Abstract
Inventia se refera la un procedeu de preparare a pirolului substituit, cu formula generala I . în care R1 reprezinta hidrogen, alchil inferior, fenil eventual substituit cu grupe alchil inferior, cu aloxi, cu benziloxi, cu halogeni, cu hidroxi, cu nitro; R2 reprezinta o grupa acil alifatica, benzoul eventual substituit cu halogen, alcoxi, carbalcoxi, carboxi, carbamil, metil-carbamil, fenilcarbamil, R3 reprezinta hidrogen, alchil inferior , carbalcoxi carbalcoximetil, trifluormetil, carboxi, carbamil, carbozoil, sau R2 si R3 reprezinta împreuna o grupa -CO-(CH2)n în care n are valorile 2,3 sau 4, prin tratarea unui alfa-amino-nitril cu formula generala II: în care R1 are semnificatiile aratate mai sus, cu un compus cu formula generala III:în care R2 si R3 au semnificatiile aratate de mai sus, iar x reprezinta o grupa -C=C- sau -CH2-CO-, reactantii fiind luati în raport molar circa 1 : 1, la o temperatura cuprinsa între temperatura de reflux a amestecului de reactie, în prezenta acidului p-toluen-sulfonic, apoi produsul de reactie izolat este dizolvat în alcool anhudru, la temperatura camerei, iar produsul de reactie poate fi eventual trecut sub forma de clorhidrat în mod cunoscut. The invention relates to a process for the preparation of the substituted pyrrole of the general formula I. wherein R 1 represents hydrogen, lower alkyl, phenyl optionally substituted by lower alkyl, alkoxy, benzyloxy, halogen, hydroxy, nitro; R2 represents an aliphatic acyl group, the benzyl optionally substituted by halogen, alkoxy, carbalkoxy, carboxy, carbamyl, methylcarbamoyl, phenylcarbamoyl, R3 represents hydrogen, lower alkyl, carbalkoxycarboxoxymethyl, trifluoromethyl, carboxy, carbamyl, carbozoyl, or R2 and R3 represents together with a group -CO- (CH2) n wherein n is 2,3 or 4 by treatment of an alpha-aminonitrile of general formula II: wherein R1 has the meanings indicated above with a compound of general formula III : wherein R2 and R3 are as defined above and x represents a group -C = C- or -CH2 -CO-, the reactants being taken in a molar ratio of about 1: 1 at a temperature between the reflux temperature of of the reaction mixture in the presence of p-toluenesulfonic acid, then the isolated reaction product is dissolved in anhydrous alcohol at room temperature and the reaction product may optionally be passed in the form of the hydrochloride in a known manner.
