、
(2)配列番号2: Arg-Arg-Trp-Gln-Trp-Arg-Met-Lys-Lys-Tyr、
(3)配列番号3: Lys-Arg-Trp-Gln-Trp-Arg-Leu-Tyr、又は

新規抗菌性ペプチド
技術分野
[oooi] 本発明は、新規なペプチドに関し、特に、殺菌及び Z又は発育阻止作用を有する、 新規なペプチド及び該ペプチドを用 ヽた組成物等に関する。
背景技術
[0002] ヒトに病原性を有する真菌の多くは二相性であり、即ち、環境条件に応じて酵母形 と、菌糸形の両者を取り得る。一部は二相性でないものもある。酵母型は卵円形の単 細胞の状態であり、出芽により分裂増殖する。菌糸型は、円筒形の細胞が縦に連な つたり、あるいは多核性の 1個の細胞が長く延びて菌糸と呼ばれる糸状の構造が枝分 かれたりしたりする状態を示すものである。
[0003] このような真菌として、カンジダ、タリプトコッカス、ァスペルギルスなどを挙げることが できる。カンジダは、正常人の口腔咽頭、消ィ匕器などに常在し、タリプトコッカスは、は とやネズミなどの糞、土の中などに多く含まれるほ力、室内のごとき湿気の多い場所 に多く存在し、空気に運ばれて人と接触している。ァスペルギルスも昔カゝら鳥の糞や 土の中などに見つかつている力 最近では病院内のエアコンダクト、フィルタなどに多 く見つかつている。これらの真菌は、正常人に対する感染能力は低いが、感染すると 外毒素、内毒素、プロテアーゼなどの組織破壊物質を放出する。これらの真菌の多く は、抗癌剤、免疫抑制剤、ステロイド剤、広域抗生剤などの投与を受けている患者に 発生し、 日和見感染としての性格を有している。正常人ではこれら真菌に対して細胞 性免疫が十分に働き、感染防御に役立っているが、上記患者ではそれが低下してい るために感染を許すことになる。特に、悪性疾患患者、臓器移植患者あるいは集中 治療室で治療を受ける患者等では深在性カンジダ症をはじめとする深在性真菌症あ るいはカンジダ血症などを招き、重篤な転帰をとることが多!、。
[0004] このような真菌によって引き起こされる真菌症の治療薬としては、ポリェン系のアン ホテリシン Bが知られている。また、フルシトシン、イミダゾール系のケトコナゾール、ミ コナゾールなどの抗真菌剤も知られて 、る。

発明が解決しょうとする課題
[0005] しかしながら、これらの抗真菌剤は、用量依存的な腎毒性などの副作用が強いという 問題点がある。特に、カンジダなどの真菌のエルゴステロールに作用起点を持つ抗 真菌剤があり、該抗真菌剤が生体細胞膜のコレステロールなどのステロール類と類 似しているために、副作用を生じるという問題がある。また、臓器機能の低下した患者 に対しては、抗真菌剤の正常細胞に及ぼす影響が大きすぎて、従来の抗真菌剤の 使用が困難である。また、特に、 non-albicansカンジダ属による感染には、抗真菌剤 の効果が非常に低いことが報告されている。さらに、 1970年後半ころよりポリェン系に 対する耐性菌の出現が報告されるようになり,またこの近年はァズール系の抗真菌剤 に対する耐性機構が存在する耐性菌が存在することが明らかとなった。したがって、 臓器機能の低下した患者に対しても、副作用が問題とならず、かつ有効に抗菌効果 を奏する抗真菌剤があれば望ましい。しかし、このような抗真菌剤はこれまで知られ ていない。
[0006] 上記目的を達成するために、発明者らは、生体や牛乳に存在する抗菌性ペプチドを 鋭意研究した結果、抗真菌剤として有力な機能を有する新規ペプチドである Lys-Ar g- Leu- Phe- Arg- Arg- Trp- Gin- Trp- Arg- Met- Lys- Lys- Tyr (以下、このペプチドを「J H8194」と称する)を見出し、既に特許第 3472821号を取得している。
[0007] そこで、本発明の目的は、臓器機能の低下した患者に対しても、副作用が問題となら ず、更に高い抗菌効果を奏することが出来る新たなペプチドを提供することにある。 課題を解決するための手段
[0008] 本発明者は、上記 JH8194のアミノ酸配列: Lys- Arg- Leu- Phe- Arg- Arg- Trp- Gin- Tr D- Are- Met- Lvs- Lvs- Tvrにお 、て、下線で示した 3つのアミノ酸残基を除去すること で、全く予想外にその抗菌性が非常に高まることを見出し、本発明を完成した。更に
、JH8194のアミノ酸配列に基づき各種の改変体ペプチドも作成した。
[0009] 即ち、本発明は以下の各態様に係る。
[1]以下のアミノ酸配列から成るペプチド:
( 1 )配列番号 1: Lys-Arg-Leu-Phe-Arg-Arg-Trp-Gln-Trp-Arg-Tyr (「JH8194Y」 )、
(2)配列番号 2: Arg-Arg-Trp-Gln-Trp-Arg-Met-Lys-Lys-Tyr (「8194F」 )、
(3)配列番号 3 : Lys- Arg- Trp- Gin- Trp- Arg- Leu- Tyr(「8194W」)、又は
(4)配列番号 4: Lys-Arg-Leu-Phe-Lys-Tyr (「8194-6」 )。
[2]上記 1記載のペプチドから成る、抗真菌剤。
[3]真菌が、カンジダ、タリプトコッカス、又はァスペルギルスである、上記 2記載の抗 真菌剤。
[4]上記 1記載のペプチドを活性成分として有効量含む、真菌に対する殺菌及び Z 又は発育阻止作用を有する組成物。
[5]医薬組成物である上記 4記載の組成物。
[6]洗浄剤、抗かび剤、又は、うがい薬である上記 4記載の組成物。

発明の効果
[0010] 本発明のペプチドは 6個〜 11個のアミノ酸力も構成される低分子量分子である為 に、合成上でコスト面等で非常に有利である。更に、他に報告されている抗菌性のぺ
プチドゃ抗真菌剤と比較して非常に低濃度で、真菌に対して優れた抗菌効果ならび に殺菌効果を有しており、医療用ある 、は研究用の抗菌薬として応用した場合に大 きな利点があると考えられる。

発明を実施するための最良の形態
[0012] 本発明の新規ペプチドは、 6個〜 11個のアミノ酸から成るペプチドであり、ペプチド 合成法などの常套手段を用いて製造することができる。上記でいう Lysはリジン、 Arg はァノレギニン、 Leuはロイシン、 Pheはフエニノレアラニン、 Serはセリン、 Tyrはチロシン、 Metはメチォニンを示す。以下の実施例で示されるように、本発明の新規ペプチドは 、真菌に対する極めて優れた殺菌及び Z又は発育阻止作用を有する。従って、本発 明のペプチドは抗真菌剤として有用である。
[0013] 本発明のペプチドは、当業者に周知の任意のペプチド合成法で取得することがで きる。例えば、本発明のペプチドは、液相法及び固相法のいずれの方法も使用する ことができる。ここで、一般に、液相法は反応を溶液状態で行い反応混合物から生成 物を単離精製し、この生成物を中間体として次のペプチド伸長反応に用いる方法で ある。又、固相法は、反応溶媒に不溶の固相担体にアミノ酸を結合させ、このアミノ酸 に準じ縮合反応を行 、ペプチド鎖を伸長させて 、く方法である。
[0014] ペプチドの化学合成は、カルボキシル基を保護したアミノ酸にアミノ基を保護したァ ミノ酸を脱水縮合させ、ペプチド結合を形成させ、次にアミノ保護基を除去後、遊離し たァミノ基に次のアミノ基保護アミノ酸を順次、 C末端から N末端に向かって一つずつ 延長していく方法が基本である。脱水縮合反応では、カルボキシル基を活性ィ匕して、 結合させようとするアミノ基と反応させる。この活性化には、ジシクロへキシカルボジィ
ミド (DCC)法、活性エステル法、酸無水物法、アジド法等があるがその反応性の高さ とラセミ化その他の副反応を考慮して選ばれる。縮合反応時の副反応を防止するた めにアミノ酸のアミノ基、カルボキシル基、側鎖 (R)の官能基には保護基が導入される 。これらの保護基は、縮合反応の条件で安定であり、必要なときには速やかに除去さ れるものが好ましい。また、ァミノ基の保護基とカルボキシル基の保護基とは互いに 選択的に除去されることが好ましい。
[0015] ァミノ基の保護基としては、例えばべンジルォキシカルボ-ル (Bz)、 t-ブチルォキシ カルボ-ル (Boc)、 p-ビフエ-ルイソプロピロォキシカルボ-ル、 9 フルォレ-ルメチ ルォキシカルボ-ル (Fmoc)等が挙げられる。カルボキシ基の保護基としては、たとえ ばアルキルエステル、ベンジルエステル等を形成し得る基が挙げられる。但し、固相 法の場合は、 C末端のカルボキシル基はクロ口トリチル榭脂、クロルメチル榭脂、ォキ シメチル榭脂、 P アルコキシベンジルアルコール榭脂等の担体に結合している。縮 合反応は、カルポジイミド等の縮合剤の存在下、あるいは N 保護アミノ酸活性エス テル又はペプチド活性エステルを用いて実施する。
[0016] 縮合反応終了後、保護基は除去されるが、固相の場合はさらにペプチドの C末端と 榭脂との結合を切断する。さらに、本発明のペプチドは通常の方法に従い精製され る。たとえば、イオン交換クロマトグラフィー、逆相液体クロマトグラフィー、ァフィ-ティ 一クロマトグラフィー等が挙げられる。こうして合成したペプチドは、エドマン分解法で C-末端力もアミノ酸配列を読み取るプロテインシークェンサ一、 GC— MS等でそのァ ミノ酸配列を分析し確認することができる。
[0017] 本明細書において、「真菌」とは、例えば、カンジダ、タリプトコッカス、ァスペルギルス 、白癬菌等の当業者に公知の任意の属に含まれる菌を包含する。
[0018] 本発明の組成物は本発明のペプチドを活性成分として有効量含み、従って、真菌に 対する殺菌及び Z又は発育阻止作用を有する。本発明組成物の用途は以下に記載 するように多岐に亘る。更に、本発明は、該ペプチド、該ペプチドから成る抗真菌剤、 又は該組成物等を投与することから成る、真菌に対する殺菌及び z又は発育阻止方 法にも係る。
[0019] 例えば、本発明の組成物が医薬組成物である場合には、真菌に対する殺菌及び Z
又は発育阻止作用を奏効するに十分な量の本発明のペプチドを有効量として含有 させること力 S出来る。例えば、 0. 001〜0. 01重量0 /0、好ましくは、 0. 005〜0. 01重 量%の割合で含有させることができる。力かる範囲としたのは、 0. 001重量%未満と する本発明のペプチドの効果を得られない場合があり、 0. 02重量%以上では、溶 血が起こらな 、からである。
[0020] 本発明の医薬組成物は、当業者に公知の任意の投与経路、例えば、経口投与、非 経口投与、及び直腸内投与で投与することが出来るが、経口投与が好ましい。本発 明の医薬組成物に含まれる本発明ペプチドの投与量は、投与方法、患者の症状、 年齢等により異なるが、血液量が成人で約 5Lと仮定すると、通常 1回 1〜: LOmgZkg、 好ましくは、 5〜: LOmg/kgを 1日当たり 1回〜複数回 (約 5〜7回)である。
[0021] 本発明の医薬組成物には、活性成分である本発明のペプチドのほかに、製剤用担 体又は補助剤として当業者に公知の任意の物質が含まれていても良い。尚、かかる 物質は本発明のペプチドと実質的に反応しない物質であることが必要である。
[0022] このような製剤用担体又は補助剤の具体的としては、例えば、乳糖、ブドウ糖、マン ニット、デキストリン、シクロデキストリン、でん粉、ショ糖、メタ珪酸アルミン酸マグネシ ゥム、合成ケィ酸アルミニウム、カルボキシメチルセルロースナトリウム、ヒドロキシプロ ピルデンプン、カルボキシメチルセルロースカルシウム、イオン交換榭脂、メチルセル ロース、ゼラチン、アラビアゴム、ヒドロキシプロピルセルロース、ヒドロキシメチルセル ロース、ポリビュルピロリドン、ポリビュルアルコール、軟質無水ケィ酸、ステアリン酸マ グネシゥム、タルク、トラガント、ベントナイト、ビーガム、酸化チタン、ソルビタン脂肪酸 エステル、ラウリル硫酸ナトリウム、グリセリン、脂肪酸グリセリンエステル、精製ラノリン 、グリセ口ゼラチン、ポリソルベート、マクロゴール、植物油、ろう、流動パラフィン、白 色ワセリン、フルォロカーボン、非イオン性界面活性剤、プロピレンダルコール、水等 が挙げられる。
[0023] 本発明の医薬組成物は投与経路、患者の症状、年齢等に応じて、任意の剤型をとる ことが可能である。例えば、錠剤、カプセル剤、顆粒剤、散剤、シロップ剤、懸濁剤、 坐剤、軟膏、クリーム剤、ゲル剤、貼付剤、注射剤等が挙げられる。これらの製剤は 常法に従って調製することが出来る。なお、液体製剤にあっては、適用に際して、水
又は他の適当な溶媒に溶解又は懸濁する形であってもよい。また錠剤、顆粒剤を、 周知の方法でコーティングしてもよい。注射剤の場合には、本発明のペプチドを水に 溶解させて調製されるが、必要に応じて生理食塩水あるいはブドウ糖溶液に溶解さ せてもよぐまた緩衝剤や保存剤を添加してもよい。
[0024] 更に、本発明組成物は、洗浄剤、抗かび剤、うがい薬などの各種用途を有する。
[0025] 例えば、本発明の組成物を洗浄剤、抗かび剤又はうがい薬等として使用する場合に は、洗浄する対象の医療機器、真菌による汚染の程度等に応じて、該組成物中の本 発明ペプチドの最適量を適宜設定することが出来る。例えば、洗浄剤及びうがい薬 中などに添加して使用する場合、洗浄剤中に含まれる本発明のペプチドの濃度を、 0 .1〜数十 Mの範囲とすることができる。このような低濃度であっても十分抗菌効果を 発揮することができるからである。本発明のペプチドの濃度は、好ましくは、 0.3〜5 Mの範囲であり、更に好ましくは、 0.6〜2.5 Mの範囲である。但し、上記の範囲に特 に限定されることはない。
[0026] 尚、本発明の医薬組成物に準じて、上記の各種組成物中にも、活性成分である本発 明のペプチドのほかに、適当な担体又は補助剤として当業者に公知の任意の物質 が含まれていても良い。
[0027] 更に、本発明のペプチドから成る抗真菌剤は、例えば、コンタクトレンズ等の各種医 療用具'機器の材料 (原料)物質に含有させることによって、例えば、抗菌性コンタ外 レンズのような、本発明のペプチドを含有し抗菌性が付与された医療用具'機器を作 製することも出来る。尚、このような本発明の医療用具'機器は、例えば、前記材料( 原料)物質と本発明のペプチドを適当な方法で混合したり、又は、材料 (原料)物質を 成型した後、それらの表面を適当な方法 (例えば、塗布又は噴霧)を用いて本発明の ペプチドで被覆すること等の当業者に公知の任意の手段によって、上記材料 (原料) 物質に本発明のペプチドを含有させることにより、容易に作成することが出来る。 実施例
[0028] 以下、実施例に則して本発明を更に詳しく説明する。尚、本発明の技術的範囲は これらの記載によって何等制限されるものではない。尚、本明細書中で引用される技 術文献の内容は、本明細書の開示内容の一部と見なされる。
[0029] 抗菌性試験 1
本発明ペプチドの抗菌効果を確認するために、カンジダに対する抗菌作用を調べた 。本発明ペプチドは常法のアミノ酸合成法により、島津製のペプチドシンセサイザー を使って行った。カンジダの菌株は、イギリスのグラスゴーデンホスピタルによって、患 者の口腔内から分離されたものを供与された。このカンジダ菌を対象として、本発明 ペプチドの抗菌効果を調べた。 Edgertonら(1998)の方法により、 C. albicansの菌液( 濃度 1.8 X 105 cfo/mL)を 20マイクロリットルと、等量のペプチド溶液(濃度 0.325 μ Μ〜 25 μ Μ)とを混和し、 90分間 37°Cで反応させた後, Yeast Nitrogen Base培地を添カロし て反応を止めた。その後、サブロー寒天培地上に播種し、 37°Cで 48時間培養後に、 生菌数をコロニーカウントによって求め、ペプチド溶液の代わりに ImMリン酸緩衝液 を用いた対照溶液と菌液を反応させた場合のコロニー数 (100%)と比較したときの残存 率 (%)を算出した。
[0030] 得られた結果を図 1に示す。本発明のペプチド (JH8194Y)の抗菌実験結果(〇)を、 J H8194 ( * )、 His5:ヒスタチン 5 (DSHEKRHHGYKRKFHEKHHSHRGY) (♦)、 LFB:ゥ シラクトフエリン(FKCRRWQWRMKKLGAPSITCVRRAF) (國)、 LFB frag (FKCRRWQ WRMKKLG) (▲)及び Dhvar 4 :オランダ ACTAで合成されたヒスタチン改変体(KRLF KKLLFSLRKY) ( X )の抗菌実験結果と比較した。図 1の縦軸は、カンジダ菌の残存 率を示す。横軸は、本ペプチドの濃度を示す。唾液中に存在する塩基性抗菌性ぺプ チドのヒスタチンファミリーのうち最も抗菌性が強いとされるでは、 0-50 μ Μの範囲で 1 00%の致死率を得られなかった。
[0031] 図 1から明らかなように、ラタトフエリンやラタトフエリシン Βと比較して非常に高い抗菌 性を示した JH8194の場合、 2.5 μ Μで 100%の致死率を示した力 本発明ペプチドの 場合は更にその 2分の 1に当たる 1.25 Μで 100%の致死率を示した。抗真菌剤の 中で作用が非常に強いとされるアンホテリシン Βの最小殺滅濃度 (Minimal Fungicidal Concentration (MFC))力^ μ Μであることからも、本発明のペプチドはこれと比較し ても極めて低濃度において真菌に対して非常に強い抗菌効果を有することが確認さ れた。

本発明の他のペプチドについての抗菌性も、抗菌性試験 1と同様に調べた。得られ た結果を図 2に示す。

ヒト赤血球を用い,各ペプチドを最終濃度が 0.325— 100 Mの範囲で調整し, Nikaw a H, Fukushima H, Makihira b, Hamada T, bamaranayake LP. Fungicidal effect of thr ee new synthetic cationic peptides against Candidaaloicans. Oral Dis. 2004 Jul ;10(4) :221-8.に記載の方法で行った。その結果,市販の抗真菌剤アンホテリシン Bが 5 M 以上で溶血を引き起こしたのに対し、本発明のペプチドでは少なくとも 100 μ Μまで 溶血は認めなかった(図 3)。
[0034] 毒件試験 2
ヒト腸骨由来骨髄間葉系幹細胞 (Cambrex Bio Science Walkers ville, Inc., Walkersvill e, MDより購入)(Product Code: PT- 2501. LotNumber:4F0218)は CD29、 44、 105、 16 6がポジティブで、 CD14、 34、 45がネガティブであること、および骨、軟骨、脂肪への 分化能をもつことが確認されている:以下「MSC」と略する。)を、 Dulbecco' s Modified Eagle Media (Sigma Co. St. Louis, Mo) (最終濃度 100 unit/mLのペニシリン G、最終 濃度 100 g/mLの硫酸ストレプトマイシン、最終濃度 0.0085%のアンホテリシン B (以 下、抗生物質液: GIBCO社)及び 10%牛胎児血清 (FBS)を含む)(以下、「培地 A」と略 する)にて 2回継代培養したものを凍結保存した。各実験には凍結保存した細胞を毎 回融解し、培地 Aにて培養したものを実験に用いた。上記 JH8194,JH8194Y, 8194F, 8194Wに示されるアミノ酸配列力もなる塩基性抗菌性ペプチドを用いて、ヒト間葉系 幹細胞に対する細胞増殖効果について検討した。ヒト間葉系幹細胞は、上記 DME M培地に 10%の子ゥシ胎仔血清を添カ卩したものに各ペプチドを 1もしくは 2 μ Μ添カロ し,細胞増殖培地として 5%CO下, 37°Cで培養した。また、細胞数は、コールター

カウンタ一により測定し,細胞増殖がペプチド非添加のコントロールとくらべ有意差が あるかないかで細胞毒性を判定した。その結果、本発明のペプチドは上記ヒト細胞の 増殖には有意な影響は与えな 、ことが示された (図 4)。
産業上の利用可能性
[0035] 本発明のペプチドは、他に報告されている抗菌性のペプチドや抗真菌剤と比較して
、非常に低濃度で、真菌に対して優れた抗菌効果ならびに殺菌効果を有しており、 医療用あるいは研究用の抗菌薬として応用した場合に大きな利点があると考えられ る。更に、本発明のペプチドは 6個〜 11個のアミノ酸力も構成される低分子量である 為に、合成上のコスト面等で産業上非常に有利である。

(1)酉 ti列番 1: Lys- Arg- Leu- Phe- Arg- Arg- Trp- Gin- Trp- Arg- Tyr、
(2)配列番号 2: Arg- Arg- Trp- Gin- Trp- Arg- Met- Lys- Lys- Tyr、
(3)配列番号 3 : Lys- Arg- Trp- Gin- Trp- Arg- Leu- Tyr、又は
(4)配列番号 4: Lys- Arg- Leu- Phe- Lys- Tyr。
[2] 請求項 1記載のペプチドから成る、抗真菌剤。
[3] 真菌が、カンジダ、タリプトコッカス、又はァスペルギルスである、請求項 2記載の抗真 菌剤。
[4] 請求項 1記載のペプチドを活性成分として有効量含む、真菌に対する殺菌及び Z又 は発育阻止作用を有する組成物。
[5] 医薬組成物である請求項 4記載の組成物。
[6] 洗浄剤、抗かび剤、又は、うがい薬である請求項 4記載の組成物。
[7] 真菌が、カンジダ、タリプトコッカス、又はァスペルギルスである、請求項 4〜6の!、ず れか一項に記載の医薬組成物。
[8] 請求項 1記載のペプチドを含有し抗菌性が付与された医療用具,機器。
[9] 請求項 1記載のペプチドを投与することから成る、真菌に対する殺菌及び Z又は発 育阻止方法。
[10] 請求項 2記載の抗真菌剤を投与することから成る、真菌に対する殺菌及び Z又は発 育阻止方法。
[II] 請求項 4記載の組成物を投与することから成る、真菌に対する殺菌及び Z又は発育 阻止方法。
[12] 真菌が、カンジダ、タリプトコッカス、又はァスペルギルスである、請求項 9〜: L 1のい ずれか一項に記載の方法。
