Description

口服配製物內可含有各種類型的酵素以作為治療劑。例如,可含有酵素以水解口服多糖類或抑制細菌的生長。更明確而言,例如,已報告口服配製物內可利用如變構酶(mutanase)、右旋糖苷酶(dextranase)、木瓜酵素(papain)及1,3-葡聚糖酶移除牙垢沈澱物。
通常,天然的酵素易受到環境變化如pH、溫度和離子濃度的破壞,而因此需採取保護酵素的方法以維持擬用於口腔保健產品內之酵素的效力。為改善酵素的性能,曾建議使用酵素穩定劑,例如螯合劑和抗氧化劑。此類酵素穩定劑的實施例為EDTA、葡萄糖酸鈉、亞硫酸鈉、金屬沒食子酸(gallates)、錫酸鈉及抗壞血酸。
然而,在酵素穩定劑中含亞硫酸離子的亞硫酸氫鹽物質對某些人具有毒性和/或經常使口腔保健產品改變口感的性質。因此,在進一步處理組成物和在消費者使用之前最好為移除或減少酵素組成物內的亞硫酸氫鹽物質。雖然可經由習知技術如鹼萃取和藉由氧化劑的中和作用移除亞硫酸氫鹽物質,但是這些技術通常對口腔組成物內酵素的催化活性產生不利影響。因此,亟需提出一種可移除亞硫酸氫鹽物質而對組成物之酵素活性不造成不良影響的不同方法。迄今,仍無在中性、非氧化條件下從酵素組成物移除亞硫酸氫鹽物質的簡易方法。
根據本發明,提供一種從含亞硫酸氫鹽物質和一或多種酵素的組成物移除亞硫酸氫鹽物質的方法。此方法包括使該組成物接觸含至少一醛功能基的化合物以形成醛-亞硫酸氫鹽複合物;該醛-亞硫酸氫鹽複合物可從組成物中被移除。
在一具體實施例中,該方法包括使該組成物接觸含至少一醛功能基如醛糖的化合物以形成醛-亞硫酸氫鹽複合物然後從該組成物分離該醛-亞硫酸氫鹽複合物,其中該酵素係選自由木瓜酵素、鳳梨酵素(bromelain)、絲胺酸蛋白酵素、胰凝乳蛋白酶(chymotrypsin)、無花果蛋白酶(ficin)、葡萄糖氧化酶、半乳糖氧化酶、乳糖過氧化酶、乳鐵蛋白(lactoferrin)、溶菌酶、脂肪分解酶、鹼性蛋白酶(alcalase)、糖解酶、葡萄糖澱粉酶、右旋糖苷酶、變位酶(mutase)、單寧酶(tannase)及脂肪酶所構成的群組。
在另一具體實施例中,提供一種從含亞硫酸氫鹽物質和酵素的組成物移除亞硫酸鹽物質的方法,其中該方法包括使該組成物接觸具有至少一醛官能基的基質以形成醛-亞硫酸氫複合物然後從該組成物分離該結合基質的亞硫酸氫鹽。
在又另一具體實施例中,進一步提供一種實質上全無亞硫酸氫鹽物質的組成物。獲得該酵素組成物的方法係藉由使該組成物接觸含至少一醛官能基的化合物以形成醛-亞硫酸氫鹽複合物然後從該組成物分離該醛-亞硫酸氫鹽複合物。
本發明係關於從含一或多種酵素的組成物移除亞硫酸氫鹽物質的方法。更明確而言,本發明係關於一種移除亞硫酸氫鹽離子的方法,其藉由使酵素組成物接觸含至少一醛官能基的化合物以形成醛-亞硫酸氫鹽複合物然後從該組成物分離該亞硫酸氫鹽複合物。同樣,本發明係關於一種無亞硫酸氫鹽酵素組成物,其製備係藉由使酵素組成物接觸含至少一醛官能基的化合物以形成醛-亞硫酸氫鹽複合物然後從該組成物分離該亞硫酸氫鹽複合物。
本發明提供一種從酵素組成物移除亞硫酸氫鹽物質的方法,其係利用具有至少一可與亞硫酸氫鹽物質形成複合物之醛官能基的化合物然後從該組成物分離該醛-亞硫酸氫鹽複合物。此方法較佳為在實質上中性、非氧化條件下進行,因而可獲得含實質上不降解之酵素的實質上無亞硫酸氫鹽組成物。
此處”降解”一詞意指相對未經萃取亞硫酸氫鹽物質之酵素組成物而言降低酵素活性。根據本發明,在亞硫酸氫鹽與醛的接觸步驟之後以及其後分離亞硫酸氫鹽-醛複合物的步驟或上述二者步驟之後較佳為保留超過50%、超過80%或超過90%的酵素活性。
“亞硫酸氫鹽”一詞意指含化學鍵相結合或化學複合物中一成分之亞硫酸氫鹽離子的化合物,例如亞硫酸鈉或其他亞硫酸鹽。
為了從一酵素組成物移除亞硫酸氫鹽,本發明利用含至少一醛官能基的化合物。”醛類”被定義為一種具有R-CHO構造的羰基化合物,其中該R基可為芳族或脂族基。為獲得含實質上不降解酵素之實質上無亞硫酸氫鹽的組成物,可選擇在中性、非氧化條件下與亞硫酸氫鹽物質形成複合物的醛。本發明之醛可為具有低分子量的單官能基醛,例如低於約1000道耳頓。該醛可為一醛糖。
實質上無亞硫酸氫鹽之組成物可藉由從組成物分離亞硫酸氫鹽-醛複合物而獲得。此類分離方法可藉由技術中習知的分離方法如化學或物理方法或結合化學和物理方法而完成。可用於本發明之分離方法的實施例包括,但不侷限於,過濾、擴散差、管柱分離和磁珠分離法,以及藉由定位法或隔離法。
此外,該分離法可藉由使組成物連接至附著醛的物體或物件而形成複合物,然後移除已附著醛-亞硫酸氫鹽複合物的物體或物件。或者,為獲得具有實質上不降解酵素的組成物,在亞硫酸氫鹽物質和酵素組成物的全部接觸步驟中可維持實質上中性、非氧化的條件然後分離該亞硫酸氫鹽-醛複合物。例如,可在維持離子強度之下將酵素溶液的pH維持在約5至約8的範圍內。
根據本發明之移除亞硫酸氫鹽的方法可被應用於含任何類型酵素的組成物。本發明可有效地被用於處理作為接觸人類或動物之皮膚或黏膜的酵素組成物。例如,其可為口腔保健產品或個人保健產品內的任何一種酵素組成物。此類酵素的實施例包括,但不侷限於,糖解酶如葡萄糖澱粉酶;萃取自天然果實的產品如蛋白酶;糖解酶如α-澱粉酶、β-澱粉酶和單寧酶,以及脂肪酶如植物性脂肪酶、胃脂肪酶和胰脂肪酶。準備藉由本發明處理之組成物內的酵素較佳為植物性來源。可利用本發明之方法的酵素係選自由α-和β-澱粉酶;右旋糖苷酶;變構酶;天然酵素如木瓜酵素(來自木瓜)和鳳梨酵素(來自鳳梨);絲胺酸蛋白酵素如胰凝乳蛋白酶;無花果蛋白酶;鹼性蛋白酶;溶菌酶;果膠酶(pectinase)和葡聚糖酶所構成的群組。
根據本發明一具體實施例中,提供一種方法其利用含至少一醛官能基的基質而可更輕易地移除亞硫酸氫鹽-醛複合物。利用含至少一醛官能基之基質的方法可在與不利用基質下移除亞硫酸氫鹽之方法的相同條件下進行。具有醛官能基的基質可為例如選自習知由多孔和非多孔珠、薄膜、細粒和顆粒所構成者。在另一具體實施例中,含至少一醛官能基的基質可被分散於一支撐物上以幫助分離亞硫酸氫鹽-醛複合物。本發明可使用任何種類之技術中習知的支撐物。支撐物的實施例包括,但不侷限於,管柱、容器、過濾器、海綿和凝膠。
在又另一具體實施例中,提供一種藉由本發明移除亞硫酸氫鹽之方法所製備之實質上無亞硫酸氫鹽物質的酵素組成物,其中該酵素組成物的製備係藉由使組成物接觸含至少一醛官能基之化合物而形成醛-亞硫酸氫鹽複合物然後從該組成物分離該醛-亞硫酸氫鹽複合物。由於本發明的酵素組成物係在中性、非氧化條件下所製備,因此實質上不減低組成物內之酵素的活性。該酵素較佳為可維持至少約90%的活性。藉由本發明方法所製造的酵素組成物實質上不含有亞硫酸氫鹽之物質,因此可有效被用於口腔保健或個人保健產品內,例如潔齒劑、皮膚保健產品及頭髮和指甲護理用品。
本發明雖然就特定實施例進行說明,但是熟習本技藝之人士將瞭解在本發明的範圍內可作出各種的改良。
Claims (14)
- 一種從含亞硫酸氫鹽物質和酵素之組成物移除亞硫酸氫鹽物質的方法,該方法包括使該組成物接觸醛糖以形成醛-亞硫酸氫鹽複合物,因而可從該組成物移除該醛-亞硫酸氫鹽複合物。
- 如申請專利範圍第1項之方法,其中該方法進一步包括從該組成物分離該醛-亞硫酸氫鹽複合物。
- 如申請專利範圍第2項之方法,其中該分離法係選自由過濾、擴散差、管柱分離和磁珠分離法所構成的群組。
- 如申請專利範圍第1項之方法,其中該醛糖係在約5至約8的pH範圍與組成物相結合。
- 如申請專利範圍第1項之方法,其中該酵素在形成該醛-亞硫酸氫鹽複合物的步驟之後實質上不被降解。
- 如申請專利範圍第1項之方法,其中該酵素在分離該醛-亞硫酸氫鹽複合物的步驟之後實質上不被降解。
- 如申請專利範圍第1項之方法,其中該酵素係取自植物來源。
- 如申請專利範圍第1項之方法,其中該酵素係選自由木瓜酵素、鳳梨酵素、絲胺酸蛋白酵素、胰凝乳蛋白酶、無花果蛋白酶、葡萄糖氧化酶、半乳糖氧化酶、乳糖過氧化酶、乳鐵蛋白、溶菌酶、脂肪分解酶、鹼性蛋白酶、糖解酶、葡萄糖澱粉酶、右旋糖苷酶、變位酶、單寧酶及脂肪酶所構成的群組。
- 如申請專利範圍第2項之方法,其中在該分離步驟之後超過90%的酵素活性未被降解。
- 一種從含亞硫酸氫鹽和酵素之組成物移除亞硫酸氫鹽的方法,該方法包括使該組成物接觸具有醛糖的基質而形成一醛-亞硫酸氫鹽複合物,因而可從該組成物分離出結合基質的亞硫酸氫鹽。
- 如申請專利範圍第10項之方法,其中該方法進一步包括從該組成物分離該結合基質的亞硫酸氫鹽。
- 如申請專利範圍第10項之方法,其中該基質係選自多孔和非多孔珠、薄膜、細粒和顆粒。
- 如申請專利範圍第10項之方法,其中該基質係分散於一支撐物上。
- 如申請專利範圍第13項之方法,其中該支撐物係選自由管柱、容器、過濾器、海綿和凝膠所構成的群組。
