Description
Foreliggende oppfinnelse vedrører kondenserte 3-pyridintia-zolotio forbindelser. Disse forbindelsene er l-karba(l-detia)-cefem antibiotika og kan anvendes for behandling av infeksiøse sykdommer i mennesker og dyr.
l-carba(1-detia)cefem antibiotika har det bicykliske ringsys-temet representert ved følgende formel der nummererings-systemet er det som vanligvis blir anvendt i det vilkårlige cefam nomenklatursystemet
l-karba(1-detia)cefemer blir referert til heri som l-karba-3-cefem-4-karboksylsyrer eller nummererte derivater derav.
Fremstilling av 1-karbacefalosporiner (eller l-karba(detia)-3-cefemer) og C-3 substituerte metylderivater derav er beskrevet av Christensen et al., i US patent nr. 4.222.866. Eirata et al., i U.K. patentsøknad nr. 2.041.923, beskriver
en fremgangsmåte for fremstilling av 3-H og 3-halo 1-karba-cef alosporiner , mens Hatanaka et al., Tetrahedron Letters, 24, nr. 44, s. 4837-4838 (1983) beskriver en fremgangsmåte for fremstilling av et 3-hydroksy-(±)-l-karbacefalosporin.
Innen området antibakteriell terapi er det nødvendig med nye kjemoterapeutiske midler. Mutante stammer som er resistente overfor eksisterende antibakterielle midler opptrer ofte. Spesielt mange stammer av Staph. aureus og Staph. epi (såkalte meticillinresistente Staph. (MRS)) er blitt økende resistente overfor tilgjengelige antibakterielle midler (se f.eks. Phillips, I., og Cookson, B., J. Appl. Bacteriology 67(6), 1989). For å imøtese dette kravet er det blitt utført betraktelig forskning for å fokusere på slike nye midler.
De antibakterielle midlene er nyttige overfor mange forskjellige gram-positive og gram-negative bakterier. Forbindelsene ifølge foreliggende oppfinnelse er spesielt nyttige mot disse meticillin-resistente Staph. organismene.
Foreliggende oppfinnelse tilveiebringer forskjellige 3-tiazolotio-l-karba(1-detia)-3-cefemer som er nyttige som antibakterielle midler. Foreliggende oppfinnelse tilveiebringer spesielt 7p<->(2-aminotiazol-4-yl)oksimino- (eller alkoksimino)acetylamino l-karba(1-detia)-3-eventuelt-substi - tuert-tiazolotio-3-cefem-4-karboksylsyrer som er nyttige som antibakterielle midler, spesielt for behandling av meticillin-resistent Staphlococci.

Utgangsmaterialet (A), (der R er metyl), 2-(trityl)amino-a-(metoksy-imino)-4-tiazol eddiksyre kan bli fremstilt fra det tilsvarende frie aminet (tilgjengelig fra Aldrich Chemical Co., Inc., 940 West Saint Paul Avenue, Milwaukee, Wisconsin 53233) ved anvendelse av velkjent metodologi innen p<->laktam området. Utgangsmaterialet (B), eller benzhydryl 7-amino-l-karba( det ia)-3-trifluormetansulfonyloksy-3-cefem-4-karboksylat kan bli fremstilt ved anvendelse av kjent metodologi som beskrevet i Evans, et al., US patent nr. 4.673.737, inkorporert heri som referanse.
I Skjema 1 kan syrekloridet til (A) bli fremstilt ved kjent metodologi, f.eks., ved omsetning med fosforylklorid, og omsatt med det frie aminet (B) for å danne 7-acyl-3-triflat
(C). Tiazolotiogruppen kan deretter bli innført ved å omsette triflate (C) med en forbindelse med formel
i nærvær av en base såsom NaH. Sluttproduktet (1) kan deretter bli fremstilt ved å fjerne amino og karboksy-beskyttende grupper. I ovennevnte skjema blir CF3CO2H/-
(CH3CH2)3SiH anvendt for å fjerne trityl og benzhydryl-grupper. Fagfolk innen området p<->laktam kjemi vet at andre beskyttelsesgrupper kan være effektive. Man kan videre også innføre tiazolotiofunksjonen inn i 3-posisjon til cefem-kjernen (B) før innskudd av 7-acylfunksjonen for å tilveiebringe nyttige mellomprodukter som angitt i formel (2) nedenfor.
Forbindelser med formel
der A og A' sammen danner en gruppe med formlene og Y er nitrogen kan "bli fremstilt ifølge skjema (2).
I Skjema (2) blir 3-aminopyridin acylert med di-t-butyldikarbonat for å innføre t-butoksykarbonyl (t-BOC) beskyttende gruppe. (Det er å bemerke at to andre pyidinotiazolotio merkaptaner kan bli fremstilt ved kjent metodologi ved anvendelse av andre aminoplyridinisomerer.) t-BOC beskyttet 3-aminopyridin blir deretter behandlet med n-butyllitium i tetrahydrofuran etterfulgt av elementær svovel (Sg), etterfulgt av behandling med mettet ammoniumklorid. Det resulterende 3-t-butoksykarbonylamino-4-tia-pyridinet blir behandlet med en blanding av eddiksyre og HC1 for å tilveiebringe 3-amino-4-merkaptopyridin hydroklorid. Det ønskede 5-pyridino-tiazolo tiomerkaptan kan deretter bli fremstilt ved å behandle denne forbindelsen med karbondisulfid under basiske betingelser.
Når A og Å' tas sammen for å danne en gruppe med formel
kan det ønskede tiolet med formlen bli fremstilt som følger i Skjema (3) nedenfor.
I ovennevnte skjema blir 2-klor-3-nitropyridin behandlet med kaliumisotiocyanat for å tilveiebringe 2-isocyanato-3-nitropyridin, som deretter blir hydrolysert for å tilveiebringe 2-merkapto-3-nitropyridin. 3-nitro mellomproduktet blir deretter redusert ved behandling med SnClg/HCl for å tilveiebringe 2-merkapto-3-amino-pyridin. Ønsket pyridino-tiazolotio merkaptan blir deretter fremstilt ved basekata-lysert kondensasjon med CS2 (KOH/CB^OH/CSg/I^O).
En antibiotisk effektiv mengde er en mengde mellom omtrent 25 mg og omtrent 2 g. Forbindelsen, saltet eller esteren kan bli administrert i en enkeltdose eller i multiple doser iløpet av dagen. Behandlingen kan fortsette i 1 uke til 10 dager eller lenger avhengig av infeksjonsvarigheten. Den bestemte dosen og regimet kan avhenge av slike faktorer som vekt og alder til pasienten, den bestemte forårsakende organismen, alvorligheten til infeksjonen, generell helsetil-stand til pasienten og toleransen til den enkelte overfor antibiotika. l-karba(1-detia)cefem kan bli administrert parenteralt, subkutant eller rektalt. Som med andre p<->laktam antibiotika kan de bli anvendt profylaktisk for å forhindre infeksjoner etter utsetting eller før mulig utsetting, f.eks., preopera-tivt. Antibiotikaet kan bli administrert ved konvensjonelle metoder, f.eks. ved sprøyte eller ved intravenøst drypp.
Farmasøytisk akseptable salter som angitt ovenfor kan være nyttige former av antibiotika for fremstilling av antibio-tiske formuleringer.
De farmasøytiske formuleringene omfatter en antibiotisk effektiv ikke-toksisk mengde av en forbindelse representert ved Formel (1) eller et farmasøytisk akseptabelt ikke-toksisk salt derav, og en farmasøytisk akseptabel bærer.
Parenterale formuleringer av det antibakterielle midlet for injeksjon blir formulert med vann-for-injeksjon, Ringers oppløsning, fysiologisk saltvann og glukoseoppløsning. Antibiotikaet kan også bli administrert i en intravenøs væske ved dryppe-metoden.
For parenteral anvendelse kan det antibakterielle midlet med Formel (1) eller et farmasøytisk akseptabelt salt derav bli fremstilt fortrinnsvis i tørr krystallinsk pulverform eller som et lyofilisert pulver og fylt inn i beholdere. Slike beholdere kan inneholde mellom omtrent 100 mg og omtrent 2 gram antibiotika pr. beholder.
Mellomprodukter har Formel (2)

I Formel (2) refererer betegnelsen "karboksy-beskyttende gruppe" til ett av esterderivatene av karboksylsyregruppen som vanligvis blir anvendt for å blokkere eller beskytte karboksylsyregruppen mens reaksjonene blir utført på andre fuksjonelle grupper på forbindelsen. Eksempler på slike karboksylsyrebeskyttende grupper omfatter 4-nitrobenzyl, 4-metoksybenzyl, 3,4-di-metoksybenzyl, 2,4-dimetoksybenzyl, 2,4,6-trimetoksybenzyl, 2,4,6-trimetylbenzyl, pentametylben-zyl, 3,4-metylendioksybenzyl, benzhydryl, 4,4'-dimetoksybenz-hydryl, 2,2',4,4'-tetrametoksybenzhydryl, t-butyl, t-amyl, trityl, 4-metoksytrityl, 4,4'-dimetoksytrityl, 4,4',4"-trimetoksytrityl, 2-fenylprop-2-yl, trimetylsilyl, t-butyldi-metylsilyl, fenacyl, 2,2,2-trikloretyl, e-(trimetylsilyl)-etyl, P-(di(n-butyl)metyldilyl)-etyl, p-toluensulfonyletyl, 4-nitrobenzylsulfonyl-etyl, allyl, cinnamyl, l-(trimetyl-silylmetyl )prop-l-en-3-yl, og lignende deler. Typene av karboksybeskyttende grupper som blir anvendt er ikke kritisk dersom den derivatiserte karboksyl syren er stabil overfor betingelsen ved påfølgende reaksjon(er) på andre posisjoner av molekylet og kan bli fjernet ved hensiktsmessige punkter uten å ødelegge det gjenværende av molekylet. Det er spesielt viktig ikke å utsette det karboksybeskyttende molekylet for sterke nukleofile baser eller reduserende betingelser ved anvendelse av høyt aktiverte metallkatalysa-torer såsom Raney nikkel. (Slike grove fjerningsbetingelser bør også unngås ved fjerning av aminobeskyttende grupper diskutert heri.) Foretrukne karboksylsyre-beskyttende grupper er allyl, benzhydryl og p-nitro benzylgrupper. Lignende karboksy-beskyttede grupper anvendt innen cefalosporin, penicillin og peptidområdet kan også bli anvendt for å beskytte en karboksygruppe. Ytterligere eksempler på disse gruppene finnes i E. Forbindelsene med Formel (2) kan bli fremstilt ved metodologien som beskrevet i Skjema (1) ovenfor som viser 3-triflatdelen med den ønskede tiolen med formel
ved anvendelse av en 7-beskyttet amino-l-karba(1-detia)-3-trifluormetansulfonyloksy-3-cefem-4-(beskyttet karboksy)-kjerne.
Sluttproduktene (1) kan deretter bli fremstilt fra mellompro-duktene med Formel (2) ved avspaltning av 7-amino funksjonen etterfulgt av acylering med en ønsket acylgruppe, og påfølg-ende fjerning av amino/karboksy beskyttende gruppe.
Den følgende eksperimentelle delen tilveiebringer ytterligere eksempler av forskjellige aspekter av foreliggende oppfinnelse.
Eksperimentell del
Fremstilling 1
3-( t- butyloksykarbonyllamino pyridin
En 76.13 g (0.81 mol) prøve av 3-aminopyridin ble løst opp i 500 ml vann, sammen med 150 ml t-butanol og 34 g (0.85 mol) NaOH, avkjølt i et isbad, og behandlet med 200 g (0.92 mol) di-t-butyldikarbonat. Etter omtrent 2.5 dager ble ytterligere 100 g di-t-butyl dikarbonat tilsatt. Reaksjonsblandingen ble deretter heilt inn i en etylacetat/vannblanding. Den organiske fasen ble separert og den gjenværende vandige fasen ble ekstrahert med etylacetat. Kombinerte organiske porsjoner ble tørket over vannfri natriumsulfat, konsentrert i vakuum og renset via flammekromatografi for å tilveiebringe 97 g (80$) av tittelforbindelsen.

Fremstilling 2
3-( t- butyloksykarbonyl) amino- 4- merkaptopyridin
En 10 g (51.5 mmol) prøve av 3-(t-butyloksykarbonyl)aminopyridin ble løst opp i 110 ml tetrahydrofuran og avkjølt til -78°C under nitrogen. En 80 ml (128 mmol, 1.6 M i heksaner) prøve av n-butyllitium ble deretter tilsatt i to porsjoner. Reaksjonsblandingen ble deretter plassert i et aceton/isbad for å muliggjøre at det resulterende faste stoffet ble oppløst. Etter omtrent 2 timer ble reaksjonsblandingen avkjølt til -78°C og behandlet med 2 g (7.8 mmol) elementær svovel. Etter omtrent en halv time ble reaksjonsblandingen varmet til romtemperatur og ble stoppet med en mettet NH4CI oppløsning. Opparbeidning og flammekronratografi (50$ heksan/etyl acetat) tilveiebragte 5.24 g (4556) av tittelforbindelsen.

Fremstilling 3
3- amino- 4- merkapto- pvr i din hydroklorid
En 13.78 g (0.06 mol) prøve av 3-(t-butyloksykarbonyl)amino-4-merkapto pyridin ble løst opp med eddiksyre (250 ml) og tilsatt til en iskald oppløsning av £3N HC1 i eddiksyre som var blitt dannet ved bobling av HCl(g) gjennom iseddiksyre (100 ml). Etter omtrent 4 timer ble det resulterende faste stoffet filtrert, vasket med dietyleter og tørket i vakuum for å tilveiebringe 10.4 g (~100#) av tittelforbindelsen.

Fremstilling 4
2- merkapto- 5- pyridinotiazol
En 13 g (0.198 mol) prøve av kaliumhydroksidet ble løst opp i 32 ml vann og 154 ml metanol. Denne oppløsningen ble deretter behandlet med 3.8 ml (0.063 mol) CS2, etterfulgt av en 10.4 g (0.06 mol) prøve av 3-amino-4-merkaptopyridin hydroklorid. Etter omrøring ved tilbakeløp over natt ble reaksjonsblandingen behandlet med avfargende karbon og filtrert gjennom Hyflo Super Cel™. Filtratet ble surgjort med eddiksyre og forårsaket at et fast stoff ble dannet. Det resulterende faste stoffet ble tørket i vakuum ved 50°C i omtrent 3 timer og ved romtemperatur i omtrent 2.5 dager for å tilveiebringe 8.19 g (81$) av tittelforbindelsen.

Benzhydryl 7- p- fenoksyacetylamino- l- karba( 1- detia)- 3- tri-fluormetansulfonyloksy- 3- cefem- 4- karboksylat
Tittelforbindelsen kan bli fremstilt ved fremgangsmåten til Evans et al., US patent nr. 4.637.737, inkorporert heri som referanse. Utveksling av ovennevnte amino og karboksybeskyttende grupper eller anvendelse av alternativer kan bli utført ifølge metodologien som er velkjent innen p<->laktamområdet. Se f.eks. Protective Groups in Organic Syntesis, av Theodora W. Greene, New York, John Wiley & Sons, 1981.
Eksempel 1
7 p - f( 2- aminotiazol- 4- yl) - ( Z)- metoks lmino- acetyl1 amino- 1-karba( l- detia)- 3- r2-( 5- pyridinotiazolotio)~ l- 3- cefem- 4-karboksylsyre
A. Benzhydryl- 7p- amino- l- karba( 1- detla)- 3- trifluormetansul-fonyloksy- 3- cefem- 4- karboksylat
En 50 g (79 mmol) prøve av benzhydryl 7p<->fenoksyacetylamino-l-karba(detia)3-trifluormetansulfonyloksy-3-cefem-4-karboksylat ble løst opp i 500 ml CHgClg, avkjølt i et isvannbad og behandlet med 7.7 ml (95 mmol) pyridin, etterfulgt av 18.2 g (87 mmol) PCI5. Etter omtrent fire timer ble ytterligere 7.7 ml pyridin og 18.2 g PCI5 tilsatt. Reaksjonsblandingen ble deretter overført via sprøyte til en oppløsning av 80 ml isobutyl alkohol og 1000 ml CH2CI2 avkjølt i et isvannbad. Etter omrøring i omtrent 20 min. ved romtemperatur ble reaksjonsblandingen fortynnet med vann. Den vandige fasen ble deretter separert og ekstrahert med CEtøC^- Kombinerte organiske porsjoner ble vasket med mettet NaHCOs oppløsning, saltvann og tørket over vannfri MgSO^ Den resulterende organiske fasen ble konsentrert til omtrent 500 ml og anvendt som angitt i del B nedenfor.
B. Benzh<y>dr<y>l 7p- r2-( trifenylmetyl) aminotiazol- 4- yl)-( Z )-metoksiminoacetyl] amino l- karba( 1- detia)- 3- trifluormetansul-fonyloksy- 3- cefem- 4- karboksylat
En 35 g (79 mmol) prøve av 2-(trifenyl-metyl)aminotiazol-4-yl-(Z )-metoksiminoeddiksyre ble løst opp i 11 CEtøC^t avkjølt i et is/acetonbad og behandlet med 8.1 ml (74 mmol) N-metyl-morfolin og deretter 6.9 ml (74 mmol) fosforholdig oksy-klorid. Etter omtrent 20 minutter ble materialet fra del A ovenfor tilsatt, etterfulgt av 16.3 ml (148 mmol) N-metylmor-folin. Reaksjonsblandingen ble oppvarmet til romtemperatur og deretter fortynnet med saltvann. Den vandige fasen ble deretter separert, ekstrahert med CH2CI2 og de kombinerte organiske reaksjonene ble vasket med saltvann, tørket og renset via kolonnekromatografi for å tilveiebringe 12.65 g av tittelforbindelsen som angitt i (B) ovenfor.
C . Benzhydryl 7g-[ 2 -{ trifenylmetyl) amino- tiazol- 4- yl)-( Z)-met ok simino acetyl] am i no- 1- kar ba ( 1- detia) - 3-\ 2 -{ 5- pyridino-tiazolotio) 1- 3- cefem- 4- karboksylat
En (20 mg, 60$ suspensjon) 12 mg (0.5 mmol) prøve av NaH ble vasket med heksaner, suspendert i 5 ml tetrahydrofuran og behandlet med 84 mg (0.5 mmol) 2-merkapto-5-pyridinotiazol. Den resulterende blandingen ble oppvarmet for å oppnå oppløsning og deretter overført i tre porsjoner til en oppløsning av en 461 mg (0.5 mmol) prøve av materialet fremstilt i del B ovenfor, løst opp i 5 ml tetrahydrofuran. Reaksjonsblandingen ble deretter tilbakestrømmet, avkjølt, fortynnet med etylacetat og vasket sekvensielt med IN HC1 (IX), mettet NaHCOs oppløsning (IX) og saltvann. Etter tørking over vannfri MgS04 ble råproduktet renset ved anvendelse av kolonnekromatografi (75$ etylacetat/heksan som elueringsmiddel) for å tilveiebringe 350 mg (74$) av tittelforbindelsen ovenfor.

D. Avspaltning for å tilveiebringe tittelforbindelsen.
En 350 mg (0.37 mmol) prøve av produktet fra del C ovenfor ble løst opp i en blanding av 5 ml trifluoreddiksyre og 2 ml trietylsilan og omrørt i omtrent 10 min. Reaksjonsblandingen ble deretter fortynnet med omtrent 40 ml toluen og blandingen ble konsentrert til en rest i vakuum. Revers fase C-^g kolonnekromatografi (17$ CH3CN/H20) ga 33.3 mg av tittelforbindelsen.

Fremstilling 6
2- isotiocyanato- 3- nitro pyridin
En 10 g prøve av 2-klor-3-nitropyridin, en 8 g prøve av kaliumisotiocyanat og 75 ml eddiksyre ble kombinert og tilbakestrømmet i 2 t. Reaksjonsblandingen ble deretter avkjølt og heilt inn i 400 ml is/E20. Det resulterende faste stoffet ble vasket med vann, på ny løst opp i etylacetat og vasket (4x) med vann. Etylacetatoppløsningen ble deretter behandlet med aktivert karbon, tørket over vannfri Na2S04, filtrert og avdampet til tørrhet for å tilveiebringe 3.72 g av tittelforbindelsen.
Smp. = 115° - 118°C.

Fremstilling 7
2- merkapto- 3- nitropyridin
En 50 ml prøve av etanol ble behandlet med 612 mg natrium ved redusert temperatur (isbad) under vesentlige vannfrie betingelser. Reaksjonsblandingen ble deretter behandlet med en 3.6 g (0.02 mol) prøve (i porsjoner) av tittelforbindelsen ifølge fremstilling 6. Reaksjonen ble omrørt i 2 timer, fortynnet med 250 ml EtøO og avdampet i vakuum. Det resulterende faste stoffet ble filtrert ut og fjernet. Oppløsningen ble deretter surgjort med eddiksyre til pH =4.5 og gul-røde krystaller ble dannet. Tittelforbindelsen ble filtrert ut, vasket med vann og tørket under vakuum over en dessikant for å tilveiebringe 1.1 g (smp. = 185° - 7°C (dek)).

Fremstilling 8
2- merkapto- 3- aminopyridin
En 100 ml prøve av konsentrert HC1 (aq) ble avkjølt i et isbad og behandlet med 100 g (0.53 mol) SnCl2- Reaksjonsblandingen ble deretter behandlet med en 14 g (0.11 mol) prøve av tittelforbindelsen fra fremstilling 7, i porsjoner, og omrørt i 3 timer.
Reaksjonsblandingen ble deretter avdampet til et fast stoff, løst opp ill HgO og behandlet med ItøSÉg) i 30 min, med oppvarming over et dampbad. Det resulterende faste stoffet ble filtrert ut, vasket med varm EgO og fjernet. Kombinerte vandige porsjoner ble avdampet for å tilveiebringe et fast stoff. Det resulterende faste stoffet ble spaltet (2x) med varm konsentrert NH4OH. Det resulterende faste stoffet ble filtrert og fjernet og NH4OH oppløsningen ble avdampet for å tilveiebringe et vått fast stoff som deretter ble mobilisert i H2O. Den resulterende gul/grønne tittelforbindelsen ble filtrert, vasket med H20 og tørket i vakuum ved 40°C over en dessikant.
Utbytte = 4.20 g (30#)

Fremstilling 9
2- merkapto- 7- pyridinotiazol
En 2.8 g (8536) prøve KOE ble løst opp i 16 ml E20 og 50 ml metanol. En 2.6 g prøve av CSg ble deretter tilsatt og vasket med 30 ml metanol. En 4 g (23.8 mmol) prøve 2-merkapto-3-aminopyridin ble tilsatt og reaksjonsblandingen ble tilbakestrømmet over natt. Etter avkjøling ble reak-sj onsblandingen behandlet med aktivert karbon og filtrert gjennom Super Cel™, med vasking av Super Ce™ stykket med en liten mengde metanol. Oppløsningen ble deretter surgjort til pH = 5.5 med eddiksyre. Tittelforbindelsen presipiterte fra denne oppløsningen som et gult fast stoff og ble tørket ved 60°C over en dessikant.
Utbytte = 3.29 g

Etyl( 2-( tr i fenyImet. yl)- aminotiazol- 4- yl)- 2- brometvl- l- vl-oksiminoacetat
En 9.88 g (0.02 mol) prøve etyl-(2-(trifenylmetyl)aminotiazol-4-yl)oksiminoacetat ble løst opp i 20 ml N,N'-dimetylfor-mamid og behandlet med 8.28 g (0.06 mol) pulverformet kaliumkarbonat. Etter en halv times omrøring ble 17.3 ml 1,2-dibrometan tilsatt og reaksjonsblandingen ble omrørt over natt under argon. Reaksjonsblandingen ble deretter heilt inn i 100 ml CH2Cl2/200 ml H20. Det vandige laget ble på ny ekstrahert med CH2C12. Den kombinerte CH2C12 fasen ble vasket med H2O og saltvann, tørket over vannfri MgS04, filtrert og avdampet i vakuum for å tilveiebringe en olje. Væskekromatograf i (2556 heksan/CE^C^) tilveiebragte 7.16 g (63.456) av tittelforbindelsen.

Fremstilling 12
( 2-( trif enylmetyl ) aminotiazol- 4- yl)- 2- fluoret- l- yl-oksiminacetinsyre
En 2.5 g (5 mmol) prøve av tittelforbindelsen av preparering 11 ble løst opp i 20 ml etanol og 5 ml (10 mmol) 2N NaOH. Etter omrøring i 2 timer ved 50°C krystalliserte natrium-saltet av syren. Dette faste stoffet ble oppslemmet i H20/CHC13 og surgjort med IN HC1. Det vandige laget ble ekstrahert på ny med CECI3 og den kombinerte CHCI3 fasen ble deretter vasket med vann, saltvann og tørket over vannfri Na2S04- CHCI3 fasen ble deretter avdampet i vakuum for å tilveiebringe 1.52 g (63. 956) av tittelf orbindelsen som et skum.
