発明の効果
本発明の遺伝子構築物は、動物細胞に導入することにより、酸素濃度のみならず、 病態を初期の段階力 重症に至るまでの過程をその重篤度に応じてリアルタイムで 観察することができる。例えば、図 11では、 1.0 X 104個のルシフェラーゼ発光腫瘍細 胞が検出可能であることを明らかにした。実際の腫瘍サイズでは、 1mmの腫瘍に最小 でも 4 X 105個の腫瘍細胞があることを考えると、本発明により lmmよりもかなり小さい 腫瘍まで in vivoで観察可能であることが明らかである。したがって、本発明の遺伝子 構築物を導入することにより得られたトランスジエニック非ヒト哺乳動物に固形がんが 形成された場合、癌の病態イメージングを的確に行うことができる。また、この癌の動 物モデルに抗癌剤の候補となる化合物を投与して癌の状態を観察することにより、癌 に対する有効性を判定することが出来、薬物のスクリーニング系や物理的治療法 (例
えば、手術、放射線療法、温熱療法)の評価系としても有用である。
[0011] 本発明は、放射線治療ゃ抗癌剤に耐性を示す腫瘍内低酸素環境の基礎研究、腫 瘍内の酸素環境に影響を及ぼす薬剤の薬効評価を好適に行うことができる。
[0012] 例えば、本発明は、虚血性疾患の病態イメージングおよび治療法の開発、自然発 生癌のモデルマウスでの病態イメージングに特に有用である。
また、癌以外に、狭心症、心筋梗塞、脳梗塞などの虚血性疾患、糖尿病の合併症の 末梢循環障害或いは糖尿病性腎症、動脈硬化に関連する疾患、血管の結紮或いは 組織の圧迫などによる人工的な虚血或いは血流の低下などの虚血に関与する病態 のイメージングが可能である。また、胃潰瘍、十二指腸潰瘍、呼吸器系の慢性炎症モ デル (例えば、タバコ或いはディーゼル、ガソリンの排気ガスの強制吸入により引き起 こすことができる)、或いは各種臓器、組織に対し起炎物質を投与することによる炎症 性の病態、外傷などの広範な病態のイメージングができる可能性もあると考えられる
[0013] 本発明の遺伝子構築物を組み込むことで、臓器な 、し組織の病態の変化を、動物 に侵襲を与えることなく評価できる。従って、本発明は複数の薬物の投与スケジユー ルなどを含めた種々の治療法の開発に非常に有用である。

[0018] ヒト子宮頸癌由来細胞株 HeLaの total RNAを、 ISOGENE (-ツボンジーン)を用いて 抽出した。これを铸型とし AMV reverse transcriptase XLを用いて逆転写反応を行う ことによってヒト HIF-1 α遺伝子の cDNAを獲得した。この cDNAを铸型とし、表 1に示 す 10種のプライマー(ODD- Bg FO, - Fl, - F2, - F3,- F4、および ODD- Nco- R0, - Rl, -R2, -R3, - R4)の- Fと- Rを組み合わせて PCRを行い、系統的に欠失させた ODD領 域をコードする DNA断片を獲得した。
[0019] これらの DNA断片を Bglll及び Ncolで消化することによって 5'末端および 3'末端を それぞれ Bglll及び Ncol消化末端にし、 pGL3/3-0を Bglll及び Ncol処理して調製した プラスミドベクターに組込んだ。本遺伝子組換えによって、 ODD領域の各種欠失 DN A断片(0-0、 0-1、 0-2、 0-3、 0-4、 1-0、 1-2、 1-3、 1-4、 2-0、 2-3、 2-4、 3-0、 3-4、 4-0 )と、 NLS、及びルシフェラーゼが融合した遺伝子を発現するプラスミド (それぞれ pGL 3/0—0、 pGL3/0— 1、 pGL3/0— 2、 pGL3/0— 3、 pGL3/0— 4、 pGL3/l— 0、 pGL3/l— 2、 pGL 3/1-3、 pGL3/l- 4、 pGL3/2- 0、 pGL3/2- 3、 pGL3/2- 4、 pGL3/3- 0、 pGL3/3- 4、 pGL 3/4-0)を構築した。
[0020] 24穴培養ディッシュに HeLa細胞をまき(10000 cells/well)、 16時間培養した後に上 記プラズミド (0.4 g/well)をそれぞれ Polyfect Transfection Reagent (QIAGEN)にて導 入した。この時ゥミシィタケルシフェラーゼを恒常的に発現するプラスミド pRL/CMV ( Promega)もインターナルコントロールとして同時に導入した(0.04 g/well)。遺伝子導 入力も 24時間後に培地を交換し、その後さらに 18時間にわたって有酸素、または低 酸素条件化 (酸素濃度く 0.02%)にて培養した。培地を吸引後、 100 mlの Passive Lysis Buffer (Promega)にて細胞抽出液を収集し、付属の説明書に従ってデュアルルシフ エラーゼアツセィを行った。
[0021] 実験の結果、 ODD/3-0ドメイン配列を融合した時に、最大の酸素濃度依存的タン ノ ク質分解効率が得られることがわ力つた。
[図 2]5HREプロモーター活性の低酸素応答性制御 低酸素特異的なプロモーターを 用いることで、図 10に示したような ODDを介した分解系の限界を十分解決することが できる。
5HRE-Lucを導入した細胞をグラフ(上)に示した時間有酸素(口)または低酸素(國
)で培養した後、細胞内で発現したルシフェラーゼの活性を測定した。データは 3回 の実験の結果を平均と SDで示した。下の図は、上述の低酸素で培養した細胞内で の HIF-1 aのタンパク質をウェスタンブロット法で調べた結果である。 ODDを介する分 解機構が、 western blotで示した HIF-1 aのタンパク質と同様に起こるということを考え ると、 5HREを用いることにより、有酸素状態の組織でのレポーター遺伝子の発現を 力なり押さえることができると考えられる。更に、低酸素状態の組織での発現誘導は、 時間が長くなるにつれて (慢性ィ匕することにより) 100倍以上に増加し、イメージングも それにつれて増強されていくことが期待される(実際 in vivoイメージングで増強されて いる図が図 6)。
[図 3]既存レポーターとの比較 (ルシフェラーゼ活性の低酸素 Z有酸素比とバックダラ ゥンド) 5HREプロモーターだけを用いた時と 5HREに ODDを組み合わせた時のレポ 一ター遺伝子の発現を比較した。特に誘導比 (低酸素/有酸素)(上グラフ)と有酸素 での発現 (バックグラウンド)(下グラフ)の比較を行った。その結果、 5HREに ODDを 組み合わせたレポーターの方が、ノ ックグラウンドが小さくなり、その結果誘導比が著 しく向上する事がわかった。上段; PGL3/5HRE- Lucもしくは pGL3/5HREp- NLS- OD D-Luciferase (pGL3/5HRE- ODD- Luc)を、 pRL-CMVとともに HeLa細胞に導入し、低 酸素/有酸素条件化で培養した後に、デュアルルシフ ラーゼアツセィを行った。低 酸素条件化および有酸素条件化における「ゥミシィタケルシフ ラーゼの活性に対す る蛍ルシフェラーゼの活性の比」をそれぞれ算出し、次に有酸素条件化における比 に対する低酸素条件化での比を算出し、 Induction Ratioとした。下段;上記処理後の 有酸素条件化におけるホタルルシフ ラーゼの活性 (実測値)をグラフにした。細胞 抽出液の代わりに Passive Lysis Bufferを用いてルシフェラーゼアツセィを実施し、 Bac kgroundを測定した。

一だけを用いた時と 5HREに ODDを組み合わせた時のレポーター遺伝子の発現を比 較した。低酸素状態力 有酸素状態に環境が変化したときのレポーターの変化を調 ベた。
[0023] 24穴培養ディッシュに HeLa/5HRE- Luc、および HeLa/5HRE- ODD- Luc細胞を播 種(1 X 104 cells/well)し 16時間培養した後、 18時間にわたって低酸素処理を行った。 細胞を低酸素チャンバ一力 取り出した直後にシクロへキサミド含有培地に培地交 換し、 0、 10、 30、 60分間にわたって 5% C02インキュベーターで培養をすることによつ て再酸素処理をした。その後培地を吸引し、 Passive Lysis Buffer (Promega Co.)を 10 0 1/well添加して- 80 °Cにて凍結した後、融解することによって細胞抽出液を収集 した。 20 1の細胞抽出液を用いルシフェラーゼアツセィ (Promega)を行った。
[0024] 再酸素化から 0、 10、 30、 60分後にルシフェラーゼ活性を測定し、 0分におけるルシ フェラーゼ活性に対する各測定時間におけるルシフェラーゼ活性の比を算出して Rel ative Luciferase Activityとした。その結果、 5HREに ODDを組み合わせた時の方が、 レポーターの減少が速やかで、よりリアルタイムに環境の変化を捕らえることができる ことがわかった。
[図 5]固形腫瘍内低酸素がん細胞のモニタリング (移植) 低酸素環境下におかれた 細胞をどれくらい感度良くモニターすることができるかを調べる目的で、 5HRE-mp-N LS-ODD-Luciferase遺伝子構築物を安定に組み込んだヒトがん細胞をヌードマウス の下腿部の皮下に移植してルシフェラーゼによる発行をモニターした。 1 X 106ヒトが ん細胞をヌードマウスの右足(circle)に皮下移植した。移植直後は有酸素環境下に あるため見えない (左図)。次の日には既にイメージが見えており、 3日後には腫瘍と しては観察されないものの、イメージは非常にはっきり観察される。固形腫瘍内の低 酸素がん細胞内のルシフェラーゼ発光を捕らえている (右図)
[図 6]虚血処理によるルシフェラーゼ発光の増強 低酸素によるレポーター遺伝子の 発現誘導を比較する目的で、 5HREプロモーターだけのレポーター遺伝子を組み込 んだ細胞で作った腫瘍と、 5HREに ODDを組み合わせたレポーター遺伝子を組み込 んだ細胞で作った腫瘍で、結紮により血流を悪くして低酸素状態を作ったときの発現 誘導を比較した。
[0025] HeLa/EF— Lucを左下腿部皮下に、 HeLa/5HRE- Luc (上段 a- d)または HeLa/5HR E-ODD-Luc (下段 e-h)を右下退部皮下に移植した。右下腿を結紮後、 0、 2、 4、 8時 間後にルシフェラーゼ発光量を測定した。その結果、レポーター遺伝子の発現誘導 にはいずれの遺伝子構築物でも発現誘導の早さに大きな差が無いことがわ力つた。 また、両足に腫瘍細胞を移植し、右足だけの血流を悪くすると、縛った方だけィメー ジの増強がみられた。
[図 7]虚血処理後再酸素化のルシフェラーゼ発光の推移 次に再酸素化によるレポ 一ターの発現変化を比較する目的で、図 6と同様に結紮により血流を悪くして低酸素 状態を作った腫瘍の結紮を解き、血流を回復した後のレポーターの発現変化を比較 した。
[0026] 左下腿に HeLa/EF-Luc (internal control),右下腿に HeLa/5HRE- Luc (右上段)ま たは HeLa/5HRE-ODD- Luc (右下段)の細胞を移植した。右下腿を 18時間にわたつ て結紮した後ルシフェリンを iv投与した。麻酔後に結紮を解き、その 0、 5、 15、 25、 35 分後にルシフ ラーゼ発光量を測定した。
[0027] 各測定時間における左下腿のルシフェラーゼの発光量に対する右下腿の発光量 の比を算出し、さらに 0分の値に対する各測定時間の値の比を算出して Relative Luci ferase Activityとした。その結果、培養細胞を用いた実験(図 4)と同様に、 ODD力 S無 い場合は、再酸素化されてもイメージは変わらないが、 in vivoでも 5HREに ODDを組 み合わせた時の方が、レポーターの減少が速やかで、よりリアルタイムに環境の変化 を捕らえることができることがわ力 た。
[図 8]トランスジエニックマウス 図 7では、 5HRE- mp- NLS- ODD- Luciferaseを安定に 組み込んだ細胞を用いて実験を行ったが、上記遺伝子構築物を全身の細胞に安定 に組み込んだトランスジエニックマウスでも、同様な現象が観察されるか否かを調べる 目的で、トランスジエニックマウスを用いた実験を行った。
[0028] トランスジエニックマウスの右下腿を 16時間に渡って結紮した。ルシフェリン投与後 1 1分経過した時点からイソフルランガスにて麻酔し、 13分の時点で撮像した (-2 min)。 ルシフェリン投与後 15分で結紮を解き、右下腿を再酸素化 (reoxygenation)した。再 酸素化した時点を基準 (0 min)として、以降 2、 5、 8、 10、 12、 15分で撮像した。その後
直ちにルシフェリンを追加投与し、さらに 17、 20、 23、 26、 30分の時点で撮像した。そ の結果、縛った足でのみルシフェリンの発光が観察され、発光は結紮を解除後、経 時的に速やかに消失し、その後のルシフェリンの再投与でも発光は観察されなかつ た。トランスジエニックで低酸素状態がモニターできた事を示して 、る。

[図 10]細胞内に導入されたタンパク質量と ODDドメインを介した酸素濃度依存的タ ンパク質分解効率の関連を調べる目的で、 NLS- ODD - β -galactosidaseタンパク質

に細胞膜を通過して細胞内に自由に蛋白を入れることが出来る膜透過ドメイン (PTD) を融合させたタンパク質を濃度を変えて細胞培養液に表示して 、るユニット数に相当 する /3 -galactosidase活性をもつタンパク質を培地に添加して、有酸素状態(20%)と 低酸素状態(1 %)で 24時間培養した。その後、細胞をホルマリン固定し、 PBSで 3回 洗い、 j8 -galactosidaseの基質を含んだ溶液に細胞を反応させることで、 j8 - galactosi daseタンパク質の活性(つまり 13 -galactosidaseタンパク質の細胞内残存量)を有酸素 と低酸素状態で比較した。 1.25 unit/mlまでのタンパク質を添加した時は、明らかに 低酸素状態の細胞でのみ安定ィ匕したタンパク質が確認できるが、それ以上濃度があ がると有酸素状態でもタンパク質の安定ィ匕がみられ、ある一定量以上のタンパク質が 細胞内に存在すると ODDを介した分解機構の限界を超えて、もはやタンパク質を分 解することが出来ないことを示している。このことは、恒常的なプロモータ一- NLS-OD D-レポーターという遺伝子構築物を使っても、レポーター遺伝子が沢山発現する組 織ではレポーターが残存して低酸素に起因する病態イメージングを正確に行えない ことを示唆している。
[図 11]A :図に示した数の HeLa/EF- Luc細胞を含む細胞懸濁液 100 μ 1と、 50 μ 1のル シフェリン溶液 (0.1mg/ml)とを各ゥエルにて混合した。その後直ちに IVIS-200システ ム(Xenogen)に化学発光をイメージングした。その結果、 103個オーダーのルシフェラ ーゼ発現細胞を可視化できることが明らかになった。 B: (A)の実験結果を元に、各ゥ エルの化学発光量を Igor Pro 4.09A (Xenogen)を用いて定量した。その結果、 7.8 X 1 02個の細胞から生じる化学発光量力 ¾.9 X 102個の細胞からの化学発光量と比較して
有意に大きいことが明らかになった(p〈0.05)。この結果は IVIS-200システムによって 1 02個オーダーの細胞が生じるルシフェラーゼの発光を検出できることを示して 、る。 ( 平均士 SD; n=2) C :ヌードマウス背部皮下に 1.0 X 105個(左)、 1.0 X 104個(右)の HeL a/EF-Luc細胞を移植し、翌日に IVIS-200システムを用いて化学発光を検出した。 In vivoにおいて、少なくとも 1.0 X 104個のルシフェラーゼ発光細胞を検出できることが明 らかになつた。
[図 12]ヒト子宮頸癌由来細胞株 HeLaの total RNAを铸型とし AMV reverse transcripta se XLを用いて逆転写反応を行うことによってヒト HIF-1 a遺伝子の cDNAを獲得した 。この cDNAを铸型とし、表 1に示すプライマーを ODD- Bg F3と A588- Nco- anti、 OD D— Bg卜 F3と A593— Nco— antiゝ ODD— Bgト F3と T598— Nco— antiゝ Q553— Bgト senseと A588 - Nco- anti、 L557- Bg卜 senseと A588- Nco- antiの組み合わせで PCRを行 ヽ、これらの D NA断片を Bglll及び Ncolで消化することによって 5 '末端および 3 '末端をそれぞれ Bgl II及び Ncol消化末端にし、 pGL3/3-0を Bglll及び Ncol処理して調製したプラスミドべク ターに組込んだ。本遺伝子組み換えによって、 ODD領域の各種欠失 DNA断片(HIF -1 aのアミノ酸番号で 548- 588、 548-593、 548-598、 553-588、 557-588)と、 NLS、及 びルシフェラーゼが融合した遺伝子を発現するプラスミド (それぞれ PGL3/548-588、 PGL3/548- 593、 pGL3/548- 598、 pGL3/553- 588、 pGL3/557- 588)を構築した。これ らのプラスミドを用いて、図 1と同じアツセィ法でルシフェラーゼ活性を測定した。使用 したプラスミド DNAは、 pGL3プロモーターベクター、 pGL3/3- 0、 pGL3/3- 4、 pGL3/54 8-588、 pGL3/548-593、 pGL3/548-598、 pGL3/553-588、 pGL3/557-588であり、図 中ではそれぞれ pGL3、 548-603 (3-0)、 548-583 (3-4)、 548-588、 548-593、 548-598 、 553-588、 557-588と表記した。結果として、 ODDをルシフェラーゼと融合させた本 図中の全ての融合タンパク質は酸素濃度依存的な安定性制御を受けることが明らか になった。中でも、 ODD553-588の領域をルシフェラーゼと融合させることによって、 0 DD3-0(548-603)を融合させた場合と同等の強い酸素濃度依存性を獲得出来ること が明らかになった。
発明を実施するための最良の形態
本発明の特徴は、低酸素或いは病態組織を非常に初期の段階でイメージングするこ
とができ、病態の解明或いは病気の治療薬を非常に効率的にスクリーニングできる にある。
[0030] 例えば図 5は、僅か 1 X 106個のガン細胞をヌードマウスに移植後 1日で癌をィメー ジングすることができ、 3日後の腫瘍の観察が不可能な段階において非常に鮮明な イメージングを行えることを示している。同様に図 11は僅か 1 X 104個のガン細胞を 検出することが出来ることを示している。
また、本発明の遺伝子構築物を使用すると、病態の重症度 (例えば酸素濃度、ガン 細胞の数)に応じて適切なイメージングが行われる(図 6〜8)。
[0031] さらに、病態モデル動物と交配させた本発明のトランスジエニックマウスは、当該病 態のイメージングを行うことができ、発症時期の特定、発症部位の特定、病変部位の 経時的観察、治療方法の開発、治療効果の経時的観察などに威力を発揮する。

大きく低下する。特許文献 1と 2は、レポーター遺伝子産物と低酸素誘導性プロモー タ(HRE)の組み合わせを開示する。低酸素状態を確認する場合、 HREとレポーター 遺伝子を組み合わせて低酸素時の発現量をできるだけ高くすることが従来行われて きた。ここに ODDを組み合わせると、レポーター遺伝子が一部分解して発現量が低 下するが、誘導比が大きくなるため、結果として鮮明な病態部位のイメージングが可 能になることを本発明者は見出した。低酸素時において、レポーター遺伝子の発現 量を増加させるエレメントである低酸素誘導性プロモータ(例えば HRE)とレポーター 遺伝子の発現量を低下させる ODDを組み合わせて、病態部位のイメージング効率を 向上させ得ることは、本発明で初めて明らかにされた。

実施例
[0065] 以下、本発明を実施例に基づきより詳細に説明する。
(1) 組み換え DNA (5HRE- mp- NLS- ODD- Luciferase)の構築
本発明のトランスジエニックマウスが有する組み換え DNAを含む発現ベクターは、 次のような手順で構築された。 pCH/3-Οプラスミド(Harada et al. 2002. Cancer Res.) を铸型とし、 NLS— Nco— sense primer; 5,— AAC CAT GGC GCC TAA GAA GAA GA G GAA G— 3,、および ODD— Nco— anti primer; 5' -AAC CAT GGT CTG CTG GAA TAC TGT AAC TG- 3,を用いて NLS- ODDの DNA断片(NLS- ODD548- 603をコード する DNA断片)を PCR法で増幅した。これを Ncolで消化後、 5HRE-hCMVmp-Lucプ ラスミド(Shibata et al. 2000. Gene Ther.;以降 pGL3/5HRE- Luc)の Ncol部位に挿入 することによって、 5HREプロモーターの制御下で NLS- ODD- Luciferase融合タンパク 質を発現するプラスミド pGL3/5HREp-NLS- ODD- Luciferaseを構築した。このプラスミ ドおよび PGL3/5HRE- Lucプラスミドの Kpnl- Xbal断片を、 pEF/myc cyto (Invitrogen) の Kpnl- Xbal部位に挿入することによってそれぞれ pEF/5HREp- ODD- Luc、 pEF/5H RE-Lucを構築した。
[0066] 恒常的にルシフェラーゼタンパク質を発現するプラスミドベクターは、次のような手 順で構築された。 Luc- Bam- sense primer 5' -AAG GAT CCA CCA TGG AAG ACG CCA AA - 3,、および Luc- RV- anti primer 5,- TTG ATA TCT TAC ACG GCG AT C TTT CC— 3,を用い、 pGL3プロモーターベクターを铸型として PCRを行うことによつ
て、ルシフェラーゼ遺伝子の cDNAを獲得した。これを BamHIおよび EcoRVで消化後 、 pEF6/Myc-His B (Invitrogen)プラスミドの BamHI- EcoRV部位に挿入し、恒常的に ルシフェラーゼを発現するプラスミド pEF/Lucを構築した。
(2)細胞培養
ヒト子宮頸癌由来 HeLa細胞を ATCCより入手し、ペニシリン 100 unit/ml,ストレプト マイシン 100 μ g/ml,及び 10% Fetal Bovin Serumを添カ卩した Dulbecco ' s Modified E agle Medium : D— MEMで培養した。
(3)レポーター遺伝子導入細胞の単離
リン酸カルシウム変法によって HeLa細胞に pEF/Luc、 pEF/5HRE- Lucおよび pEF/5 HRE-ODD- Lucを導入した。各プラスミドがゲノム DNAに安定に組み込まれた細胞株 を獲得する目的で、遺伝子導入翌日から 10日間に渡って 400 mg/mlの G418を含む 選択培地で培養し、形成したコロニーを単離した。 pEF/Lucを導入して単離したクロ ーンのうち恒常的にルシフェラーゼを発現するクローン(HeLa/EF-Luc)を実験で使 用した。一方、 HeLa細胞に PEF/5HRE- Lucまたは PEF/5HRE-ODD- Lucを導入して 得たそれぞれのクローンのうち、低酸素処理によるルシフェラーゼの誘導能が高いク ローン(HeLa/5HRE- Luc、および HeLa/5HRE- ODD- Luc)を実験で使用した。
(4)ルシフェラーゼ活性の測定
5HREを含む HIF-1依存性プロモーターの低酸素応答性を確認するために以下の ような実験を行った。 24穴培養ディッシュに HeLa/5HRE- Luc細胞を播種し(1 X 10* ce lls/well) 16時間培養した。その後有酸素条件 (20%O )または低酸素条件(1%0以下

)で 0, 1 , 2, 4, 8および 16時間培養した。その後、細胞を Phosphate Buffered Saline ( PBS)で洗净後、 100 1 Passive Lysis Buffer (Promega, Madison, Wisconsin)で溶解 し、 10 μ 1のノレシフェラーゼ活'性を luciferase assay kit (Promega)で測定し、 net count を図 2のグラフに示した。各々の細胞内で発現している HIF-1 aの蛋白を western bl ot法で確認した(図 2の下)。具体的には、上述した方法で調整した細胞を低酸素条 件で培養し、 0, 1 , 2, 4, 8および 16時間後、細胞を低酸素チャンバ一内で直接 100 1の 1 X loading bufferに溶解し、そのうち 20 μ 1を 7.5% SDS-polyacrylamide gel を用いて電気泳動法で展開し、 PVDF膜(Amersham Biosciences, Piscataway, NJ)に
転写し、膜上の HIF- laタンパク質を monoclonal anti- HIF- la antibody (BD Bioscie nce Pharmingen, San Diego, CA)と anti mouse IgG horseradish peroxidase linked wh ole antibody (Amersham Bioscience)を用いて標識し、 ECL— PLUS system (Amersham Bioscience)を用いて標識タンパク質を確認した。結果は、図 2に示した。
[0067] レポーター遺伝子がホタルルシフェラーゼのみの時と、 NLS-ODDを融合させたホタ ルルシフェラーゼの低酸素応答性を比較する目的で以下の実験を行った。 24穴培 養ディッシュに HeLa細胞をまき(10000 cells/well)、 16時間培養した後に pGL3、 pGL 3/5HRE- Lucゝまたは pGL3/5HREp- NLS- ODD- Luciferase (0.4 g/well)を Polyfect Transfection Reagent (QIAGEN)にて導入した。この時ゥミシィタケルシフェラーゼを 恒常的に発現するプラスミド pRL/CMV (Promega)もインターナルコントロールとして同 時に導入した(0.04 /z g/well)。遺伝子導入力も 24時間後に培地を交換し、その後さら に 18時間にわたって有酸素、または低酸素条件化 (酸素濃度く 0.02%)にて培養した。 培地を吸引後、 100 mlの Passive Lysis Buffer (Promega)にて細胞抽出液を収集し、 付属の説明書に従ってデュアルルシフェラーゼアツセィを行った。結果は、図 3に示 した。
[0068] プロメガのデュアルアツセィシステムは、図 1, 3の実験のみ使用した。これは、各々 の構築物をコードする遺伝子を一時的に発現させるために、遺伝子の導入効率によ る差を補正するためにゥミシィタケルシフェラーゼをコードする遺伝子を一定量同時 に導入し、両方のルシフェラーゼ(ホタルルシフェラーゼとゥミシィタケルシフェラーゼ )活性を測定して、その比で活性を評価するためである。
[0069] その他の実験では、単にホタルルシフェラーゼ (Firefly Luciferase)の発現量を活性 で測定して評価した。

[0073] 24穴培養ディッシュに HeLa/5HRE- Luc、および HeLa/5HRE- ODD- Luc細胞を播 種(1 X 104 cells/well)し 16時間培養した後、 18時間にわたって低酸素処理を行った。 細胞を低酸素チャンバ一力 取り出した直後にシクロへキサミド含有培地に培地交 換し、 0、 10、 30、 60分間にわたって 5% C02インキュベーターで培養をすることによつ て再酸素処理をした。その後培地を吸引し、 Passive Lysis Buffer (Promega Co.)を 10 0 1/well添加して- 80 °Cにて凍結した後、融解することによって細胞抽出液を収集 した。 20 1の細胞抽出液を用いホタルルシフェラーゼ活性をルシフェラーゼアツセ ィキット (Promega)で調べた。 0分におけるルシフェラーゼ活性に対する、各測定時間 におけるルシフェラーゼ活性の比を算出し、 Relative Luciferase Activityとした。

(5)腫瘍モデルの作成
細胞をトリプシン処理後、冷 PBSで洗浄し、 PBSにより(1 X 106 cells/ 100 μ 1)の濃度 に懸濁した。この細胞懸濁液を 6週齢、雌の BALB/c nu/nuマウスの下腿皮下に移植 した。
[0075] 得られたモデルマウスのイメージングの結果を図 5に示す。
(0ノ in vivo imaging
HeLa/EF- Lucを左下腿部、 HeLa/5HRE- Lucまたは HeLa/5HRE- ODD- Lucを右下 腿部に皮下移植した。移植 10日後に右下腿を結紮し、その直後にイソフルランガスを 充填した麻酔装置 (Xenogen Co)にマウスを入れた。麻酔から 0、 2、 4、 8時間後にル シフェリンを 50 mg/kg ivにて投与し IVIS-100システム(Xenogen Co.)を用いてルシフ エラーゼによる化学発光を検出した。得られた画像を IGOR Pro,V4.0.6.1(Wavemetric s, LakeOswego,OR)を用いて解析した。
[0076] 結果を図 6に示す。
[0077] 右下腿部を結紮してから 18時間後にルシフェリンを 50 mg/kg ivにて投与した。投与
力 12分経過した時点でイソフルランガスを充填した麻酔装置 (Xenogen Co)にマウ スを入れた。その 3分後に結紮を解き、以後 0、 5、 15、 25、 35分の時点で化学発光を 検出した。

[0079] 5HRE- mp- NLS- ODD- Luciferaseのトランスジエニックマウスを用いた in vivo imaging に関しては以下の様に実施した。
[0080] まずトランスジエニックマウスの右下腿を 16時間に渡って結紮した。ルシフェリン投 与後 11分経過した時点からイソフルランガスにて麻酔し、 13分の時点で撮像した。ル シフェリン投与後 15分で結紮を解き、右下腿を再酸素化 (reoxygenation)した。再酸 素化した時点を基準(0 min)として、以降 2、 5、 8、 10、 12、 15分で撮像した。レポータ 一タンパク質の発光は経時的に小さくなり消失した。その後これまでに観察されたィメ ージが出血等による擬陽性のイメージで無いことを確認するために、直ちにルシフエ リンを追加投与し、さらに 17、 20、 23、 26、 30分の時点で撮像した。一度再酸素化され て消えてしまったレポータータンパク質は再度確認することができな力つた。

(7)遺伝子操作動物作製
遺伝子改変動物作製には、雌 B6C3F1 X雄 B6C3F1の自然交配によって得られた 受精卵 B6C3F2を使用した。当該遺伝子断片(pGL3/5HREp- NLS- ODD- Luciferase の Xhol- Sail断片、約 1.7 kbp; 5HRE-CMVmp- NLS- ODD(548- 603)- (Firefly Luciferas e gene))を電気泳動後に 0.6%ァガロースゲルでから回収'精製後にトリス 10mM、 ED TA0.1mM (T E )溶液に 500copy/pl濃度で溶解した。マウス前核期受精卵 1個当た

り 1000コピー相当量をマイクロインジェクション法(参考図書 Methods in Enzymolizy Vol. 225 Guode to Techniques in Mouse Development)で注入し、生存の確認でき た受精卵を偽妊娠 0.5dpcの ICR系統仮腹親の卵管に移植後 19日に、帝王切開で産 仔 (フアウンダートランスジエニック)を得た。得られた産仔の PCR遺伝子解析の結果、 当該遺伝子が陽性であった。該遺伝子陽性の個体を基にライン化して、本発明のト ランスジェニックマウスを榭立した。

ジーンがヘテロの交雑種 Nl雄個体とワイルド(C57BL/6J)の雌を用いて体外受精に より受精卵を作製した。得られた受精卵の融解'移植後に得られる個体は、トランスジ ーン (導入遺伝子)保有と非保有が 1: 1になる。得られたトランスジエニックマウス (HO L-A)の受精卵は、 2005年 3月 8日付で独立行政法人産業技術総合研究所 特許 生物寄託センターに「受託番号 FERM P-20451」で国内寄託され、その後、 2006年 2月 22日付で独立行政法人産業技術総合研究所 特許生物寄託センターに「受領 番号 FERM ABP-10537Jで国際寄託された。

請求の範囲
[I] レポーター遺伝子を低酸素応答性プロモーターの制御下に組み込んでなり、かつ o
DDトメイン (oxygen dependent degradation domain,をレ 一ター遺 1 子とインフレ一 ムで連結してなる遺伝子構築物。
[2] 低酸素応答性プロモーターが、 HIF-1結合ドメイン (HRE: hypoxia responsive element
)を有する請求項 1に記載の遺伝子構築物。
[3] 低酸素応答性プロモーターが、ミニマムプロモータ一 (mp)を有する請求項 1に記載の 遺伝子構築物。
[4] レポーター遺伝子がルシフェラーゼ遺伝子である請求項 1〜3のいずれかに記載の 遺伝子構築物。
[5] 核局在シグナル (NLS)をさらに含む、請求項:!〜 4のいずれかに記載の遺伝子構築 物。
[6] (HIF- 1結合ドメイン) - (mp)― (NLS) - (ODDドメイン) - (レポ一ター遺伝子)の構 造を有する請求項 5に記載の遺伝子構築物。
[7] 請求項 1〜6のいずれかに記載の遺伝子構築物を宿主細胞に導入してなる形質転 換体。
