En utførelsesform av oppfinnelsen er en forbindelse representert ved strukturformel (I) hvor Y er en kovalent binding av en substituert eller usubstituert rettkjedet hydrokarbylgruppe. I tillegg er Y sammen med begge >C=-grupper hvortil den er bundet en substituert eller usubstituert aromatisk gruppe. Fortrinnsvis er Y en kovalent binding eller - C(RR)-. Ri er en alifatisk gruppe, en substituert alifatisk gruppe, en ikke-aromatisk heterocyklisk gruppe eller en substituert ikke-aromatisk heterocyklisk gruppe, R-Ruavhengig er -H, en alifatisk gruppe, en substituert alifatisk gruppe, en ikke-aromatisk heterocyklisk gruppe, en substituert ikke-aromatisk heterocyklisk gruppe, en arylgruppe eller en substituert arylgruppe, eller Ri og Rsammen med karbon- og nitrogenatomene som de er bundet til, og/eller Rog Rsammen med karbon- og nitrogenatomene som de er bundet til, danner en ikke-aromatisk heterocyklisk ring eventuelt kondensert til en aromatisk ring. BAKGRUNN FOR OPPFINNELSEN
Mange nye medikamenter er nå tilgjengelige som vil bli anvendt av
oncologister ved behandling av pasienter med kreft. Ofte er tumorer mer mottagelige for behandling når anti-kreft medikamenter blir administrert i en kombinasjon til pasienten enn når samme medikamenter blir administrert enkeltvis og sekvensielt.
En fordel ved dette er at anti-kreftmidler ofte virker synergistisk fordi tumor-cellene angripes samtidig med midler som har multiple virkningsmåter. Således er det ofte mulig å oppnå raskere reduksjoner i tumorstørrelse ved administrering av disse medikamenter i kombinasjon. En annen fordel ved kombinasjonskjemoterapi er at tumorer mer sannsynlig blir fullstendig utryddet, og det er mindre sannsynlig å
utvikle resistens mot anti-kreft medikamentene som anvendes for å behandle pasienten.
En alvorlig begrensning ved kombinasjonskjemoterapi er at anti-kreftmidler generelt har alvorlige bivirkninger, selv når de blir administrert individuelt. For eksempel forårsaker det velkjente anti-kreftmiddel taxol neutroperi, nevropati,
mucosit, anemi, trombocytopeni, bradycardi, diaré og kvalme. Uheldigvis er toksisiteten til anti-kreftmidler generelt additiv når medikamentene blir administrert i kombinasjon. Som resultat derav blir visse typer av anti-kreft medikamenter generelt ikke kombinert. De samlede toksiske bivirkninger av anti-kreft-medikamentene som blir administrert samtidig kan medføre alvorlige begrensninger på mengdene som kan anvendes i kombinasjon. Ofte er det ikke mulig å anvende nok av kombinasjonsterapien for å oppnå de ønskede synergistiske effekter. Det er derfor et presserende behov for midler som kan øke de ønskelige tumorangreps-egenskapene til anti-kreftmidler uten ytterligere å øke deres uønskede bivirkninger.
OPPSUMMERING AV OPPFINNELSEN
Det er nå funnet at visse bis[tio-hydrazid amid]-forbindelser øker betydelig anti-kreftaktiviteten til taxol. For eksempel ble Forbindelse (1) anvendt i kombinasjon med taxol (Paclitaxel) for å behandle tumorer fremkalt hos hårløse mus med den humane brysttumorcellelinje MDA-435. Tumorvolumet var omtrent fem ganger mindre etter 24 dagers behandling hos mus som var blitt administrert 5 mg/kg taxol og 25 mg/kg forbindelse (1) enn hos mus som bare var blitt administrert 5 mg/kg taxol eller hos mus som bare var blitt administrert 50 mg/kg av forbindelse (1) (Eksempel 7). Det er også funnet at disse bis[tio-hydrazid amid]-forbindelser har minimale toksiske bivirkninger. For eksempel viste musen behandlet med taxol og Forbindelse (1) liten om overhodet noe som helst vekttap gjennom behandlingstiden (se Figur 2). Basert på disse resultater er nye forbindelser som øker anti-kreftaktiviteten til taxol, farmasøytiske preparater omfattende disse forbindelser og slike forbindelser for anvendelse som et medikament i kombinasjon med en effektiv mengde av paclitaxel eller en paclitaxelanalog for behandling av et individ med kreft beskrevet her. En annen utførelsesform av oppfinnelsen er et farmasøytisk preparat omfattende en farmasøytisk akseptabel bærer eller fortynningsmiddel og en forbindelse representert ved strukturformelen over. Fortrinnsvis omfatter det farmasøytiske preparatet en effektiv konsentrasjon av forbindelsen.
Enda en annen utførelsesform av oppfinnelsen er slike forbindelser som beskrevet over for anvendelse som et medikament i kombinasjon med en effektiv mengde av paclitaxel eller en paclitaxelanalog for behandling av et individ med kreft.
De beskrevne forbindelser øker anti-kreftaktiviteten til taxol- og taxol-analoger. I tillegg har disse forbindelser minimale toksiske bivirkninger. Følgelig er det mulig å øke effektiviteten av taxol og analoger derav når de blir anvendt i kombinasjon med de beskrevne forbindelser, selv når de høyeste tolererte doser av taxol blir nådd. Således er det forventet at kombinasjonsterapi med forbindelsene ifølge foreliggende oppfinnelse vil gi forbedrete kliniske resultater for pasienter med kreft som behandles med taxol. Ved ko-administrering av de beskrevne forbindelser med taxol er det også mulig å oppnå samme terapeutiske effektivitet som tidligere ble oppnådd med høyere doser av taxol og derved redusere bivirkningene og forbedre livskvaliteten til pasienten.

Oppfinnelsen omfatter også forbindelsene som definert over for anvendelse som et medikament i kombinasjon med en effektiv mengde av paclitaxel eller en paclitaxelanalog for behandling av kreft.
De beskrevne forbindelser er forsterkere av anti-kreft-aktiviteten til taxol og taxol-analoger. En forbindelse forsterker anti-kreft-aktiviteten til taxol eller en taxol-analog når aktiviteten til taxol eller taxol-analogen er større ved administrering i kombinasjon med forbindelsen enn ved administrering alene. Graden av økningen i aktivitet avhenger av mengden av forbindelse som blir administrert. Forbindelsene ifølge foreliggende oppfinnelse kan derfor anvendes i kombinasjon med taxol eller taxol-analoger for å behandle individer med kreft. Eksempler omfatter kolonkreft, pankreaskreft, melanom, nyrekreft, sarkom, brystkreft, eggstokk-kreft, lungekreft, magekreft, blærekreft og cervikal kreft.
Et "individ" er et pattedyr, fortrinnsvis et menneske, men kan også være et dyr med behov for veterinærbehandling, f.eks. kjeledyr (f.eks. hunder, katter), gårdsdyr (f.eks. kuer, sauer, griser, hester) og laboratoriedyr (f.eks. rotter, mus, marsvins).
For å oppnå en forbedring av anti-kreft-aktiviteten til taxol og taxol-analoger, blir en effektiv mengde av en forbindelse ifølge foreliggende oppfinnelse og en effektiv mengde av taxol eller analog av taxol administrert til individet. Med hensyn til taxol eller en analog av taxol er en "effektiv mengde" en mengde hvor anti-kreft- effekter normalt blir oppnådd. Med hensyn til en forbindelse ifølge foreliggende oppfinnelse er en "effektiv mengde" mengden hvor en større anti-kreft-effekt blir oppnådd når forbindelsen administrert sammen med taxol eller en taxol-analog sammenlignet med når taxol eller taxol-analogen blir administrert alene. Forbindelsen og taxol (eller taxol-analog) kan administreres sammen til individet som del av samme farmasøytiske preparat eller alternativt som separate farmasøytiske preparater. Når de blir administrert som separate farmasøytiske preparater, kan forbindelsen eller foreliggende oppfinnelse og taxol (eller taxol-analog) administreres samtidig eller på forskjellig tider, forutsatt at økningen av effekten av forbindelsen beholdes.
Mengden av forbindelse og taxol (eller taxol-analog) som blir administrert til individet vil avhenge av typen og alvorlighetsgraden av sykdommen eller tilstanden og av individets tilstand, så som generelle helse, alder, kjønn, kroppsvekt og toleranse overfor medikamenter. Den vil også avhenge av graden, alvorligheten og typen av kreft. Fagfolk vil være i stand til å bestemme passende doser avhengig av disse og andre faktorer. Effektive doser for taxol og taxol-analoger er velkjente og varierer typisk fra mellom ca. 1 mg/mm<2>pr. dag og ca. 1000 mg/mm<2>pr. dag, fortrinnsvis mellom ca. 10 mg/mm<2>pr. dag og ca. 500 mg/mm<2>pr. dag. Effektive mengder av en forbindelse ifølge foreliggende oppfinnelse varierer typisk mellom ca. 1 mg/mm<2>pr. dag og ca. 10 gram/mm<2>pr. dag og fortrinnsvis mellom 10 mg/mm<2>pr. dag og ca. 5 gram/mm<2>.
De beskrevne forbindelser blir administrert ad hvilken som helst egnet vei, omfattende for eksempel oralt i kapsler, suspensjoner eller tabletter eller ved parenteral administrering. Parenteral administrering kan for eksempel omfatte systemisk administrering, så som by intramuskulær, intravenøs, subkutan eller intraperitoneal injeksjon. Forbindelsene kan også administreres oralt (f.eks. dietary), topisk, ved inhalering (f.eks. intrabronkial, intranasal, oral inhalering eller intranesedråper) eller rektalt, avhengig av typen kreft som skal behandles. Oral eller parenteral administrering er foretrukne administreringsmetoder. Egnete administreirngsmetoder for taxol og taxol-analoger er velkjente på området og omfatter parenteral administrering som beskrevet ovenfor for forbindelsene ifølge foreliggende oppfinnelse. Egnete administreirngsmetoder for taxol og analoger derav er velkjente og omfatter bl. a. parenteral og oral administrering.
De beskrevne forbindelser kan administreres til individet sammen med en akseptabel farmasøytisk bærer som en del av et farmasøytisk preparat for behandling av kreft. Formulering av forbindelsen som skal administreres vil variere i henhold til administreringsveien som blir valgt (f.eks. løsning, emulsjon, kapsel). Egnete farmasøytiske bærere kan inneholde inerte bestanddeler som ikke interagerer med forbindelsen. Standard farmasøytiske formuleringsteknikker kan anvendes, så som de som er beskrevet i Remington's Pharmaceutical Sciences, Mack Publishing Company, Easton, PA. Egnete farmasøytiske bærere for parenteral administrering omfatter for eksempel sterilt vann, fysiologisk saltvann, bakteriostatisk saltvann (saltvann inneholdende ca. 0,9% mg/ml benzylalkohol), fosfat-bufret saltvann, Hank's løsning og Ringer's-laktat. Metoder for innkapslende preparater (så som i et belegg av hard gelatin eller cyklodekstrasn) er kjente på området (Baker, et al, "Controlled Release of Biological Active Agents", John Wiley and Sons, 1986). Egnede formuleringer for taxol og taxol-analoger er velkjent på området.
De beskrevne forbindelser kan fremstilles i henhold til metoder beskrevet i Eksemplene 1-12 og også i henhold til metoder beskrevet i den samtidig verserende US Provisional Application med tittelen SYNTHESIS OF TAXOL ENHANCERS, U.S. Provisional Application No. 60/304,318, innlevert 10. juli 2001.
Foreliggende oppfinnelse er illustrert ved de følgende eksempler.
EKSEMPLIFISERING
Eksempel 1.
Fremstilling av N- malonvl- bislT^- fenvl- N^ tioacetvDhvdrazidl En blanding av fenylhydrazin (30 ml) og etylmalonat (i xylen (150 ml) ble oppvarmet til tilbakeløp natten over. Reaksjonsblandingen ble avkjølt til romtemperatur. Fellingene ble oppsamlet ved filtrering og vasket med etanol, hvilket ga N-malonyl-bis(N'-fenylhydrazid) som et hvitt, fast stoff (14 g). Hydrazidet (3,4 g) ble oppslemmet i eddiksyreanhydrid (30 ml) og avkjølt i et isbad. Til dette sattes dråpevis perklorsyre (57% i vann, 3 ml). Reaksjonsblandingen gikk over til klar løsning først og stivnet deretter raskt. Etter henstand ved romtemperatur i 1 time ble eter (50 ml) tilsatt. Den resulterende oppslemningen ble filtrert og vasket med eter (2 x 00 ml), hvilket ga perkloratsaltene som et hvitt, fast stoff (5,7 g). Saltene ble tatt opp i aceton og satt som en oppslemning over 5 min til Na2S (0,6 M i vann, 90 ml) rørt ved romtemperatur. Etter 30 min ble reaksjonen surgjort med HCl(c), hvilket ga en gul oppslemning. Det faste stoffet ble oppsamlet ved filtrering og vasket med vann (20 ml) og eter (2x25 ml), hvilket ga N-malonyl-bis[N'-fenyl-N'-(tioacetyl)hydrazid] som et gråhvitt, fast stoff (3,6 g).<J>H NMR (DMSO-d6): 811,5 (m, 2H); 7,5 (m, 10 H); 3,2 (m, 2H); 2,6 (s, 3H); 2,5 (s, 3H). MS beregnet (400,1); Funnet: 423,1 (M+Na)<+>.
Eksempel 2.
Fremstilling av tiocykloheksansvre- N- fenylhydrazid
Fenylhydrazin (5,4 g, 50 mmol) ble oppløst i tørt diklormetan (50 ml) i en 250 ml rundkolbe. Di-tert-butyl-dikarbonat (10,9 g, 50 mmol) ble deretter tilsatt under røring ved 0 °C. Den resulterende løsning ble deretter rørt under tilbakeløp i 3 timer. Fjerning av de flyktige komponentene under redusert trykk ga et fargeløst, fast stoff, som ble vasket med heksan og tørket i vakuum. 10 g (utbytte 96%) av produktet ble oppnådd som et fargeløst, fast stoff, som kan anvendes i neste trinn uten ytterligere rensning. 2,5 g (12 mmol) av dette materialet ble oppløst i tørt pyridin (5 ml). Cykloheksanekarbonylklorid (2,0 ml, 15 mmol) ble deretter tilsatt langsomt ved 0 °C. Den røde løsningen ble rørt ved 0 °C i en halv time, og den resulterende gule suspensjonen ble rørt ved rt i 3 timer før den ble helt i is-H20 (100 ml). Fellingsproduktet ble oppsamlet ved filtrering og vasket grundig med H2O. Etter én omkrystallisering fra EtOH/H20 ble 3,63 g (95%) N-fenyl-N-cykloheksyl-N'-ter/-butoksykarbonylhydrazid oppnådd som et hvitt pulver; sm.p. 141-143 °C;<!>H NMR (CDCI3) 5 0,9-2,3 (m, 11H), 1,4 (s, 9H), 6,9 (br, 1H), 7,4 (m, 5H) ppm.
Til en løsning av N-fenyl-N-cykloheksyl-N'-/er/-butoksykarbonylhydrazid (1,1 g, 3,46 mmol) i diklormetan (6 ml) sattes trifluoreddiksyre (6 ml) ved 0 °C. Den resulterende løsning ble rørt ved 0 °C i en halv time. Flyktige komponenter ble deretter fjernet under redusert trykk, hvilket ga en sirup, som gikk over i et fast stoff ved henstand; dette materialet ble kort blandet med kald 2 N NaOH (5 ml) i noen få minutter ved 0 °C. Fast produkt ble deretter oppsamlet ved filtrering og omkrystallisert fra heksan, hvilket ga cykloheksansyre-N-fenylhydrazid (0,6 g, 80% utbytte) som et hvitt pulver;<J>H NMR (DMSO-d*) 8 0,8-3,2 (m, 1H), 5,3 (s, 2H), 7,0-7,7 (m, 5H); ESMS beregnet (Ci3Hi8N20): 218,3; funnet: 241,1 (M + Na)<+>.
En blanding av cykloheksansyre-N-fenylhydrazid (0,25 g, 1,15 mmol) og Lawesson's Reagens (0,46 g, 1,15 mmol) i tørt toluen (20 ml) ble rørt under
tilbakeløp i 1 time. Etter avkjøling til romtemperatur ble blandingen filtrert gjennom en kort kolonne av silikagel (5 g) som var forvasket med benzen. Fjerning av benzen ga råproduktet som et fast stoff som ble renset ved kolonnekromatografi på silikagel ved anvendelse av heksan/EtOAc (4 : 1 volum/volum) som elueringsmiddel. 0,15 g (60%) tiocykloheksansyre N-fenylhydrazid ble oppnådd som et gråhvitt, fast stoff.<J>H NMR (CDC13) 5 0,8-2,4 (m, 11H), 5,65 (br, 1H), 7,1-7,6 (m, 5H); ESMS beregnet (Ci3Hi8N2S): 234,1; funnet: 235,1 (M+H)<+>.
Eksempel 3.
Til en omrørt løsning av cykloheksansyre N-fenylhydrazid (0,1 g, 0,45 mmol) i tørr benzen (5 ml) sattes P2Ss(0,2 g, 0,45 mol). Den resulterende suspensjon ble oppvarmet til tilbakeløp i 3 timer. Etter avkjøling til romtemperatur ble blandingen fortynnet med benzen (5 ml) og filtrert gjennom en kort kolonne av silikagel (2 g), vasket med benzen og 2:1 heksan/EtOAc (15 ml hver). Filtratet og vaskevæskene ble samlet og inndampet, hvilket ga et fast stoff. Krystallisert fra heksan for å oppnå mellomproduktet tiocykloheksansyre-N-fenylhydrazid som et gråhvitt, fast stoff;;<J>H NMR (CDCI3) 5 0,8-2,4 (m, 11H), 5,65 (br, 1H), 7,1-7,6 (m, 5H); ESMS beregnet (Ci3Hi8N2S): 234,1; funnet: 235,1 (M+H)<+>.
Eksempel 4.
Cyklopropylbromid (4,8 g, 40 mmol) ble satt til en 50 ml vannfri THF-løsning inneholdende magnesiumpulver (1,1 g, 45 mmol), rørt i 30 min og tilbakeløpskokt i 30 min til. Etter at den var avkjølt, ble den klare reaksjonsløsningen satt til karbondisulfid (4 ml, 67 mmol) ved 0 °C og rørt i 30 min ved romtemperatur. Den resulterende blanding ble deretter satt til metylhydrazin (8 ml, 150 mmol) ved 0 °C og rørt i 2 timer til. Til denne løsningen sattes vann (40 ml), og det ble ekstrahert med EtOAc (60 ml x 3). 2-karboksybenzaldehyd (150 mg, 1 mmol) og karbazinsyre (132 mg, 1 mmol) i 40 ml metanol ble rørt ved romtemperatur i 4 timer. Til denne løsningen sattes Pd/C (60 mg, inneholdende 50 % H2O), reaksjonen ble holdt under H2-atmosfære i 3 timer. Reaksjonsblandingen ble filtrert, og løsningsmidlet ble avdampet. Det resulterende
residuet ble underkastet silikagelkolonnekromatografi. Forbindelse (1) øker anti-kreftaktiviteten til Paclitaxel in vivo ( Human xenograft modell: Humant brystkarsinom MDA- 435 hos hårløse mus)
Generell prosedyre for in vivo anti-tumor studium
ln vivo anti-kreft-forsterkningseffekten av nye forbindelser ble bedømt i tumor-bærende mus ved anvendelse av tumorveksthemningsforsøk. Tumorceller ble implantert ved injeksjon av en tumorcellesuspensjon subkutant i siden til en mus. Behandling av tumoren med en eksperimentell forbindelse og Paclitaxel begynte etter at tumoren var blitt etablert (volum var ca. 150 mm<3>). Dyrene påbegynte deretter et flerinjeksjons-opplegg hvor forbindelsen og Paclitaxel ble gitt ved IV administreringsvei. Tumorer ble målt to ganger i uken. I løpet av dette forsøket ble dyrene fulgt daglig etter tegn på toksisitet omfattende kroppsvektstap.
Detaljert prosedyre for MDA-435 (Humant brystkarsinom) anti-tumorstudium.
Et supplert medium ble fremstilt fra 50% DMEM/Dulbecco Modified Eagle Medium (Høy glukose), 50% RPMI 1640, 10% FBS/Føtalt bovint serum (Hybridomtestet; sterilfiltrert), 1% L-Glutamin, 1% Penicillin-Streptomycin, 1% MEM natriumpyruvat og 1% MEM ikke-essensielle aminosyrer. FBS ble oppnådd fra Sigma Chemical Co., og andre bestanddeler ble levert fra Invitrogen Life Technologies, USA). Det supplerte medium ble oppvarmet til 37° C og 50 ml medium ble satt til en 175 cm<2>vevskultur-kolbe.
Cellene anvendt i målingen var MDA-435 Humant brystkarsinom fra American Type Culture Collection. 1 medisinglass av MDA-435 celler fra cellene lagret frosset i flytende nitrogen ble tatt ut. Det frosne medisinglass med celler ble umiddelbart plassert i et 37° C vannbad og forsiktig virvlet inntil det var tint. Frysemedisinglasset ble gnidd med 70% etanol, og celler ble umiddelbart pipettert over i 175 cm<2>vevskulturkolben inneholdende supplert medium. Cellene ble inkubert natten over og mediet ble fjernet og erstattet med frisk supplert medium neste dag. Kolben ble inkubert inntil kolben var ca. 90% sammenflytende. Dette tok fra 5-7 dager.
Kolben ble vasket med 10 ml sterilt fosfatbufret saltvann (PBS) ved romtemperatur. Cellene ble trypsinisert ved tilsetning av 5 ml oppvarmet Trypsin-EDTA (Invitrogen) til kolben med celler. Cellene ble deretter inkubert i 2-3 minutter ved 37° C inntil cellene begynte å løsne fra overflaten av kolben. Et likt volum av supplert medium (5 ml) ble satt til kolben. Alle cellene ble oppsamlet i 50 ml rør og sentrifugert ved 1000 OPM i 5 minutter ved 20° C. Supernatanten ble dampet av, og celleklumpen ble gjenoppslemmet i 10 ml supplert medium og cellene ble tellet. 1-3 million celler/kolbe ble podet i 5-7 vevskulturkolber (175 cm<2>). Hver kolbe inneholdt 50 ml supplert medium. Kolbene ble inkubert inntil de var ca. 90% sammenflytende. Overføring av cellene ble gjentatt inntil nok celler var blitt dyrket for tumorimplantasjon.
Prosedyren ovenfor for trypsinisering og sentrifuging av cellene ble fulgt. Supernatanten ble dampet av og celleklumpen ble gjenoppslemmet i 10 ml steril PBS, og cellene ble tellet. Cellene ble sentrifugert og deretter gjenoppslemmet med passende volum steril PBS for injeksjon av riktig antall celler som var nødvendig for tumorimplantasjon. I tilfellet MDA-435 ble 100 million celler oppslemmet med 2,0 ml steril PBS til en sluttkonsentrasjon på 50 million celler/ml for å injisere 5 million celler i 0,1 ml/mus.
Mus (CD-1 nu/nu) ble levert fra Charles River Laboratories: nomenklatur: Crl:CD-l-nuBR, alder: 6-8 uker. Musen fikk akklimatisere i 1 uke før de ble anvendt i en eksperimentell prosedyre.
Implantasjon av MDA-435 tumorcellesuspensjonen fant sted i corpus adiposum hos CD-1 nu/nu hunnmus. Dette fettlegemet blir lokalisert i ventrale abdominale viscera hos musen. Tumorceller ble implantert subkutant i fettlegemet lokalisert i den høyre kvadrant av abdomen ved skjøten av os coxae (lyskeben) og os femoris (femur). 5 millioner MDA-435 celler i 0,1 ml steril PBS ble injisert ved anvendelse av 27 G (12,5 mm) nål. MDA-435 tumorer utviklet seg 2-3 uker etter implantasjon.
Stamløsninger av forbindelse ble fremstilt ved oppløsning av forbindelsen i en 50:50 blanding av EtOH og Cremophor EL (Polyoksyl 35 Ricinusolje, BASF, Tyskland). Denne stamløsning i 50%EtOH / 50%CrEL ble ultralydbehandlet i et ultralydvannbad inntil alt pulver var oppløst.
Formulering av doseringsløsning for forbindelsesadministrering: Stamløsningen av forbindelsen ble fortynnet 1:10 med D5W (5% Dekstrose i vann, Abbott Laboratories, USA).: 1) 2,0 ml 2,5 mg/ml doseringsløsning av Forbindelse (1) ble fremstilt ved fortynning 0,2 ml av en 25 mg/ml Forbindelse stamløsning med 1,8 ml 100% D5W; og 2) en doseringsløsning omfattende 1,5 mg/ml Paclitaxel (oppnådd fira Sigma Chemical Co.) og 2,5 mg/ml Forbindelse (1) ble oppnådd ved blanding av 0,2 ml av en 50%EtOH/ 50% CrEL stamløsning inneholdende 25 mg/ml Forbindelse (1) og 15 mg/ml Paclitaxel med 1,8 ml av en 100% D5W løsning. Den ferdige formulering for doseringsløsningen var 5% EtOH, 5% CrEL, 4,5 % Dekstrose og 85,5% vann.
Doseringsløsningen (Doseringsvolum: 0,01 ml/gram =10 ml/ kg) ble injisert intravenøst i musen som bærer MDA-435 human bryst-tumor.

RESULTATER
Figur 1 viser virkningene av Forbindelse (1) på forsterkning av anti-tumor-aktivitet av Paclitaxel (Taxol). Som det kan sees fra Figur 1 forbedret Forbindelse (1) betydelig anti-tumoraktiviteten til Paclitaxel på human brysttumor MDA-435 hos hårløse mus. Figur 2 viser virkningene av Forbindelse (1) og Paclitaxel på kroppsvekten av hårløse mus som bærer MDA-435 human brysttumor. Som det kan sees fra Figur 2, forbedret Forbindelse (1) betydelig anti-tumoraktiviteten til Paclitaxel uten å øke toksisiteten.
Skjønt foreliggende oppfinnelse er spesielt vist og beskrevet med referanser til foretrukne utførelsesformer derav, vil det være klart for fagfolk på området at forskjellige endringer i form og detaljer kan foretas uten at man går utenfor omfanget av foreliggende oppfinnelse som omfattet av de vedlagte krav.
