Description
Foreliggende oppfinnelse er rettet mot fremstilling av en ny gruppe av eksitatoriske aminosyreantagonister.
Oppfinnelsen angår således en analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser av formelen:
hvori T er representert ved en binding eller C1.4-alkylen; X og Y er hver uavhengig representert ved et halogenatom; R og Rx er hver uavhengig representert ved -0-(CH2)p-NR5R6; p er representert ved et helt tall fra 1 til 4; R5 og R6 er hver uavhengig representert ved C^-alkyl, eller danner sammen med det til-støtende nitrogenatom en morfolinogruppe; og farmasøytisk akseptable addisjonssalter derav.
Som anvendt i foreliggende beskrivelse, angir:
a) uttrykket "halogen" et fluor-, klor- eller brom-atom;
b) uttrykkene "lavere alkylgruppe og C1.4-alkyl" an-
gir en forgrenet eller rettkjedet alkylgruppe inneholdende fra
1 til 4 karbonatomer, slik som metyl, etyl, n-propyl, iso-propyl, n-butyl, isobutyl etc;
c) uttrykkene "lavere alkoksygruppe og C1_4-alkoksy" angir en rettkjedet eller forgrenet alkoksygruppe inneholdende
fra 1 til 4 karbonatomer, slik som metoksy, etoksy, n-prop-
oksy, isopropoksy, n-butoksy, isobutoksy etc;
d) uttrykket "C1_4-alkylen" angir en forgrenet eller rettkjedet alkylengruppe inneholdende fra 1 til 4 karbon-
atomer, slik som metylen, etylen, n-propylen, isopropylen, n-butylen, isobutylen etc;
e) uttrykket "aminoester" angir følgende substi-
tuent: -0-(CH2)p-NR5R6; f) uttrykket "funksjonaliseringsreaksjon" angir en forestring, amidering etc; g) uttrykket "farmasøytisk akseptable addisjonssalter" angir enten et basisk addisjonssalt eller et syreaddisjonssalt.
Uttrykket "farmasøytisk akseptable, basiske addisjonssalter" er beregnet på å gjelde hvilke som helst ikke-toksiske, organiske eller uorganiske, basiske addisjonssalter av forbindelsene representert ved formel I eller hvilke som helst av deres mellomprodukter. Illustrative baser som danner egnede salter innbefatter alkalimetall eller jordalkalimetall-hydroksider, slik som natrium-, kalium-, kalsium-, magnesium-eller bariumhydroksider; ammoniakk, et alifatisk, alisyklisk eller aromatisk, organisk amin, slik som metylamin, dimetyl-amin, trimetylamin og pikolin. Enten mono- eller dibasiske salter kan dannes med disse forbindelser.
Uttrykket farmasøytisk akseptable syreaddisjons-salter" er beregnet på å gjelde et hvilket som helst ikke-toksisk, organisk eller uorganisk syreaddisjonssalt av base-forbindelsene representert ved formel I, eller hvilket som helst av dets mellomprodukter. Illustrative uorganiske syrer som danner egnede salter, innbefatter saltsyre, hydrobromsyre, svovelsyre og fosforsyre, og sure metallsalter, slik som natriummonohydrogenortofosfat og kaliumhydrogensulfat. Illustrative organiske syrer som danner egnede salter, innbefatter mono-, di- og trikarboksylsyrene. Eksempler på slike syrer er f.eks. eddiksyre, glykolsyre, melkesyre, pyruvsyre, malonsyre, ravsyre, glutarsyre, fumarsyre, eplesyre, vinsyre, sitronsyre, askorbinsyre, maleinsyre, hydroksymaleinsyre, benzosyre, hydroksybenzosyre, fenyleddiksyre, kanelsyre, salisylsyre, 2-fenoksybenzosyre, p-toluensulfonsyre, og sulfonsyrer, slik som metansulfonsyre og 2-hydroksyetansulfonsyre. Slike salter kan eksistere i enten en hydratisert eller hovedsakelig vannfri form. Generelt er syreaddisjonssaltene av disse forbindelser løselig i vann og forskjellige hydrofile organiske løsnings-midler, og som sammenlignet med deres frie baseformer generelt utviser høyere smeltepunkter.
Enkelte av forbindelsene av formel I eksisterer som optiske isomerer. Enhver henvisning i foreliggende beskrivelse til en av forbindelsene representert ved formel I er ment å omfatte enten en spesifikk optisk isomer eller en blanding av optiske isomerer. De spesifikke optiske isomerer kan separeres og gjenvinnes ved teknikker kjent innen faget, slik som kroma-tografi på kirale, stasjonære faser, oppløsning via kiral saltdannelse og etterfølgende separasjon ved selektiv krystal-lisering, eller enzymatisk hydrolyse under anvendelse av stereoselektive esteraser, som kjent innen faget.
Som indikert ved nærvær av X- og Y-substituentene, skal stillingene 4- og 6- av indolringen være substituert med et halogenatom. Stillingene 4- og 6- kan være substituert med samme halogenatom eller forskjellige halogenatomer (dvs. et 4,6-diklor-, et 4-brom-6-klorderivat etc. skal betraktes som å være innen rammen for kravene).
2- og 3-stillingene av indolringen er substituert med en aminoester (dvs. -0-(CH2)p-NR5R6).
T er representert ved C^-alkylen. Dette C1.4-alkylen kan være enten lineært eller forgrenet. R3 og R4 kan være representert ved samme substituent eller forskjellige substituenter. Likeledes kan R5 og R6 være representert ved samme substituent eller forskjellige substituenter.
Det foretrekkes at T er representert ved en etylen-gruppe.
Analogifremgangsmåten ifølge oppfinnelsen er kjenne-tegnet ved at en forbindelse av formelen
hvori Pr og Pr<1> uavhengig betegner hydrogen eller en egnet forlatende gruppe, fortrinnsvis C1.4-alkyl/ og hvor X, Y og T har de ovenfor angitte betydninger, underkastes funksjonali-sering ved omsetning med overskudd av ammoniakk eller et mono-eller dialkylamin svarende til den ønskede R- eller R1-substituent, eller ved omsetning med en alkohol av formelen ROH, hvori R er som ovenfor angitt.
Reaksjonen er angitt nedenfor i reaksjonsskjema I:
Som angitt i reaksjonsskjema I, kan forbindelsene av formel I fremstilles ved å underkaste en av de 4,6-disubsti-tuerte indoler av struktur 1 en egnet funksjonaliseringsreaksjon som innfører den egnede funksjonalitet ved 2- og/eller 3-stilling av indolkjernen, og derved danner en av de ønskede forbindelser av formel I. I struktur (1) er X, Y, T og Z som i formel I, og Pr og Pr' betegner hver uavhengig grupper, slik som hydrogen eller fortrinnsvis C1.4-alkyl eller andre aktive esterforlatende grupper kjent innen faget. Den egnede indol av struktur 1 for anvendelse som et utgangsmateriale er et hvori X, Y, T og Z er representert ved de samme substituenter som ønskes i sluttproduktet av formel I.
Funksjonaliseringsreaksjonene kan utføres under anvendelse av teknikker vel kjent innen faget. Eksempelvis kan esterfunksjonaliteter adderes til 2- og/eller 3-stillingene av indolkjernen av struktur 1 under anvendelse av et utall av forestringsteknikker. En egnet forestringsteknikk omfatter å bringe den egnede forbindelse av struktur 1, hvori Pr og Pr' er C^-alkylfunksjoner, i kontakt med et overskudd av en alkohol av formelen ROH, hvori R er det samme som i formel I og er representert ved den samme funksjonalitet som ønskes i sluttproduktet (forestringsteknikk nr. 1). Reaksjonen utføres typisk i nærvær av et overskudd av en base, slik som kalium-karbonat. Reaksjonen utføres typisk ved en temperatur varierende fra romtemperatur til tilbakeløpstemperaturen i et tidsrom varierende fra 1 time til 24 timer. Etter at reaksjonen er fullført, kan det ønskede produkt av formel I gjenvinnes ved organisk ekstraksjon. Det kan deretter renses ved flashkromatografi og/eller omkrystallisering, som kjent innen faget. Egnede kromatografiske løsningsmidler innbefatter 5 % metanol i triklormetan. Egnede omkrystalliseringsløsnings-midler innbefatter etylacetat/heksan.
Andre egnede forestringsteknikker omfatter å bringe en forbindelse i henhold til struktur 1, hvori Pr og Pr' er representert ved H, i kontakt med et overskudd av en alkohol av formelen ROH, hvori R er det samme som i formel I og er representert ved den samme funksjonalitet som ønskes i sluttproduktet (forestringsteknikk nr. 2). Reaksjonen utføres typisk i nærvær av en overskuddsmengde av trifenylfosfin og en overskuddsmengde av dietyldiazodikarboksylat (DEAD). Reaksjonen utføres typisk ved et temperaturområde fra 25 °C til 35 °C i et tidsrom varierende fra 1 til 6 timer. Det ønskede produkt av formel I kan gjenvinnes og renses som angitt ovenfor.
Amider kan også lett fremstilles ved å bringe en forbindelse av struktur 1, hvori Pr og Pr' er C^-alkyl, i kontakt med et overskudd av ammoniakk eller et mono- eller dialkylamin svarende til den ønskede R- eller R^substituent, ved en temperatur på fra 0 °C til 100 °C i et tidsrom varierende fra 1 til 48 timer, i et inert løsningsmiddel, slik som tetra-hydrofuran. De resulterende amidderivater av formel I kan deretter isoleres og renses ved teknikker vel kjent innen faget.
Som det lett fremgår for fagmannen, hvis R og Rx ikke begge er representert ved samme funksjon i sluttproduktet, vil det da være nødvendig å utføre funksjonaliseringsreaksjonene på en sekvensvis måte under anvendelse av egnede beskyttende grupper, slik som dem som er beskrevet i Protection Groups in Organic Synthesis, av T. Greene. Dette kan gjøres under anvendelse av teknikker vel kjent innen faget.
Én egnet sekvensvis forestringsteknikk er vist nedenfor i reaksjonsskjema II.
I trinn A underkastes indolderivater av struktur 1, hvori X, Y, Z og T er som ovenfor angitt og Pr er etyl, mens Pr' er H, som vist, en omestringsreaksjon som innfører den ønskede estergruppe ved 2-stillingen av indolkjernen. Denne omestring utføres på samme måte som angitt ovenfor for forestringsteknikk nr. 1. Det resulterende produkt kan gjenvinnes og renses som angitt i reaksjonsskjema I. I trinn B innføres den ønskede estergruppe i 3-stillingen av indolkjernen. Dette kan utføres ved å underkaste produktet fra trinn A en forestrings-reaksjon under anvendelse av de teknikker som er angitt ovenfor for forestringsteknikk nr. 2. Det resulterende produkt kan også gjenvinnes og renses som angitt i reaksjonsskjema I. Andre sekvensvise reaksjoner kjent innen faget er også like egnet.

De antagoniserer effekten som eksitatoriske aminosyrer har på NMDA-reseptorkomplekset. De binder preferen-sielt til det strykninufølsomme glysinbindingssete assosiert med NMDA-reseptorkomplekset. De er anvendbare ved behandling av et utall av sykdomstilstander. Forbindelsene utviser antikonvulsive egenskaper og er anvendbare ved behandling av epilepsi. De er anvendbare ved behandling av grand mal-anfall, petit mal-anfall, psykomotor-iske anfall, autonome anfall etc. Én metode for demonstrering av deres antiepileptiske egenskaper er ved deres evne til å inhibere de anfall som forårsakes ved administrering av kino-linsyre. Denne testen kan utføres på følgende måte. En gruppe inneholdende 10 mus administreres 0,01-100 ug testforbindelse intraperitonealt i et volum på 5 ul saltvann. En andre kontrollgruppe inneholdende et rikt antall mus administreres et likt volum saltvann som kontroll. Cirka 5 minutter senere administreres begge grupper 7,7 pg kinolin-syre intraperitonealt i et volum på 5 ul saltvann. Dyrene observeres i 15 minutter deretter med hensyn til tegn på kloniske anfall. Kontrollgruppen vil ha en statistisk høyere grad av kloniske anfall enn testgruppen. En annen metode for demonstrering av de antiepileptiske egenskaper av disse forbindelser er ved deres evne til å inhibere audiogene konvulsjoner i DBA/2 mus. Denne testen kan utføres på følgende måte. Typisk administreres en gruppe av fra 6-8 DBA/2J audiogent mottakelige hannmus fra ca. 0,01 ug til ca. 10 ug av testforbindelsen. Testforbindelsen administreres intracerebralt i den laterale ventrikkel av hjernen. En andre gruppe av mus administreres et likt volum av en salt-vannskontroll ved samme rute. Fem minutter senere anbringes musene individuelt i glasskår og eksponeres for en lydstimulus på 110 desibel i 30 sekunder. Hver mus observeres under lyd-eksponeringen med hensyn til tegn på anfallsaktivitet. Kontrollgruppen vil ha en statistisk høyere forekomst av anfall enn gruppen som mottar testforbindelsen. Forbindelsene av formel I er anvendbare for å forhindre eller minimere skaden som nervevev inneholdt innen CNS lider av ved eksponering for enten iskemiske, traumatiske, hypoksiske eller hypoglykemiske tilstander. Representative eksempler på slike iskemiske, hypoksiske, traumatiske eller hypoglykemiske tilstander innbefatter slag eller cerebrovasku-lære ulykker, sjokk, hyperinsulinemi, hjertestans, drukninger, kvelning og neonatalt, anoksisk traume. Forbindelsene skal administreres til pasienten innen 24 timer etter starten av den hypoksiske, iskemiske, traumatiske eller hypoglykemiske tilstand for at forbindelsenes effektivt skal minimere den CNS-skade som pasienten vil oppleve. Forbindelsene er også anvendbare ved behandling av neurodegenerative sykdommer, slik som Huntingtons sykdom, Alzheimers sykdom, senildemens, glutarsyreacidemi type I, multiinfarktdementia, og neuronal skade assosiert med ukon-trollerte anfall. Administrering av disse forbindelser til en pasient som opplever en slik tilstand, vil tjene til enten å forhindre pasienten i å oppleve ytterligere neurodegenerering eller nedsette graden ved hvilken neurodegenereringen finner sted.
Som det fremgår for fagmannen, vil forbindelsene ikke korrigere enhver CNS-skade som allerede har oppstått som
resultat av en sykdom, fysikalsk skade eller mangel på oksygen eller sukker. Som anvendt i foreliggende søknad, angir uttrykket "behandle" forbindelsenes evne til å forhindre ytterligere skade eller forsinke graden ved hvilken enhver ytterligere
skade finner sted.
Forbindelsene utviser en anxiolytisk effekt og er således anvendbare ved behandling av angst. Disse anxioiytiske egenskaper kan demonstreres ved deres evne til å blokkere nød-skrik i rottevalper. Denne testen er basert på det fenomen at når en rottevalp fjernes fra dens kull, vil den utsende en ultralydvokalisering. Det ble funnet at anxioiytiske midler blokkerer disse vokaliseringer. Testmetodene er blitt beskrevet av Gardner, C.R., Distress vocalization in rat pups: a simple screening method for anxiolytic drugs, J. Pharmacol. Methods, 14:181-187 (1985), og Insel et al., Rat pup ultra-sonic isolation calls: Possible mediation by the benzo-diazepine receptor complex. Pharmacol. Biochem. Behav., 24:1263-1267 (1986).
Forbindelsene utviser også en analgesisk effekt og er anvendbare ved regulering av smerte.
For å utvise disse terapeutiske egenskaper må forbindelsene administreres i en mengde tilstrekkelig til å inhibere den effekt som de eksitatoriske aminosyrer har på NMDA-reseptorkomplekset. Doseområdet ved hvilket disse forbindelser utviser denne antagonistiske effekt kan variere vidt, avhengig av den bestemte sykdom som skal behandles, strengheten av pasientens sykdom, pasienten, den bestemte forbindelse som administreres, administreringsrute og nærvær av andre under-liggende sykdomstilstander i pasienten etc. Typisk utviser forbindelsene deres terapeutiske effekt ved et doseområde på fra ca. 0,1 mg/kg/dag til ca. 50 mg/kg/dag for hvilken som helst av de sykdommer eller tilstander som er angitt ovenfor. Gjentatt daglig administrering kan være ønskelig og vil variere i henhold til de ovenfor angitte betingelser.
Forbindelsene ifølge oppfinnelsen kan administreres ved et utall ruter. De er effektive hvis de administreres oralt. Forbindelsene kan også administreres parenteralt (dvs. subkutant, intravenøst, intramuskulært, intraperitonealt eller intratekalt).
Farmasøytiske preparater kan fremstilles under anvendelse av teknikker vel kjent innen faget. Typisk blandes en antagonistisk mengde av forbindelsen med en farmasøytisk akseptabel bærer.
For oral administrering kan forbindelsene formuleres i faste eller væskeformige preparater, slik som kapsler, piller, tabletter, pastiller, smelter, pulvere, suspensjoner eller emulsjoner. Faste enhetsdoseringsformer kan være kapsler av vanlig gelatintype inneholdende f.eks. overflateaktive midler, smøremidler og inerte fyllstoffer, slik som laktose, sukrose og maisstivelse, eller de kan være preparater med for-lenget frigivelse.
I en annen utførelsesform kan forbindelsene av formel I tabletteres med konvensjonelle tablettbaser, slik som laktose, sukrose og maisstivelse i kombinasjon med bindemidler, slik som akasie, maisstivelse eller gelatin, oppbrytende midler, slik som potetstivelse eller alginsyre, og et smøre-middel slik som stearinsyre eller magnesiumstearat. Væskeformige preparater fremstilles ved oppløsning av den aktive bestanddel i et vandig eller ikke-vandig, farmasøytisk aksep-tabelt løsningsmiddel som også kan inneholde suspenderings-midler, søtningsmidler, smaksgivende midler og konserveringsmidler, som kjent innen faget.
For parenteral administrering kan forbindelsene opp-løses i en fysiologisk akseptabel farmasøytisk bærer og administreres enten som en løsning eller en suspensjon. Eksempler på egnede farmasøytiske bærere er vann, saltvann, dekstroseløsninger, fruktoseløsninger, etanol eller oljer av animalsk, vegetabilsk eller syntetisk opprinnelse. Den farma-søytiske bærer kan også inneholde konserveringsmidler, buffere etc, som kjent innen faget. Når forbindelsene administreres intratekalt, kan de også være oppløst i cerebrospinalvæsken, som kjent innen faget.
Som anvendt i foreliggende søknad, angir:
a) uttrykket "pasient" varmblodige dyr, slik som f.eks. marsvin, mus, rotter, katter, kaniner, hunder, aper,
sjimpanser og mennesker;
b) uttrykket "behandle" angir forbindelsenes evne til enten å lindre, dempe eller nedsette progresjonen av
pasientens sykdom;
c) uttrykket "neurodegenerering" angir en progressiv død og forsvinning av en populasjon av nerveceller som oppstår
på en måte som er karakteristisk for en bestemt sykdomstil-stand og fører til hjerneskade.
Forbindelsene av formel I kan også blandes med enhver inert bærer og anvendes i laboratorieutprøvninger for å bestemme konsentrasjonen av forbindelsene innen serum, urin etc. av pasienten, som kjent innen faget.
Neurodegenerative sykdommer er typisk assosiert med tap av NMDA-reseptorer. Således kan forbindelsene av formel I anvendes i diagnostiske prosedyrer for å avhjelpe legen med diagnosen av neurodegenerative sykdommer. Forbindelsene kan merkes med avbildende midler kjent innen faget, slik som iso-tope ioner, og administreres til en pasient for å bestemme hvorvidt pasienten utviser et nedsatt antall av NMDA-reseptorer, og graden ved hvilken dette tap finner sted.
De etterfølgende eksempler er angitt for ytterligere å illustrere oppfinnelsen. De skal ikke betraktes som å be-grense oppfinnelsen på noen måte.
Eksempel 1
2- dimetylaminoetyl- 3- r2-( 2- dimetvlaminoetoksvkarbonyl)- 4, 6-diklorindol- 3- vllpropionat
0,4 g (1,12 mmol) etyl-3-(2-karboksyetyl-4,6-diklor-indol-3-yl)propionat ble oppløst i 2 ml dimetylaminoetanol. Til denne løsning ble tilsatt 2,31 g (2,24 mmol) kaliumkarbo-nat. Reaksjonskolben ble forseglet og oppvarmet til 70 °C under kraftig omrøring. Etter 24 timer ble reaksjonsblandingen avkjølt til romtemperatur, ble filtrert og fortynnet med 75 ml etylacetat. Det organiske lag ble vasket med vann og mettet
NaCl, ble tørket (MgS04) og konsentrert i vakuum. Residuet ble anbrakt på en silikagelflashkolonne og ble eluert med 5 % CH3-OH/CHCI3. Etterfølgende omkrystallisering av det isolerte produkt (etylacetat/heksan) gav fargeløse krystaller (220 mg, 44 %): sm.p. 95-97 °C.

Analyse beregnet for C20H27Cl2N3O4:
C 54,06; H 6,12; N 9,46.
Funnet: C 53,97; H 6,07; N 9,38.
Eksempel 2
2-( 4- morfolinyl) etyl- 3- T 2-( 2-( 4- morfolinyl) etoksykarbonyl)-4, 6- diklorindol- 3- yllpropionat
Til 0,65 g (4,97 mmol) 2-N-morfolinoetanol og 0,96 g (3,64 mmol) trifenylfosfin i 10 ml tørt THF ble det dråpevis tilsatt 0,50 g (1,66 mmol) 3-(2-karboksy-4,6-diklorindol-3-yl)propionsyre og 0,63! g (3,64 mmol) dietylazodikarboksylat (DEAD) i 10 ml THF. Etter 5 timer ble THF fordampet, og den resulterende fargeløse olje ble tatt opp i 50 ml CH2C12. Sili-kagel ble tilsatt til løsningen, og fordampning av løsnings-midlet gav det urene produkt absorbert på silikagelen. Dette ble tilsatt til en flashkolonne og eluert med 1 % metanol i kloroformløsning, resulterende i separasjon av 2-(4-morfolinyl)etyl-3-[2-(2-(4-morfolinyl)etoksykarbonyl)-4,6-diklor-indol-3-yl]propionat fra trifenylfosfinoksid og dihydrodietyl-azodikarboksylat. Fordampning av løsningsmidlet gav 0,68 g (78 % utbytte) av 2-(4-morfolinyl)etyl-3-[2-(2-(4-morfolinyl )-etoksykarbonyl)-4,6-diklorindol-3-yl]propionat som et hvitt, fast materiale. Dette materialet ble omkrystallisert fra en blanding av etylacetat og heksan under dannelse av 0,54 g (62 % utbytte) 2-(4-morfolinyl)etyl-3-[2-(2-(4-morfolinyl)-etoksykarbonyl)-4,6-diklorindol-3-yl]propionat som et hvitt, krystallinsk materiale, sm.p. 110-112 °C.

2- dietvlaminoetyl- 3- f2-( 2- dietylaminoetoksykarbonvl)-4,6-diklorindol- 3- ynpropionat
Metoden beskrevet ved fremstilling av 2-(4-morfolinyl)etyl-3-[2-(2-(4-morfolinyl)etoksykarbonyl)-4,6-diklor-indol-3-yl]propionat ble anvendt ved syntesen av 2-dietyl-aminoetyl-3-[2-(2-dietylaminoetoksykarbonyl)-4,6-diklorindol-3- yl]propionat under anvendelse av 0,21 g (2,40 mmol) dietyl-aminoetanol, 0,46 g (1,76 mmol) trifenylfosfin, 0,24 g (0,80 mmol) 3-(2-karboksy-4, 6-diklorindol-3-yl)propionsyre og 0,31 g (1,76 mmol) DEAD. Etter flashkromatografi ble det . isolert 0,24 g (61 %) 2-dietylaminoetyl-3-[2-(2-dietylamino-etoksykarbonyl ) -4, 6-diklorindol-3-yl]propionat som en blek, gul olje.
