Abstract
En termofil mikroorganisme mangler laktatdehydrogenaseaktivitet og inneholder fortrinnsvis en aktiv pyruvatformatlyasereaksjonsvei. Den termofile mikro-organismen inneholder et gen som koder for en NAD-koblet formatdehydrogenase. Genet som koder for en NAD-koblet formatdehydrogenase er fortrinnsvis en kodon-optimalisert versjon av genet som koder for en varmestabil NAD-koblet formatdehydrogenase. DNA-konstruksjoner muliggjør stabil ekspresjon av genet som koder for en NAD-koblet formatdehydrogenase i den termofile mikroorganismen. DNA-konstruksjonene bygger på anvendelse av en innsettingssekvens for oppnå stabil ekspresjon eller rekombinasjon for innsetting av genet som koder for en NAD-koblet formatdehydrogenase inn i laktatdehydrogenasegenet slik at utslåing av genet og ny funksjonalitet oppnås i et enkelt trinn. Mikroorganismene er anvendbare for fermentering av sukkere for fremstilling av etanol.
