Abstract
Invenţia se referă la un procedeu de obţinere prin biosinteză a glucoamilazelor cu o tulpină selecţionată de Aspergillus awamori, care constă în aceea că, microorganismul se cultivă submers, în sistem discontinuu, în mediu nutritiv conţinând făină de porumb 2...3 g%, făină de soia 1...2 g%, extract de porumb 2...4 g% cu 48...50% s.u. şi supliment de săruri minerale KHPO0,1...0,5 g% şi (NH)SO0,1...0,5 g%, urmată de obţinerea preparatului enzimatic, prin filtrarea soluţiei enzimatice native pe filtrul Nutsche cu strat de perlită pe pânză petrol şi concentrarea fazei lichide rezultate, sub vacuum, la temperaturi cuprinse între 35 şi 40°C, pentru a se evita fenomenul de inactivare termică, parametrii indicaţi pentru desfăşurarea bioprocesului fiind temperatura 28...31°C,aerarea 0,1...0,3 1/1 min, iniţial, şi ridicată lent, pe parcursul fermentaţiei, până la 0,4...0,6 1/1 min, agitarea 200...300 rpm, pH-ul care rezultă după sterilizarea mediului este de 4,7...5,2, iar soluţiile enzimatice native se stabilizează prin adaos de CaCl0,1 g/%.
