Description
Hypercholesterolemia er kjent for å være en av hovedrisikofaktorene for ischemisk cardiovaskulær sykdom, slik som arteriosclerosis. Gallesyresekvestranter er blitt anvendt for å behandle denne tilstand, og disse synes å være moderat effektive, men må forbrukes i store mengder, dvs. flere gram av gangen, og de er ikke særlig velsmakende.

Fremstilling av de naturlig forekommende forbindelser og deres semi-syntetiske analoger fører til en blanding av lactonet og dihydroxysyreformer. Mevacor<®> markedsføres i lactonform, og det er således av stor betydning å anvende en høy utbyttegivende metode for lactonisering av den frie syre eller saltform. Tidligere ble lactonisering av den frie syre eller ammoniumsaltet utført ved oppvarming av disse sub-strater i et nøytralt organisk løsningsmiddel slik som toluen, ethylacetat eller isopropylacetat ved eller nær til-bakeløpstemperaturen. Lactoniseringen katalyseres ved nærvær av en syre. Den nødvendige surhet oppstår enten via den om-givende surhet av substratet i seg selv, eller ved tilsetning av en sterkere syre for å bevirke lactonisering ved en lavere temperatur.
Lactoniseringen er en likevektsprosess, og for å oppnå et høyt utbytte av lactonproduktet må det anvendes midler for å skyve likevekten til lactonsiden av ligningen:
Ved de tidligere kjente prosedyrer tvinges lactoniseringen til tilnærmet fullførelse ved å fjerne reaksjonsbiproduktene (vann, ammoniakk) fra reaksjonsblandingen ved hjelp av azeo-trop destillasjon og/eller nitrogenspyling. Fjerningen av vannet, og ammoniakk når det gjelder ammoniumsaltet, forskyver likevektsstillingen til lactonsiden.
Tidligere lactoniseringsprosedyrer utviste flere ulemper. Typisk virket hydroxysyresubstratet som syrekatalysator, og etter som substratet ble forbrukt, avtok reak-sjonshastigheten, hvilket krevet lengre reaksjonstid og muliggjorde økt biproduktdannelse. Under reaksjonsbeting-eisene ble produktet 3-hydroxylacton utsatt i lengre peri-oder for den frie syre, hvilket fører til forøkede mengder av en dimer (1) som resulterer fra en forestringsreaksjon mellom 3-hydroxylgruppen av 3-hydroxylactonet og den frie syre.
Den ovenfor angitte dimer-urenhet har vist seg å være vanskelig å separere fra det ønskede lactonderivat selv med omhyggelige omkrystalliseringsteknikker. Nærværet av dimeren nedsetter selvsagt det totale utbytte og renheten av lactonproduktet. Forsøk på å minimere dannelsen av dimeren har ført til anvendelse av høy fortynning i lactonis-eringsreaksjonen, men denne teknikk går imidlertid på bekost-ning av reaksjonens effektivitet.
En andre urenhet, resulterende fra dehydrering av 3-hydroxylgruppen på lactonringen, ble også observert under anvendelse av de tidligere kjente lactoniseringsbetingelser. Denne urenhet ble likeledes bare ueffektivt fjernet ved om-krystallisering, noe som resulterte i reduserte utbytter. Foreliggende oppfinnelse unngår dette problem.
Foreliggende oppfinnelse angår en ny fremgangsmåte for omdannelse av mevinsyre HMG-CoA-reduktaseinhibitorer eller analoger derav til et lacton. Foreliggende fremgangsmåte muliggjør en forskyvning av lactoniseringslikevekten ved kontinuerlig og selektiv fjerning av lactonproduktet fra reaksjonsblandingen. For å anvende denne nye løsning i foreliggende lactoniseringslikevekt var det nødvendig å identifisere et reaksjonsmedium hvori lactonproduktet ville være mindre løselig enn det ulactoniserte utgangsmateriale, og finne en selektiv krystalliseringsmetode for fjerning av lactonet uten bibeholdelse av prosessurenheter eller utgangsmateriale.
Oppfinnelsen angår således en fremgangsmåte for fremstilling av forbindelser av formel II:
hvori R er sekundær butyl eller 1,1-dimethylpropyl og a og b er begge dobbeltbindinger eller hvor en av a og b er en enkeltbinding, eller hvor både a og b er enkeltbindinger, hvilken fremgangsmåte er kjennetegnet ved at
A) en forbindelse av formel (I):
hvori R, a og b er som ovenfor angitt, behandles med en blanding av et vann-blandbart organisk løsningsmiddel, vann og en syrekatalysator under en inert gassatmosfære i 2-3 timer ved 20-25 °C, og B) reaksjonsblandingen behandles med ytterligere vann for å utfelle lactonproduktet (II) som en krystallinsk masse.
Spesifikt innbefatter foreliggende oppfinnelse be-handling av den frie hydroxysyre eller ammonium- eller metall-saltderivat av en mevinsyre eller analog derav i et eddiksyre-medium, eller annet vannblandbart, organisk løsningsmiddel som utviser en tilstrekkelig løselighetsforskjell mellom hydroxysyren og lactonet, og en sterk syrekatalysator. Etter at den frie hydroxysyre-lacton-likevekt er blitt oppnådd, tilsettes vann gradvis i en mengde tilstrekkelig til å bevirke fullstendig krystallisering av lactonet fra reaksjonsmediet. Denne fjerning av lactonet forskyver kontinuerlig likevekten til lactonsiden og driver således lactoniseringen tii full-førelse. I foreliggende prosedyre fjernes lactonet kontinuerlig fra reaksjonsmediet etter som det dannes, hvilket minimerer lactonets eksponering overfor reaksjonsbetingelsene og minimerer således dets mulighet for ytterligere reaksjon under dannelse av dimerer. Det oppnås således et lacton-produkt fritt for de urenheter som kompliserer de tidligere kjente lactoniseringsprosedyrer.
Foreliggende oppfinnelse kan illustreres som:
hvori R er som ovenfor angitt ; og
a og b er begge dobbeltbindinger, eller en av a og b er en enkeltbinding, eller både a og b er enkeltbindinger.
Hovedfordelene som oppnås ifølge foreliggende oppfinnelse sammenlignet med den kjente teknikk, er økt prosess-produktivitet og produktrenhet. De tidligere kjente prosedyrer utføres i et sterkt fortynnet medium (0,1 M) for å minimere dimerdannelse. Foreliggende oppfinnelse muliggjør lactonisering ved meget høyere konsentrasjoner (~0,24 M), hvilket markert forbedrer produktiviteten. Enn videre krever den kjente prosedyre et ytterligere trinn hvori løsningsmidlet konsentreres før isolering av lactonet, mens lactonet ifølge foreliggende prosedyre isoleres direkte fra reaksjonsblandingen. Igjen utviser foreliggende prosedyre en større effektivitet enn den kjente teknikk.
Med hensyn til produktrenhet ga den kjente metode et halvrent produkt som inneholdt fra 0,4 til 0,8% dimer-forurensning som var vanskelig å fjerne, mens nivåene av denne forurensning under foreliggende lactoniseringsbetingelser er redusert til mindre enn 0,2%.
De spesifikke hydroxysyrer eller salter avledet der-fra som anvendes ifølge oppfinnelsen, har struktur (I):
hvori R er som ovenfor angitt;
a og b er begge dobbeltbindinger eller en av a og b er en enkeltbinding, eller både a og b er enkeltbindinger.
De anvendte løsningsmidler er vannblandbare, organiske løsningsmidler. Det bestemte løsningsmiddel bestemmes av karakteristikaene for krystallisering, dvs. løselighetsfor-skjellen mellom hydroxysyre eller -saltet og lactonet, krystalliseringsselektiviteten og krystalliseringskinetikken i den vann-organiske løsningsmiddelblanding. Egnede løs-ningsmidler innbefatter eddiksyre, acetonitril, aceton og methanol, fortrinnsvis eddiksyre, acetonitril eller aceton.
Lactoniseringsgraden avhenger av styrken og konsen-trasjonen av syrekatalysatoren. En hvilken som helst uorganisk eller organisk syre med surhet tilstrekkelig til å katalysere dannelsen av et lacton, kan anvendes. Eksempler på egnede syrer som kan anvendes, er maursyre, fosforsyre, trifluoreddiksyre, svovelsyre, saltsyre, perklorsyre, p-toluensulfonsyre og methansulfonsyre.
Lactoniseringen kan utføres over et temperaturområde på 20 til 30°C, fortrinnsvis 20-25°C. Det er kritisk at temperaturen ikke tillates å stige over 30°C da dette fører til økt biproduktdannelse.
Lactoniseringen utføres under en inert gassatmosfære
i 2 til 3 timer.
Foretrukne metallkationer er kationer av alkali-metaller slik som natrium eller kalium, kationer av jord-alkalimetaller slik som calcium eller magnesium, eller kationer av andre metaller slik som aluminium, jern, sink, kopper, nikkel eller kobolt. Alkalimetallkationene, jordalkalimetall-kationene og aluminiumkationene er foretrukne, og natrium-, calcium- og aluminiumkationene er de mest foretrukne.
Det organiske løsningsmiddel er eddiksyre, acetonitril eller aceton, og den sure katalysator er trif luoreddiksyre eller methansulfonsyre. Når R er 1,1-dimethylpropyl, er det organiske løsningsmiddel fortrinnsvis eddiksyre, og syrekatalysatoren er trif luoreddiksyre eller methansulf onsyre, fortrinnsvis methansulfonsyre. Når R er sek.-butyl; er det organiske løsningsmiddel aceton, eddiksyre eller acetonitril, og syrekatalysatoren er trifluoreddiksyre eller methansulfonsyre.
De etterfølgende eksempler illustrerer oppfinnelsen.
Eksempel 1
Fremstilling av 6(R)-[2-[8(S)-(2,2-dimethylbutyryloxy)-2(S),6(R)-dimethyl-1,2,6,7,8,8a(R)-hexahydronafthyl-1( S)] ethyl]- 4( R)- hydroxy- 3, 4, 5, 6- tetrahydro- 2H- pyran- 2- on
12,5 g (97,9 vekt% renhet ved HPLC, 27,0 mmol) ammonium-7-[1,2,6,7,8,8a(R)-hexahydro-2(S),6(R)-dimethyl-8(S)-(2,2-dimethylbutyryloxy)-l(S)-nafthyl]-3(R),5 (R)-dihydroxyheptanoat ble omrørt med en blanding av 40 ml eddiksyre, 20 ml vann og 3,84 g (33,7 mmol, 2,5 ml) trifluoreddiksyre under en nitrogenatmosfære i 3 timer ved 23-26°C.
Etter 3 timer ble den første porsjon vann (10 ml) tilsatt på en gang. Fra dette punkt ble tilsetningen av den andre porsjon av vann (15 ml) utført jevnt over en periode på 3 timer. Lactonproduktet begynte å krystallisere ut i løpet av dette tidsrom.
Den siste porsjon vann (35 ml) ble tilsatt i løpet av 1 time. Prøven ble deretter omrørt i ytterligere 2 timer. Trifluoreddiksyren ble nøytralisert med konsentrert ammoniumhydroxyd (5,0 ml, 1,35 g NH3, 79 mmol) som ble langsomt tilsatt under avkjøling av blandingen. Reaksjonsblandingen ble deretter omrørt i 1 time, og produktet ble filtrert og vasket med ca. 100 ml eddiksyre-vannblanding (1:2 v/v), etterfulgt av vasking med 100 ml vann. Filterkaken ble tørket til konstant vekt i vakuum ved 35°C under en langsom nitrogenspyling under dannelse av tittelforbindelsen i 98,0% renhet (HPLC). Nivået av dimer var <0,2%.
Eksempel 2
Fremstilling av•6(R)-[2-[8(S)-(2,2-dimethylbutyryloxy)-2(S),6(R)-dimethyl-1,2,6,7,8,8a(R)-hexahydronafthyl-1( S)] ethyl]- 4( R)- hydroxy- 3, 4, 5, 6- tetrahydro- 2H- pyran- 2- on
12,5 g (95,4 vekt% renhet ved HPLC, 27,6 mmol) ammonium-7-[ 1, 2, 6, 7, 8, 8a (R) -hexahydro-2 (S) , 6 (R).-dimethyl-8(S)-(2,2-dimethylbutyryloxy)-1(S)-nafthyl]-3(R),5(R)-dihydroxyheptanoat og 0,075 g BHA (butylert hydroxyanisol) ble suspendert i en blanding av 40 ml eddiksyre og 20 ml vann ved 22-25°C under nitrogen. 2,94 g (2,00 ml, 30,6 mmol)
MSA (methansulfonsyre) ble tilsatt til den ovenfor angitte suspensjon.
Etter 2 timer ble 10 ml vann tilsatt i én porsjon. Fra dette tidspunkt ble tilsetningen av den andre porsjon vann (15 ml) utført jevnt over et tidsrom på 3 timer. Den siste porsjon vann (35 ml) ble tilsatt i løpet av 1 time,
og prøven ble omrørt i ytterligere 1 time. Methansulfon-syren ble nøytralisert med 4,0 ml (1,35 g NH^, 63,2 mmol) ammoniumhydroxyd som ble tilsatt langsomt under avkjøling av reaksjonsblandingen. Reaksjonsblandingen ble deretter om-rørt i 1 time, hvorpå produktet ble filtrert og vasket med ca. 100 ml eddiksyre-vannblanding (1:2 v/v), etterfulgt av vasking med 100 ml vann. Filterkaken ble tørket til konstant vekt i vakuum ved 35°C under en langsom nitrogenspyling under dannelse av tittelforbindelsen med 96,5% renhet (HPLC). Nivået av dimer var <0,2%.
Eksempel 3
Fremstilling av 6(R)-[2-[8(S)-(2-methylbutyryloxy)-2(S) ,6(R)-dimethyl-1,2,6,7,8,8a(R)-hexahydronafthyl-1(S)]ethyl]-4(R)-hydroxy- 3, 4, 5, 6- tetrahydro- 2H- pyran- 2- on
Tittelforbindelsen ble fremstilt ved å følge prose-dyren beskrevet i eksempel 1 eller eksempel 2, men under anvendelse av ammonium-7-[1,2,6,7,8,8a(R)-hexahydro-2(S),6(R)-dimethyl-8(S)-(2-methylbutyryloxy)-1(S)-nafthyl]-3(R),5(R)-dihydroxyheptanoat som forbindelsen som skulle lactoniseres. Alternativt kan aceton eller acetonitril anvendes som det organiske løsningsmiddel, men med et nøytralt løsningsmiddel ble syren nøytralisert i nærvær av en pH-sonde til pH 6.
Claims (3)
1. Fremgangsmåte for fremstilling av forbindelser av formel II:
hvori R er sekundær butyl eller 1,1-dimethylpropyl og a og b er begge dobbeltbindinger eller hvor en av a og b er en enkeltbinding, eller hvor både a og b er enkeltbindinger, karakterisert ved at A) en forbindelse av formel (I):
hvori R, a og b er som ovenfor angitt, behandles med en blanding av et vann-blandbart organisk løsningsmiddel, vann og en syrekatalysator under en inert gassatmosfære i 2-3 timer ved 20-25 °C, og B) reaksjonsblandingen behandles med ytterligere vann for å utfelle lactonproduktet (II) som en krystallinsk masse.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at den sure katalysator er trifluoreddiksyre eller methansulfonsyre.
3. Fremgangsmåte ifølge krav 1, karakterisert ved at den sure katalysator er methansulfonsyre.
