201100108 六、發明說明: 【發明所屬之技術領域】 本發明係有關一種美白劑,以及一種使皮膚美白之美容 方法’具體而言,係有關一種含有選自由4_順-羥基脯胺 酸、與其衍生物及/或鹽所組成之群組的一種或兩種以上 之化合物的美白劑,以及包含投與該美白劑的步驟之使皮 膚美白之美容方法。 【先前技術】 皮膚之色素即黑色素,係由存在於表皮之基礎膜附近之 黑色素細胞所產生,其係以環抱入表皮細胞之形態與表皮 細胞一起移動至皮膚表面,最終以皮垢自皮膚剝落。皮膚 之黑化或色素斑、雀斑等之色素異常,係因表皮内之黑色 素之異常增加、沉積所生,被指稱與紫外線、女性荷爾 蒙、遺傳因子有關。為此,藉由抑制黑色素生成而減輕皮 膚色素沉積之美白劑已由業者所開發(專利文獻1及2、非 專利文獻1及2)。 例如’還原劑之對苯二酚具有美白效果,作為紫外線所 ia·成之色素斑之形成的預防劑,已被指定為緩效藥品。 又’對於作為黑色素合成之最初二階段之反應的觸媒之酵 素赂胺酸酶具有阻礙活性之化合物,據信可作為有用之美 白劑。迄今為止,作為美白劑已有龍膽酸甲酯、苯并味 唑、間苯二酚等之化合物已被開發。 先前技術文獻 專利文獻 147105.doc 201100108 專利文獻1 :美國專利第6852747號 專利文獻2 :美國專利申請公開第2009/0023792號 非專利文獻 非專利文獻 1 : Mutagenesis, 5:263-266 (1990). 非專利文獻 2 : Biochem. Pharmacol.,57:663-672 (1999) 【發明内容】 發明所欲解決之問題 然而’對苯二酚已知有皮膚刺激性等之副作用,且對於 中國倉鼠V79細胞具有突變原活性(非專利文獻丨)。龍膽酸 甲醋雖被認為無突變原活性,但分子量為16 8,因此,分 子量較低、合成步驟數少,且製造成本低廉之美白劑之開 發’乃有所必要。此外,含相關美白劑之醫藥品、化妝品 及食品,以及含投與該美白劑之步驟的使皮膚美白之美容 方法之開發,也屬有其必要者。 解決問題之技術手段 本發明提供一種美白劑,其含有選自由4_順-羥基脯胺 酸、與其衍生物及/或鹽所組成之群組的一種或兩種以上 之化合物。 本發明之美白劑中,上述4-順-羥基脯胺酸可為4_順_L_ 經基脯胺酸及/或4-順-D-羥基脯胺酸。 本發明提供一種含本發明美白劑之化妝料組合物。 本發明之化妝料組合物可為皮膚外用劑。 本發明提供一種含本發明美白劑之食品組合物。 本發明提供一種含本發明美白劑之醫藥品組合物。 147105.doc 201100108 本發明之醫藥品組合物可為皮膚外用劑。 本發明另提供一種使皮膚美白之美容方法,其包含將選 自由4-順-羥基脯胺酸、與其衍生物及/或鹽所組成之群組 的一種或兩種以上之化合物投與之步驟。 本發明使皮膚美白之美容方法中,上述4_順-羥基脯胺 酸可為4-順-L-羥基脯胺酸及/或4_順_D_羥基脯胺酸。 本發明使皮膚美白之美容方法中,投與上述化合物之步 驟可為對於皮膚表面之外用。 本發明使皮膚美白之美容方法中,投與上述化合物之步 驟可為經口適用。 本發明又k供一種選自由4-順-經基脯胺酸、與其衍生 物及/或鹽所組成之群級的一種或兩種以上之化合物在製 造用以治療伴隨有皮膚色素異常之疾患的醫藥品組合物之 用返。在製造本發明醫藥品組合物之用途中,上述4_順-經 基脯胺酸可為4-順-L-羥基脯胺酸及/或4_順_D_羥基脯胺 酸。 羥基脯胺酸依羥基之位置在3位置或4位置(位置異構 物)、或其立體異構物為順式或反式(幾何異構物)、脯胺酸 為D體或L體(光學異構物),而存在有8種立體異構物。羥 基脯胺酸在自然界通常係以4-反_L-羥基脯胺酸存在,據知 其他立體異構物之存在極為限定。以下茲表示出-羥基 脯胺酸之幾何異構物及光學異構物之結構式。 147105.doc 201100108 [化1]

G 本發明之醫藥品組合物’除了 4_順_羥基脯胺酸單體、肛 順,基脯胺酸之鹽及/或在生體内可藉藥物代謝酵素或其 他物質放出4_順,基脯胺酸之衍生物以外,以無損4_順_ 減捕胺酸之美白效果為條件,進而可含—種或兩種以上 之治療其他疾患有效之成分及/或藥學上容許之添加物。 上述添加物包含稀釋劑及膨脹劑、結合劑及接著劑、滑 :丨、流動促進劑、可塑劑、崩解劑、载體溶媒、緩衝劑、 者色料、香料、甘味料、防腐劑及安定劑、吸附劑、業者 既知之其他醫藥品添加劑,但不受此限制。 【實施方式】 以下說明之本發明之實施例僅為例示之目的,並未限定 本發明之技術範圍。本發明之技術範圍僅由申請專利範圍 之§己載所限定。 實施例1 細胞培養 細胞係採小鼠B16惡性黑色腫瘤細胞。上述細胞係以 ΐχίο5個/孔之方式接種於6孔盤,並使用添加有1〇%胎牛血 清之市售培養基(Eagle MEM,日水製藥)。將上述細胞在 147l05.doc 201100108 37°C下於C〇2及飽和水蒸氣氛圍下培養之。 試藥 本發明實施例中所用之羥基脯胺酸中,4-順-L-羥基脯胺 酸、4-順-D-羥基脯胺酸及4-反-L-羥基脯胺酸係自Sigma-Aldrich Japan株式會社購入,4_反-D_羥基脯胺酸係自 Bachem公司購入。 羥基脯胺酸之添加 為研討羥基脯胺酸之細胞增殖及黑色素量減低之效果, 係在進行上述24小時細胞培養後,準備4_反丄_羥基脯胺 酸、4-反-D-經基脯胺酸、4-順-L-羥基脯胺酸及4-順-D-經 基脯胺酸之各種濃度之;辰縮水溶液,添加同一體積後,作 3曰培養並用於實驗。又,對照實驗中係添加上述體積之 水0 細胞數定量 添加試驗物質溶液,3日後將培養基吸引除去,添加含 10%之 alamarBlue(商標 Biosource,Biosource.International) 溶液之Eagle,s-MEM培養基,在37t下令其反應。3〇分鐘 後,以激勵波長544 nm、測定波長59〇 nm測定螢光,將其 值作為細胞數之相對值,算出相對試驗物質無添加群(僅 添加溶媒)之試驗物質添加群的細胞數比率(%細胞數)〇 % 細胞數愈南,表示細胞毒性低,未滿8〇%時,未滿8〇%時 判斷「有毒性」。 細胞數定量之結果 圖1A-D(有陰影之棒)’係表示以各種濃度經添加4_反丄_ 147l05.doc 201100108 羥基脯胺酸、4-反-D-羥基脯胺酸、4-順-L-羥基脯胺酸及 4-順-D-羥基脯胺酸之小鼠B16惡性黑色腫瘤細胞的基準組 設為100%時之細胞數調查實驗結果。圖1A係表示以各種 濃度添加4-反-L-羥基脯胺酸後設基準組為1 〇〇%時之細胞 數,3,3 3 ppm為 95%,11.1 ppm為 103%,33.3 ppm為 99%, 333 ppm為101%,且2690 ppm為94%。圖1B係表示以各種 濃度添加4-反-D-羥基脯胺酸後設基準組為100%時之細胞 數,3.33 ppm 為 98%,11.1 ppm 為 100%,33.3 ppm 為 〇 101%,333 ppm為 105%,且 2690 ppm為 109%。圖 1C係表 示以各種濃度添加4-順-L-羥基脯胺酸後設基準組為100% 時之細胞數,3.33 ppm為 93%,11_1 ppm為 92%,33.3 ppm 為84%。圖1D係表示以各種濃度添加4-順-D-羥基脯胺酸後 設基準組為100%時之細胞數,3.3 3 ppm為97%,11.1 ppm 為 98°/。’ 33.3 ppm為 101%,333 ppm為 92%。由以上結果可 知’基準組設為1〇〇時之細胞數,4_反_L_羥基脯胺酸及4_ ❹ 反-D-經基脯胺酸之情況下至2690 ppm為止,4-順-L-羥基 脯胺酸之情況下至33.3 ppm為止,4_順_〇_羥基脯胺酸之情 況下至333 ppm為止之濃度下未見變化,對小鼠b16惡性黑 色腫瘤細胞之細胞生成並無影響。 黑色素之定量 將培養基吸引除去’使用緩衝液(磷酸緩衝液5〇 , pH 6.8)作3次洗淨後,添加i N Na〇H溶解細胞, 測定475 nm之吸光度。將該值作為黑色素量之相對值,算出相對試 驗物質無添加群(只添加溶媒)之試驗物質添加群之黑色素 147105.doc 201100108 量比率(%)。此黑色素量比率愈低,意位著黑色素生成抑 制效果愈高。 黑色素量之評估 使用上述測定法所得之黑色素量之測定值,將經添加4_ 反_L_羥基脯胺酸、4-反-D-羥基脯胺酸、4-順-L-羥基脯胺 酸及4-順-D-羥基脯胺酸之細胞的黑色素量,以對照實驗 之細胞之黑色素量除算算出百分率,以之作為黑色素量 率。 黑色素定量之結果 圖1A-D(塗黑之棒),係表示調查以各種濃度經添加4_反_ L-髮基脯胺酸、4-反-D-經基脯胺酸、4-順-L-經基脯胺酸 及4-順-D-羥基脯胺酸之小鼠B1 6惡性黑色腫瘤細胞的黑色 素量之減低效果的實驗結果。圖丨八係表示以各種濃度添加 4-反-L-羥基脯胺酸後之黑色素量率,3 33 ppm為ι〇3〇/〇, 11.1 ppm為 108。/。,33.3 ppm為 105%,333 ppm為 104%,且 2690 ppm為107%。圖1B係表示以各種濃度添加4_反_〇_經 基脯胺酸後之黑色素量率,3.33卯爪為”%,U1卯爪為 990/。’ 33.3 ppm為 99%,333 ppm為 101%,且 2690 ppm為 110¾。圖1C係表示以各種濃度添加4-順_l-經基脯胺酸後 之黑色素量率 ’ 3.33 ppm為 92%,li.i ρρπι為 83%,33.3 ppm為30%。圖1D係表示以各種濃度添加順_D-經基鋪胺 酸後之黑色素量率,3·33 ppm為100%,n」ppm為98%, 33.3 ppm為96% ’ 333 ppm為32%。由以上結果顯示,4_反_ L-起基脯胺酸及4-反-D-幾基脯胺酸之情況下,並無專色素 147105.doc -12- 201100108 量減低之效果。又,4-順羥基脯胺酸及4-順_D_羥基脯 胺酸之情況下,則顯示具有濃度依存性之黑色素量減^之 效果。 根據4-反-L-羥基脯胺酸與4_順_D_羥基脯胺酸之混合液 之細胞增殖及黑色素定量試驗 4-反-L-羥基脯胺酸與4_順〇_羥基脯胺酸之混合液,係 將4_反-L-羥基脯胺酸與4-順-D-羥基脯胺酸以等量之方式 調製供實驗。細胞培養、細胞數及黑色素之定量及評估, 係與前述實施例同樣地進行。 混合液之細胞增殖試驗之結果 圖2A-C(有陰影之棒),係表示調查以各種濃度經添加‘ 反-L-羥基脯胺酸、4-順-D-羥基脯胺酸、與兩者之混合液 之小鼠B16惡性黑色腫瘤細胞之細胞增殖的實驗效果。各 實驗條件之誤差棒係表示以同一條件重複兩次之實驗結果 的測定值之標準偏差。圖2A係表示以各種濃度添加4_反_ Q L_經基脯胺酸後設基準組為100%時之細胞數,33.3 ppm為 95〇/〇,50 ppm為 100% ’ 1〇〇 ppm為 98%,333 ppm為 102%。 圖2B係表示以各種濃度添加4-順_D-羥基脯胺酸後設基準 . 組為1〇〇%時之細胞數,33.3 ppm為97%,50 ppm為93%, 100 ppm為95%,333 ppm為89%。圖2C係表示以各種濃度 添加兩者之混合液後設基準組為1 〇〇%時之細胞數,66.6 ppm為 97% ’ 100 ppm為 100%,200 ppm為 98%,666 ppm為 100%。由以上結果可知,基準組設為i 〇〇%時之細胞數, 4-反-L-羥基脯胺酸與4-順-D-羥基脯胺酸之混合液的濃度 147I05.doc •13· 201100108 為66.6 ppm至666 ppm下未見變化’顯然對小鼠B16惡性黑 色腫瘤細胞之細胞增殖並無影響。 混合液之黑色素定量之結果 圖2 A-C(塗黑之棒),係表示調查以各種濃度經添加4_反_ L-羥基脯胺酸、4-順-D-羥基脯胺酸、兩者之混合液之小鼠 B16惡性黑色腫瘤細胞的黑色素量之減低效果的實驗結 果。圖2A係表示以各種濃度添加4 -反-L-經基脯胺酸後之 黑色素量率 ’ 33.3 ppm為 103%,50 ppm為 104%,100 ppm 為108%,333 ppm為112%。圖2B係表示以各種濃度添加4_ 順-D-經基脯胺酸後之黑色素量率,33 3 ppm為9〇%,5〇 ppm為 85%,1〇〇 卩?111為 72。/。,333 ppm為 32。/。。圖 2C係表示 以各種濃度添加兩者之混合液後之黑色素量率,66 6 ppm 為 97%,1〇〇 卩卩瓜為 93¾,200 ppm 為 86%,666 ppm 為 57%。由以上結果可知,4_順_!)•羥基脯胺酸自33 3卯爪顯 示具有濃度依存性之顯著性黑色素合成抑制效果。然而, 經添加兩者之混合液者,雖然以相同於單獨添加4_順-D_ 羥基脯胺酸的量添加,但並未顯示顯著之黑色素量之抑 制。 各種混合比率之混合液之細胞增殖及黑色素量之減低試驗 使用4-反-L-羥基脯胺酸與4_順_D_羥基脯胺酸之以不同 比率調製之混合液,實施細胞增殖及黑色素量之減低試 驗。細胞培養、細胞數及黑色素之定量及評估,係與前述 實施例同樣地進行。 各種混合比率之混合液之細胞增殖試驗之結果 I47105.doc •14- 201100108 圖3(有陰影之棒),係表示調查以各種濃度經添加4_反_ L-經基脯胺酸、4-順-D-羥基脯胺酸、與上述混合液之小鼠 B16惡性黑色腫瘤細胞之細胞增殖的實驗效果。各實驗條 件之誤差棒係表示以同一條件重複兩次之實驗結果的測定 值之標準偏差。圖3A係表示以各種濃度添加4-反-L-羥基 脯胺酸後設基準組為100%時之細胞數,333 ppm為1〇〇〇/0,
