遺伝物質を支持体に固定し、細胞に導入するための組成物および方法 技術分野
[0001] 本発明は、遺伝物質の固定技術に関する。より詳細には、本発明は、固相支持体 に生体分子などの物質を固定するための方法およびそれに関するキット、組成物な どに関する。
背景技術
[0002] マイクロタイタープレートなどの支持体に DNAなどの遺伝物質を固定して種々の分 析を行う試みが広く行われてレ、る。
[0003] 従来支持体に生体分子を固定するやり方としては、例えば、共有結合を介する方 法、疎水性相互作用、吸着、シラン化などの固相の表面処理などの方法がある。
[0004] 例えば、 Nikiforov et al.は、陽イオン性界面活性剤を媒体としたオリゴマーの ポリスチレンゥエルへの結合を利用する方法を開示した (非特許文献 1)。
[0005] Nagata et al.は、未知量のクローニング DNAを 0· 1M MgClの存在下でマ

イクロタイターゥエルに結合させる技術を開示した(非特許文献 2)。 Dahlen, P. et al.は、マイクロタイターゥエル中でのサンドイッチハイブリダィゼーシヨンにおいて、 クローニング捕捉 DNAをゥエルに吸着させることを記載してレ、る(非特許文献 3)。 N aClを媒体としたオリゴマーのポリスチレンゥエルへの結合が Nikiforov Nikiforov et al. (非特許文献 4)により検討されている。
[0006] しかし、これらの方法は、固定工程において、一定の工数を必要とするカ 特殊な 処理を必要とすることから、より簡便な生体分子などの物質の固定方法への需要は 大いにある。
[0007] また、固定後に徐放することが望ましい場合も多レ、。そのような場合として、例えば、 マイクロタイタープレートなどのプレート上でいったん生体分子を固定した後、アツセ ィにおいてアツセィ溶液にそのプレートを浸した後にその固定された生体分子が徐 々に放出することが必要な場合などが挙げられる。あるいは、細胞などの生体に対し て固定後に親和性を保持することが求められる場合もあるが、従来の方法では達成
されていないか、あるいは固定後の生体への親和性が大いに低減することが多ぐ満 足な固定方法が開発されていないのが現状である。
[0008] また、本発明者らは、支持体上に生体物質を固定するために、塩を介在させる方法 を見出したが、当該分野においては、支持体の上で簡易、遺伝物質を支持体に固定 し、細胞に導入することを可能にさせるようなことが求められている。特に、複数の遺 伝物質を任意に支持体 (例えば、アレイなど)上で混合することで、種々のパターンの アレイを作成することができれば、細胞に関する開発スピードは格段に上昇すること が期待されることから、そのような技術の登場が待ち望まれてレ、る。

発明の開示
発明が解決しょうとする課題
[0009] 上記現状に鑑み、本発明では、遺伝物質を支持体に固定し、細胞に導入する方法 を開発することを課題の一つとした。本発明はまた、支持体への固定後に、特定の溶 液に溶解したときに固定された生体分子などの物質が徐放され、あるいは細胞など の生体への親和性が実質的に保持または改善されるような効果を達成する固定方 法を開発することも課題の一つとした。特に、複数の遺伝物質を支持体上で任意の パターンで配置し、その後細胞に導入することができるような技術を開発することを目 的とした。
課題を解決するための手段
[0010] 上記課題は、 DNA、 RNA、ポリペプチドなどの遺伝物質を、それとは反対の電荷 を有する物質と、塩 (有機塩、無機塩 (例えば、培地に含まれるリン酸カルシウムなど の無機塩))とを、支持体上で混合することによって、予想外に、簡便に支持体に固 定され、その後細胞に取り込まれ、細胞において発現することを本発明者らが見いだ したことによって解決された。本発明者らはまた、上記構成によって、固定された遺伝
物質が、固定された後、アツセィ溶液などの溶液に支持体 (例えば、プレート)を浸す と徐放されることを予想外に見いだした。本発明者らはまた、上記構成を用いると、生 体 (例えば、組織、細胞)に対する親和性が保持または改善されることを見いだした。 このように、本発明は、以下を提供する。
1.遺伝物質を支持体に固定し、細胞に導入するためのキットであって、
a)遺伝物質と、
b)上記遺伝物質とは反対の荷電を有する荷電性物質と、
c)塩と、
を含み、
上記遺伝物質と上記荷電性物質と塩とを固相支持体上で混合することを指示する 指示書を備える、キット。
2.上記遺伝物質と、荷電性物質と、塩とからなる群より選択される少なくとも 2つの物 質は、混合されて提供される、項目 1に記載のキット。
3.上記遺伝物質は、少なくとも 2種類存在する、項目 1に記載のキット。
4.上記遺伝子物質は、 DNA、 RNA、 PNAおよびそれらの組み合わせからなる群よ り選択される、項目 1に記載のキット。
5.上記荷電性物質は、細胞親和性を有する、項目 1に記載のキット。
6.上記荷電性物質は、生体分子を含む、項目 1に記載のキット。

[0014] 遺伝物質を、支持体 (例えば、アレイ、プレートなど)上で任意のパターンで混合し、 細胞に導入し、遺伝子発現させることが可能となった。従って、複数の遺伝子を同時
に細胞に導入した場合などの細胞における影響を調べることが可能となった。細胞 は、医療技術、医薬開発などにおいて重要なツールであり、そのツールを種々開発 できることから、その産業上における有用性も評価されるべきである。
図面の簡単な説明
[図 1]図 1は、実施例 4における代表的な結果例である。図 1は、複合体化していない DNA (上)と、正に荷電した物質と複合体化させた DNA (下、 NP10処理)とを対比 した結果を示す。固相に対して複合体を添加した直後(0分)ならびに、添加の 1分後 、 5分後および 10分後に細胞含有液をカ卩えた後のトランスフエクシヨンの様子を示す
[図 2]図 2は、実施例 4のヒト間葉系幹細胞でのプリント(重ね打ち)の場合の結果例を 示す。固相基板上に pEGFP-Nlベクターをプリントし、乾燥後、 LipofectAMINE2000 を同一座標にプリントし乾燥させた。その後、フイブロネクチン溶液を同一座標に重 ねてプリントし乾燥させた。これに、間葉系幹細胞を播種し、 2日間培養を行った後に 、蛍光イメージスキャナにて画像を取得した。
[図 3]図 3には、実施例 5における HeLa— K細胞にトランスフエタトした後 3日目の遺伝 子発現の様子を示す。固相基板上に pEGFP-Nlと pDsRed2-l, pEGFP-Nlと pPUR6iGFP272ベクターをそれぞれプリントし、乾燥後、 LipofectAMINE2000を同一 座標にプリントし乾燥させた。その後、フイブロネクチン溶液を同一座標に重ねてプリ ントし乾燥させた。これに、 HeLa_K細胞を播種し、 3日間培養を行った後に、蛍光ィメ ージスキャナにて画像を取得した。
[図 4]図 4には、実施例 5における HeLa— K細胞にトランスフエタトした後 6日目の遺伝 子発現の様子を示す。固相基板上に pEGFP-Nlと pDsRed2-l, pEGFP-Nlと pPUR6iGFP272ベクターをそれぞれプリントし、乾燥後、 LipofectAMINE2000を同一 座標にプリントし乾燥させた。その後、フイブロネクチン溶液を同一座標に重ねてプリ ントし乾燥させた。これに、 HeLa-K細胞を播種し、 6日間培養を行った後に、蛍光ィメ ージスキャナにて画像を取得した。

[図 7B]図 7Aおよび図 7Bは、液相トランスフヱクシヨンと SPTAの比較を示す結果で ある。図 7Bは、図 7Aの示すデータに対応する、 EGFPを発現する細胞の蛍光画像 である。図 7Bにおいて、白丸で示された領域は、プラスミド DNAを固定化した領域 を示す。プラスミド DNAを固定化した領域以外の領域では、細胞が固相に固定化さ れたにもかかわらず、 EGFPを発現する細胞は観察されなかった。 白棒は、 500 x m を示す。従来の液相トランスフエクシヨンと、 SPTAとの間の比較が示される。上の液 相トランスフエクシヨンと固相トランスフエクシヨン(下)とでは、 Α)測定された蛍光/ m m2の 5種の細胞株について、図 7Aに対応する。 EGFP発現細胞の蛍光写真であり 、白色の円状のものは、プラスミド DNAプリント領域に当たる。この領域の外の領域 の細胞は、 EGFPを発現している力 プリント領域以外の領域にも蛍光を発していな い細胞が付着している。

scrambleRNAをもちいた。その後、、 LipofectAMINE2000を各遺伝子プリント同一座 標にプリントし乾燥させた。その後、フイブロネクチン溶液を同一座標に重ねてプリン トし乾燥させた。これに、 HeLa_K細胞を播種し、 2日間培養を行った後に、蛍光ィメー ジスキャナにて画像を取得した。
[図 10]図 10は、実施例 7における RNAiトランスフエクシヨンアレイのトランスフエクショ ン結果を細胞ごとに示す。各レポーターの蛍光輝度を画像解析によって定量ィ匕した 後に、ネガティブコントロールであるスクランブル RNAをプリントした各レポーターの輝 度に対する比率を算出した。これを、各レポーター ·各細胞に対してすベて行った結 果を示している。
[図 11]図 11は、実施例 8における RNAiトランスフエクションアレイのトランスフエクショ ン結果を示す。固相基板上に表 3に示す各レポーター遺伝子発現ユニット PCR断片 を一遺伝子あたり 4スポットになるようプリントし、乾燥後、各転写因子に対しての siRNA (28種類)を対応したレポーター遺伝子がプリントされている座標上にプリントし 、乾燥させた。また、コントロールとして、 EGFPに対する siRNAを用レヽ、ネガティブコン トロールとしては、スクランブル RNAをもちいた。その後、、 LipofectAMINE2000を各遺 伝子プリント同一座標にプリントし乾燥させた。その後、フイブロネクチン溶液を同一 座標に重ねてプリントし乾燥させた。これに、各細胞を播種し、 2日間培養を行った後 に、蛍光イメージスキャナにて画像を取得した。

[0017] 以下、本発明を説明する。本明細書の全体にわたり、単数形の表現は、特に言及 しない限り、その複数形の概念をも含むことが理解されるべきである。また、本明細書 において使用される用語は、特に言及しない限り、当該分野で通常用いられる意味 で用いられることが理解されるべきである。

以下に本明細書において特に使用される用語の定義を列挙する。
[0019] 本明細書において使用される用語「物質」は、当該分野において用いられる最も広 義な意味と同じ意味で含まれ、正または負に荷電することができるものを含む。
[0020] 本明細書において「遺伝物質」とは、遺伝情報を担う任意の物質をいう。そのような 物質としては、例えば、 DNA、 RNA、 PNA、タンパク質などが挙げられるがそれらに 限定されない。 DNAは、 cDNA、ゲノム DNA,人工 DNA、 RNAiなど任意の形態 のものであり得る。 RNAは、 mRNA、 tRNA、 rRNA、 RNAiなどであってもよい。

そのような物質には、正に荷電した物質および負に荷電した物質などが挙げられる がそれらに限定されない。

電した物質は、複合体を形成することができる物質であることが有利である。そのよう な複合体を形成することができる正に荷電した物質としては、例えば、カチオン性ポリ マーのようにある程度の分子量を有する物質、あるいは、カチオン性脂質のように特 定の溶媒 (例えば、水、水溶液など)中においてある程度溶解せずに残存することが できる物質などが挙げられるがそれらに限定されない。そのような好ましい正に荷電 した物質としては、例えば、ポリエチレンィミン、ポリ Lリシン、合成ポリペプチドもしくは それらの誘導体などが挙げられるがそれらに限定されなレ、。あるいは、正に荷電した 物質としては、ヒストン、合成ポリペプチドなどのような生体分子が挙げられるがそれら に限定されない。そのような好ましい正に荷電した物質の種類は、複合体を形成する パートナーである負に荷電した物質の種類に応じて変動する。好ましい複合体形成 パートナーを選択することは、当業者には容易であり、そのような選択は、当該分野 において周知の技術を用いて行うことができる。そのような好ましい複合体形成パー トナーの選択においては、種々のパラメータを考慮することができる。そのようなパラメ ータとしては、例えば、電荷、分子量、疎水性、親水性、置換基の性質、 pH、温度、 塩濃度、圧力などの種々の物理的パラメータ、化学的パラメータなどが挙げられるが それらに限定されない。

のインクジェット装置が使用される。液体チャンバ一に接続された圧電素子に電圧を 加えることにより、圧電素子の体積の変化によってチャンバ一内の液体力 チャンバ 一に接続された、キヤビラリ一から液滴となって射出される。射出される液滴の大きさ は、キヤピラリーの径、圧電素子の体積変化量、液体の物理的性質によって決定され るが、一般には、直径が 30 μ πι程度である。圧電素子を用いたインクジェット装置は、 このような液滴を ΙΟΚΗζ程度の周期で射出することができる。このようなインクジェット 装置を使った DNAアレイ製造装置は、インクジェット装置と DNAアレイ基板とを相対運 動させることにより、 DNAアレイ上の所望のスポットに所望の液滴を滴下することがで きる。インクジェット装置を使った DNAアレイ製造装置には、大きくわけて 2種類ある。 1 つはただ 1台のインクジェット装置を用いた DNAアレイ製造装置であり、もう 1つはマル チヘッドのインクジェット装置を用いた装置である。ただ 1台のインクジェット装置を用 レ、た DNAアレイ製造装置は、オリゴマー末端の保護基を除去する試薬を所望のスポ ットに滴下する構成になっている。所望の塩基を導入したレ、スポットの保護基を、この インクジェット装置を用いて除去して活性な状態にした後、 DNAアレイ全体に所望の 塩基の結合反応操作を実施する。この際、インクジェット装置からの試薬の滴下によ つて、末端が活性化したオリゴマーを持つスポットのみに所望の塩基が結合する。こ の後、新たに付加した塩基の末端を保護する操作を行う。次に、保護基を除去する スポットを変更してこの操作を所望のヌクレオチド配列が得られるまで繰返す。一方、 マルチヘッドのインクジェット装置を用いた DNAアレイ製造装置は、各塩基を含む試 薬毎にインクジェット装置を用意することによって、各スポット毎に所望の塩基を直接 結合させることができる構成になっており、前述した 1台のインクジェット装置を用いた DNAアレイ製造装置よりも高レ、スループットが得られる。あらかじめ合成したオリゴヌク レオチドを基板に固定化させる方法のうち、メカニカルマイクロスポッティング技術は、 ステンレス製のピンの先端についたオリゴヌクレオチドを含む液体を機械的に基板上 に押し付けて固定化していく技術である。この方法で得られるスポットは、 50 300 μ m程度になる。マイクロスポッティング後には、 UV光による固定化等の後処理が行わ れる。
本発明の方法に使用される吹き付ける工程としては、インクジェット方式 (バブルジ
エツト(登録商標)方式を含む)、 PIN法などが挙げられる。好ましくは、バブルジェット

図 1一 2にトランスフエクシヨンの結果の一部を示す。図 1は、最初から混合した場合 (上にコントロール、下に混合物)を示す。固相に対して複合体を添加した直後(0分) ならびに、添加の 1分後、 5分後および 10分後に細胞含有液をカ卩えた後の複合体の 溶出の様子を示す。図 2は、実施例 2の手順に従ってプリントした支持体における、ヒ ト間葉系幹細胞におけるトランスフエクシヨン効果を示す。示されるように、プリントを連 続的に行った (重ね打ち)の場合でも、混合物と同様の遺伝物質の細胞導入効果が 示された。
[0162] この結果から明らかなように、混合して塩をカ卩えてから支持体に固定された系に加 えて、別個にプリントした場合でも、固定がなされ、 DNAが徐放されることも明らかに なった。このような効果は、従来の技術では明らかでなかったことであり、これにより、 プレート、チップなどで任意の遺伝子を任意の組み合わせで細胞内に導入して観察 することができるという技術が初めて確立されたことになる。また、細胞導入の効果に ついても、重ね打ちの順序に関係ないことが明らかになった。
[0163] また、このほかの本発明の塩を固定に使用した場合でも、トランスフエクシヨンが起こ つていたことが判明した。これは、トランスフエクシヨンが起こるために必要な DNAの 細胞への移入が、いずれの例でも起こっていたことを示す。従って、本発明の支持体 は、細胞親和性および細胞導入効果を維持または改善していたことになる。また、実 施例 1一 2における放出のデータに加え、トランスフエクシヨンが起こっていたことは固 定された遺伝物質が活性を保持していたことを示すことになる。

用した系では、単塩を用いた系よりもトランスフエクシヨン効率が高力 たことから、固 定の際に細胞に適合する塩類の組み合わせを用いることが、細胞親和性および遺 伝物質の細胞導入効果の維持または改善にも適切であることがわかる。

多様な種類の細胞に分化するヒト間葉系幹細胞 (hMSC)の能力は、組織再生およ び組織復活を標的化する研究にとって特に興味深レ、ものになっている。特に、これら の細胞の形質転換体についての解析は、 hMSCの多能性を制御する因子を解明す る上で、関心が高まっている。 hMSCの研究における重大な障害は、所望の遺伝物 質を用いたトランスフエクシヨンが不可能 (または、非常に困難)な点にある。
[0179] これを達成するための従来の方法は、ウィルスベクターまたはエレクト口ポレーショ ンのいずれかの技術を含む。本発明者らが開発した複合体一塩という系を用いること により、種々の細胞株(hMSCを含む)に対して高レ、トランスフエクシヨン効率ならびに 密集したアレイ中での空間的な局在の獲得を可能にする固相系トランスフエクシヨン が達成された。固相系トランスフエクシヨンの概略を、図 5に示す。

[0185] これらの目的の達成を明確に確立するために、上述のように本発明者らは、 5種類 の異なる細胞株(HEK293、 HeLa、 NIH3T3、 HepG2および hMSC)を、本発明 者らの方法論(固相系でのトランスフエクシヨン)および従来の液相系トランスフエクシ ヨンの両方を用いて一連のトランスフエクシヨン条件下で研究した。 SPTAの場合、相 互夾雑を評価するために、本発明者らは、チェック模様のパターンでガラス支持体上 にプリントした赤色蛍光タンパク質 (RFP)および緑色蛍光タンパク質 (GFP)を使用 し、一方、従来の液相系トランスフエクシヨンを含む実験の場合 (ここで、本来、トラン スフェクト細胞の自発的な空間的分離は達成され得ない)、本発明者らは、 GFPを使 用した。レ、くつかのトランスフエクシヨン試薬を評価した: 4つの液体トランスフエクショ ン試薬(Eff ectene、 TransFastTM, TfxTM_20、 LopofectAMINE 2000)、 2 つのポリアミン(SuperFect、 PolyFect)、ならびに 2つの型のポリイミン ietPEI ( X 4)および ExGen 500)。

[0187] し力、し、細胞をトランスフエタトするのが困難な場合(例えば、 hMSCおよび HepG2 )において SPTA方法論に最適化した条件を用いることによって、本発明者らは、細 胞の特徴を維持しながら、トランスフヱクシヨン効率が 40倍まで増加したことを観察し た(上述のプロトコルおよび図 4C一 Dを参照のこと)。結果を、図 7Bに示す。
[0188] hMSCの特定の場合(図 8)、最良条件は、ポリエチレンィミン(PEI)トランスフヱク シヨン試薬の使用を含んだ。予想したように、高レ、トランスフヱクシヨン効率を実現する ための重要な因子は、ポリマー内の窒素原子(N)の数とプラスミド DNA内のリン酸残 基(P)の数との間の電荷バランス(NZP比率)、ならびに DNA濃度である。一般的 に、 NZP比率および濃度における増大は、トランスフエクシヨン効率の増大を生じる 。並行して、本発明者らは、 hMSCの溶液トランスフエクシヨン実験における高い DN Aおよび高い N/P比率の場合に、細胞生存率の有意な低下を観察した。これら 2つ の拮抗因子に起因して、 hMSCの液相系トランスフエクシヨンの効率は、かなり悪い 非常に低い細胞生存率 (N/P比率 > 10で観察された)であった。しかし、 SPTAプ 口トコノレは、細胞生存率にも細胞形態にも有意に影響を与えることなぐ非常に高い( 固体支持体に固定された) N/P比率および DNA濃度を許容し (おそらぐ細胞膜に 対する固体支持体の安定化効果に起因する)、従って、このことがおそらぐトランス フエクシヨン効率の劇的な改善の原因となっている。 SPTAの場合、 10の N/P比率 が最適であることが見出され、細胞毒性を最小化しながら十分なトランスフエクシヨン レベルを提供する。 SPTAプロトコルにおいて観察されたトランスフエクシヨン効率の 増大に関するさらなる理由は、高い局所的な DNA濃度 Zトランスフヱクシヨン試薬濃 度(これは、液相系トランスフヱクシヨン実験において使用される場合は細胞死を生成 する)の達成である。
[0189] チップ上での高レ、トランスフエクシヨン効率の達成のための重要な点は、使用される コーティングに使用する物質の種類である。ガラスチップを使用した際のコーティング 剤として、 PLLがトランスフエクシヨン効率および相互夾雑の両方に関して、最良の結
果を提供することを発見した。フイブロネクチンコーティングしない場合、少数のトラン スフェクト体を観察した (他のすべての実験条件は一定に保った)。完全に確立した わけではないが、フイブロネクチンの役割はおそらぐ細胞接着プロセスを加速し (デ ータは示していない)、ゆえに、表面を離れた DNA拡散が可能になる時間を制限す るということである。
[0190] 低い相互夾雑: SPTAプロトコルで観察されたより高レ、トランスフエクシヨン効率は別 として、本技術の重要な利点は、別個に分離された細胞アレイの実現であり、その各 位置では、選択した遺伝子が発現する。本発明者らは、フイブロネクチンでコーティン グしたガラス表面上に、 JetPEI (「実験プロトコル」を参照のこと)およびフイブロネクチ ンと混合した 2つの異なるレポーター遺伝子(RFPおよび GFP)をプリントした。得ら れたトランスフエクシヨンチップを適切な細胞培養に提供した。最良であると見出され た実験条件下において、発現された GFPおよび RFPは、それぞれの cDNAがスポッ トされた領域に局在した。相互夾雑はほとんど観察されなかった(図 6)。しかし、フィ ブロネクチンまたは PLLの非存在下において、相互夾雑は重大な障害であり、そし てトランスフエクシヨン効率は、有意に低かった(図 7)。このことは、細胞接着および支 持体表面から離れて拡散するプラスミド DNAの相対的な割合が、高いトランスフヱク シヨン効率および高い相互夾雑の両方に対して重要な因子であるという仮説を立証 する。
[0191] 相互夾雑のさらなる原因は、固体支持体上のトランスフエクシヨン細胞の移動性で あり得る。本発明者らは、数個の支持体上での細胞接着速度(図 6C)およびプラスミ ド DNAの拡散速度の両方を測定した。その結果は、最適条件下において DNA拡 散はほとんど生じなかった。しかし、高い相互夾雑条件下では、細胞接着が完了する までの時間に、相当の量のプラスミド DNAが固相表面から拡散して枯渴した。
[0192] この確立された技術は、経済的な高スループットの遺伝子機能スクリーニングの状 況において特に重要である。実際に、必要とされる少量のトランスフエクシヨン試薬お よび DNA、ならびに全プロセス(プラスミドの単離から検出まで)を自動化する可能性 は、上記の方法の有用性を増大する。
[0193] 結論として、本発明者らは、複合体一塩を用いた系で、 hMSCトランスフヱクシヨンァ
レイを好首尾に実現した。このことは、多能性幹細胞の分化を制御する遺伝子機構 の解明など、固相系トランスフエクシヨンを利用した種々の研究における高スループッ ト研究を可能にすることになる。固相系トランスフエクシヨンの詳細な機構ならびに高 スループットのリアルタイム遺伝子発現モニタリングに対するこの技術の使用に関す る方法論は種々の目的に応用可能であることが明らかになった。
