1340165 玖、發明說明: 【發明所屬之技術領域】 本發明係關於一作爲食品、營養補充劑、化妝用或醫藥 組合物中之成份的油之製造方法,該油係長鏈多元不飽和 必需脂肪酸(LC-PUFA)之來源。 【先前技術】 種含有LC-PUFA(例如花生四烯酸(ARA)、二十二碳六 烯酸(DHA)、二十碳五烯酸(EpA)或雙同γ亞麻酸(DHGLA)) 之油可自一生物基質發酵液中獲得。爲自該生物基質獲得 該油’已使用有機溶劑(例如己烷)或超臨界流體萃取法。一 般而言,該油已可藉由用己烷滲濾經乾燥之生物基質自生 物基質中萃取。 此一使用有機溶劑之萃取法闡釋於,舉例而言,W〇 9737032、W0 9743362或出版物「分散科學與技術雜誌」 中之製備PUFA之生物技術方法」(journal 〇f Dispersi〇n Science and Technology,1〇, 56U579, 1989「Bi〇techn〇1〇gical processes f<pr the production of PUFAs」)中。 該技術具有數項缺點: -在使用熱溶劑萃取或蒸餾溶劑階段,LC_pUFA可能在 與氧接觸時進行降解。 •完全去除油或殘餘生物基質_所含之溶劑需要在高溫 下實施熱處理。 -而且,溶劑(例如己烷)可溶解生物基質的實際上構成雜 質之非三醯基甘油成份。 87409 1^40165 溶劑蒸發後所獲得之粗製油應進—步進行包含脫腰、用 鹼中和、脫色、脫蝶及脫臭的幾個精製階段以至少邹分去 除雜質。此意謂該高度不飽和油係暴露於可促進會与響其 品質之物理化學反應之條件下。舉例而言,脫色劑=成 -種共軛雙鍵系統且藉由與經氧化之甘油醋發生化學反應 形成降解産物。 … -種不使用溶劑萃取生物基質之方法㈣吾人所孰知, 舉例而言可自ΕΡ·Α_1178118中得知。根據該方法,可藉由 製造該生物基質之水懸浮液並藉由離心分離含有所需:之 油相與水相來避免使用溶劑。該水相含有自生物基質獲得 之大量水溶性物質及細胞壁碎片。該方法之—缺點係所獲 得之粗製油受到大量雜質(舉例而言,因存在水所致之蛋白 質殘餘物、極性類脂)之污染。然後應藉由習知用於精製植 物油之方法精製此一粗製油。 本發明之目的係藉由提供一種製造含有一或多種衍生自 生物基質之多元不飽和脂肪酸之安定油之方法來避免先前 支《之缺點’所述脂肪酸以具有高産量之純化態三醯基甘 ’由形式存在且其中該油進行最低限度之降解。 【發明内容] 本發明係關於. lc-pufa(具體而 (DHGLA)、二十二 之安定油之方法。 種用於製造含有呈三酿基甘油形式之 言’花生四烯酸(ARA)、雙同γ亞麻酸 碳六烯酸(DHA)或二十碳五烯酸(ΕΡΑ)) 〇亥方法之特徵在於:壓榨-或多種自微生物(尤其爲真菌 87409 或微澡)培養物所獲得之含有酸ARa、DHGLA、DHA或EPA 之幹態生物基質,以獲得一 一次壓榨油及一餅;及用一吸 附劑處理以此方式獲得之油;以及在經控制之條件下將其 脫臭。 以此方式獲得之油不需要化學精製。使用一吸附劑(舉例 而言一妙酸鹽)處理並在溫和條件下脫臭足以獲得—可用 作營養補充劑之經純化且穩定之油。 根據該方法之一較佳具體實施例, -使該餅與一加至一食品組合物中之載劑油接觸,且壓 榨該載劑油與餅之混合物以獲得二次壓榨油,且 -將έ亥等壓榨油以不同比例混合以獲得一適於具體應用 之LC-PUFA濃度。該混合物然後進行(適當時)物理精製 與脫臭。 較佳地’該油所含LC-PUFA以重量計不超過10%。因此 ,該油在其生産期間對氧化作用更不敏感,而先前技藝之 含有LC-PUFA之油之情況並非如此。 根據本發·明之一主要方面,其一重要有利性質爲可獲得 含有呈三醯基甘油形式之LC-PUFA之新穎油。 根據本發明之另一方面,本發明係關於含有上述油之食 品、化妝用或醫藥産品、營養補充劑或動物飼料。 根據本發明之另一方面,本發明係關於含有自該方法獲 得之生物基質殘餘物之動物飼料,具體而言寵物飼料。 【貫施方式】 油之製造藉由直接壓榨一含有LC_PUFA2生物基質且産 87409 1340165 生一次壓榨油實施。爲增加LC-PUFA之産量,使自直接壓 榨獲得之生物基質餅與一載劑油接觸且壓榨該混合物以獲 得二次壓榨油。然後將該兩種壓榨油以不同比例混合以獲 得一適於特定應用之LC_PlJFA濃度。該混合物然後在適當 條件下進行物理精製,由此可獲得所需之油。 在本發明中,物理精製旨在減少磷脂及游離脂肪酸且可 理解爲意指一在不使用酸及無中和反應之狀況下之脫膠處 理。 舉例而言,吾人觀察到,倘使爲—含Ara之生物基質, 則s玄一次壓榨油已不含無大多數游離脂肪酸及磷脂,因此 ,端視所需純度,其可不需實施脫膠處理。 另一方面,舉例而言,倘使爲含DHA之生物基質,則必 須實施此一脫膠處理以降低,具體而言,磷脂以便獲得所 而純度。 所獲得之油適於應用於食品(具體而言,嬰兒調配物)中或 用作_營養補充劑。其亦可用於化妝用或醫藥產品中。而 且,所獲得·之生物基質殘餘物亦爲該方法之產物,i可在 不經後續處理之狀況下直接升級,舉例而言作爲動物飼料 ’具體而言指寵物飼料。 此一油之製造可藉由簡單塵榨乾生物基質實施。較佳地 :將該載劑油與生物基質之第,榨殘餘物混合,且隨 後稭由壓榨自非脂類固體中分離出該油。 較佳者係降低乾燥生物 且藉此釋放油。在該較 爲增加所獲得LC-PUFA之産量, 基質粒徑,以破碎微生物之細胞壁 87409 -9- 1340165 佳具體實施例中,可降低壓搾餅之粒徑以增加載劑油與生 物基質殘餘物間之接觸表面積。此適於使用各種方法實施 ,舉例而言: -可於載劑油之存在下研磨生物基質餅; -可在與載劑油混合前碾壓生物基質餅; -可於載劑油之存在下高壓處理該生物基質餅,且所獲 得之油然後可藉由壓榨及最終過濾自生物基質餅中分 離; -可使用能降解細胞壁之酶處理生物基質或餅。 因爲該載劑係一種油,故與生物基質餅接觸後所獲得之 油含有極少量之磷脂、游離脂肪酸、色素、聚合物及其他 自生物基質獲得或衍生之非三醯基甘油物質。此意謂根據 本發明之方法可構成一種用於製造含有lc_pufa之穩定純 化油之選擇性方法。無需藉由在本發明之前所用的具侵害 性且麻煩之方法(例如脫膠、中和、脫蠟及脫色階段)來純化 含L C - P U F A之不飽和油。 根據本曼明,壓榨油可進行(適當時)使用一處理劑(例如 ,一矽酸鹽)之精製步驟。該使用處理劑之處理可在^與栽♦ 油接觸期間或於生成壓榨油後(舉例而言,在過濾期間)實^1 。最後,該等油進行一脫臭階段,舉例而言在—相對 溫度下藉由蒸汽蒸餾或分子蒸餾實施。結果係所獲得又低 含有特別少的反式脂肪酸。 .之冰 該方法不使用有機溶劑且,由於該作業係在氮氣層下 在存在天然或添加至載劑油中之生育酚或生* 0且 ’ 月二烯齡之狀 87409 •10· 況下實施’故心職在整個過程中未發生氧化性降解。 除所獲得之油之品質外’本方法之另—優點在於下述事 貫:生物基質殘餘物未受到有機溶劑污染且其因此可在不 經後續處理之狀況下首描^ α 、‘ ’舉例而言用於動物飼料中 ’具體而言指寇物飼料。 下文所述洋細方法旨在以非限制性實例形式閣述—含 ARA2,由、^DHA之油及一含ARA及DHA之油之製造。 用於將其他LC-PUFA(例如,DHGLA或EPA)自合適之生物基 質轉移至一載劑油之作業條件極爲相似。 在該方法之較佳實施例中,該油藉由下述方法獲得:將 生物基貝之乾壓梓油與藉由混合載劑油與乾生物基質壓梓 殘餘物並藉由壓榨自固體組份中分離所獲得之油混合。爲 增加LC-PUFA之納人量,需藉由高壓處理、酶法破裂微生 物細胞,或藉由研磨或碾壓來降低生物基質乾粒徑。 所用研磨步驟可爲先前技藝中已知之多種技術之一。舉 例而言,生物基質可經礙壓,較佳在低溫下,然後使其與 mm爲-變化形 < ’可於該載劑油之存在下研 磨該生物基質。爲盡可能將aLC_PUFA之破壞降至最小, 研磨條件應較溫和。因此,較佳者係於載劑油之存在下且 在惰性蒙氣下(舉例而言在一氮氣流中)研磨該生物基質。 然後,含有LC-PUFA之油藉由過濾或壓榨自生物基質餅 中刀離,較佳在兩壓下,然後實施—最終過濾以去除生物 基質殘餘物之微粒。 據觀察,當生物基質粒徑減小時,LC_pUFA之納入量增 87409 -11 - 1340165 加,舉例而言,當9〇。/。之顆粒的粒徑小於25〇微米時, PUFA之納入量大於9〇〇/0。 舉例而言,可使用一球磨機或一膠體磨。待考慮之參數 爲研磨時間、生物基質粒徑、研磨溫度、生物基質量與载 劑油量之比率。 研磨時間對粒徑有影響且後者亦受研磨溫度之影響。因 此,貫際上,較佳者係將粒徑指定爲一研磨階段之關鍵參 數。因此,理想狀況爲90%之顆粒的粒徑小於5〇〇微米較 佳爲90%之顆粒的粒徑小於3〇〇微米且更佳爲9〇%之顆粒的 粒徑小於200微米》 所選擇之研磨溫度高於該載劑油之熔點,且其較佳介於 20至80°C之間。 所選擇之生物基質與載劑油之重量比可確定最終油之 LC-PUFA含量。因此’舉例而言,每7〇份載劑油選擇儿份 生物基質以使經轉換之油中<LC_PUFA含量至少爲3 。 用作載劑之油可係任何可作爲人類食品之油或油混合物 。加入該專品組合物中之油或混合物較佳使用可增加 PUFA之含量者。對於-嬰兒配方而言,尤其應提及高油酸 向曰葵油(HOSFO)、向日蔡油(SFO)'大豆油、㈣油精及 中鏈三醯基甘油(MCT’ ±要含有飽和C8_C|〇脂月方酸三酿基 甘油)之使用。 該方法之下-階段在於藉由任何習知方法(例如,壓榨、 過濾或離心)分離剩餘之生物基質殘餘物。爲此,較佳使用 高壓下之壓榨作業。 87409 -12- 1340165 藉由精濾所獲得之油應無不溶性細顆粒。該作業可藉由( 適當時)將該油置於一作爲助濾劑之礦物吸附劑(例如,代卡 利特(dicalite))中實施。 最後,將經過濾之油予以脫臭以去除揮發性物質。此可 藉由任何已知之方法實施,其限制條件爲應使用溫和條件 以溫和作用於LC-PUFA。亦可使用,舉例而言,蒸汽蒸餾( 較佳在真空下)或分子蒸顧。 所獲得之油可以原有狀態或以與其他油(例如,魚油或沙 拉油)之混合物形式或另一選擇爲以沙拉醬或蛋黃醬中之鲁 乳/夜形式用於人類食用之食品組合物中。其可爲一用於青 少年或成年人之營養乳、一用於早産兒及未完全斷奶的足 月嬰兒之嬰兒調配物或一用於幼兒之改良乳之成份。 其可加入一用於經口攝取之營養性或補充組合物中。 其可加入一用於經口、經腸或非經腸攝取或用於局部、 皮膚病學或眼科應用之醫藥組合物中。 其可構成一用於一化妝用、局部或經口組合物之成份。 农後,其可構成用於一龍物食品(舉例而言一乾或濕食品癱 或一乳狀液)之組份。 分離出油後之生物基質殘餘物可較佳用於動物飼料中, 特別係寵物飼料中。 下述貝例可闡釋本發明。其中’除非另有說明,份數及 百分比皆係以重量計。 游離脂肪酸(FFA)之百分率使用IUPAC 2 2〇1方法確定; 87409 •13- 1340165 ARA含量使用IUPAC 2.3 04方法確定;且 磷(P)含量使用NI C12-1976-SSOG方法確定。 實施例1 :使用一液壓機壓榨一令ARA之生物基質 設備 :
