Description
Antibiotikum CC-1065, ( 7bR, 8aS)-7-[[1,6-dihydro-4-hydroksy-5-metoksy-7- [(4,5,8, 8a-tetrahydro-7-metyl-4-okso-cyklopropan[c]pyrrol[3,2-e]indol-2( 1H)-yl)karbonyl]benzo[l,2-b:4,3-b']dipyrrol-3(2H)-yl]karbonyl]-1, 6-dihydro-4-hydroksy-5-metoksybenzo[1,2-b:4,3-b']dipyrrol-3(2H)-karboksamid, er beskrevet og krevd i US patentskrift nr. 4.169.888 sammen med en fremgangsmåte for fremstilling av antibiotikum CC-1065 ved hjelp av aerobe fermenterings fremgangsmåter, og utvinning av antibiotikum CC-1065.
I The Journal of Antibiotics, 1985, 38, s. 746, rap-porterte D.G. Martin et al. at eddiksyre adderes over spiro-cyklopropylcykloheksadienylsystemet (SCPCH) i CC-1065, hvorved det fenoliske eddiksyreprodukt (AAP), 7-[[7-[[1-[(acetyloksy)-metyl]-1,6-dihydro-5-hydroksy-8-metylbenzo[l,2-b:4,3-b']di-pyrrol-3(2H)-yl]karbonyl]-1,6-dihydro-4-hydroksy-5-metoksy-benzo[1,2-b:4,3-b']dipyrrol-3(2H)-yl]karbonyl]-1,6-dihydro-4-hydroksy-5-metoksybenzo[l,2-b:4,3-b']dipyrrol-3(2H)-karboks-amid, fås. AAP ble testet in vitro og in vivo og funnet å være svakere enn CC-1065 med en faktor på IO<3>til IO<4>, avhengig av det bestemte testsystem, og var derfor tilbøyelig til å bort-lede oppmerksomhet fra addukter av SCPCH-systemet som anvendbare antitumormidler eller som forløperlegemidler til CC-1065-analoger.
I J. Am. Chem. Soc., 103, nr. 18, 1981, publiserte W. Wierenga en "Synthesis of the Left-Hand Segment of the Anti-tumor Agent CC-1065".
EP-søknad 0 154 445 (publisert 11.09.85) åpenbarer forskjellige analoger av antibiotikum CC-1065, inkludert forbindelser med formel EP-I og EP-II (se generelt formelskjema ifølge EP 0 154 445), hvor Rx i formel EP-II er CH3-, -CH2Ph, CH=CHCH2-,<->CH2SCH3, -CH2OCH3, -CH2OCH2CH2OCH3, -CH2CC13, -CH2CH2-Si(R2)3eller H, hvor Ph er fenyl; R er alkyl (C^Cs), fenyl eller H; R2. er C^Cj-alkyl, fenyl eller hydrogen og er ikke nødvendigvis det samme som R i én forbindelse; R3er alkyl(C1-C5), fenyl eller H; og X er Cl, Br, I eller OS02R40, hvor R40 er C-L-Cj-alkyl, fenyl, tolyl, bromfenyl, nitrofenyl eller trifluormetyl. De O-beskyttede forbindelser med formel EP-II er kjemisk stabile og lar seg bare fjerne under bestemte kjemiske betingelser. Når forbindelsene ifølge formel EP-II er O-avbe-skyttet, kan de imidlertid ringsluttes til forbindelsene ifølge EP-I.
EP-søknad 0 154 445 beskriver også CPl-dimerer bundet sammen ved hjelp av -CO-(CH2)nl-C0-, hvor ni er 2-12, og CPI-dimerer bundet sammen ved hjelp av kjeden -C(0)-(-Rn- )-C(0)-X7-(-CH2CH2-<X>7)n4-C(0)-(Rn-)-C(0)-, hvor Rn= CH2CH2eller CH=CH, og X7= 0 eller NH, og n4 = 1-4, og HC1- og Mel-saltene for X7
= NH.

Oppsummering av oppfinnelsen
Ved analogifremgangsmåten ifølge oppfinnelsen til-veiebringes to nye, syntetisk erholdte forbindelser, forbindelsene 1 og 2 (se generelt formelskjema). Forbindelsene er blitt påvist å ha nyttige områder for antitumoraktivitet i standard laboratoriedyrtester. Forbindelsene fremstilt ifølge oppfinnelsen lar seg erholde ved hjelp av kjemiske fremgangsmåter vist i reaksjonsskjemaene 1-6 og nærmere beskrevet i eksemplene.
Nærmere beskrivelse av oppfinnelsen
Ifølge oppfinnelsen er det således tilveiebrakt en analogifremgangsmåte for fremstilling av de terapeutisk aktive forbindelser
[S-(R<*>,R<*>)]-6,6'-[karbonylbis(imino-lH-indol-5,2-di-karbonyl) ] bis [8 - ( klormetyl )-3, 6, 7, 8-tetrahydro-l-metylbenzo-[l,2-b:4,3-b']dipyrrol-4-ol] (forbindelse 1) og
[7bR-[2(7'bR*,8'aS*),7bR*,8aS*] ]-2,2 * -[karbonylbis-(imino-lH-indol-5,2-dikarbonyl)]bis[l, 2,8,8a-tetrahydro-7-metyl-cyklopropan[c]pyrrol[3,2-e]indol-4(5H)-on] (forbindelse 2).
Analogifremgangsmåten er kjennetegnet ved at den passende 4,5,8,8a-tetrahydro-4-oksocyklopropan[c]pyrrol(3,2-e)indol-analog (formel B)
hvor Y er -COR, hvor R er valgt fra C^-C^-alkyl og fenyl som eventuelt er substituert med 1, 2 eller 3 C1-C4-alkyl, C1-C3-alkoksy, halogen, C1-C3-alkyltio, trifluormetyl, C2-C6-dialkylamino eller nitro; -C(0)NHS02R4, hvor R4er valgt fra Ci-C^-alkyl, fenyl som eventuelt er substituert med 1, 2 eller 3 C1-C4-alkyl, C1-C3-alkoksy, halogen, C^-Cj-alkyltio, trifluormetyl, C2-C6-dialkyl-amino eller nitro, naftyl som eventuelt er substituert med 1 eller 2 C1-C4-alkyl, C1-C3-alkoksy, halogen, trif luormetyl, C2-C6-dialkylamino, C^-C^-alkyltio eller nitro, -C(0)NR2R3, hvor R2og R3, som er like eller forskjellige, er valgt fra hydrogen, C^-C^-alkyl eller fenyl som eventuelt er substituert med 1, 2 eller 3 C1-C4-alkyl, C1-C3-alkoksy, halogen, C^-C-j-alkyltio, trif luormetyl, C2-C6-dialkylamino eller nitro, med det forbehold at både R2og R3ikke kan være fenyl eller substituert fenyl;
omsettes med biskarboksylsyrereagenset og så med den samme 4,5,8,8a-tetrahydro-4-oksocyklopropan[c]pyrrol-(3,2-e)indol-analog med formel B ovenfor, hvoretter Y-beskyttelsesgruppene fjernes slik at forbindelse 1 ovenfor fås, og eventuelt omdan-nes den erholdte forbindelse til forbindelse 2 ovenfor.
Forbindelsene 1 og 2 fremstilles lett ved å omsette den passende 4,5,8,8a-tetrahydro-4-oksocyklopropan[c]pyr-rol(3,2)indol-analog (formel B) med biskarboksylsyrereagenset og så med den samme 4,5,8,8a-tetrahydro-4-oksocyklopropan[c]-pyrroi(3,2-e)indol-analog (formel B), som illustret i reak-sjons skjemaene 1, 2, 2a, 3, 4, 5 og 6, under anvendelse av de følgende reaksjonsbetingelser: Trinnene 1 og 4: Utføres i oppløsning av 1 mg til 100 mg/ml i slike oppløsningsmidler som etylacetat, metylenklorid eller 1,2-dikloretan, med en gassformig syre ved 1-5 M, slik som HC1, eller HBr eller en sterk organisk syre, slik som CF3CO3H. Reaksjonen forløp ved 0-50°C. Produktet som et arain-salt ble isolert ved avdamping av oppløsningsmiddel og syre. Reaksjonen kan gjennomføres i 10 min til 48 timer.
Trinn 2: Reaksjonen forløp i polart oppløsnings-middel, slik som DMF, DMA eller THF, ved -10 til 50°C. Forhold mellom CPI-rest:biskarboksylsyre:dehydratiseringsmiddel, fortrinnsvis 2:1:2. Dehydratiserings- eller koblingsreagens kan være et karbodiimid, slik som dicykloheksylkarbodiimid (DCC) eller etyldimetylaminopropylkarbodiimid (EDC) eller andre amid-dehydratiseringsmidler kjent innen teknikken. Omsetningen kan foretas fra 10 min til 72 timer.
Trinnene 5 og 7: Det samme som trinn 2, men forholdet mellom CPI-rest:syre: dehydratiseringsreagens er fortrinnsvis 1:1:1.
Trinnene 3, 8, 9, 11, 14, 15 og 17: Omsetningen foretas i vandig, organisk base. De foretrukne betingelser er acetonitril:vann:trietylamin i et forhold på 1:1:1. Forholdet mellom disse reagensene kan endres til 1:2:2 til 5:1:1. Også oppløsningsmidlene kan endres slik at det i stedet for acetonitril kan brukes DMF, DMA, N-metylpyrrolidon, THF etc. Også den organiske base kan varieres til andre tertiære, organiske aminer, slik som 1,5-diaza-bicyklo[3.3.3]oktan (DABCO), eller etyldiisopropylamin. Temperaturen er vanligvis 0-50°C, og reaksjonen forløper vanligvis i 5 min til 10 timer.
Trinn 6: Avbeskyttelsen avhenger av beskyttelses-gruppen R. Dersom den er t-butyl, så kan gruppen fjernes ved lignende syrebehandling som i trinn 1. Dersom R = benzyl, kan det fjernes ved hydrogenolyse med f.eks. Pd-katalysator i etylacetat eller THF, eller ved hjelp av ammoniumformiat, metanol, vann og Pd/C. Dersom R=SiR3, så kan den fjernes ved hjelp av syre eller fluoridioner. Disse metodene er godt kjent innen teknikken.
Trinnene 5a og 7a: Reaksjonene kan utføres i aprotiske, polare oppløsningsmidler. Det er foretrukket med pyridin, DMF, DMA, THF eller metylenklorid. Når oppløsningsmidlene er forskjellige fra pyridin, kan det tilsettes en slik base som trietylamin. Omsetningen utføres ved et forhold mellom CPI-rest og aktivert karbonylforbindelse på ca. 1:1. Omsetningen kan utføres ved -50°C til 100°C og i fra 5 min til 72 timer.
Trinnene 10 og 16: Reagensene blandes i et forhold mellom CPI-rest og aktivt karbonyl på ca. 2:1. Omsetningen kan utføres i polare, aprotiske oppløsningsmidler, slik som THF, DMF, DMA, pyridin, acetonitril etc. Omsetningen kan utføres
ved -50°C til 100°C og i 10 min til flere uker.
Trinnene 12, 13, 18 og 19: Forhold mellom CPI-rest og aktiv karbonylrest på ca. 1:1. Ellers utført som trinn 10.
I de etterfølgende eksempler ble NMR-spektraldata målt i DMS0-d6med mindre annet er angitt. IR-spektra ble målt fra Nujol-"mulls". Oppløsninger for UV-spektra ble fremstilt ved å oppløse det faste stoff i noen få dråper DMF eller dimetylacetamid (DMA) og fortynne med 95 % etanol eller metanol med mindre annet er angitt. TLC-data er gitt for "Analtech Uniplates" av silikagel GF.
Eksempel 1
Fremstilling av [S -(R*,R*)]-6,6'-[karbonylbis(imino-lH-indol-5, 2-dikarbonyl)]bis[8-(klormetyl)-3,6,7,8-tetrahydro-l-metyl-benzoTl, 2- b:4, 3- b' ldipyrrol- 4- oll ( forbindelse 1)
Del A - fremstilling av 5,5'-(karbonyldiimino)bis-lH-indol-2-karboksylsyredietylester
En oppløsning av 5-aminoindol-2-karboksylsyreetyl-ester (6,82 g, 33,4 mM) og 4-dimetylaminopyridin (19 mg,
0,15 mM) oppløst i 80 ml THF ble avkjølt til -98°C i et ned-frosset metanolbad. Til dette ble det tilsatt 7 ml (40,2 mM) diisopropyletylamin, etterfulgt av 1,93 M fosgen i 8,5 ml (16,4 mM) toluen. Reaksjonsblandingen ble omrørt ved -98°C i 1 time og så ved -78°C i 1 time. Den ble så lagret ved -13"C over natten. Reaksjonen ble så stanset med vann og reaksjonsblandingen fortynnet med vann og etylacetat. Vannlaget ble fraskilt og på nytt ekstrahert to ganger med etylacetat. De kombinerte etylacetatlag ble vasket med N HC1, hvorved man fikk en blanding som separeres ved filtrering. Det frafUt-rerte, faste stoff ble funnet å være et rent produkt, 5,5'-(karbonyldiimino)bis-lH-indol-2-karboksylsyredietylester (6,38 g, 88 %). Filtratet ble tørket og oppkonsentrert, hvorved man fikk 1,49 g av et mørkt, fast stoff. Dette materialet ble kromatografert over silikagel (100 g) eluert med 10 % DMF i toluen. 20 ml fraksjoner ble samlet opp. Produkt ble funnet i fraksjonene 23-50 ved hjelp av TLC. Oppkonsentrering av disse fraksjonene under høyvakuum ga en rest som ble utkrys-tallisert fra DMF-metanol. Det ble derved erholdt ytterligere
0,80 g produkt.

Del B - fremstilling av 5,5'-(karbonyldiimino)bis-lH-indol-2-karboksylsyre
Indoldimerdiesteren, 5,5'-(karbonyldiimino)bis-lH-indol-2-karboksylsyredietylester (7,16 g, 16,5 mM), ble behandlet med 300 ml pyridin og 60 ml N NaOH. Den resulterende blanding ble omrørt i 24 timer ved 25°C, 7 timer ved 50-60°C og ytterligere 48 timer ved 25°C, på hvilket tidspunkt TLC indikerte at omsetningen var avsluttet. Reaksjonsblandingen ble oppkonsentrert ved 25°C under høyvakuum nesten til tørr-het. Resten ble surgjort med 3 N HC1 og det resulterende faste stoff samlet opp ved filtrering. Det filtrerte faste stoff ble oppløst i DMF (100 ml) ved ca. 100°C. Oppløsningen ble så fortynnet med 400 ml metanol og 10 ml vann. Oppløsningen ble av-kjølt og det krystallinske produkt, 5,5'-(karbonyldiimino)bis-lH-indol-2-karboksylsyre, samlet opp ved filtrering (5,11 g, 82 %).
NMR: 6 7,07 (d, 2 H, J = 9 Hz); 7,28 (dd, 2 H, J = 1,8 Hz, J = 9 Hz); 7,40 (d, 2 H, J = 9 Hz); 7,89 (d, 2 H, J = 1,8 Hz); 8,59 (bs, 2 HO); 11,65 (d, 2 H, J = 1,5 Hz).
IR: 3400-2300, 1680, 1530, 1225 cm'<1>.
UV: Xmaks= 294 nm (e = 23000).
MS (FAB) m/z 379 (M + H), 362, 331, 253, 176, 158, 132.
TLC: Rf = 0,33 i (40-60-2) DMF-toluen-eddiksyre.
Del C - omsetning av (S)-1-(klormetyl)-1,6-dihydro-5-hydroksy-8-metylbenzo[1,2-b:4,3-b']dipyrrol-3(2H) - karboksylsyre-1,1-dimetylester [(Boe)CPI-fenolklorid] med hydrogenklorid, etterfulgt av kondensasjon med 5, 5'-( karbonyldiimino) bis- lH- indol- 2- karboksylsvre
(S)-1-(klormetyl)-1,6-dihydro-5-hydroksy-8-metyl-benzo[l,2-b:4,3-b']dipyrrol-3(2H)-karboksylsyre-1,1-dimetyl-ester [(Boe)CPI-fenolklorid] (128 mg, 0,38 mM) ble oppløst i 6 ml etylacetat, og oppløsningen ble behandlet med en ny-fremstilt oppløsning av mettet HC1 i etylacetat (6 ml). Reaksjonsblandingen ble omrørt i 1 time, hvoretter TLC viste at utgangsmaterialet var fullstendig forsvunnet. Reaksjons-oppløsningen ble konsentrert under vakuum og ført tilbake til atmosfæretrykk under nitrogen. Den resulterende rest ble på nytt inndampet to ganger fra metylenklorid, hvorved man fikk ( S)-l-(klormetyl)-5-hydroksy-8-metyl-l, 2,3, 6-tetrahydrobenzo-[ 1, 2-b:4,3-b']dipyrrol-hydroklorid (CPI-fenolklorid-hydro-kloridsalt). Dette materialet ble behandlet med en oppløsning av disyren, 5,5'-(karbonyldiimino)bis-lH-indol-2-karboksylsyre (78 mg, 0,21 mM), i 4 ml dimetylacetamid. l-etyl-3-(3-dimetyl-aminopropyl)-karbodiimid-hydroklorid (EDC, 68 mg, 0,08 mM) ble så tilsatt som et fast stoff. Etter 45 min ble en andre mengde EDC tilsatt (68 mg). Omrøring ble fortsatt i 50 min.
Reaksjonsoppløsningen ble utfelt med vann og det faste stoff sentrifugert. Det ble vasket med 0,1 N HC1, 0,5 % NaHC03og vann, med sentrifugering og fjerning av vannfasen hver gang. Resten ble inndampet under vakuum ved mindre enn 20 torr i løpet av 2 dager. Det urensede produkt ble adsorbert fra DMF på "Celite" og kromatografert på en silikagelkolonne (15 g) under eluering med 30 % DMF i toluen. Fraksjoner å 10 ml ble samlet opp. [S-(R*,R*)]-6,6'-[karbonylbis( imino-lH-indol-5,2-dikarbonyl)]bis[8-(klormetyl)-3,6,7,8-tetrahydro-l-metylbenzo-[1, 2-b:4,3-b']dipyrrol-4-ol] ble funnet i fraksjonene 4-16 (115,1 mg fast stoff, 74 % utbytte basert på BocCPI). NMR, IR,
UV, FAB.

Eksempel 2
Fremstilling av [7bR-[2(7'bR<*>,8'aS<*>),7bR<*>,8aS<*>]]-2,2'-[karbo-nylbis(imino-lH-indol-5,2-dikarbonyl)]bis[1,2,8,8a-tetrahydro-7-metyl-cyklopropan[c]pyrrol[3,2-e]indol-4(5H)-on] ( forbindelse^)
[S-(R<*>,R<*>)]-6,6'-[karbonylbis(imino-lH-indol-5,2-di-karbonyl )]bis[8-(klormetyl)-3,6,7,8-tetrahydro-l-metylbenzo-[l,2-b:4,3-b']dipyrrol-4-ol] (80 mg, 0,1 mM) ble behandlet med 10 ml acetonitril, 3 ml vann og 3 ml trietylamin. Reaksjonsblandingen ble omrørt under nitrogen ved værelsestemperatur. Etter 40 min ble produktet som var utfelt, samlet opp ved filtrering. Filtratet ble fortynnet med vann og etylacetat og en andre mengde produkt samlet opp ved filtrering. De faste stoffene ble slått sammen, hvorved man fikk [7bR-[2( 7'bR*,8'-aS<*>),7bR<*>,8aS<*>]]-2,2'-[karbonylbis(imino-lH-indol-5,2-dikarbo-nyl )]bis[1,2,8,8a-tetrahydro-7-metyl-cyklopropan[c]pyrrol [3, 2-e]indol-4(5H)-on] (38 mg, 52 %).

Forbindelsene fremstilt ifølge oppfinnelsen er særlig anvendbare som antitumormidler. Eksempler på forbindelser med formel I utviser antitumoraktivitet i P388-leukemimus, og utviser også betydelig aktivitet i L1210-leukemi- og B16-melanom-murintest-systemer. Disse murintestsystemene er forut-sigbare for klinisk anvendbare humanantitumormidler (se f.eks. A Geldin et. al., European J. Cancer, Vol. 17, s. 129-142, 1981; J.M. Vendetti, Cancer Treatment Reports, Vol. 67, s. 767-772, 1983 og J.M. Vendetti et al, Advances in Pharmacology and Chemotherapy, Vol. 20, s. 1-20, 1984), og forbindelsene fremstilt ifølge foreliggende oppfinnelse vil derfor være anvendbare ved kontroll og behandling av sensible, neoplastiske (kreft)sykdommer, inkludert sensible leukemier, hos mennesker når de f.eks. gis intravenøst i doser på 0,001 ug/kg til ca. 10 mg/kg kroppsvekt pr. dag, idet den nøyaktige dose avhenger av alderen, vekten og tilstanden til pasienten, og av admini-strer ingshyppigheten .
Forbindelsene 1 og 2 er effektive når de ad-ministreres intravenøst (i.v.) i væskeoppløsninger ved bolusinjeksjon eller ved infusjon. De foretrukne doser er 0,01 pg/kg til 1000 ug/kg ved bolusinjeksjon og 0,0002-20 ug/kg/min ved infusjon. Den nøyaktige dose vil variere, avhengig av den bestemte forbindelse samt alderen, vekten, administrerings-måten og den fysiske tilstanden til pasienten, og av admini-strer ingshyppigheten .
Illustrerende in vivo og in vitro L1210-testdata for forbindelsene 1 og 2 er gjengitt i tabellene 1 og 2.
Tabell 3 viser data hvor [S-(R<*>,R<*>)]-6,6'-[karbonylbis(imino-lH-indol-5, 2-dikarbonyl) ]bis[8-(klormetyl)-3,6,7,8-tetrahydro-l-metylbenzo[1,2-b:4,3-b']dipyrrol-4-ol] (forbindelse 1) sammenlignes med (7bR)-N-[2-[(4,5,8,8a-tetrahydro-7-metyl-4-oksocyklopropan[c]pyrrol[3,2-e]indol-2(lH)oyl)karbonyl]1H-indol-5-yl]-2-benzofurankarboksamid (U-73.975) i de andre murine systemer hvor forbindelsene er blitt undersøkt sammen. Forbindelse 1 er minst like aktiv som forbindelsen U-73.975 i hvert system, og noen, slik som den subkutane L1210-analyse, utviser en bedre aktivitet over flere doseområder. Forbindelse 1 er, på samme måte som forbindelse U-73.975, blitt funnet å ikke forårsake senere død.
In vivo biologiske data for L1210 viser at [S-( R*,R*)]-6,6'-[karbonylbis(imino-lH-indol-5, 2-dikarbonyl)]bis-[8-(klormetyl)-3,6,7,8-tetrahydro-l-metylbenzo[1,2-b:4,3-b' ]-dipyrrol-4-ol] (forbindelse 1) er den mest aktive analog ved at den er svært helbredende ved 15 ug/kg. Resultatene angitt i
tabellene 1 og 4 ble oppnådd ved å bruke standard, godt kjente fremgangsmåter (In Vivo Cancer, Models, NIH-publikasjon nr. ' 84-2635, 1984) .
T/C % henviser til midlere livslengde for behandlede
mus dividert med midlere livslengde for kontrollmus x 100.
Forbindelsene 1 og 2 kan anvendes som anti-bakterielle midler. Disse forbindelsene kan anvendes til å kontrollere proliferasjonen av ømfintlige mikrober i forskjellige miljøer under anvendelse av standard mikrobiologiske teknikker. Slike miljøer omfatter tannlegeutstyr forurenset med S. aureus o.l, og laboratoriebenker i et mikrobiologisk laboratorium som kan rengjøres med et preparat som inneholder ca. 1-10 % (vekt/volum) av forbindelse 1 eller 2.
Tabell I i J. Am. Chem. Soc. 1989, 111, s. 6429 er en sammenstilling av cytotoksisitets- og kryssbindingsdata for "fleksible" CPI-dimerer, det vil si forbindelser hvor kjede-bindingen er -C(0)-(CH2)n-C(0)-. Se f.eks. EP Al 154445 og AU B2 617304. I fotnote (20) på side 6429 i litteraturartikkelen er det rapportert at forbindelse 4g (n=8) ved 3,1 ug økte livslengden til mus som hadde P388-leukemi med mer enn 150 %.
Når det gjelder godtakbarheten av forbindelse U-73975 som en representativ sammenligningsforbindelse fra teknikkens stand, skal det bemerkes:
In vivo-dataene i Tabell A viser at flere av de "fleksible" dimerene, nemlig forbindelsene 4b (n=3), 4c (n=4), 4d (n=5) og 4g (n=8) (identifisert i J. Am. Chem. Soc., 1989, 111, 6429-6431) er sterkere antitumormidler (i.p. P388 tumor/i.p. legemiddel, Q4Dx3) enn U-73975. De "fleksible" dimerer er imidlertid ikke så virkningsfulle som U-73975. Det vises til det etterfølgende Appendix som viser struktur-formlene til hver av de "fleksible" dimerer som det er henvist til i tabellene A, B og C.
Ved de mer umedgjørlige tumormodeller ble den samme trend sett. Hos mus behandlet i.p. med L1210 og med en eneste i.v.-dose legemiddel, er de "fleksible" dimerer sterkere, men mindre aktive enn U-73975. Se Tabell B.

Anmerkning: Den cyklopropylringåpnede analog 4g, det vil si
+ 8 +<*>2HC1, var i det vesentlige inaktiv og forholdsvis ikke-toksisk ved B16.
En sammenstilling av data for forbindelsen [7bR-[2(7bR<*>), 7bR<*>]]-2,2'-(1,10-diokso-l,10-dekandiyl)bis-[1,2,8,8a-tetrahydro-7-metyl-cyklopropan[c]pyrrol[3,2-c]indol-4(5)-on]; [4g (n=8)] er vist i Tabell D.
T/C henviser til midlere livslengde for behandlede mus dividert med midlere livslengde for kontrollmus x 100.
På bakgrunn av de data som viser at U-73975 er mer virkningsfull enn de "fleksible" dimerer (generisk beskrevet) i de anførte publikasjoner, burde anvendelsen av forbindelsen U-73975 som en sammenligningsforbindelse i tabellene 1 og 3 kunne godtas.
Claims (1)
- Analogifremgangsmåte for fremstilling av de terapeutisk aktive forbindelser [S-(R*,R*)]-6,61 -[karbonylbis( imino-lH-indol-5,2-di-karbonyl ) ]bis[8-( klormetyl) -3, 6, 7, 8-tetrahydro-l-metylbenzo-[l,2-b:4,3-b']dipyrrol-4-ol] (forbindelse 1) og [7bR-[2(7'bR<*>,8'aS<*>),7bR<*>,8aS<*>]]-2,2'-[karbonylbis-(imino-lH-indol-5,2-dikarbonyl)]bis[1,2,8,8a-tetrahydro-7-metyl-cyklopropan[c]pyrrol[3,2-e]indol-4(5H)-on] ( forbindelse 2), karakterisert vedat den passende 4,5,8,8a-tetrahydro-4-oksocyklopropan[c]pyrrol(3,2-e)indol-analog (formel B)hvor Y er -COR, hvor R er valgt fra Cx-C^-alkyl og fenyl som eventuelt er substituert med 1, 2 eller 3 C\-C4-alkyl, Ci-Ca-alkoksy, halogen, C^-C^-alkyltio, trif luormetyl, C2-C6-dialkylamino eller nitro; -C(0)NHS02R4, hvor R4er valgt fra C^-C^o-alkyl, fenyl som eventuelt er substituert med 1, 2 eller 3 C^-C^-alkyl, Ca-C3-alkoksy, halogen, C1-C3-alkyltio, trifluormetyl, C2-C6-dialkyl-amino eller nitro, naftyl som eventuelt er substituert med 1 eller 2 C^-C^-alkyl, C^-Ca-alkoksy, halogen, trif luormetyl, C2-C6-dialkylamino, C^-C^-alkyltio eller nitro, -C(0)NR2R3, hvor R2og R3, som er like eller forskjellige, er valgt fra hydrogen, C^-C^o-alkyl eller fenyl som eventuelt er substituert med 1, 2 eller 3 C^-C^-alkyl, C^-C^-alkoksy, halogen, Cj-C3-alkyltio, trif luormetyl, C2-C6-dialkylamino eller nitro, med det forbehold at både R2og R3ikke kan være fenyl eller substituert fenyl; omsettes med biskarboksylsyrereagenset og så med den samme 4,5,8,8a-tetrahydro-4-oksocyklopropan[c]pyrrol-(3,2-e)indol-analog med formel B ovenfor, hvoretter Y-beskyttelsesgruppene fjernes slik at forbindelse 1 ovenfor fås, og eventuelt omdan-nes den erholdte forbindelse til forbindelse 2 ovenfor.
