Description
Foreliggende oppfinnelse angår nye, racemisk eller optisk aktive, halogenerte 15-hydroxy-E-homoeburnan-derivater av generell formel (Ia) og/eller (Ib),
hvori R er en C-^_g alkylgruppe og hvor X er halogen, og syreaddisjonssalter derav, og ennvidere en fremgangsmåte for fremstilling av disse nye forbindelser.
De nye forbindelser som ovenfor definert, fremstilles ifølge oppfinnelsen ved al; et racemisk eller optisk aktivt 15-hydroxy-E-homoeburnan-derivat av generell formel
(II):
hvori R er. som ovenfor angitt, eller et syreaddisjonssalt derav, behandles med et halogeneringsmiddel. Om ønsket kan forbindelsene av generell formel (Ia) og (Ib) som dan-
nes ved reaksjonen, separeres fra hverandre, og deretter kan enhver av forbindelsene omdannes til dets syreaddisjons-
salt og/eller oppløses om ønskes.
De nye forbindelser ifølge oppfinnelsen kan an-
vendes som mellomprodukter ved fremstilling av farmasøytisk aktive forbindelser, slik som halovincaminsyreestere. De nye forbindelser ifølge oppfinnelsen utviser også verdi-
fulle biologiske aktiviteter.
I forbindelsene av generell formel (Ia) og (Ib)
kan.R betegne en rettkjedet eller forgrenet C^_g alkyl-
gruppe, slik som methyl, ethyl, n-propyl, isopropyl, n-
butyl, sek.-butyl, tert.-butyl, n-pentyl, isopentyl, n-hexyl eller isohexyl-gruppe. R er fortrinnsvis ethyl eller n— butylgruppe.
X kan betegne alle de fire halogenatomer, dvs.
fluor, klor, brom og jod, fortrinnsvis brom.
Utgangsmaterialene av generell formel (II) kan fremstilles etter den metode som er beskrevet i Tetrahedron 33,.1903 (1977).
Utgangsmaterialene av generell formel (II) halo-generes med reaktanter som er i stand til å innføre et halogenatom i den umettede ring uten samtidig å erstatte 15-hydroxygruppen med halogen. Det foretrekkes å anvende ele-mentære halogenforbindelser som halogeneringsmidler.
Ifølge en foretrukket metode ifølge oppfinnelsen fremstilles forbindelser av generell formel (Ia) og (Ib)
hvori X betegner brom. De tilsvarende utgangsmaterialer av generell formel (II) bromeres fortrinnsvis med elementært brom, men andre bromeringsmidler som fører til dannelse av den ønskede bromforbindelse, kan også anvendes.
Bromeringen utføres i et inert, organisk løsnings-middel eller løsningsmiddelblanding. Blant løsningsmidler som er egnet i dette trinn, kan eksempelvis nevnes følgende: ikke-polare organiske løsningsmidler slik som halogenerte alifatiske hydrocarboner (f.eks. kloroform, diklormethan, diklorethan etc.), envidere polare organiske løsningsmidler slik som organiske syrer (f.eks. iseddik, propionsyre, etc).
I enkelte tilfeller er det foretrukket å utføre bromeringen i nærvær av en Lewis-syre. Som Lewis-syre kan det eksempelvis anvendes jernklorid, zinkklorid, aluminium-klorid, tinnklorid, antimontetraklorid, bortrifluorid etc.
Bromeringen kan utføres ved temperaturer på fra
20 til 40° C, fortrinnsvis ved romtemperatur.
Forholdet mellom de dannede stereoisomerer ved reaksjonen avhenger av graden av bromtilførsel.
Ved bromering av en .forbindelse av generell formel (II) erholdes en blanding av forbindelser av generell formel (Ia) og (Ib). De to stereoisomere bromderivater kan separeres fra hverandre ved kjente metoder, slik som krystal-lisering, saltdannelse og separasjon, preparativ skiktkromatografi etc. Det foretrekkes å anvende Merck PF254+3<g>6 9ra<^e silicagel som absorbent i den preparative skiktkromatografi. Forskjellige løsningsmiddelkombinasjoner kan anvendes som driv- og elueringsmidler.
Forbindelsene av generell formel (Ia) og/eller (Ib) kan omsettes med forskjellige syrer for å danne de respektive syreaddisjonssalter. Blant syrer som er anvendbare i denne reaksjon, skal nevnes følgende: uorganiske syrer slik som hydrogenhalogenider (f.eks. saltsyre eller hydrogenbromid), svovelsyre, fosforsyre, salpetersyre, perhalogensyrer (f.eks. perklorsyre), organiske carboxylsyrer slik som maursyre, eddiksyre, propionsyre, glycolsyre, maleinsyre, hydroxy-maleinsyre, fumarsyre, salicylsyre, melkesyre, kanelsyre, benzoesyre, fenyleddiksyre, p-aminobenzoesyre, p-hydroxy-benzoesyre, p-aminosalicylsyre etc., alkylsulfonsyrer slik som methansulfonsyre, ethansulfonsyre etc., cycloalifatiske sulfonsyrer slik som cyclohexylsulfonsyre, arylsulfonsyrer, slik som p-toluensulfonsyre, nafthylsulfonsyre, sulfanylsyre etc., aminosyrer slik som aspartinsyre, glutaminsyre etc.
Om ønsket kan de racemiske forbindelser av generell formel (Ia) og/eller (Ib) oppløses på kjent måte for å danne de respektive-optisk aktive derivater. Optisk aktive sluttprodukter kan også erholdes når en optisk aktiv for bindelse av generell formel (II) anvendes som utgangsmate-riale.
Om ønsket kan de racemiske eller optisk aktive forbindelser av generell formel (Ia) og/eller (Ib), såvel som syreaddisjonssaltene derav, underkastes ytterligere rensetrinn slik som omkrystallisering fra et hensiktsmessig valgt løsningsmiddel eller løsningsmiddelblanding. Løs-ningsmidlene eller løsningsmiddelblandingene som anvendes i dette trinn, velges i henhold til løseligheten og krystalli-sasjonsegenskapene til de materiale som skal renses.
Fremgansmåten ifølge oppfinnelsen gir sluttproduk-tene i former som er lette å identifisere. IR-spektre, NMR-spektre og massespektre for de fremstilte forbindelser er i overensstemmelse med den angitte struktur.
Oppfinnelsen illustreres ytterligere i de etter-følgende eksempler.
Eksempel 1 (-)- 9- brom- 14- oxo- 15- hydroxy- E- homoeburnane-( 3a, 17a) og
(-)- ll- brom- 14- oxo- l5- hydroxy- E- homoeburnane-( 3a, 17a)
a) 1,1 g jernkloridhexahydrat ble tilsatt til en løsning av 1,30 g (4,01 mmol) (-)-14-oxo-l5-hydroxy-E-homo-burnane-(3a,17a) som smelter vec 193 - 195° C, i 15 ml iseddik, og 5 ml av en 1 molar bromløsning i iseddik ble dråpe-vis tilsatt til blandingen ved romtemperatur under konstant omrøring. Bromløsningen ble innført langsomt, i en hastighet på 0,5 ml/min. Når addisjonen var fullført ble blandingen omrørt i ytterligere 9 timer ved romtemperatur. Når reaksjonen avtok ble 200 ml vann tilsatt til suspensjonen, og pH på den resulterende blanding ble justert til 5 med 25 % vandig ammoniakk. Blandingen ble ekstrahert med 100, 80 og 60 ml diklormethan. De organiske løsninger ble kombinert, blandet med 100 ml vann og pH på blandingen ble justert til 10 med 25 % vandig ammoniakk. Tofaseblandingen ble ristet, deretter ble diklormethanfasen fraskilt, vasket tre ganger med 100 ml vann hver gang, tørket over vannfritt fast magnesiumsulfat og filtrert, hvorpå filtratet ble fordampet til tørrhet i vakuum.
Det 1,4 g tørre residuum som ble erholdt, ble underkastet preparativ tynnskiktskromatografi. PF254+366 silicagelplater med et areal på 20 x 20 cm og en tykkelse på 1,5 mm ble anvendt som absorbent, og en 10:3 blanding av benzen og acetonitril ble anvendt som løsningsmiddel. De eluerte substanser ble krystallisert fra methanol.
0,15 g (11 %) (-)-9-brom-14-oxo-15-hydroxy-E-homoeburnan-(3a,17a) ble isolert fra den øvre flekk. Substansen
• smelter ved 202 - 203° C. Den empiriske formel av produktet
er C20H23BrN2O2(molvekt: 403.33).
IR spektrum (KBr): ^maks 3410 cm-1 (-OH), 1685 cm"1 (amid -CO).
. NMR spektrum (deuterokloroform). 6 - 0,97 (t, 3H,
CH-j) , 7,21 - 8,64 (m, 3H, aromatiske protoner).
C-^g-H = 7,51 ppm, J^^ 12 = 7,8 Hz (ortho)
Cj^-H = 7,21 ppm, = 7,6 Hz (ortho)
C12-H = 8,64 ppm, J1Q 12 = 1,9 Hz (meta)
Massespektrum (m/e): 404, 403, 402, 401, 376,
374, 360, 358, 347, 345, 332, 330, 317, 315, 303, 301, 277,
275, 180, 167, 153, 140.
0,63 g (46,1 %) (-)-ll-brom-14-oxo-15-hydroxy-E-homoeburnane-(3a,17a) ble isolert fra den midterste flekk. Materialet smelter ved 195 - 197^ C. Den empiriske formel
av produktet er C20H23BrN2O2(molvekt: 403,33).
IR spektrum (Kbr): Xmaks3350 cm"<1>(-OH),
1680 cm"1 (amid -CO).
NMR spektrum (deuterokloroform): 6 = 0,95 (t, 3H,
CH3), 7,25 - 8,69 (m, 3H, aromatiske protoner) ppm.

0,2 g av utgangsmaterialet, (-)-14-oxo-l5-hydroxy-E-homoeburnane-(3a, 17a) ble gjenvunnet fra den nedre flekk.
b) 1,1 g jernkloridhexahydrat ble tilsatt til en løsning av 1,3 g (4,01 mmol) (-)-14-oxo-15-hydroxy-E-homoeburnane-(3a,17a) som smelter ved 193 - 195° C, i 15 ml is-
eddik, og 5 ml av en 1-molar bromløsning i iseddik ble til-
satt til blandingen i én enkelt porsjon ved romtemperatur under konstant omrøring. Reaksjonsblandingen ble omrørt ved. romtemperatur i ytterligere 9 timer, hvorpå den ble fortyn-
net med 200 ml vann og pH på den vandige fase ble justert til 5 med 25 % vandig ammoniakk. Blandingen ble deretter ekstrahert med 100, 80 og 60 ml diklormethan. De organiske faser ble kombinert, blandet med 100 ml vann og pH på den vandige fase ble justert til 10 med 25 % vandig ammoniakk. Blandingen ble ristet, diklormethanfasen ble fraskilt, vasket tre ganger med 100 ml vann hver gang, tørket over vannfritt fast magnesiumsulfat, filtrert, hvorpå filtratet ble fordampet til tørrhet i vakuum. Det erholdte 1,5 g tørre residuum ble under kastet preparativ tynnskiktskromatografi. ^254+36<6>silicagelplater med et areal på 20 x 2,0 cm og tykkelse på
1,5 mm ble anvendt som adsorbent, og en 10:3 blanding av benzen og acetonitril ble anvendt som løsningsmiddel. De eluerte substanser ble krystallisert fra methanol.
0,2134 g (13,2 \) f+)-9-brom-14-oxo-15-hydroxy-E-homoeburnan-(3a,17a) ble isolert fra den øvre flekk. Denne substans (produktet med den høyere Rf-verdi) er identisk med forbindelsen med høyest R^-verdi fremstilt som beskrevet i punkt a).
0,6684 g ]41,4 %) (+)-ll-brom-14-oxo-15-hydroxy-E-homoeburnan-(3a,17a), et produkt med lavere R^-verdi, ble isolert fra den midterste flekk. Denne substans er identisk med forbindelsen med middels R^-verdi fremstilt som beskrevet iDunkt a) .
9-brom-14.15-dioxo-E-homoeburnan-(13a,17a) og 11-brom-14,15--dioxo-E-homoeburnan-(3a,17a) ble også dannet ved reaksjonen i en mengde på ca. 5 %. Disse forbindelser ble identifisert ved tynnskiktskromatografi.
Eksempel 2
(+)- 3( S), 17( S)- 9- brom- 14- oxo- 15- hydroxy- E- homoeburnan og (+)- 3( S), 17( S)- ll- brom- 14- oxo- 15- hydroxy- E- homoeburnan
1,10 g jernkloridhexahydrat ble tilsatt til en
løsning av 1,45 g (4,02 mmol) (+)-3-(S) ,17 (S)-14-oxo-15-hydroxy-E-homoeburnan-hydroklorid (sm.p. 24 0 - 24 2° C;
[cx]^° = +37,8°, c = 1 %, i pyridin) i 15 ml iseddik, og 5 ml av en 1-molar bromløsning i iseddik ble langsomt tilsatt med en hastighet på 0,5 ml/min, til løsningen ved romtemperatur under konstant omrøring. Når tilsetningen var fullført ble blandingen omrørt i 9 timer ved romtemperatur. Når reaksjonen var avsluttet ble blandingen fortynnet med 200 ml vann og pH på blandingen ble justert til 5 med 25 % vandig ammoniakk. Den resulterende løsning ble ekstrahert med 100, 80 og 60 ml diklormethan. De organiske faser ble kombinert, blandet med 100 ml vann og pH på den vandige fase ble justert til 10 med 25 % vandig ammoniakk. Blandingen ble ristet, diklormethanfasen ble fraskilt, vasket tre ganger med 100 ml vann hver gang, tørket over fast vannfritt magnesiumsulfat, filtrert hvorpå filtratet ble fordampet i vakuum.
Det erholdte 1,4 g tørre residuum ble underkastet preparativ tynnskiktskromatograf i . PF254+366 s:'-l:'-ca9elpla~ ter med et areal på 20 x 20 cm og en tykkelse på 1,5 mm ble anvendt som absorbent, og en 10:3 blanding av benzen og acetonitril ble anvendt som løsningsmiddel. De eluerte substanser ble krystallisert fra methanol.
Substansen med høyere R^-verdi er ( + )-3 (S) ,17(S)-9pbrom-14-oxo-15-hydroxy-E-homoeburnan. Denne forbindelse ble erholdt i et utbytte på 0,2 g (12,3 %) og smelter ved 104 - 105° C. Den empiriske formel for substansen er C20H23BrN2°2 (molvekt: 403,33).
IR spektrum (KBr): X , 3380 cm"1 (-OH),
1690 cm (-C0).
• [cc]p° = +43,7° (c = k %, i kloroform).
Substanden med lavere R^-verdi er ( + )-3(S) , 17(S)-ll-brom-14-oxo-15-hydroxy-E-homoeburnan. Denne forbindelse ble erholdt i et utbytte på 0,8 h (49,4 %) og smelter ved 117 - 119° C. Den empiriske formel for substansen er C20H23BrN2°2 (molvekt: 403,33).

Claims (3)
1. Fremgangsmåte for fremstilling av racemisk eller optisk aktive halogenerte 15-hydroxy-E-homoeburnan-derivater av generell formel (Ia) og/eller (Ib),
hvori R er en C^ _g alkylgruppe og X er halogen, og syre-addis jonssalter derav, karakterisert ved at et racemisk eller optisk aktivt 15-hydroxy-E-homoeburnan-derivat av generell formel (II)
hvori R er som ovenfor angitt, eller et syreaddisjonssalt derav, behandles med et halogeneringsmiddel og om ønsket,
at forbindelsene av generell formel (Ia) og (Ib) dannet i reaksjonen, separeres fra hverandre, og at deretter enhver av forbindelsene omdannes til dens syreaddisjonssalt og/eller oppløses om- ønsket.
2- Fremgangsmåte ifølge krav 1, karakterisert ved at elementært brom anvendes som halogeneringsmiddel.
3. Fremgangsmåte ifølge krav 1, karakterisert ved at halogeneringen utføres i nærvær av en Lewis-syre.
