Abstract

 本発明の目的は、アクチビンの活性は阻害せずに、GDF-8の活性を特異的に阻害することができるフォリスタチンの変異体を提供することである。本発明によれば、(a)フォリスタチンドメインIを含み(b)式(I):Cys−(X1)a−Cys−(X2)b−Cys−(X3)c−Cys−(X4)d−Cys−(X5)e−Cys−(X6)f−Cys−(X7)g−Cys−(X8)h−Cys−(X9)i−Cys(式中、X1、X2、X3、X4、X5、X6、X7、X8およびX9は、それぞれ独立して同一または異なった、システイン以外の天然に存在するアミノ酸残基を表し、aは2~6、bは3~7、cは7~11、dは0~4、eは1~6、fおよびgは8~12、hは4~8、iは11~15の整数をそれぞれ表す。)で表されるアミノ酸配列であって、かつ配列番号2または配列番号29のアミノ酸配列と50%以上の相同性を有する配列を含まず、かつ(c)アクチビンの活性の阻害と比較して、GDF-8の活性を選択的に阻害する、ことを特徴とするフォリスタチン変異体ポリペプチドが提供される。

ァクチビンは、 トランスフォーミング増殖因子 (transforming growth factor- 、 TGF-3) スーパーファミリーに属し、 赤芽球系細胞分化誘導、 下垂 体からの卵胞刺激ホルモン (FSH) 分泌および FSHによる黄体形成の促進、 神経細 胞生存維持作用、 インスリン産生促進作用など多機能性の成長'分化因子であ る。
増殖 Z分化因子一 8 (growth/differentiation factor- 8、 GDF- 8) は、 発生中 および成体の骨格筋に特異的に発現している、 ァクチビンサブフアミリーに属す る TGF-i3スーパーファミリーの一員として発見され (ネイチヤー (Nature) ,
(イギリス) , 1997年, 第 387卷, 第 6628号, p. 83— 90) 、 ミ ォスタチン (myostatin) とも呼ばれる。 GDF- 8は筋体積を負に制御する機能を持 ち、 GDF-8欠損マウスでは筋体積が野生型マウスの 2〜 3倍に増加する (ネイチ ヤー (Nature) , (イギリス) , 1997年, 第 387卷, 第 6628号, ρ· 83— 90) 。

フォリスタチンのアミノ酸配列解析から、 N末端側より N末端ドメイン、 フォ リスタチンドメイン I、 フオリスタチンドメイン II、 フオリスタチンドメイン III という 4つのドメインからなることが明らかとされている (プロシーデイング ズ ·ォブ, ·ザ ·ナショナル 'アカデミー 'ォブ ·サイェンシズ ·ォブ ·ザ ·ュナ ィテッド ·ステーッ ·ォブ ·ァメリ力 (Proceedings of the National Academy of Sciences of the United States of America) , (米国) , 1 988年, 第 8 5卷, 第 1 2号, ρ· 42 1 8— 422 2) 。 フオリスタチンドメインは、 ァ クチビンやその他の増殖因子との結合に関与すると考えられている。
近年、 GDF-8 が様々な疾患に関与することを示唆する報告がなされている。 Zimmers らはヌードマウス大腿部に GDF - 8安定発現細胞を移植すると、 骨格筋お ょぴ脂肪の減少という悪液質の症状が誘導されることを示した (サイエンス (Science) , (米国) , 200 2年, 第 2 9 6卷, 第 5 5 72号, p. 1 48 6- 1488) 。 体重減少を伴う HIV感染者の筋肉中には、 抗 GDF- 8抗体陽性の タンパク質が増加していることも報告されている ( プロシーデイングズ*ォ ブ.ザ.ナショナル ·アカデミー ·ォブ ·サイェンシズ ·ォブ ·ザ ·ュナイテツ ド ·ステーッ ·ォブ,ァメリカ (Proceedings of the National Academy of Sciences of the United States of America) , (米国) , 1 9 98年, 第 9 5 卷, 第 25号, p. 1 4938— 14 943) 。 デュシェンヌ型筋ジストロフィ 一のモデルマウスである mdxマウスと GDF- 8欠損マウスを交配して得られるマウ スは、 mdxマウスと比較して筋量 ·筋機能ともに改善していることなども報告さ れている (アナルズ 'ォブ 'ニューロロジー (Annals of Neurology) , (米 国) , 200 2年, 第 5 2卷, 第 6号, p. 83 2— 8 3 6) 。 また、 GDF- 8欠 損マウスでは、 骨格筋の増加と共に脂肪量の減少が見られることや、 糖尿病モデ ルマウスにおいて GDF-8遺伝子を欠損させることにより、 脂肪蓄積およびダルコ ース代窗す異常が抑制されることから、 肥満や Π型糖尿病の発症や病態悪化への 関与も示唆されている (ザ ·ジャーナル 'ォブ 'クリ二カル ·ィンべスティゲー シヨン (The Journal of Clinical Investigation) , (米国) , 200 2年, 第 1 0 9卷, 第 5号, p. 5 9 5— 60 1) 。
以上から、 GDF - 8の活性を阻害することにより、 これらの GDF- 8が発症または病 態の進行に寄与する疾患の治療を行うことができると考えられ、 しかも、 副作用 軽減のためには、 ァクチビンの活性を阻害せず、 GDF- 8の活性を選択的に阻害す ることが好ましい。 GDF- 8の活性を阻害する物質としては、 フォリスタチン (米 国特許出願公開第 2002/0 1 5 7 1 26号明細書、 及ぴ特表 2002— 50 6044号公報) 、 抗 GDF- 8抗体 (米国特許第 60 9 6 506号明細書) 、 GDF- 8 プロペプチド (国際公開第 02ノ09 64 1号パンフレット) 、 ドミナントネガ ティブ GDF- 8変異体 (国際公開第 0 1 53 3 50号パンフレット、 及ぴ国際公 開第 00/43 78 1号パンフレツト) 、 GDF- 8抗原 (米国特許第 636 920 1号明細書、 国際公開第 0 1/058 20号パンフレット、 及び国際公開第 9 9 /42 5 73号パンフレツト) 、 GDF- 8レセプターおょぴ抗 GDF- 8レセプター抗体 (国際公開第 02Z102 4号パンフレツト) 、 および Smad2および Smad3リン 酸化阻害剤 (国際公開第 02/055077号パンフレット) などが知られてい る。 発明の開示
本発明は、 下垂体 FSH分泌、 黄体成熟、 赤芽球系分化誘導、 インスリン分泌な どの重要な機能を担っているァクチビンの活性は阻害せずに、 悪液質、 筋ジスト ロフィ一、 肥満およぴ II型糖尿病の発症およびあるいは病態悪化に寄与している ことが示唆されている GDF- 8の活性をァクチビンの活性の阻害と比較して選択的 に阻害することができるフォリスタチンの変異体を提供することを解決すべき課 題とした。 '

本 明における変異体を作製する場合の、 元になるフォリスタチンはどの動物 種のものでもよいが、 哺乳類または鳥類のフォリスタチン、 例えばヒ ト、 マウ ス、 ラット、 ゥシ、 ブタ、 ヒッジ、 ゥマ、 ニヮトリ等のフオリスタチンが好まし く、 特にヒトフオリスタチンが好ましい。 また、 mRNAのスプライシングの違いあ るいは翻訳後のプロセシングにより、 C末端のアミノ酸配列が異なる分子がある が、 どれでもよい。 具体的には、 配列番号 4の 30〜344番目のアミノ酸配列から なるヒ トフオリスタチン、 NCBIの蛋白質データベース登録番号 P47931のアミノ酸 配列の 30〜344番目の配列からなるマウスフォリスタチン、 NCBIの蛋白質データ ベース登録番号 NP— 032072のァミノ酸配列の 30〜343番目の配列からなるマウスフ オリスタチン、 NCBIの蛋白質データベース登録番号 P21674のアミノ酸配列の 30〜 344番目の配列からなるラットフオリスタチン、 NCBIの蛋白質データベース登録 番号 P50291のァミノ酸配列の 30〜344番目の配列からなるゥシフオリスタチン、 NCBIの蛋白質データベース登録番号 P10669のァミノ酸配列の 30〜344番目の配列 からなるプタフ.ォリスタチン、 NCBIの蛋白質データベース登録番号 P31514のアミ ノ酸配列の 30〜344番目の配列からなるヒッジフオリスタチン、 NCBIの蛋白質デ ータベース登録番号 062650のァミノ酸配列の 30〜344番目の配列からなるゥマフ オリスタチン、 NCBIの蛋白質データベース登録番号 NP— 990531のアミノ酸配列の 29〜343番目の配列からなるニヮトリフオリスタチン、 これらのフォリスタチン の C末端の 27ァミノ酸または 12ァミノ酸が欠失した配列からなるポリぺプチド、 また、 天然に存在する、 これらのポリペプチドの遺伝子のアレル変異や 1塩基多 型の遺伝子がコードするポリぺプチドがあげられる。
本発明において、 フォリスタチンドメインは、 フォリスタチン内に 3回繰り返 される、 保存された 10個のシスティンを含む、 Cys - (Xaa)4 - Cys - (Xaa)5-Cys - (Xaa) 9— 10— Cys一 (Xaa)ト 2一 Cys— (Xaa) 3一 Cys— (Xaa 9— 10— Cys— (Xaa) 10一 Cys一 (Xaa) 6一し ys一 (Xaa)13- Cysで示される約 70ァミノ酸の配列 (Xaaは任意のァミノ酸配列を表す) からなる領域をいい、 N末端側から、 それぞれ、 ブオリスタチンドメイン I (以 下、 FSIと略す) 、 フオリスタチンドメイン II (以下、 FSIIと略す) およぴフォ リスタチンドメイン III (以下、 FSIIIと略す) という。 例えば、 配列番号 4で表 されるヒトフオリスタチン前駆体のアミノ酸配列 (当該アミノ酸配列をコードす る塩基配列を配列番号 3に示す) において、 95〜164番目の領域が FSI、 168〜239 番目の領域が FSII、 245〜316番目の領域が FSIIIとなる。 また、 N末端ドメイン
(以下、 FSNと略す) は、 フオリスタチンの N末端から FSIまでの領域をいい、 例 えば、 配列番号 4で表されるヒトフオリスタチン前駆体のァミノ酸配列におい て、 30〜94番目の領域をいう。 図 1にヒト、 マウス、 ラット、 ゥシ、 ブタ、 ヒッ ジ、 ゥマ、 ニヮトリの各フオリスタチンのアミノ酸配列と FSN、 FSI、 FSII, FSIIIの各ドメィンの位置、 FSIおよび FSIIのコンセンサス配列を示した。
本発明のフォリスタチン変異体ポリべプチドは、 ァクチビンの活性の阻害と比 較して GDF-8の活性を選択的に阻害するフォリスタチン変異体ポリべプチドであ り、 FSIを含み、 式 ( I ) : C y s— (X1) a— C y s— (X2) ¾_C y s _
(X3) Cy s - (X4) d— Cy s— (X5) e - Cy s— (X6) f-Cy s - (X7) g-Cy s - (X8) h-Cy s - (X9) ,-C y s (式中、 X1、 X 2、 X3、 X4、 X5、 X6、 X7、 X8および X9は、 それぞれ独立して同一または 異なった、 システィン以外の天然に存在するアミノ酸残基を表し、 aは 2〜6、 bは 3〜7、 cは 7〜: L l、 dは 0〜4、 eは.1〜6、 f および gは 8〜1 2、 hは 4〜8、 iは 1 1〜1 5の整数をそれぞれ表す。 ) で表されるァミノ酸配列 であって、 かつ配列番号 2または配列番号 2 9のアミノ酸配列と 50%以上、 好ま しくは 60%以上、 より好ましくは 80°/0以上、 さらに好ましくは 95%以上の相同性 を有する配列 (以下、 FSII相同配列とよぶ) を含まない配列であることを特徴と する。 ここで、 「システィン以外の天然に存在するアミノ酸残基」 とは、 ァスパ ラギン、 ァスパラギン酸、 ァラニン、 ァノレギニン、 イソロイシン、 グノレタミン、 グノレタミン酸、 グリシン、 セリン、 スレオニン、 チロシン、 トリプトファン、 ヒ スチジン、 パリン、 フエニノレアラニン、 プロリン、 メチォニン、 リジンおよび口 イシンのいずれかのアミノ酸残基を意味する。 配列番号 2のアミノ酸配列は、 各 哺乳類およぴニヮトリのフォリスタチンの FSIIのコンセンサスなアミノ酸配列で ある。 配列番号 2 9のアミノ酸配列は、 マウスのフォリスタチン関連遺伝子 (follistatin related gene FLRG) がコードする FLRGタンパク質の、 2つある フォリスタチンドメイン様の配列の 2番目のフォリスタチンドメイン様の配列で あり、 配列番号 2の配列と 61%の相同性を有する。 FSII相同配列には配列番号 2 のァミノ酸配列、 配列番号 2 9のァミノ酸配列も含まれる。 FSII相同配列として は、 例えば、 上記式 (I ) で表されるアミノ酸配列において、 (X 1 ) aが配列 番号 2の 2〜5位のアミノ酸配列または配列番号 2 9の 2〜5位のアミノ酸配 列、 (X 2) bが配列番号 2の 7〜1 1位のアミノ酸配列または配列番号 2 9の 7〜 1 1位のアミノ酸配列、 (X 3) cが配列番号 2の 1 3〜2 1位のアミノ酸 配列または配列番号 2 9の 1 3〜 2 1位のァミノ酸配列、 (X 4) dが配列番号 2の 2 3 ~ 2 4位のァミノ酸配列または配列番号 2 9の 2 3〜 2 4位のァミノ酸 配列、 (X 5) eが配列番号 2の 2 6〜 2 8位のァミノ酸配列または配列番号 2 9の 2 6〜2 9位のアミノ酸配列、 (X 6 ) fが配列番号 2の 3 0〜3 9位のァ ミノ酸配列または配列番号 2 9の 3 1〜 4 0位のァミノ酸配列、 (X 7 ) gが配 列番号 2の 4 1〜5 0位のアミノ酸配列または配列番号 2 9の 4 2〜5 1位のァ ミノ酸配列、 (X 8) hが配列番号 2の 5 2〜 5 7位のァミノ酸配列または配列 番号 2 9の 5 3〜5 8位のアミノ酸配列、 (X 9) iが配列番号 2の 5 9〜7 1 位のァミノ酸配列または配列番号 2 9の 6 0〜 7 2位のァミノ酸配列である配列 があげられる。 FSII相同配列を含まない配歹 Uであるフォリスタチン変異体としては、 例えば、 FSII全体を欠失させた変異体、 FSIIの 1つ以上のシスティン残基を含む一部の領 域を欠失させた変異体、 FSIIの 1つ以上のシスティン残基を欠失または他のァミ ノ酸に置換した変異体、 FSIIの内部に、 FSII相同配列を含まない 10アミノ酸以上 のアミノ酸配列を挿入した変異体などがあげられる。 FSIを含み、 FSII相同配列 を含まない配列であれば、 FSIおよび FSII以外の領域のアミノ酸配列について は、 アミノ酸の欠失、 付加、 置換の位置、 数は限定されない。 アミノ酸の変異 (即ち、 アミノ酸の欠失、 付加または置換) の数は好ましくは 1〜5 0個であ り、 より好ましくは 1〜3 0個であり、 さらに好ましくは 1〜2 0個であり、 さ らに好ましくは 1〜1 0個であり、 特に好ましくは 1〜5個である。 また、 フォ リスタチン変異体ポリペプチドが、 ァクチビンの活性の阻害と比較して、 GDF-8 の活性を選択的に阻害することができる限りは、 FSIのァミノ酸配列において も、 アミノ酸の欠失、 付加または置換が存在してもよい。 このような FSIのアミ ノ酸配列におけるアミノ酸の変異の数は好ましくは 1〜1 0個であり、 より好ま しくは 1〜8個であり、 さらに好ましくは 1〜5個である。 本発明のフォリスタ チン変異体ポリペプチドとしては、 さらに FSIIIも欠失していることが好まし い。 また、 FSIの数は 1つでもよいが、 2つ以上含むことが好ましい。 また、 FSN を含んでもよい。
本発明のフォリスタチン変異体ポリペプチドとしては、 FSI、 FSI -FSI, FSI— FSI— FSIII、 FSN— FSI、 FSN—FSI _FSI、 FSN— FSI—FSI—FSIII等のドメイン構 造のポリぺプチド、 これらのポリぺプチドの N末端または C末端に他のポリぺプ チドを融合させたポリべプチドがあげられる。 各ドメイン間は直接または 1〜50 個、 好ましくは 1〜10個、 より好ましくは 1〜5個のアミノ酸からなるリンカ一 ポリペプチドでつながれる。 リンカ一ポリペプチドのアミノ酸配列は、 システィ ンを含まなければいかなる配列でもよい。 N末端または C末端に融合.させるポリ ペプチドの配列は、 上記の FSII相同配列を含まなければ、 いかなる配列でもよ く、 1アミノ酸でもよい。 N末端または C末端に融合させるポリペプチドと'して は、 ィムノグロブリンの定常領域、 プロテイン A、 プロテイン G、 グリーン 'フ ノレ才レツセント 'プロテイン、 β -ガラク トシダーゼ、 クロラムフエニコーノレ ' ァセチルトランスフェラーゼ、 マルトース結合蛋白質、 グルタチオン S -トラン スフエラーゼ、 チォレドキシン、 ポリヒスチジン、 Sペプチド、 FLAGペプチド、 HAェピトープぺプチド、 mycェピトープぺプチドなどをあげることができる。
FSIの配列としては、 配列番号 4の 95〜164番目のアミノ酸配列、 および各哺乳 類およびニヮトリのフォリスタチンの FSIのコンセンサス配列である配列番号 1 のアミノ酸配列をあげることができる。 FSIIの配列としては、 配列番号 4の 168 〜239番目のアミノ酸配列、 および各哺乳類およびニヮトリのフォリスタチンの FSIIのコンセンサス配列である配列番号 2のァミノ酸配列をあげることができ る。 FSNの配列としては、 例えば配列番号 4の 1〜94番目のアミノ酸配列をあげ ることができる。 FSIIIの配列としては、 例えば配列番号 4の 245〜316番目のァ ミノ酸配列をあげることができる。
本発明のフオリスタチン変異体ポリぺプチドの具体例としては、 配列番号 6、 8または 1 0のアミノ酸配列からなるポリペプチドをあげることができる。
本発明のフォリスタチン変異体ポリぺプチドは、 修飾されたポリぺプチドでも よい。 本発明で言う 「修飾」 という用語は、 化学的修飾及び生物学的修飾を含め て最も広義に解釈しなければならない。 修飾の例としては、 例えば、 アルキル化、 エステル化、 ハロゲン化、 又はアミノ化などの官能基導入、 酸化、 還元、 付加、 又は脱離などによる官能基変換、 糖化合物 (単糖、 二糖、 オリゴ糖、 若しくは多 糖) 又は脂質化合物などの導入、 リン酸化、 ビォチン化、 又はポリエチレンダリ コールでの修飾などを挙げることができるが、 これらに限定されることはない。 上記した本発明のポリぺプチドは遊離形態であってもよいが、 酸付加塩又は塩 基付加塩として提供されてもよい。 酸付加塩としては、 例えば、 塩酸塩、 硫酸塩、 硝酸塩、 リン酸塩などの鉱酸塩;又は、 p-トルエンスルホン酸塩、 メタンスルホ ン酸塩、 クェン酸塩、 蓚酸塩、 マレイン酸塩、 酒石酸塩などの有機酸塩を挙げる ことができる。 塩基付加塩としては、 例えば、 ナトリウム塩、 カリウム塩、 カル シゥム塩、 マグネシウム塩などの金属塩;アンモニゥム塩;メチルアンモニゥム 塩、 トリェチルアンモニゥム塩などの有機アンモユウム塩を挙げることができる。 グリシンなどのアミノ酸と塩を形成する場合もあり、 また、 分子内で対イオンを 形成することもある。
さらに、 これらのポリペプチド又はその塩は、 水和物又は溶媒和物として存在 する場合がある。 本発明のポリペプチドは複数の不斉炭素を有しているが、 各不 斉炭素の立体は特に限定されないが、 アミノ酸残基は L -アミノ酸であることが 望ましい。 これらの不斉炭素に基づく光学活性体、 ジァステレオ異性体などの立 体異性体、 立体異性体の任意の混合物、 ラセミ体などはいずれも本発明の範囲に 包含される。
以下に、 本発明のフォリスタチン変異体の作製方法、 活性評価方法おょぴその 使用方法について説明する。
2 . フオリスタチン変異体ポリペプチドの製造方法
( 1 ) フォリスタチン変異体ポリぺプチドをコ一ドする DNAの構築
1 . に記載した条件に基づき、 ァクチビンの活性の阻害と比較して、 GDF - 8を 選択的に阻害するフォリスタチン変異体ポリべプチドの構造を設計し、 該ポリ.ぺ プチドをコ一ドする DNAを以下のようにして構築する。
まず、 フオリスタチン全体または設計したブオリスタチン変異体ポリペプチド の構成に必要なフォリスタチンの領域を含む部分断片をコードする DNAを取得す る。 各種哺乳類およぴニヮトリのフォリスタチン cDNAまたはゲノム DNAの塩基配 列は公知であるので、 これらの配列の情報をもとにしたプライマーを用い、 各種 哺乳類またはニヮトリの精巣から単離した cDNAを铸型とした PCRを行うことによ り、 該哺乳類またはニヮトリのフオリスタチン cDNAの任意の部分断片を取得でき る。 また、 精巣 cDNAライブラリーのスクリーニングによりブオリスタチン cDNAを クローニングし、 該 cDNAを铸型としてもよい。 各種哺乳類およぴニヮトリのフォ リスタチン cDNAまたはゲノム DNAの塩基配列としては、 NCBIのヌクレオチドデー タベース登録番号厘— 013409 (ヒ トフオリスタチン cDNA) 、 雇— 008046 (マウスフ オリスタチン cDNA) 、 丽— 012561 (ラットフオリスタチン cDNA) 、 丽— 175801 (ゥ シフオリスタチン cDNA) 、 M63123 (ヒッジフオリスタチン cDNA) 、 AB010829 (ゥ シフオリスタチン cDNA) 、 M19529 (ブタフオリスタチンゲノム DNA) 、 丽_205200
(ニヮトリフオリスタチン cDNA) 等に記載された塩基配列をあげることができ る。 取得したい領域の 5'端の 15〜30塩基の配列、 および 3'端の 15〜30塩基の配列 と相補的な配列を含む DNAを合成し、 プライマーとする。 PCRの両プライマーの 5, 端には、 部分断片の連結やベクターへの挿入に用いるため、 適当な制限酵素の認 識配列を付力 Πしておくのが好ましい。 cDNAの調製や PCRは、 Sambrook, J. and Russel, D. W. , Molecular Cloning: A Laboratory Manual, 3rd ed. , Cold Spring Harbor Laboratory Press (2001)等の実験書の記載にしたがって、 行う ことができる。 必要に応じて、 得られた DNAを鎵型として同様の操作を行い、 設 計したフォリスタチン変異体ポリべプチドの構成に必要な領域をコードする該 DNAの部分断片を取得する。
設計したフォリスタチン変異体ポリぺプチドの構造に基づいて、 得られた部分 断片をフレームが合うように連結させ、 必要に応じて、 他のポリペプチドをコー ドする DNAと連結させたり、 終止コドンをコード領域の 3'端に付加したり、 変異 を導入することにより、 該フォリスタチン変異体ポリペプチドをコードする DNA を構築することができる。
( 3 ) で記述するように、 形質転換細胞の培養時に、 ブオリスタチン変異体ポ リぺプチドを細胞外に分泌させる場合は、 分泌蛋白質の前駆体の N末端に存在す るシグナルぺプチドを、 設計したフォリスタチン変異体ポリべプチドの N末端に 付加したポリべプチドをコ一ドする DNAを構築する。 シグナルぺプチドとして は、 例えば、 フオリスタチンめシグナルペプチドを用いることができる。 フオリ スタチンのシグナルぺプチドをコ一ドする DNAは、 上記のブオリスタチン変異体 ポリべプチドの構成に必要な FSIを含むフォリスタチン断片をコードする DNAを取 得する際に、 フォリスタチンのシグナルべプチドを含むような断片を増幅するこ とにより、 取得することができる。
分泌させない場合は、 設計したフォリスタチン変異体ポリべプチドの N末端が メチォニンでない場合には、 フォリスタチン変異体ポリべプチドをコ一ドする領 域の前に開始コドン (atg) を付加する。
例えば、 FSN- FSI - FSIというドメイン構造を有するフォリスタチン変異体ポリ ペプチドを分泌させる場合は、 まず PCRで、 シグナルペプチド- FSN- FSIを含む領 域をコードするフォリスタチン cDNAの部分断片 Aおよび FSIを含む領域をコード するフォリスタチン cDNAの部分断片 Bをそれぞれ取得する。 得られた cDNA断片 A の下流に cDNA断片 Bを連結させることにより、 シグナルぺプチド- FSN- FSI- FSIと いうドメィン構造を有するフォリスタチン変異体ポリペプチドをコードする DNA を作製することができる。
また、 FSN-FSI- FSI - FSIIIというドメイン構造を有するフォリスタチン変異体 ポリペプチドを分泌させる場合は、 PCRで、 FSIIIを含む領域をコードするフオリ スタチン cDNAの部分断片 Cを取得し、 上記の cDNA断片 Aの下流に cDNA断片 Bを連 結させ、 その下流にさらに cDNA断片 Cを連結させることにより、 シグナルぺプチ ド- FSN - FSI- FSI - FSIIIというドメイン構造を有するフォリスタチン変異体ポリぺ プチドをコ一ドする DNAを作製することができる。
( 2 ) フォリスタチン ¾異体発現べクターの作製と宿主細胞への導入
( 1 ) で作製したフォリスタチン変異体ポリペプチドをコードする DNAを宿主 . 細胞に応じた適当な発現用ベクターに挿入して、 ブオリスタチン変異体発現べク ターを作製する。
宿主細胞としては、 細菌等の原核生物、 酵母等の真核微生物、 動物細胞、 昆虫 細胞、 植物細胞等、 フオリスタチン変異体ポリペプチドを発現できるものであれ ばいずれも用いることができるが、 フオリスタチン変異体発現べクタ一を導入し た細胞 (以下、 形質転換細胞ともいう) が発現するフオリスタチン変異体ポリべ プチドが正しい高次構造を保ち、 GDF-8特異的な阻害活性を保持するためには、 動 物細胞や昆虫細胞などで発現させるのが好ましい。 フオリスタチン変異体ポリペプチドをコードする DNAを挿入する発現用べクタ一 としては、 上記宿主細胞において自立複製が可能、 または染色体中への組込みが 可能で、 宿圭細胞中で機能するプロモーターを含有しているものを用いることが できる。 また、 大腸菌 (Escherichia coli) を用いたベクターの調製のために、 大腸菌用の内在性プロモーターを含む薬剤耐性遺伝子、 大腸菌中での複製に必要 な複製開始点を含むことが好ましい。 大腸菌用の薬剤耐性遺伝子としては、 大腸 菌由来のアンピシリン耐性遺伝子 (i3 -ラクタマーゼ遺伝子) 、 テトラサイクリン 耐性遺伝子、 カナマイシン耐性遺伝子等があげられる。 大腸菌での複製に必要な 複製開始点としては、 PBR322の複製開始点、 colElの複製開始点等があげられる。 宿主細胞が、 動物細胞の場合、 フオリスタチン変異体ポリペプチドをコードす る DNAを挿入する発現用ベクターとしては、 動物細胞中で機能するプロモーターお よびポリアデニルイ匕シグナルを有するプラスミ ドがあげられる。
プロモーターとしては、 動物細胞中で機能するものであればいずれも用いるこ とができ、 例えば、 ヒ トサイトメガロウィルス (CMV) の IE (immediate early) 遺伝子のプロモーター、 SV40の初期プロモーター、 モロニ一マウス白血病ウィル スのロング 'ターミナル . リピート (LTR) 、 ラウス肉腫ウィルスの LTR、 単純へ ルぺスウィルス (HSV) のチミジンキナーゼ (TK) 遺伝子のプロモーター、 レトロ ウイ レスのプロモーター、 ヒートショックプロモーター、 SR CKプロモーター、 メ タロチォネインのプロモーター等をあげることができる。 上記プロモーターと共 に、 ヒ ト CMVの IE遺伝子のェンハンサ一等も用いることができる。 プロモーターは、 その下流にフォリスタチン変異体ポリべプチドをコ一ドする DNAを揷入できる、 適 当な制限酵素サイトを有することが好ましい。
ポリアデニル化シグナルとしては、 いかなるポリアデュル化シグナルも利用で きるが、 例えば、 SV40初期遺伝子のポリアデュル化シダナル、 ゥサギ グロビン 遺伝子のポリアデニル化シグナル、 ゥシ成長ホルモン遺伝子のポリアデ二ル化シ グナル等を用いることができる。
さらに動物細胞用の薬剤耐性遺伝子の発現ュニット (プロモーターの下流に薬 剤耐性遺伝子とポリアデ二ルイ匕シグナルを順に連結した構造) を有していてもよ い。 動物細胞用の薬剤耐性遺伝子としては、 ネオマイシン耐性遺伝子、 ハイグロ マイシン耐性遺伝子、 プラストサイジン耐性遺伝子、 ピューロマイシン耐性遺伝 子、 ゼォシン耐性遣伝子等があげられる。 プロモーターとポリアデュル化シグナ ルは上記のものを用いることができる。 薬剤耐性遺伝子の発現ュ-ットを有する 発現ベクターを用いた場合、 薬剤存在下で形質転換細胞を培養することにより、 一過的でなく安定発現する形質転換細胞を得ることができる。
ジヒドロ葉酸レダクターゼ (dhfr) 遺伝子の発現ュニットを含む発現用べクタ 一を用い、 dhfr遺伝子を欠失している動物細胞を宿主細胞とし、 メトトレキセー ト存在下で形質転換細胞を培養し、 メ トトレキセートの濃度を段階的に上昇させ ることにより、 dhfr遺伝子と共に導入したフォリスタチン変異体ポリぺプチドを コ一ドする DNAのコピー数'を増幅させ、 フオリスタチン変異体ポリぺプチドの発現 量の高い形質転換細胞を得ることができる。
また、 動物細胞での複製に必要な複製開始点を有していてもよい。 複製開始点 としては、 SV40の複製開始点やェプスタイン ·パー · ウィルスの複製開始点 oriP を用いることができる。 SV40の複製開始点を有するプラスミドベクターは、 SV40 のラージ T抗原を発現する宿主細胞中ではコピー数が增加するため、 目的の遺伝子 を髙発現させる上で好適である。 oriPを含むベクターは、 ェプスタイン 'バー ' ウィルスの EBNA—1 (Epstein— Barr virus nuclear antigen— 1) を発現する宿主中 では宿主の染色体に組み込まれず、 染色体外に維持される。
以上のような、 動物細胞の発現用ベクターとしては、 pcDNA3 〔インビトロジェ ン (Invitrogen). 社製〕 、 pcDNA3. 1 (+) (インビトロジェン社製) 、 pSI 〔キアゲ ン (QIAGEN) 社製〕 、 pCI- neo (キアゲン社製) 、 PAGE107、 PAGE103、 pAMo
[J. Biol. Chem. , 268, 22782 (1993)〕 等をあげることができる。
( 1 ) で作製したフォリスタチン変異体ポリペプチドをコードする DNAを、 上記 の動物細胞の発現用ベクターのプロモーターの下流に挿入して、 フォリスタチン 変異体ポリぺプチド発現べクターを作製する。 宿主細胞としては、 サル腎臓細胞株 COS - 1 (ATCC番号: CRL - 1650) および COS - 7 (ATCC番号: CRL- 1651) 、 チャイニーズノヽムスタ卵巣細胞株 CH0 - Kl (ATCC番号: CCL-61) および CHO/dhFr- (ATCC番号: CRL - 9096) 、 ヒ ト Bリンパ球細胞株
Namalwa (ATCC番号: CRL - 1432) 、 ヒ ト腎臓細胞株 HEK293 (ATCC番号: CRL- 1573) 、 ヒ ト子宮頸部上皮細胞株 HeLa (ATCC番号: CCL- 2) 、 マウス胎児細胞株 NIH/3T3 (ATCC番号: CRL - 1658) 等をあげることができる。
糸且換えベクターの導入方法としては、 動物細胞に DNAを導入する方法であればい ずれも用いることができ、 例えば、 エレクト口ポレーシヨン法 〔Cytotechnology, 3, 133 (1990)〕 、 リン酸カルシウム法 (特開平 2- 227075) 、 リボフヱクシヨン法
[Proc. Natl. Acad. Sci. USA, 84, 7413 (1987)〕 、 Virology, 52, 456 (1973) に記載の方法等をあげることができる。 また、 市販の DNA導入試薬を用いることも できる。 該 DNA導入試薬として、 トランスファスト (TransFast;プロメガ社製) 、 リボフエタトァミン 2000 (Lipofectamine 2000;インビトロジェン社製) 、 スー パーフエクト (SuperFect;キアゲン社製) 、 ポリフエタト (PolyFect;キアゲン 社製) 等をあげることができる。
昆虫細胞を宿主として用いる場合には、 例えば、 Baculovirus Expression Vectors, A Laboratory Manual, W. H. Freeman and Company, New York (1992;、 Molecular Cloning : A Laboratory Manual, 3rd ed. , Cold Spring Harbor
Laboratory Press (2001)、 Bio/Technology, 6, 47 (1988)等に記载された方法に よって、 フォリスタチン変異体ポリぺプチドを発現する昆虫細胞を得ることがで さる。
すなわち、 組換え遺伝子導入ベクターおょぴバキュロウィルスを昆虫細胞に共 導入して昆虫細胞培養上清中に組換えウィルスを得た後、 さらに組換えウィルス を昆虫細胞に感染させ、 ポリぺプチドを発現する昆虫細胞を得ることができる。 該方法において用いられる遺伝子導入べクタ—としては、 例えば、 pVL1392およ ぴ PVL1393 (ともにファーミンジェン社製) 、 pBlueBac4. 5 (インビトロジェン社 製) 等をあげることができる。 パキュロウィルスとしてば、 例えば、 ャガ科昆虫に感染するウィルスであるァ ゥトグラファ ·力リフォ /レニ力 ·ヌクレア一 ·ポリへドロシス · ウイ/レス
(Autographs californica nuclear po丄 yhedrosis virusノ 等を用いること力5でき る。
昆虫細胞としては、 Spodoptera frugiperda由来の Sf9および Sf21 (インビト口 ジェン社製) 、 Trichoplusia ni由来の High 5 (インビトロジェン社製) 、 カイコ (Bombyx mori) 由来の Bm5 (インビトロジェン社製) および N4等の細胞株をあげ ることができる。
組換えウィルスを調製するための、 トランスファーベクターと上記バキュロウ ィルスの共導入方法としては、 例えば、 リン酸カルシウム法 (特開平 2- 227075) 、 リポフエクシヨン法 〔Proc. Natl. Acad. Sci. USA, 84, 7413 (1987) ] 等をあげ ることができる。
細菌等の原核生物を宿主細胞として用いる場合は、 発現ベクターは宿主原核生 物中で自律複製可能であり、 プロモーターおよびその下流にリポソーム結合配列 およぴフォリスタチン変異体ポリべプチドをコ一ドする DNAを揷入するクロー二 ングサイトを有するものを用いる。 必ずしも必要ではないが、 該クローニングサ- イトの直後に転写終結配列を配置する方が好ましい。 また、 形質転換体の選択の ため、 薬剤耐性遣伝子等のマーカーとなる遺伝子を発現する配列を含むようにす る。 リボソーム結合配列の下流のクローユングサイトにフオリスタチン変異体ポ リペプチドをコードする DNAを揷入する。 リポソーム結合配列と開始コドンとの 間は適当な距離 (例えば、 大腸菌宿主のベクターの場合 6〜18塩基) に調節され ていることが好ましい。
プロモーターとしては、 宿主細胞中で発現できるものであればいかなるもので もよい。 例えば大腸菌を宿主とした場合は、 rpプロモーター、 lacプロモータ 一、 PLプロモーター、 T7プロモーター、 PRプロモーター等の、 大腸菌やファージ 等に由来するプロモーター等をあげることができる。 また trpプロモーターを 2 つ直列させたプロモーター、 tacプロモーター、 T71acプロモーター、 let Iプロ モーターのように人為的に設計改変されたプロモーター等も用いることができ る。 枯草菌を宿主とした場合は、 枯草菌のファージである SP01や SP02のプロモー ター、 PenPプロモーター等をあげることができる。
発現ベクターとしては、 例えば、 pGEMEX- 1 (プロメガ社製) 、 pQE-30 (キアゲ ン社製) 、 pKYP200 [Agric. Biol. Chem. , 48, 669 (1984) ] 、 pLSAl [Agric. Biol. Chem. , 53, 277 (1989) ] 、 pGELl [Proc. Natl. Acad. Sci., USA, 82, 4306 (1985)〕 、 PTrS30 〔大腸菌 J1109/PTrS30 (FERM BP-5407)より調製〕 、 pGEX- 5X-3 (アマシャム .バイオサイェンシズ社製) 、 pET14 (ノバジヱン社製) 、 pPROTet. E (クロンテック社製) pRSET A (インビトロジヱン社製) 等を例示する ことができる。

糸且換えベクターの導入方法としては、 上記宿主細胞へ DNAを導入する方法であ ればいずれも用いることができ、 例えば、 エレク ト口ポレーシヨン法 [Nucleic Acids Res. , 16, 6127 (1988)〕 、 カルシウムイオンを用いる方法 〔Proc. Natl. Acad. Sci. USA, 69, 2110 (1972); Gene, 17, 107 (1982)〕 、 プロ トプラスト 法 〔特開昭 63- 248394 ; Mol. Gen. Genet. , 168, 111 (1979)〕 等をあげることが できる。
酵母を宿主細胞として用いる場合の発現ベクターとしては、 宿主酵母で転写を 行なうプロモーター、 転写の終止配列おょぴ酵母での形質転換マーカーとなる遣 伝子、 たとえば薬剤耐性遺伝子や TRP1、 HIS3、 LEU2等のアミノ酸合成系の遺伝子 を発現できる配列を含有しているものが用いられる。 また、 発現ベクターの作製 や維持を容易にするため、 大腸菌内でも自律複製と遺伝子導入マーカーとなる薬 剤耐性遺伝子を発現できるものが好ましい。
プロモーターとしては、 酵母中で転写を行なえるものであればいずれのものを 用レヽてもよく、 例えば Saccharomyces cerevisiaeのァノレコーノレテヒ ドロゲナーゼ 遺伝子 ADH1、 ガラクトース代謝系遺伝子 GAL1や GAL10等のプロモーター、 酸性フ ォスファターゼ遺伝子 PH05プロモーター、 フォスフォグリセレートキナーゼ遺伝 子 PGKプロモーター、 グリセルアルデヒド 3リン酸デヒドロゲ^ "一ゼ遺伝子 GAPプ 口モーター、 ヒートショック蛋白質遺伝子プロモーター、 α接合因子遺伝子 MF a 1プロモーター、 銅メタ口チォネイン遺伝子 CUP1プロモーター、 Pichia pastoris のアルコールォキシダーゼ遺伝子 A0X1のプロモーター等が用いられる。
宿主細胞としては、 サッカロマイセス属、 シゾサッカロマイセス属、 ピヒア属 等に属する酵母菌株をあげることができ、 具体的には、 Saccharomyces cerevisiae^ Schizosaccharomvces pombe、 Pichia nastoris等 ¾■あ ^ こと X)で きる。
組換えベクターの導入方法としては、 酵母に DNAを導入する方法であればいず れも用いることができ、 例えば、 エレク ト口ポレーシヨン法 [ Methods Enzymol. , 194, 182 (1991)〕 、 スフエロプラスト法 〔Proc. Natl. Acad. Sci. USA, 81, 4889 (1984)〕 、 酢酸リチウム.法 〔 J. Bacteriol., 153, 163 (1983)〕 等をあげることができる。
( 3 ) 形質転換細胞の培養とブオリスタチン変異体ポリぺプチドの精製 ( 2 ) で作製した形質転換細胞が大腸菌等の原核生物あるいは酵母等の真核生 物である場合、 該細胞を培養する培地はは、 該細胞が資化し得る炭素源、 窒素源、 無機塩類等を含有し、 形質転換体の培養を効率的に行える培地であれば天然培地、 合成培地のいずれを用いてもよい。 炭素源としては、 該生物が資化し得るもので あればよく、 グルコース、 フラクトース、 スクロース、 これらを含有する糖蜜、 デンプンあるいはデンプン加水分解物等の炭水化物、 酢酸、 プロピオン酸等の有 機酸、 エタノール、 プロパノールなどのアルコール類等を用いることができる。 窒素源としては、 アンモニア、 塩化アンモニゥム、 硫酸アンモ-ゥム、 酢酸アン モニゥム、 リン酸アンモニゥム等の無機酸もしくは有機酸のアンモニゥム塩、 そ の他の含窒素化合物、 並びに、 ペプトン、 肉エキス、 酵母エキス、 コーンスチー プリカ一、 カゼイン加水分解物、 大豆粕および大豆粕加水分解物、 各種発酵菌体、 およびその消化物等を用いることができる。 無機塩としては、 リン酸水素二カリ ゥム、 リン酸二水素カリウム、 リン酸マグネシウム、 硫酸マグネシウム、 塩化ナ トリウム、 硫酸第一鉄、 硫酸マンガン、 硫酸銅、 炭酸カルシウム等を用いること ができる。
培養は、 通常振盪培養または深部通気攪拌培養などの好気的条件下で行う。 培 養温度は 1 5〜 4 0 °Cがよく、 培養期間は、 通常 1 6〜 9 6時間である。 培養中 p Hは 3 . 0〜9 . 0に保持する。 p Hの調整は、 無機または有機の酸、 アル力 リ溶液、,尿素、 炭酸カルシウム、 アンモニアなどを用いて行う。 必要に応じて、 培養期間中にアンピシリンゃテトラサイタリン等の抗生物質を培地に添加しても よい。 プロモーターとして誘導性のプロモーターを用いた発現ベクターで形質転 換した微生物を培養するときには、 必要に応じてィンデューサーを培地に添加し てもよい。 例えば、 1 a cプロモーターを用いた発現ベクターで形質転換した微 生物を培養するときにはイソプロピルチオガラクトシド (I P T G) 等を、 t r― 且プロモーターを用いた発現べクターで形質転換した微生物を培養するときには インドールアクリル酸等を培地に添加してもよい。
( 2 ) で作製した形質転換細胞が動物細胞である場合、 該細胞を培養する培地 は、 一般に使用されている RPill 1640培地 〔J. Am. Med. Assoc. , 199, 519 (1967)〕 、 イーグル (Eagle) の MEM培地 [Science, 122, 501 (1952) ] 、 ダルべ ッコ改変イーグル培地 [Virology, 8, 396 (1959)〕 、 199培地 〔Proc. Soc. Exp. Biol. Med. , 73, 1 (1950)〕 、 MEM培地、 EX- CELL 301培地 (JRHバイオサイェンシ ズ社製) またはこれら培地にゥシ胎児血清等を添カ卩した培地等を用いることがで きる。 培養は、 通常 pH 6〜 8、 30〜40°C、 5 %C02存在下等の条件下で 1〜 7日間 行う。 また培養中必要に応じて、 カナマイシン、 ペニシリン等の抗生物質を培地 に添加してもよい。
( 2 ) で作製した形質転換細胞が昆虫細胞である場合、 該細胞を培養する培地 としては、 一般に使用されている T M- FH培地 (ファーミンジェン社製) 、 Sf-900 II SFM培地 (インビトロジェン社製) 、 EX-CELL 400、 EX-CELL 405 (JRHパイォサ イエンシズ社製) 、 グレースの昆虫培地 [Nature, 195, 788 (1962)〕 等を用いる ことができる。 培養条件は pH 6〜7、 培養温度は 25〜30°C、 培養時間は通常 1〜 5日間が好ましい。 また、 培養中必要に応じて、 ゲンタマイシン等の抗生物質を 培地に添加してもよい。
フォリスタチン変異体ポリペプチドは、 細胞外に分泌させても、 分泌させずに 細胞内に発現させてもよいが、 細胞外に分泌させる方が好ましい。 細胞外へ分泌 させる場合は、 (2 ) でフオリスタチン変異体ポリペプチドをコードする DNAとし て、 設計.したフォリスタチン変異体ポリべプチドの N末端にシグナルぺプチドを 付加したポリペプチドをコードする DNAを用いて発現ベクターを作製し、 形質転換 細胞を取得する。 分泌時には、 シグナルペプチドは切断されるため、 培地中には シグナルぺプチドが除かれたフォリスタチン変異体ポリべプチドが蓄積する。 以上のように培養することにより、 形質転換細胞を培養した培地または形質転 換細胞中に、 発現したフォリスタチン変異体ポリペプチドが蓄積する。 該培地ま たは該形質転換細胞から、 以下のようにして、 フオリスタチン変異体ポリぺプチ ドを単離 ·精製することができる。
細胞外にフォリスタチン変異体ポリべプチドが分泌される場合には、 培地を回 収する。 形質転換細胞が浮遊性の場合は、 遠心分離やろ過により、 培地と細胞を 分離して培地を回収する。 回収した培地から、 硫酸化セル口ファインカラムを用 いたアブイ-ティークロマトグラフィーにより、 効率的に精製することができる。 また、 その他に通常のタンパク質の精製に用いられる精製方法、 例えば、 ゲル濾 過、 イオン交換クロマトグラフィー、 逆相カラムクロマトグラフィーおよび限外 濾過等を組み合わせて行い、 精製することができる。 また、 フオリスタチンの FSI を特異的に認識する抗体を用いたァフイエティークロマトグラフィーによっても 精製することができる。
また、 フオリスタチン変異体ポリペプチドが、 他のポリペプチドとの融合蛋白 質である場合、 融合させた他のポリペプチドと特異的に結合する物質を用いたァ フィニティークロマトグラフィーを利用して精製することもできる。 例えば、 IgG の定常領域との融合蛋白質にした場合はプロテイン Aゃ抗 IgG抗体を固定ィ匕した力 ラム、 プロテイン Aとの融合蛋白質にした場合は IgGを固定ィ匕したカラム、 FLAGぺ プチド、 Sぺプチド、 HAェピトープぺプチド、 mycェピトープぺプチドとの融合蛋 白質にした場合は、 それらのペプチドに対する抗体を固定化したカラム、 ポリヒ スチジンとの融合蛋白質にした場合は、 ニッケルイオンを固定化したカラム、 グ ルタチオン一 S—トランスフェラーゼとの融合蛋白質にした場合は、 グルタチォ ンを固定化したカラムをそれぞれ利用したァフィ二ティークロマトグラフィーに より精製することができる。
フォリスタチン変異体ポリべプチドが細胞内に発現した場合は、 形質転換細胞 を回収する。 回収した細胞をプロテアーゼ阻害剤を含む適当な緩衝液中でホモジ ェナイザーや超音波処理等により破碎した後、 遠心分離により上清を回収し、 細 胞溶解液を得る。 得られた細胞溶解液から、 培地からの単離'精製法と同様の手 法により、 フオリスタチン変異体ポリぺプチドを単離 ·精製することができる。 また、 公知の方法 [J. Biomolecular MR, .6, 129、 Science, 242, 1162
(1988)、 J. Biochem. , 110, 166 (1991)〕 に準じて、 in vitro転写 ·翻訳系を用 いてフォリスタチン変異体ポリぺプチドを生産することもできる。 精製したフォリスタチン変異体の分子量は、 SDS -ポリアクリルアミ ドゲル電気 泳動 〔SDS- PAGE; Nature, 227, 680 (1970)〕 やウェスタンブロッテイング法
(Antibodies : A Laboratory Manual, Cold Spring Harbor Laboratory, Chapter 12 (1988)、 Monoclonal Antibodies : Principales and Practice, Academic Press Limited (1996) ) などで測定することができる。
3 . 本発明のフォリスタチン変異体の GDF-8選択的阻害の確認
本発明のフォリスタチン変異体ポリべプチドが 「GDF- 8の活性を選択的に阻害 する」 とは、 「ァクチビンの活性の阻害と比較して、 GDF-8の活 1"生を選択的に阻 害する」 ことを意味する。 「ァクチビンの活性の阻害と比較して、 GDF - 8の活性 を選択的に阻害する」 とは、 請求項 3に記載の方法で、 同じ濃度のフオリスタチ ン変異体ポリべプチドについて、 ァクチビンの活性の阻害と GDF - 8の活性の阻害 を測定したときに、 GDF-8の活性は 50%以上、 好ましくは 60%以上、 より好まし くは 70%以上、 さらに好ましくは 80%以上阻害するのに対し、 ァクチビンの活性 の阻害は 50%未満、 好ましくは 30%以下、 より好ましくは 10%以下であるか、 さ らに好ましくは阻害せず、 了クチビンの活性の阻害と比較して、 GDF - 8の活性を より強く阻害することを意味する。
本発明のフォリスタチン変異体ポリぺプチドは、 ァクチビンの活性の阻害と比 較して、 .GDF- 8の活性を選択的に阻害するフォリスタチン変異体ポリべプチドで あり、 ァクチビンの活性は阻害せず、 GDF- 8の活性を選択的に |3且害するものが好 ましい。 以下のように、 本発明のブオリスタチン変異体ポリペプチドの活性 (ァ クチビンの活性の阻害と比較して GDF-8の活性を選択的に阻害する活性) は、 例 えば、 ァクチビンおよび GDF- 8が有する転写促進活性をレポーターァッセィによ り測定することにより確認できる。
レポータープラスミ ド(CAGA) 12-MLP- Luc ΓΕΜΒ0 J. , 17, 3091 (1998)〕 を導入 した動物細胞を作製する。 (CAGA) 12-MLP-Lucは、 CAGAボックスと呼ばれるァクチ ビンおょぴ GDF- 8に対する応答配列 (AGCCAGACA) を上流に 12個揷入したアデノウ ィルスの主要後期プロモーターを有し、 該プロモーターの下流にレポーター遺伝 子としてホタルルシフェラ-一ゼ遺伝子を連結した構造を有する。 レポータープラ スミ ドの導入は、 2に記載の方法で行うことができる。 得られた形質転換細胞 に、 (a ) GDF- 8、 ( b ) ァクチビン、 (c ) フォリスタチン変異体ポリべプチ ド、 (d ) GDF- 8およぴフオリスタチン変異体ポリペプチド、 (e ) ァクチビン およびフォリスタチン変異体ポリペプチドをそれぞれ添加した場合、 (f ) 何も 添加しない場合について、 ルシフェラーゼ活性を測定する。 形質転換細胞は、
( f ) の何も添加しない場合と比較して、 (a ) GDF- 8および (b ) ァクチビン をそれぞれ添加した場合には、 転写の活性ィ匕を受け、 ルシフェラーゼ活性が上昇 する。 また、 (c ) フオリスタチン変異体ポリペプチドを添加した場合にはルシ フェラーゼ活性の上昇はみられない。 本発明のフォリスタチン変異体ポリべプチ ドが GDF- 8の活性を選択的に阻害することは、 (I) ( a ) GDF- 8を添加した場合 と比較して、 (d ) GDF- 8およぴフオリスタチン変異体ポリペプチドを添加した 場合には、 ルシフヱラーゼ活性が低下し、 (c ) フォリスタチン変異体ポリぺプ チドだけを添加した場合あるいは (e ) 何も添加しない場合のルシフェラーゼ活 性に近くなるのに対し、 (II) ( a ) ァクチビンを添加した場合と比較して、
( d ) ァクチビンおよぴフォリスタチン変異体ポリべプチドを添加した場合に は、 ルシフェラーゼ活性の低下が見られないか、 ルシフヱラーゼ活性の低下が、 GDF - 8の場合の活性の低下よりも、 明らかに弱いことにより確認できる。
4 . 本発明のフォリスタチン変異体ポリペプチドを含む医薬組成物
本発明のフオリスタチン変異体ポリべプチド、 該ポリべプチドを投与した生体 内で発現するための該ポリぺプチドをコードするポリヌクレオチドおよび該ポリ ヌクレオチドを含むベクターは、 ァクチビンの活性の阻害と比較して、 GDF - 8の 活性を選択的に阻害するので、 GDF - 8が発症あるいは進行に関与する症状や疾患 の治療や予防に用いることができ、 しかも、 ァクチビンの阻害による副作用が少 ないことが期待される。 このような症状や疾患としては、 例えば、 疾患や老化、 寝たきり等に伴う骨格筋の減少や萎縮、 皮下脂肪や内臓脂肪等の脂肪の増加、 悪 液質、 筋萎縮を伴う疾患、 II型糖尿病、 肥満および後天性免疫不全症候群などを あげることができる。 筋萎縮を伴う疾患としては、 デュシェンヌ型筋ジストロフ ィ一等の遺伝性の筋ジストロフィー、 および筋萎縮性側索硬化症、 脊髄小脳変性 症おょぴパーキンソン病等の神経の変性により二次的に筋萎縮を呈する疾患等を あげることができる。
本発明のフォリスタチン変異体ポリぺプチド、 該ポリぺプチドを投与した生体 内で発現するための該ポリペプチドをコードするポリヌクレオチドおよび該ポリ ヌクレオチドを含むベクターは、 GDF - 8の選択的阻害剤、 骨格筋増加剤、 脂肪低 下剤、 筋萎縮症状の治療剤又は予防剤、 ならびに悪液質、 筋萎縮を伴う疾患、 Π 型糖尿病、 肥満または後天性免疫不全症候群の治療剤又は予防剤として単独で用 いることも可能ではあるが、 通常は各種の医薬製剤として提供するのが望まし い。 また、 それら医薬製剤は、 動物または人に使用されるものである。
本発明に係わる医薬製剤は、 上記の活性成分またはその薬理学的に許容される 塩を単独で、 あるいは任意の他の治療のための有効成分との混合物として含有す ることができる。 また、 それら医薬製剤は、 活性成分を薬理学的に許容される一' 種またはそれ以上の担体と一緒に混合し、 製剤学の技術分野においてよく知られ ている任意の方法により製造される。
投与経路としては、 予防または治療に際し最も効果的なものを使用するのが望 ましく、 経口または、 例えば静脈内などの非経口をあげることができるが、 非経 口が好ましい。
投与形態としては、 例えば錠剤、 注射剤などがあげられる。
経口投与に適当な例えば錠剤などは、 乳糖、 マンニットなどの賦形剤、 澱粉な どの崩壌剤、 ステアリン酸マグネシウムなどの滑沢剤、 ヒドロキシプロピルセル ロースなどの結合剤、 脂肪酸エステルなどの界面活性剤、 グリセリンなどの可塑 剤などを添加剤として用いて製造できる。
非経口投与に適当な製剤は好ましくは受容者の血液と等張である活性ィ匕合物を 含む滅菌水性剤からなる。 例えば、 注射剤の場合は、 塩溶液、 ブドウ糖溶液また は塩水とブドウ糖溶液の混合物からなる担体などを用いて注射用の溶液を調製す る。 また、 これらの非経口剤においても経口剤で例示した賦形剤、 崩壊剤、 滑沢 剤、 結合剤、 界面活性剤、 可塑剤および希釈剤、 防腐剤、 フレーバー類などから 選択される 1種もしくはそれ以上の補助成分を添加することもできる。
さらに、 脂質、 ヒアルロン酸、 温度感受性ポリマー、 難水溶性高分子、 又は界 面活性剤などを適宜選択して添加することにより、 ぺプチド吸収の持続化を,図る こともできる。
上記の活性成分またはその薬理学的に許容される塩は、 上記の目的で用いる場 合、 通常、 全身的または局所的に、 経口または非経口の形で投与される。 投与量 および投与回数は、 投与形態、 患者の年齢、 体重、 治療すべき症状の性質もしく は重篤度などにより異なるが、 通常経口の場合、 成人 1人あたり、 1回につき 0. 01〜: 1000 mg、 好ましくは 0. 05〜500 mgの範囲で、 1日 1回ないし数回投与す る。 静脈内投与などの非経口投与の場合、 通常成人一人当り 0. 001〜1000 mg、 好 ましくは 0. 01〜300 mgを一日一回ないし数回投与するか、 または 1日 1〜24時間 の範囲で静脈内に持続投与する。 し力 しながら、 これら投与量および投与回数に 関しては、 前述の種々の条件により変動する。
また、 GDF- 8の選択的阻害剤、 骨格筋増加剤、 脂肪低下剤、 筋萎縮症状の治療 剤又は予防剤、 悪液質、 筋萎縮を伴う疾患、 II型糖尿病、 肥満または後天性免疫 不全症候群の治療剤又は予防剤の有効成分として DNAや RNAなどの核酸を利用する 遺伝子治療剤の場合には、 該 DNAまたは RNAを単独あるいはレト口ウィルスべクタ 一、 アデノウイルスベクター、 アデノ随伴ウィルスベクターなどの適当なベクタ 一に挿入した後、 上記に記載した常法に従って製剤化、 処方および投与する方法、 あるいは非ウイルス遺伝子移入法により投与する方法を使用することができる。 糸且換えウィルスべクターは、 以下に記載の方法に従い作製することができる。 有効成分として使用する DNA (以下、 対象 DNAとも称する) の断片を調製する。 該 DNA断片をウィルスベクター内のプ モーターの下流に揷入することにより、 組換えウィルスベクターを造成する。
R Aウィルスベクターの場合には、 対象 DNAに相同な RNA断片を調製し、 それら をウィルスベクター内のプロモーターの下流に揷入することにより、 組換えウイ. ルスを造成する。 R A断片は、 2本鎖のほか、 ウィルスベクターの種類に応じて、 センス鎖もしくはアンチセンス鎖のどちらか一方の鎖を選択する。 例えば、 レト ロウィルスベクターの場合は、 センス鎖に相同な RNAを、 センスウィルスベクタ 一の場合には、 アンチセンス鎖に相同な R Aを選択する。
該組換えウィルスベクターを、 該べクタ一に適合したパッケージング細胞に導 入する。 パッケージング細胞としては、 ウィルスのパッケージングに必要な蛋白 質をコードする遺伝子の少なくとも 1つを欠損している組換えウィルスベクター の該欠損する蛋白質を補給できる細胞であればいかなるものでもよい。 例えば、 ヒト腎臓由来の HEK293細胞、 マウス繊維芽細胞 NIH3T3などを用いることができる。 パッケージング細胞で捕給する蛋白質としては、 レトロウイルスベクターの場合 はマウスレトロウイルス由来の gag、 pol、 envなと、 レノチウ ルスへクタ1 ~の 場合は HIV由来の gag、 pol、 env, vpr、 vpu、 vif、 tat, rev, nefなど、 アデノウ ィルスべクタ一の場合はアデノウイルス由来の E1A、 E1Bなど、 アデノ随伴ウィル スの場合は Rep (p5、 pl9, p40) 、 Vp (Cap) などの蛋白質があげられる。
ウィルスベクターとしては、 上記パッケージング細胞において組換えウィルス が生産でき、 標的細胞で対象 DNAを転写できる位置にプロモーターを含有してい るものが用いられる。 プラスミ ドベクターとしては MFG [Proc. Natl. Acad. Sci. USA, 92, 6733 (1995)〕 、 pBabePuro [Nucleic Acids Res. , 18, 3587 (1990)〕 、 LL— CG、 CL— CG、 CS— CG、 CLG [J. Virol. , 72, 8150 (1998)〕 、 pAdexl [Nucleic Acids Res., 23, 3816 (1995)〕 などが用いられる。 プロモーターとしては、 ヒ ト耝織中で機能するものであればいずれも用いることができ、 例えば、 サイトメ ガロウィルス (CMV) の IE (immediate early) 遺伝子のプロモーター、 SV40の初 期プロモーター、 レトロウイノレスのプロモーター、 メタ口チォネインプロモータ 一、 ヒートショック蛋白質プロモーター、 SR aプロモーターなどをあげることが できる。 また、 ヒ ト CMVの IE遣伝子のェンハンサーをプロモーターと共に用いて もよい。
ノ ッケージング細胞への組換えウィルスベクターの導入法としては、 例えば、 リン酸カルシウム法 (特開平 2- 227075) 、 リボフヱクシヨン法 〔Pro Natl. Acad. Sci. USA, 84, 7413 (1987)〕 などをあげることができる。
また、 上記組換えウィルスベクターの投与方法としては、 上記の方法に加え、 リボソームデリパリ一を用いる直接的ィン ·ビボ ( vivo)遺伝子移入と組み合 わせることにより、 患者の癌組織にウィルスベクターを指向させるようにするこ ともできる。 すなわち、 適当なサイズの対象 DNAを、 アデノウイルス 'へキソン 蛋白質に特異的なポリリジン一コンジュゲート抗体と組み合わせてコンプレック スを作製し、 得られたコンプレックスをアデノウィルスベクターに結合させるこ とにより、 ウィルスベクターを調製することができる。 該ウィルスベクターは安 定に標的細胞に到達し、 エンドソームにより細胞内に取り込まれ、 細胞内で分解 され効率的に遺伝子を発現させることができる。
本発明においては、 対象 DNAを発現するベクターは非ウイ.ルス遺伝子移入法に よっても病巣に輸送することができる。
当該分野で公知の非ウィルス遺伝子移入法には、 リン酸カルシウム共沈法

リボソームを介した膜融合一介在移入法では.リボソーム調製物を標的とする組 織に直接投与することにより、 当該組織の局所的な遺伝子の取り込みおよび発現 が可能である。 DNAを患者の癌組織に直接標的化するには、 直接 DNA取り込み技術 が好ましい。 受容体を介した DNA移入は、 例えば、 ポリリジンを介して、 蛋白質 リガンドに DNA (通常、 共有的に閉環したスーパーコイル化プラスミドの形態をと る)をコンジュゲートすることによって行う。 リガンドは、 標的細胞または糸且織 の細胞表面上の対応するリガンド受容体の存在に基づいて選択する。 当該リガン ド一DNAコンジュゲートは、 所望により、 血管に直接注射することができ、 受容 体結合および DNA_蛋白質コンプレックスの内在化が起こる標的組織に指向し得 る。 DNAの細胞内破壌を防止するために、 アデノウイルスを同時感染させて、 ェ ンドソーム機能を崩壌させることもできる。 '
5 . 本発明のフオリスタチン変異体ポリぺプチドをコードするポリヌクレオチ ドを含む外来遺伝子を導入したトランスジヱニック非ヒト哺乳動物
本発明のフォリスタチン変異体ポリペプチドをコードするポリヌクレオチドを 含む外来遺伝子を非ヒト哺乳動物に導入することにより、 トランスジエニック非 ヒト哺乳動物を作製することができる。 上記トランスジエニック非ヒト哺乳動物 においては、 本発明のフォリスタチン変異体ポリべプチドが過剰発現しており、 好ましくは骨格筋量が増大していることを特徴とする。 当該トランスジエニック 非ヒト哺乳動物は、 骨格筋の形成機構の研究や、 骨格筋の異常を伴う疾患の研究 の際に使用するモデル動物として有用である。
本明細書で言う 「非ヒト哺乳動物」 の具体例.としては、 マウス、 ラット、 モル モット、 ハムスター、 ゥサギ、 ィヌ、 ネコ、 ヒッジ、 ブタ、 ャギ、 ゥシまたはサ ルなどが挙げられ、 好ましくは、 マウス、 ラット、 モ 'ルモット、 ハムスター、 ゥ サギなどのげつ歯類動物であり、 最も好ましくは、 マウスである。
以下、 本発明のトランスジエニック非ヒト哺乳動物の作出方法について説明す る。
( 1 ) 導入遺伝子の調製
本発明のフオリスタチン変異体ポリペプチドをコードする D N Aを対象動物に 導入する際には、 導入の対象動物の細胞内で当該 D NAを発現させうるプロモー ターの下流に当該 D N Aを連結した遺伝子構築物を用いるのが一般に有利である。 導入させる遺伝子は、 常法により、 例えば、 下記の通り調製することができる。 導入させる遺伝子の全長相補 D N Aをもとにして、 必要に応じて、 該ポリぺプ チドをコードする部分を含む適当な長さの D N A断片を調製し、 次いで該 D N A 断片、 または全長相補 D NAを適当な発現ベクターのプロモーターの下流に挿入 することにより、 組換えベクターを作製する。
動物へ導入させる遺伝子の発現ベクターとしては、 大腸菌由来のプラスミド、 枯草菌由来のプラスミド、 酵母由来のプラスミド、 λファージなどのバクテリオ ファージ、 モロ-一白血病ウイ/レスなどのレトロゥイノレス、 ワクシニア'ウイノレス またはバキュロウィルスなどの動物ウィルスなどが用いられる。 これらの中でも、 大腸菌由来のプラスミド、 枯草菌由来のプラスミドまたは酵母由来のプラスミド などが好ましく、 大腸育由来のプラスミドが特に好ましい。
本発 で用いるプロモーターの具体例は本明細書中上記した通りである。 本発 明で用いる発現ベクターは、 トランスジエニック哺乳動物において目的とするメ ッセンジャー R N Aの転写を終結する配列 (一般にターミネタ一と呼ばれる) を 有していることが好ましく、 例えば、 ウィルス由来、 各種哺乳動物および鳥類由 来の各遺伝子に含まれる同機能を有する配列を用いて遺伝子発現を操作すること ができる。 好ましくは、 シミアンウィルスの SV40 ターミネタ一などがよく用い られる。 その他、 目的遺伝子をさらに高発現させる目的で、 各遺伝子のスプライ シングシグナル、 ェンハンサー領域、 真核生物遺伝子のィントロンの一部をプロ モーター領域の 5' 上流、 プロモーター領域と翻訳領域間あるいは翻訳領域の 3, 下流 に連結することも目的により可能である。
( 2 ) トランスジエニック非ヒト哺乳動物の作製
本発明のトランスジヱニック非ヒ ト哺乳動物は、 ブオリスタチン変異体ポリべ プチドをコードする D NAを対象動物に導入することによって作製できる。 具体 的には、 該遺伝子を対象となる非ヒト哺乳動物の受精卵、 胚性幹細胞、 体細胞、 精子、 未受精卵へ導入することによって、 目的とするフォリスタチン変異体ポリ ぺプチドをコ一ドする D N Aが胚芽細胞を含むすべての細胞の染色体上に組み込 まれたトランスジエニック非ヒト哺乳動物を得ることによって作出できる。 受精 卵、 胚性幹細胞、 体細胞、 精子、 未受精卵における該遺伝子の導入は、 対象非ヒ ト哺乳動物の胚芽細胞および体細胞を含む全ての細胞の染色体上に存在するよう に確保されることが好ましい。 遺伝子導入後の作出動物の胚芽細胞において該導 入遺伝子が存在することは、 作出動物の子孫がその胚芽細胞および体細胞の全て に該導入遺伝子を有することを意味する。
個体の選択は、 導入遺伝子産物の発現を蛋白質あるいは mR N Aレベルで確認 することによって行うことができる。 選択された個体の遺伝子を受け継 、だこの 種の動物の子孫は、 該導入遺伝子産物を有する。 導入遺伝子を相同染色体の両方 に持つホモザィゴート動物を取得し、 この雌雄の動物を交配することによりすべ ての子孫が該遺伝子を穸定に保持し、 また、 該遺伝子を有することを確認して、 通常の飼育環境で繁殖継代することができる。
受精卵あるいは胚性幹細胞への遺伝子導入によるトランスジヱニック動物の作 製は、 マニピユレ一ティング 'マウス 'ェンブリオ第 2版、 Gene Targeting, A Practical Approach, IRL Press at Oxford University Press (1993)ヽ バイォ マニュアルシリーズ 8, ジーンターゲッティング, E S細胞を用いた変異マウス の作製, 羊土社 (1995)、 発生工学実験マニュアル, トランスジエニック 'マウス の作り方, 講談社 (1987)等に記載された方法を用い、 哺乳類動物細胞への遺伝 子導入と核移植によるトランスジエニック動物の作製は、 Cibelliら (Science, 280, 1256, 1998) 、 Schniekeら (Science, 278, 2130, 1997)、 Baguisiら (Nature Biotechnology, 17, 456, 1999) 等によって報告された方法を用い、 精子への遺伝子導入によるトランスジェニック動物の作製は、 Perryら (Science, 284, 1180, 1999) や Wakayamaら (Nature Biotechnology, 16, 639, 1998)によ つて報告された方法を用い、 未受精卵への遺伝子導入と体外受精によるトランス ジエニック動物の作製は、 Chanら (Pro Natl. Acad. Sci. USA, 95, 14028, 1998) によって報告された方法を用いて行うことができる。
6 . 家畜の改良への応用
本発明のフォリスタチン変異体ポリぺプチド、 該ポリぺプチドを投与した生体 内で発現するための該ポリペプチドをコードするポリヌクレオチドおよび該ポリ ヌクレオチドを含むベクターをゥシ、 ブタ、 ヒッジ、 ニヮトリ等の非ヒト動物に 投与することにより、 該非ヒト動物の骨格筋を増加させ、 脂肪を低下させること ができる。 また、 5に記載した方法と同様にして、 本発明のフォリスタチン変異 体ポリペプチドをコードする D N Aを導入したトランスジエニック非ヒト動物を 作製することにより、 、 該非ヒト動物の骨格筋を増加させ、 脂肪を低下させるこ とができる。 特に非ヒト動物が家畜である場合、 以上のようにして肉の量が多く、 脂肪の少ない家畜を作製することができるので、 肉の生産量を増加でき、 また脂 肪の少ない良質肉の生産をすることができる。 また骨格筋の増加から、 家畜の労 働力を改良することができる。
以下、 本発明の実施例を示すが、 これにより本発明の範囲が限定されるもので はない。 実施例
実施例 1 :フォリスタチン変異体ポリペプチド発現ベクターの作製
ヒト精巣 cDNAライブラリー 〔クロンテック (Clontech) 社製〕 から、 ブタフォ リスタチン前駆体 cDNAをプロープとしたハイブリダィゼーシヨンによりクローン ィ匕された、 ヒ トフオリスタチン前駆体 cDNA [Proc. Natl. Acad. Sci. USA, 85, 4218, (1988)〕 を、 プラスミド pUC19 (宝酒造社製) 中の EcoRI部位と Xbal部位の 間に挿入し、 フォリスタチン変異体用の铸型 PUC19/FSを作製した。
0. 2 謹 ol/l dNTPs、 1 讓 ol/l塩化マグネシウムを含む反応液に、 フオリスタチ ン変異体用の铸型を 100 ng、 そしてプライマーとして、 配列番号 11および配列番 号 12に示した塩基配列を有する合成 DNA 〔ジヱンセット (Genset) 社製〕 をそれ ぞれ 10 pmolずつ加え、 さらに 2. 5単位の Taqポリメラーゼ (TaKaRa Ex Taq;宝酒 造社製) を添加して、 合計 25 とし、 PCRを行った。 反応条件は、 94°C 30秒 間、 74°C 15秒間、 72°C 30秒間のサイクルを 3サイクル、 94°C 30秒間、 70°C 15 秒間、 72°C 30秒間のサイクルを 3サイクル、 94°C 30秒間、 66°C 15秒間、 72°C 30秒間のサイクルを 3サイクル、 94°C 30秒間、 62°C 15秒間、 72°C 30秒間のサイ クルを 3サイクル、 94°C 30秒間、 58°C 15秒間、 72°C 30秒間のサイクルを 3サイ クル、 94°C 30秒間、 54°C 15秒間、 72°C 30秒間のサイクルを 12サイクル、 その 後、 72°C 10分間を 1サイクルで行った。 該反応液を 1 %ァガロースゲル電気泳動 にて分画し、 約 0. 25kbの遺伝子増幅断片 Aを回収した (図 2 ) 。 遺伝子増幅断片 A がコードするァミノ酸配列には、 配列番号 4の 95〜168番目の配列が含まれ、 ヒ トフオリスタチンの FSIを含む。
プライマーとして、 配列番号 13および配列番号 14に示した塩基配列を有する合 成 DNA (ジェンセットネ土製) をそれぞれ 10 pmolずつ用いて、 上記の遺伝子増幅断 片 Aと同様の条件で PCRを行い、 反応液を 1%ァガロースゲル電気泳動にて分画 · し、 約 0. 6kbの遺伝子増幅断片 Bを回収した (図 3 ) 。 遺伝子増幅断片 Bがコード するアミノ酸配列には、 配列番号 4の 1〜166番目の配列が含まれ、 ヒトフオリス タチンの FSNおよび FSIを含む。
得られた遺伝子増幅断片 Aおよび Bをそれぞれ、 pGEM-T Easy TAクローニングキ ット 〔プロメガ (Promega) 社製〕 を用いて添付の説明書に従って、 pGEM- T Easy ベクターに連結し、 図 2およぴ図 3に示すプラスミド pGEM- T Easy (A)および pGEM-T Easy (B)を得た。
次にプラスミド pGEM- T Easy (B)を制限酵素 EcoRI (宝酒造社製) および Hindlll (宝酒造社製) で切断して得られる増幅断片 Bを含む約 0. 59kbの EcoRI- Hindlll断片(B)およびプラスミド pBluescript SK (-) 〔ストラタジーン
(Stratagene) 社製〕 を EcoRIおよび Hindlllで切断して得られる約 2. 95kbの EcoRI- Hindlll断片を連結し、 図 4に示すプラスミド pBluescript SK (-) (B)を 調製した。
次に、 プラスミド pGEM- T Easy (A)を制限酵素 Hindlllおよび Sail (宝酒造社 製) で切断して得られる増幅断片 Aを含む約 0. 27kbの Hindlll-Sall断片 (A)、 およ ぴプラスミド pBluescript SK (-) (B)を Hindlllおよび Sailで切断して得られる 約 3. 54kbの Hind III- Sal I断片(B)を連結し、 図 5に示すプラスミド pBluescript SK (-) (A+B)を調製した。
次に、 プラスミド pBluescript SK (-) (A+B)を制限酵素 EcoRIおよび Spel (宝 酒造社製) で切断して得られる増幅断片 Aおよび Bを含む約 0. 84kbの Eco Rl-Spe I 断片 (A+B)、 および動物細胞発現用プラスミド pcDNA3 (インビトロジェン社製) を EcoRIおよび Xbal (宝酒造社製) で切断して得られる約 5. 4kbの EcoRI - Xbal断片 を連結し、 図 6に示したフオリスタチン変異体ポリぺプチド発現べクタ一 pcDNA3 (A+B)を得た。 pcDNA3 (A+B)に含まれるフォリスタチン変異体ポリぺプチドをコ 一ドする領域の塩基配列を配列番号 15に、 該ポリぺプチドのァミノ酸配列を配列 番号 16に示す。 配列番号 16の 1から 29番目のアミノ酸配列はシグナルぺプチドで あり、 ポリペプチドの分泌時には切断される。 したがって、 pcDNA3 (A+B)を導入 した動物細胞は配列番号 6のアミノ酸配列からなるフォリスタチン変異体ポリぺ プチド (以下、 FS1 - 1とよぶ) を分泌する。 FS1-1はヒトフオリスタチンの FSNお ょぴ 2つの FS Iを含むが、 FS Πおよひ TS 111は欠失しているフォリスタチン変異体 ポリペプチドである。
また、 pBluescript SK (-) (B)を EcoRIおよび Xholで切断して得られる、 増幅 断片 Bを含む約 0. 6 kbの EcoRI - Xhol断片および、.動物細胞発現用プラスミド pcDNA3を EcoRIおよび Xholで切断して得られる約 5. 4kbの EcoRI- Xhol断片を連結 し、 図 7に示すフォリスタチン変異体ポリぺプチド発現べクタ一 PcDNA3 (B)を得 た。 pd)NA3 (B)に含まれるフォリスタチン変異体ポリペプチドをコードする領域 の塩基配列を配列番号 17に、 該ポリぺプチドのァミノ酸配列を配列番号 18に示 す。 配列番号 18の 1から 29番目のアミノ酸配列はシグナルペプチドであり、 ポリ ペプチドの分泌時には切断される。 したがって、 pcDNA3 (B)を導入した動物細胞 は配列番号 8のァミノ酸配列からなるフォリスタチン変異体ポリペプチド (以 下、 FS1とよぶ) を分泌する。 FS1はヒトフオリスタチンの FSNおよび 1つの FSIを 含むが、 FSIIおよび FSIIIは欠失しているフォリスタチン変異体ポリペプチドで ある。
また、 フオリスタチン変異体用の鎵型を EcoRIで切断して得られるヒトフオリ スタチン前駆体 cDNAを含む約 1. 1 kbの EcoRI断片をプラスミド pBluescript SK (- )の EcoRI部位に挿入し、 図 8に示す pBluescript SK (-) /FSを得た。 得られたプ ラスミ ド pBluescript SK (-) /FSを、 EcoRVおよび Xholで切断して得られる約 0. 4 kbの EcoRV- Xhol断片 Cおよび、 プラスミ ド pBluescript SK (-) (A+B)を EcoRVおよ ぴ Xholで切断して得ちれる約 3. 8 kbの ficoRV- Xhol断片を連結し、 図 9に示すプラ スミド pBluescript SK (-) (A+B+C)を調製した。
次に、 pBluescript SK (-) (A+B+C)を EcoRIおよび Xholで切断して得られる、 約 1. 2 kbの EcoRI- Xhol断片おょぴ、 動物細胞発現用プラスミ ド pcDNA3を EcoRIお よび Xholで切断して得られる約 5. 4kbの EcoRI - Xhol断片を連結し、 図 1 0に示す フオリスタチン変異体ポリぺプチド発現べクタ一 pcDNA3 (A+B+C)を得た。 pcDNA3 (A+B+C)に含まれるフォリスタチン変異体ポリべプチドをコードする領域の塩基 配列を配列番号 19に、 該ポリぺプチドのァミノ酸配列を配列番号 20に示す。 配列 番号 20のの 1から 29番目のアミノ酸配列はシグナルぺプチドであり、 ポリぺプチ ドの分泌時には切断される。 したがって、 PcDNA3 (A+B+C)を導入した動物細胞は 配列番号 10のァミノ酸配列からなるフオリスタチン変異体ポリぺプチド (以下、 FSI- 1 - 3とよぶ) を分泌する。 FS1 - 1-3はヒ トフオリスタチンの FSNおよび 2つの FSIおよび FS3のほぼ全体を含むが、 FSIIは欠失しているフォリスタチン変異体ポ リペプチドである。 また、 マウス FLRG cDNA [J. Biol. Chem., 275, 40788 (2000)3 を铸型と し、 プライマーとして、 配列番号 21および配列番号 22に示した塩基配列を有する 合成 DNA (ジェンセット社製) をそれぞれ 10 pmolずつ用いて、 上記の遺伝子増幅 断片 Aと同様の条件で PCRを行い、 反応液を 1%ァガロースゲル電気泳動にて分画 し、 約 0. 3kbの遺伝子增幅断片 Dを回収した (図 1 1 ) 。 該遺伝子増幅断片がコー ドするアミノ酸配列には、 マウス FLRGの (NCBI蛋白質データベース:登録番号 BAB32663) の 165〜256番目のアミノ酸配列を含み、 マウス FLRGの FSII様ドメイン
(NCBI蛋白質データベース:登録番号 BAB32663の 169〜241番目の領域) が含まれ る。 図 1 4に示すように、 マウス FLRGの FSII様ドメインはマウスフオリスタチン の FSIIとアミノ酸配列で 56% (41/73) の相同性を、 配列番号 2のアミノ酸配列 とは 61. 6% (45/73) の相同性を有する。
得られた遺伝子増幅断片 Dを EcoRIで切断し、 pBluescript SK (-)の EcoRI部位 に挿入し、 図 1 1に示す pBluescript SK (-) (D)を得た。 得られたプラスミド pBluescript SK (-) (D)を EcoRVおよび Xholで切断して得られる約 0. 3 kbの EcoRV - Xhol断片、 およびプラスミ ド pBluescript SK (-) (A+B)を EcoRVおよび Xholで切断して得られる約 3. 8 kbの EcoRV- Xhol断片を連結し、 図 1 2に示すプラ スミ ド pBluescript SK (-) (A+B+D)を調製した。
次に、 pBluescript SK (-) (A+B+D)を EcoRIおよび Xholで切断して得られる、 約 1. 1 kbの EcoRI- Xhol断片および、 動物細胞発現用プラスミ ド pcDNA3を EcoRIお ょぴ Xholで切断して得られる約 5. 4kbの EcoRI-XhoI断片を連結し、 図 1 3に示す フオリスタチン変異体ポリぺプチド発現べクタ一 pcDNA3 (A+B+D)を得た。 pcDNA3 (A+B+D)に含まれるフォリスタチン変異体ポリぺプチドをコ一ドする領域の塩基 配列を配列番号 23に、 該ポリぺプチドのァミノ酸配列を配列番号 24に示す。 配列 番号 24の 1から 29'番目のアミノ酸配列はシグナルぺプチドであり、 ポリぺプチド の分泌時には切断される。 したがって、 pcDNA3 . (A+B+D)を導入した動物細胞は配 列番号 25のァミノ酸配列からなるブオリスタチン変異体ポリぺプチド (以下、 FS1 - 1- 2,とよぶ) を分泌する。 FS1- 1 - 2,はヒトフオリスタチンの FSNおよび 2つ の FSIを含み、 FSIIの代わりにマウス FLRGの FSII様ドメインを含むフォリスタチ ン変異体ポリぺプチドである。 実施例 2 :フオリスタチン変異体ポリペプチドの動物細胞を用いた発現と精製
( 1 ) COS - 7細胞への発現ベクターの導入と培養
実施例 1で得られたフオリスタチン変異体ポリぺプチド発現べクタ一 PcDNA3 (A+B)、 pcDNA3 (B)、 pcDNA3 (A+B+C)、 pcDNA3 (A+B+D)を用いてフォリスタチン 変異体ポリぺプチドの動物細胞での発現を以下のようにして行つた。
1 X 106細胞の COS- 7 (ATCC CRL-1651) 細胞あたり 20 gのプラスミ ド PcDNA3 (A+B)をエレクト口ポレーシヨン法 (Cytotechnology, 3, 133 (1990) ) により導 入した後、 ゥシ胎児血清 (FCS) を 10%添加したダルベッコ改変イーグル培地
[DMEM;シグマ一アルドリツチ (Sigma- Aldrich) 社製〕 10 mlに懸濁し、 10 cm 径細胞培養ディッシュ 〔コーユング (Corning) 社製〕 に播種した。 5% C02イン キュベータ一内で 37°C、 24時間培養した後、 10 mlの DMEMへ培地交換し、 3日間培 養した。 細胞培養ディッシュ 6枚で上記の培養を行い、 培養上清約 60 mlを回収 した。 pcDNA3 (B)、 pcDNA3 (A+B+C)および pcDNA3 (A+B+D)についても同様に COS - 7細胞に導入して培養を行い、 それぞれ培養上清約 60mlを回収した。
( 2 ) フオリスタチン変異体ポリペプチドの精製と検出
( 1 ) .で得られた 4種類のブオリスタチン変異体ポリペプチドそれぞれを発現 する COS- 7細胞の約 60 mlの培養上清を 20 mmol/L Tris - HCl (pH7. 2) , 0. 3 mol/L NaCl, 0. 03% 3- [ (3-コールアミ ドプロピル)ジメチルアンモニォ] - 1 -プロパンス ルホネート (CHAPS) で平衡化した 2 mlの硫酸化セル口ファインカラム (生化学 工業社製) に通塔した。 その後、 計 20 mlの 20 mmmol/L Tris-HCl (pH7. 2) , 0. 3 mol/L NaCl, 0. 03% CHAPSゝ 続いて計 6 mlの 20 醒 ol/L Tris-HCl (pH7. 2), 0. 5 mol/L NaCl, 0. 03% CHAPSでカラムを洗浄した。 その後、 計 10mlの 20mmmol/L Tris-HCl (PH7. 2) , 1. 0mol/L NaCl, 0. 03% CHAPSを用いて吸着タンパク質を力 ラムから溶出させた。 この溶出液をウルトラフリ一- 15 (Ultrafree-15) 遠心式 フィルターュニット (バイオマックス -10メンブレン装着ュニット ; ミリポア社 製) を用いて添付の説明書に従って濃縮し、 200 μ 1のフオリスタチン変異体ポリ ぺプチド溶液を得た。
ヒ トフオリスタチンの FSIの部分配列である、 配列番号 26のアミノ酸配列から なるペプチドを化学合成し、 このペプチドを抗原として、 常法 [Antibodies : A Laboratory Manual, Cold Spring Harbor Laboratory (1988) J に従ってゥサキ を免役し、 抗体価の上昇したゥサギから血清を採取し、 ゥサギ抗フオリスタチン ポリクローナル抗体とした。 該抗体のクラスは IgGであった。
得られた各フオリスタチン変異体ポリペプチドを公知の方法 [Nature, 227, 680 (1970) ] に従って SDS-PAGEに供した。 泳動タンパク質を膜 (Iramobilon P; Millipore社製) に転写し、 5%スキムミルク (ナカライテスク社製) で膜をプロ ッキングした後、 ゥサギ抗フオリスタチンポリクローナル抗体と反応させた。 検 出は、 西洋ヮサビペルォキシダーゼ標識ラット抗ゥサギ IgG抗体 〔ケミコン
(Chemicon) 社製〕 および化学発光試薬 (ECL検出試薬;アマシャム ·バイオサ イエンシズ社製) を用いて行った。 各フオリスタチン変異体ポリペプチドとも、 ゥサギ抗フオリスタチンポリクローナル抗体により検出された。
図 1 5に FS1 - 1のウェスタンブロット解析を示した。 FSI- 1は 254個のアミノ酸 からなり、 推定分子量は約 27kDである。 一方、 ウェスタンプロット解析で得られ た分子量は 31kD、 33kD、 37kDである。 FSI- 1には糖鎖付加サイトがあること力 ら、 糖鎖の付加によって分子量が大きくなつたものと考えられる。 実施例 3 : レポーターアツセィによるフォリスタチン変異体ポリペプチドのァク チビンおよび GDF-8機能阻害活性の検定
HEK293細胞 (ATCC番号: CRL- 1573) を 24ゥエルプレートに 5 X 104細胞 Zゥエル で播種し、 10%FCS、 ペニシリン (100単位/ ml) .およびストレプトマイシン (100 g/ml) を添カ卩した高グルコース含有 DMEM培地 (シグマ一アルドリッチ社製)
(以下、 血清含有 DMEM培地とする) 1 ml中で 24時間培養した。 リン酸緩衝生理食 塩水 (PBS) で洗浄し、 レポータープラスミ ド(CAGA)12-MLP- Luc [EMBO J. , 17, 3091 (1998)〕 (3/ gZウエノレ) および内部標準プラスミ ド CMV- j3 - gal (1/ gZ ゥエル) をトランスフエクションした。 トランスフエクションはプラスミ ドとカ チオン性リボソーム (Transfast;プロメガ社製) の混合液を添加した血清不含 DMEM培地 (200 μΐ) 中で、 細胞を 2時間 37°Cで培養することにより行った。 さら に血清含有 DMEM培地 (1 ml) を添加し、 37°Cで 24時間培養した。
細胞を PBSで洗浄した後、 (a) ァクチビン (10 ng/ml) 、 (b) GDF - 8 (15 ng/ml) 、 (c) ァクチビン (10 ng/ml) とフオリスタチン (100 ng/ml) 、 (d) GDF - 8 (15 ng/ml) とフオリスタチン (100 ng/ml) 、 (e) ァクチビン (10 ng/ml) とフオリスタチン変異体ポリぺプチド FS1- 1溶液 5 1、 もしくは (f ) GDF- 8 (15 -ng/ml) とフオリスタチン変異体ポリペプチド FSl-1溶液 5 μ 1を それぞれ含む血清不含 DMEM培地 200 μ 1/ゥ工ルを添加し、 37°Cで 24時間培養し た。 PBSで洗浄した後、 細胞溶角用緩衝液 Cl%(v/v)トリ トン X- 100、 15 mmol/l MgS04、 4 mmol/lエチレングリコール -ビス(2-アミノエチノレエーテノレ)- N, N, Ν' ,Ν' -4酢酸、 1 mmol/l DL-ジチオスレィ トール、 25 mmol/lグリシルグ リシン、 pH 7.0〕 を 100 μΐ/ゥエル添加し、 0°Cで 15分間放置し、 細胞溶解液を 調製した。 細胞溶解液 50 μ1をルシフェラーゼアツセィ用緩衝液 〔0.24 mmol/l ルシフェリン、 1.4 mmol/l ATP、 4.8 mmol/l KH2P04、 4.8 mmol/l MgCl2、 pH 7.4〕 105 1と混合し、 発光量をルミノメータ 〔マイクロルーマット
(MicroLumat) LP98P ;ベルトルード (BERTH0LD) 社製〕 を用いて測定した。 反 応開始から 20秒間の相対光度 (RLU) 値を積算し、 ルシフェラーゼ活性とした。 続いて細胞溶解液 35 μΐを —ガラクトシダーゼアツセィ用緩衝液 [1.6 mg/ml o—二トロフエ二ノレ—; 3 - D-ガラクトシド、 60 mmol/l Na2HP04、 40 mmol/l
NaH2P04 10 mmol/l KC1、 1 mmol/l MgCl2、 50 腿 ol/l i3—メルカプトエタノー ル〕 235μ1と混合し、 吸光度 (0D) を吸光度計. 〔マイクロプレートリーダー 'ベ ンチマーク (Microplate Reader Benchmark) ;バイオラッド (BI0-RAD) 社製〕 を用いて測定した。 以下の式によって与えられる値を標準化ルシフェラーゼ活性 とし、 転写促進活性の指標と'した。
標準化ルシフヱラーゼ活性 = RLU 0D
測定結果を図 1 6に示す。 フオリスタチンは、 ァクチビンおよぴ GDF - 8による 標準化ルシフヱラーゼ活性の上昇をいずれも 100%阻害した。 一方、 フオリスタ チン変異体ポリペプチドは、 GDF-8による標準化ルシフェラーゼ活性の上昇を . 86%阻害したのに対して、 ァクチビンによる標準化ルシフェラーゼ活性の上昇を 17%しか阻害しなかった。
フォリスタチン変異体ポリべプチド FS1、 FS1- 1- 3および FS1- 1- 2,についても、 上記の FS1-1と同様なアツセィを行った。 その結果、 図 1 7およぴ図 1 8に示す ように、 FS1、 FS1 - 1 - 3は FS1 - 1と同様に、 GDF- 8による標準ィ匕ルシフェラーゼ活性 の上昇をほぼ完全に阻害したのに対して、 ァクチビンによる標準化ルシフェラー ゼ活性の上昇はほとんど阻害しなかった。 ただし、 FS1は FS1- 1よりも、 また FS1 - 1-3は FS1よりも、 ァクチビンによる標準化ルシフェラーゼ活性の上昇を阻害する 程度がやや強く、 3種類の中では FS1- 1が最も選択的に GDF- 8の活性を阻害するこ とがわかった。 したがって、 選択的に GDF-8の活性を阻害するためには、 フオリ スタチン変異体ポリペプチドにおいて、 FSIの数は 2個以上ある方がよく、 FSII だけでなく FSIIIも欠失している方が好ましいことがわかった。 また、 FS1 - 1 - 2, は GDF-8による標準化ルシフェラーゼ活性の上昇だけでなく、 ァクチビンによる 標準ィ匕ルシフェラーゼ活性の上昇も阻害した。 したがって、 選択的に GDF- 8の活 性を阻害するためには、 ブオリスタチン変異体ポリペプチドにおいて、 FSIIおよ び FSII様ドメインを有しないことが好ましいことがわかった。 実施例 4 :ブオリスタチン変異体ポリ-ペプチドを過剰発現させたマウスの骨格筋 量の増大およぴ脂肪量の減少
実施例 1で作製した pcDNA3 (A+B)を EcoRIおよび Saclで切断して得られる、 フ ォリスタチン変異体ポリべプチド FS1 - 1をコードする DNAを含む EcoRI- Sacl断片 を、 ベクター PSP72 (プロメガ社製) の EcoRI部位おょぴ Sacl部位の間に挿入し た。 得られたプラスミドを EcoRIおよび Smalで切断して得られる、 フオリスタチ ン変異体ポリぺプチド FS 1-1をコードする DNAを含む EcoRI- Sacl断片を、 MDAF2ベ クタ一 〔Pro Natl. Acad. Sci. USA, 98, 9306 (2001)〕 のミオシン軽鎖のプ 口モーターの下流に存在する EcoRI部位おょぴ Sacl部位の間に揷入し、 プラスミ ド MDAF2- FS1- 1を得た。
MDAF2- FS1- 1の約 3. 7 kbpの Clal断片を単離し、 BDF1マウスの受精卵 221個に顕 微注入した。 状態の良好な受精卵をレシピエントマウスに移植し、 自然出産さ せ、 33匹の産仔を得た。 ゲノム DNA中に MDAF2 - FS1- 1由来の DNAが導入されている ことが確認されたトランスジヱニックマウスの系統を維持した。 該トランスジ工 ニックマウスは、 ミオシン軽鎖のプロモーターによりフオリスタチン変異体ポリ ぺプチド FS1 - 1を骨格筋特異的に過剰発現する。 このようにして得られたトラン スジエニックマウスでは、 骨格筋の明らかな増加が観察された。 例として、 14週 齢のォスのトランスジエニックマウスと野生型マウスの大胸筋、 大殿筋、 上腕二 頭筋を採取し、 重量を測定した結果を図 1 9に示す。 野生型マウスと比較してト ランスジヱニックマウスでは、 測定した全ての骨格筋の重量が増加していた。 さらに、 このようにして得られたトランスジヱニックマウスの、 皮下脂肪およ び内臓脂肪の量を測定し、 フォリスタチン変異体ポリべプチドがこれらの脂肪量 に与える影響を調べた。.皮下脂肪としては鼠径部の皮下脂肪、 内臓脂肪としては 腎臓周囲の脂肪、 それぞれの重量を測定し、 体重に対する割合 (%) を量の指標 とした。 その結果、 トランスジエニックマウスでは、 皮下脂肪量および内臓脂肪 量ともに、 野生型マウスと比較して著しく減少していることがわかった。 例とし て、 図 2 0に 12週齢のメスのトランスジエニックマウスおよび 12週齢のメスの野 生型マウスそれぞれの、 体重に対する皮下脂肪量 (%) および内臓脂肪量 (%) を示した。 野生型マウスは 3匹の平均値を示した。 また、 ォスのトランスジェニ ックマウスでも同様に、 皮下脂肪量および内臓脂肪量の著しい減少が見られた。 なお、 トランスジエニックマウスでは、 骨格筋の増加と脂肪の減少以外の異常は 観察されなかった。 実施例 5 :フォリスタチン変異体ポリペプチドとィムノグロブリン定常領域との 融合蛋白質による GDF - 8特異的な機能阻害
( 1 ) フオリスタチン変異体ポリペプチドとィムノグロプリン定常領域との融合 蛋白質の発現べクターの作製
フオリスタチン変異体ポリぺプチド FS1および FS1 - 1それぞれとィムノグロブリ ンの定常領域との融合蛋白質の発現ベクターを以下のようにして作製した。 実施例 1で作製した pGEM- T Easy (B)を铸型にして、 プライマーとして配列番 号 27および 28に示した塩基配列を有する合成 DNA (ジヱンセット社製) をそれぞ れ 10 pmolずつ用いて、 実施例 1の遺伝子増幅断片 Aと同様の条件で PCRを行い、 反応液をァガロースゲル電気泳動にて分画し、 5'端に Kpnlサイト、 3'端に BamHI サイトがそれぞれ付カ卩された FS1をコードする増幅断片を回収した。 得られた増 幅断片を Kpnlおよび BamHIで切断し、 CD5- IgGlベクター 〔Cell, 61, 1303 (1990)〕 を制限酵素 Kpnlおよび BamHIで切断して得られるヒ ト IgGl定常領域をコ ードするゲノム配列を含む Kpnl- BamHI断片と連結し、 FS1の C末端にヒト IgGlの定 常領域が融合した融合蛋白質 (以下、 FS1- Fcとよぶ) 発現プラスミド pcDM3 FS1- Fcを作製した。 CD5- IgGlベクターでは、 ベクター pcDNA3のプロモーターの下 流の Xholサイトー Xbalサイト間にヒト IgGl定常領域をコードする配列が挿入され ているの.で、 pcDNA3 FS1- Fcは、 pcDNA3のプロモーターの下流に、 FS1- Fcをコー ドする配列が揷入された構造を有する。
実施例 1で作製した pBluscript (A+B)を EcoRIおよび EcoRVで切断して得られる FS1- 1をコードする配列を含む断片を、 ベクター pSP72 (プロメガ社製) の EcoRI サイトー Smalサイト間に挿入した。 得られたプラスミドを EcoRIおよび BamHIで切 断して得られる FS1 - 1をコードする配列を含む断片を、 pBluescript SK (-)の EcoRIサイトー bamHIサイト間に挿入した。 得られたプラスミドを KpnIKpnlおよび BamHIで切断して得られる FS1 - 1をコードする配列を含む断片を、 CD5 - IgGlベクタ 一を制限酵素 Kpnlおよび BamHIで切断して得られるヒ ト IgGl定常領域をコードす るゲノム配列を含む Kpnl - BamHI断片と連結し、 FS1 - 1の C末端にヒト IgGlの定常領 域が融合した融合蛋白質 (以下、 FS1 - 1 - Fcとよぶ) 発現プラスミド PcDNA3 FS1 - 1 - Fcを作製した。 pcDNA3 FS1- 1- Fcは、 pcDNA3のプロモーターの下流に、 FS1- 1- Fcをコードする配列が揷入された構造を有する。
( 2 ) CH0- K1細胞への発現べクターの導入と培養
CH0-K1 (ATCC番号: CCL- 61) 細胞に pcDNA3 FS1- Fcまたは pcDNA3 FS:i- 1- Fcそれ ぞれをリボソーム試薬 Transfast (プロメガ社製) を用いて導入した後、 安定に 導入した遺伝子が組み込まれたシングルクローンを得た。 それぞれのシングルク ローンを FCSを 10%添力 Dした a MEM培地 10 mlに懸濁し、 10 cm径細胞培養ディッシ ュに播種した。 5% C02インキュベーター内で 37°C、 コンフルェントになるまで 培養した後、 EX- CELL 301培地 (JRHバイオサイェンシズ社製) へ培地交換して、 細胞培養ディッシュ 6枚で 1週間培養し、 培養上清約 60 mlを回収した。
( 3 ) FS1- Fcおよび FS卜 1- Fcの精製
( 2 ) で得られた FS1 - Fc、 FS1- 1-Fcそれぞれを発現する CH0-K1細胞の培養上清 に 1/10量のへパリンセル口ファイン (生化学工業社製) を加えて、 室温で 2時間 混合し、 蛋白質を吸着させた。 へパリンセル口ファインを洗浄バッファー 〔20 mmmol/L Tris-HCl (pH7. 4) , 0. 15 mol/L NaCl] で洗浄後、 溶出バッファー 〔20 mmmol/L Tris-HCl (pH7. 4) , 1. 5 mol/L NaCl] を用いて吸着蛋白質を溶出させ た。 得られた溶出液から、 ィムノグロブリンの定常領域と特異的に結合するプロ ティン A固定化レジンを用いて、 常法に従って、 FS1- Fc溶液および FS1- 1- Fc溶液 それぞれを得た。
( 4 ) FS1- Fcおよび FS1- 1- Fcのァクチビンおよび GDF- 8機能阻害活性の検定
( a ) 緩衝液のみ、 (b ) FSl-Fc (20 μ g/ml) ( c ) ァクチビン (15 ng/ml) 、 (d ) ァクチビン (15 ng/ml) と FSl—Fc (20 μ g/ml) 、 ( e ) GDF-8
(15 ng/ml) 、 ( f ) GDF-8 (15 ng/ml) と FSト Fc (10 μ g/ml) および (g ) GDF-8 (15 ng/ml) と FS1 - Fc (20 μ g/ml) それぞれの条件で、 実施例 3と同様に してレポーターァッセィにより、 FS1- Fcのァクチビンおょぴ GDF- 8機能阻害活性 の検定を行った。 図 2 1に示すように、 FSl- Fcは GDF-8による標準化ルシフェラ ーゼ活性の上昇をほぼ完全に阻害したのに対して、 ァクチビンによる標準化ルシ フエラーゼ活性の上昇はほとんど阻害しなかつた。
同様にして、 ) 緩衝液のみ、 (b ) FSl - 1- Fc (20 μ g/ml) ( c ) ァクチ ビン (15 ng/ml) 、 (d ) ァクチビン (15 ng/ml) と FS卜 1 - Fc (20 μ g/ml) 、 ( e ) GDF-8 (15 ng/ml) 、 および (f ) GDF-8 (15 ng/ml) と FSl - Fc (20 μ g/ml) それぞれの条件で、 FSl- 1-Fcのァクチビンおよび GDF- 8機能阻害活性の検 定を行った。 図 2 2に示すように、 FSl- 1- Fcは GDF- 8による標準化ルシフェラー ゼ活性の上昇をほぼ完全に阻害したのに対して、 ァクチビンによる標準化ルシフ エラーゼ活性の上昇はほとんど阻害しなかった。
以上から、 フオリスタチン変異体ポリペプチドとィムノグロプリンの定常領域 の融合蛋白質である FSl- Fcおよび FS1 - 1- Fcは、 FS1および FS1 - 1と同様に、 選択的 に GDF-8の活性を阻害できることが確認された。 産業上の利用可能性
本発明により、 ァクチビンの活性の阻害と比較して、 GDF- 8の活性を選択的に 阻害するフォリスタチン変異体ポリべプチドが提供される。 該フォリスタチン変 異体ポリペプチドは、 骨格筋の增加および脂肪の減少に用いることができる。 ま た、 筋萎縮症状の治療、 悪液質、 筋萎縮を伴う疾患、 Π型糖尿病、 肥満および後 天性免疫不全症候群の治療に用いることができる。

請求の範囲
(b) 式(I) : Cy s— (X1) a-Cy s - (X2) b— Cy S— (X3) C-C y s- (X4) d-Cy s - (X5) e-Cy s - (X6) 「Cy s _ (X7) g - Cy s - (X8) h— Cy s— (X9) i— Cy s (式中、 X1、 X2、 X3、 X4、 X5、 X6、 X7、 X8および X9は、 それぞれ独立して同一または異なった、 シス ティン以外の天然に存在するアミノ酸残基を表し、 aは 2〜6、 bは 3〜7、 c は 7〜; L l、 dは 0〜4、 eは 1〜6、 f および gは 8〜12、 hは 4〜8、 i は 11〜 15の整数をそれぞれ表す。 ) で表されるアミノ酸配列であって、 かつ 配列番号 2または配列番号 29のァミノ酸配列と 50%以上の相同性を有する配列 を含まず、 かつ
(c) ァクチビンの活性の阻害と比較して、 GDF-8の活性を選択的に阻害する、 ことを特徴とするフォリスタチン変異体ポリぺプチド。
2. フォリスタチンドメイン Iを含み、 フォリスタチンドメイン IIを欠失し た請求項 1に記載のフォリスタチン変異体ポリぺプチド。
3. さらにフォリスタチンドメイン IIIを含まない、 請求項 1または 2に記 載のフォリスタチン変異体ポリペプチド。
4. 2つ以上のフオリスタチンドメイン Iを含む、 請求項 1から 3のいずれ か 1項に記載のブオリスタチン変異体ポリぺプチド。
5. さらに N末端ドメインを含む、 請求項 1から 4のいずれか 1項に記載の フオリスタチン 異体ポリぺプチド。
6. FSI、 FSI— FSI、 FSI_FSI_FSIII、 FSN—FSI、 FSN—FSI— FSI、 又は FSN. -FSI-FSI-FSIII (ここで FSIはブオリスタチンドメイン Iを示し、 FSIIIはフォ リスタチンドメイン ΙΠを示し、 FSNは N末端ド インを示し、 各ドメイン間は直 接またはリンカーポリペプチドで連結している) から選択されるいずれかのドメ ィン構造を含む請求項 1または 2に記載のブオリスタチン変異体ポリぺプチド。
7 . フオリスタチンドメィン Iが配列番号 1のァミノ酸配列からなるポリべ プチドである請求項 1カゝら 6のいずれか 1項に記載のフォリスタチン変異体ポリ ぺプチド。
8 . 配列番号 4の 30〜166番目めアミノ酸配列を含む、 請求項 7に記載のフ オリスタチン変異体ポリぺプチド。
9 . フォリスタチンのドメインのアミノ酸配列の N末端または C末端に他の ポリぺプチドが融合している、 請求項 1力、ら 8の何れか 1項に記載のフォリスタ チン変異体ポリペプチド。
1 0 . 配列番号 6、 8または 1 0のァミノ酸配列からなるポリぺプチド。
1 1 . 請求項 1から 1 0のいずれか 1項に記載のポリペプチドをコードする ポリヌクレオチド。
1 2 . 配列番号 5、 7または 9の塩基配列を含む請求項 1 1に記載のポリヌ クレオチド。
1 3 . 請求項 1 1または 1 2に記載のポリヌクレオチドを含むベクター。
1 4 . 請求項 1 3に記載のベクターを宿主細胞に導入して得られる形質転換 細胞。
1 5 . 請求項 1 4に記載の形質転換細胞を培地中で培養し、 該細胞で発現し た請求項 1カゝら 1 0のいずれか 1項に記載のポリぺプチドを培地または該細胞中 に蓄積 せ、 培地または該細胞から該ポリぺプチドを採取することを特徴とす る、 該ポリペプチドの製造方法。
1 6 . 請求項 1から 1 0のいずれか 1項に記載のポリぺプチド、 請求項 1 1 または 1 2に記載のポリヌクレオチドまたは請求項 1 3に記載のベタターを含有 するァクチビンは阻害しなレ、GDF - 8選択的な阻害剤。
1 7 . 請求項 1から 1 0のいずれか 1項に記載のポリぺプチド、 請求項 1 1 または 1 2に記載のポリヌクレオチドまたは請 項 1 3に記載のベクターを含有 する医薬組成物。

2 2 . 筋萎縮を伴う疾患が、 遺伝性の筋ジストロフィー、 筋萎縮性側索硬化 症、 脊髄小脳変性症またはパーキンソン病である請求項 2 1に記載の治療剤また は予防剤。
2 3 . 請求項 1 1または 1 2に記載のポリヌクレオチドを含む外来遺伝子を 導入したトランスジエニック非ヒト哺乳動物。
2 4 . 非ヒト動物に請求項 1から 1 0のいずれか 1項に記載のポリべプチ ド、 請求項 1 1または 1 2に記載のポリヌクレオチドまたは請求項 1 3に記載の ベクターを投与して、 該非ヒト動物の筋肉を増加させる方法。
2 5 . 非ヒト動物に請求項 1から 1 0のいずれか 1項に記載のポリべプチ ド、 請求項 1 1または 1 2に記載のポリヌクレオチドまたは請求項 1 3に記載の ― ベクターを投与して、 該非ヒト動物の脂肪を減少させる方法。
2 6 . レポータープラスミドを有する細胞をァクチビン、 GDF-8、 ァクチビ ンと被験物質、 又は GDF- 8と被験物質の存在下で培養し、 レポーターアツセィを 行うことを含む、 被験物質のァクチビンおょぴ GDF - 8機能阻害活性を検定する方 法。
27. レポーターとしてルシフェラーゼを使用する、 請求項 26に記載の方 法。
