Nye 4-(2-pyrimidinyl)-l-piper-azinyl heterosykliske carbonylderivater av formel:. hvori n er et helt tall fra 2 til 5; X er radikalet. Ri og Rer hver methyl, eller danner sammen en tetramethylen- eller penta-methylenring; og farmasøytisk aksep-table syreaddisjonssalter derav, utviser terapeutisk aktivitet. Fremstilling av forbindelsene er beskrevet. (56) Anførte publikasjoner Europeisk (EP) patentsøknad, pbl. nr. 129128.
Description
Foreliggende oppfinnelse angår fremstilling av terapeutisk aktive 4-(2-pyrimidinyl)-1-piperazinyl heterosykliske carbonylderivater.
Angst er blitt definert som en frykt for en fremtidig hendelse. De fleste mennesker, om ikke alle, lider leilig-hetsvis av symptomer på angst som svar på passende stimuli. I enkelte individer kan disse følelser av angst eller panikk som svar på hverdagens livspress bli overveldende, som gjør individet til et uproduktivt medlem av samfunnet. Mens individuell gruppeterapi representerer den foretrukne primære behandlingsmåte, har anvendelse av kjemoterapeutiske midler vist seg å være et nyttig supplement ved behandling av angst, som derved muliggjør et alvorlig rammet individ å gjeninnta produktiv status mens han gjennomgår ledsagende psykoterapi.
Forbindelser av klassen benzodiazepiner er for tiden de valgte terapeutiske midler ved behandling av angst. I særdeleshet er klordiazepoxyd, diazepam og oxazepam vanlig anvendt. Denne klasse av forbindelser medfører en stor risiko for misbruk, særlig blant den klasse av pasienter som gjennomgår behandling. Enn videre utviser benzodiazepinene generelt uønsket sedativ virkning og gir skadelige inter-aksjoner med andre legemidler, innbefattende f.eks. alkohol.
Det er nå funnet en klasse av nye 4-(2-pyrimidinyl)-1-piperazinyl heterosykliske carbonylderivater som er anvendbare som angsthemmende midler, og som generelt er fri for de uønskede virkninger av benzodiazepinene. Når de her beskrevne forbindelser anvendes, hjelper de til å lindre slike symptomer som overdreven frykt, engstelse, uro, spenn-ing, stress, neurotisk depresjon, og er anvendbare for lind-ring av enkelte personlighetsforstyrrelser.
Foreliggende oppfinnelse angår en analogifremgangsmåte for fremstilling av terapeutisk aktive 4-(2-pyrimidinyl)-1-piperazinyl heterosykliske carbonylderivater av formel (1): hvori n er et helt tall fra 2 til 5; X er radikalet

Som det fremgår fra den ovenfor angitte formel (1), består forbindelsene fremstilt ved analogifremgangsmåten ifølge oppfinnelsen, hovedsakelig av to terminale heterosykliske deler adskilt av en alkylenkjede med fra 2 til 5 carbonatomer. Nærmere bestemt består en slik heterosyklisk del av 4-(2-pyrimidinyl)-1-piperazinylgruppen, mens den andre heterosykliske del representerer et tetrahydro-6H-l,3-oxazin-6-on.
Forbindelsene fremstilt ved fremgangsmåten ifølge oppfinnelsen, kan illustreres som følger:

De alkylengrupper hvori symbolet n er fra 2 til 4 carbonatomer, representerer de foretrukne alkylenkjede-lengder. Symbolene R, og R_ er hver methyl.
Analogifremgangsmåten ifølge foreliggende oppfinnelse er kjennetegnet ved at en alkylester av 3-[(N-alkyl)-ureido]-propionsyre med formel:
hvori n, og R2 er som ovenfor angitt, R er en lavere alkylgruppe, og Y er en egnet forlatende gruppe, omsettes
med l-(2-pyrimidinyl)-piperazin under dannelse av en alkylester av 3-[N-[w-[4-(2-pyrimidinyl)]-1-piperazinyl]-alkyl]-ureido]-propionsyre, angitte alkylester av (2-pyrimidinyl)-1- piperazinyl]-alkyl]-ureido]-propionsyre sykiiseres, og det ønskede 4-(2-pyrimidinyl)-1-piperazinyl heterosyklisk carbonylderivat isoleres derfra.
Således kondenseres en alkylester av 3-[(N-alkyl)-ureido]-propionsyre (2) med forbindelsen 1-(2-pyrimidinyl)-piperazin (3) under dannelse av en alkylester av 3-[N-[u»-[4-(2-pyrimidinyl)-1-piperazinyl]-alkyl]-ureido]-propionsyre (4). Disse alkylestere (4) isoleres generelt ikke, men til-lates å syklisere spontant under dannelse av de ønskede 2- [co-[4- (2-pyrimidinyl) -1-piperazinyl]-alkylimino]-tetra-hydro-6H-l,3-oxazin-6-oner (1). Denne reaksjonssekvens kan illustreres som følger, hvori symbolene n, R^ og R2 er som tidligere beskrevet, R er en lavere alkylgruppe, og Y be-tegner en egnet forlatende gruppe.
Anvendelse av løsningsmidler er foretrukket. Egnede løsningsmidler innbefatter ethvert ikke-reaktivt løsnings-middel, fortrinnsvis et polart løsningsmiddel som har et kokepunkt i området fra 60 til 150°C. Løsningsmidler som hensiktsmessig kan anvendes, innbefatter klorerte hydrocarboner slik som ethylenklorid, methylenklorid eller kloro-form; klorerte aromatiske forbindelser slik som 1,2,4-tri-klorbenzen eller o-diklorbenzen; etheriske løsningsmidler slik som tetrahydrofuran eller p-dioxan; aromatiske løs-ningsmidler slik som benzen, toluen eller xylen; eller alkoholiske løsningsmidler slik som ethanol eller butanol. Spesielt foretrukne løsningsmidler er de som er kjent for å aktivere nukleofile reaksjoner slik som dimethylsulfoxyd og dimethylformamid.
Som ovenfor angitt, kan alkylestrene (4) syklisere spontant under dannelse av de ønskede forbindelser (1). Denne syklisering fremmes ved nærvær av en katalytisk mengde av en sterk base slik som natrium-eller kalium-tert.-butoxyd, og i nærvær av et polart løsningsmiddel slik som dimethyl-sulf oxyd eller dimethylformamid.
Alkylestrene av 3-[(N-alkyl)-ureido]-propionsyre (2) kan fremstilles ved omsetning av en alkylester av 3-aminopropionsyre (5) med et alkylisocyanat (6) inneholdende en egnet forlatende gruppe som illustrert i det etterfølgende. Symbolene R, R^, R2, n og Y har de tidligere angitte betydninger .
Tilnærmet ekvimolare mengder av reaktanter (5) og (6) anvendes. Ettersom begge reaktanter er væskeformige ved romtemperatur, kan reaksjonen utføres ved enkel blanding. Fortrinnsvis anvendes imidlertid et egnet aprotisk løsnings-middel. Egnede aprotiske løsningsmidler innbefatter klorerte hydrocarboner slik som methylenklorid, ethylenklorid eller carbontetraklorid; aromatiske hydrocarboner slik som benzen eller toluen; etheriske løsningsmidler slik som diethyl-ether, tetrahydrofuran eller p-dioxan, og estere av organ-iske syrer slik som ethylacetat.
Reaksjonen er eksoterm og kan utføres ved romtemperatur under anvendelse av egnede kjølemidler. Avhengig av de anvendte reaktanter, deres konsentrasjon og de bestemte løsningsmidler som anvendes, kan reaksjonen utføres ved temperaturer fra +30°C til omgivende temperatur, i tidsrom varierende fra 30 min. til 3 timer. Fortrinnsvis anvendes temperaturer fra *20 til 0°C, og med reaksjonstider varierende fra 30 min. til en time. Fjerning av løsningsmidlene fra reaksjonsblandingen, generelt ved fordampning, gir de ønskede alkylestere av 3-[(N-alkyl)-ureido]-propionsyre, som deretter kan anvendes på den tidligere angitte måte.
Alternativt kan alkylestrene av 3-[N-[u>-[4-(2-pyrimidinyl ) -1-piperazinyl ]-alkyl] -ureido] -propionsyre (4) fremstilles via nukleofil kondensasjon av en alkylester av 3-isocyanopropionsyre av formel (7) med et oj-[4-(2-pyrimidinyl) -1-piperazinyl]-alkylamin (8) som vist i det etterføl-gende. Symbolene R, R^, R2 og n har de tidligere angitte betydninger.
Denne nukleofile kondensasjon utføres fortrinnsvis under anvendelse av ekvimolare mengder av alkylestrene av 3-isocyanopropionsyre (7) med u>-[4-(2-pyrimidinyl)-1-piper-azinyl ]-alkylamin (8) under hovedsakelig samme reaksjons-betingelser som for den tidligere beskrevne reaksjon, innbefattende de omvendte funksjonaliteter, dvs. omsetning av en alkylester av 3-aminopropionsyre (5) med alkylisocyanatet (6). Forbindelsene av formel (1) utviser anvendbare angst-dempende egenskaper. Anxiolytiske egenskaper indikeres under anvendelse av 5-HT1A in vitro-reseptorbindingsstudier, se Middlemiss et al., Eur. J. Pharmacol., 90, 151-3 (1983) og Glaser et al., Arch. Pharmacol., 329, 211-215 (1985). I tillegg kan de anxiolytiske egenskaper av de her beskrevne forbindelser demonstreres in vivo under anvendelse av rotte-slikketesten, som er en anerkjent dyremodell for angst anvendt innen faget, se Vogel et al., Psychopharmacologia, 21, 1-7 (1971).
De etterfølgende eksempler illustrerer oppfinnelsen.
Eksempel I
2-[ 4-[ 4-( 2- pyrimidinyl)- 1- piperazinyl]- butylimino]- tetra-hydro- 4, 4- dimethyl- 6H- l, 3- oxazin- 6- on
4- brombuty1i socyanat
En løsning av 36,2 g 5-brompentansyre i 15,7 ml thionylklorid ble omrørt i 2 timer ved romtemperatur, og over-skudd thionylklorid ble fjernet ved fordampning. Syreklorid-resten ble oppløst i 50 ml aceton, og løsningen ble langsomt tilsatt til en omrørt suspensjon av 13 g natriumazid i 15 ml vann ved 0°C. Etter omrøring i ytterligere 30 min. ble reaksjonsblandingen ekstrahert med ether. Etherlaget ble vasket flere ganger med en mettet natriumbicarbonatløsning, ble tørket over MgSO^ og fordampet under dannelse av 29 g 5-brompentanoylazid som en olje. En løsning av syreazidet i benzen ble kokt under tilbakeløpskjøling i 2 timer og ble deretter destillert under dannelse av 15,1 g 4-brombutylisocyanat, kokepunkt 110°C/15 mm.
Ethyl- 3-[( 4- brombutyl)- aminocarbonylamino]- 3- methylbutanoat
En løsning av 15,1 g 4-brombutylisocyanat i 50 ml methylenklorid ble tilsatt til en omrørt løsning av 12,3 g ethyl-3-amino-3-methylbutanoat oppløst i 50 ml methylenklorid ved +20°C. Blandigen ble omrørt i 30 min. ved +20°C og deretter i ytterligere 2 timer ved romtemperatur. Fordampning av løsningsmidlet gav et residuum av 26 g ethyl-3-[(4-brombutyl)-aminocarbonylamino]-3-methylbutanoat.
2-[ 4-[ 4-( 2- pyrimidinyl)- 1- piperazinyl]- butylimino]- tetra-hydro- 4, 4- dimethyl- 6H- l, 3- oxazin- 6- on
En blanding av 3,21 g 3-[(4-brombutyl)-aminocarbonyl-amino ] -3-methylbutanoat , 1,64 g 1-(2-pyrimidinyl)-piperazin, 1,38 g kaliumcarbonat og 50 ml dimethylformamid ble oppvar-met til 100°C i 3 timer, fikk stå over natten ved romtemperatur, ble filtrert og fordampet til tørrhet. Residuet ble kromatografert på kieselgel under anvendelse av en blanding av ethylacetat-ethanol (90:10) som elueringsmiddel, under dannelse av 2-[4-[4-(2-pyrimidinyl)-1-piperazinyl]-butyl-imino ]-tetrahydro-4,4-dimethyl-6H-1,3-oxazin-6-on, sm.p. 124-6°C som krystallisert fra vandig methanol (Rf 0,3).
Eksempel II
2-[ 4-[ 4-( 2- pyrimidinyl)- 1- piperazinyl]- butylimino]- tetra-hydro- 4, 4- dimethyl- 6H- l, 3- oxazin- 6- on
2- ethoxycarbonyl- l, 1- dimethylethylisocyanat
En løsning av 4,1 g triklormethylklorformiat i 15 ml methylenklorid ble langsomt tilsatt til en omrørt blanding av 3 g ethyl-3-amino-3-methylbutanoat, 6,6 g triethylamin og 30 ml methylenklorid ved 5°C. Blandingen ble omrørt over natten ved romtemperatur, ble kokt under tilbakeløpskjøling i 3 timer, ble avkjølt, hvorpå vannfri ether ble tilsatt. Det utfelte triethylamin-hydroklorid ble filtrert, og fil-tratet ble fordampet under dannelse av 3,5 g 2-ethoxycarbonyl-l , 1-dimethylethylisocyanat som en olje.
Ethyl- 3-[ 4-[ 4-( 2- pyrimidinvl)- 1- piperazinyl]- butyl]- amino-carbonylamino- 3- methylbutanoat
En løsning av 3,5 g av det ovenfor angitte isocyanat i 10 ml methylenklorid ble tilsatt til en omrørt løsning av 4,15 g 4-[4-(2-pyrimidinyl)-1-piperazinyl]-butylamin i 10 ml methylenklorid ved romtemperatur. En svak eksoterm reaksjon ble observert. Etter omrøring i 30 min. ble methylenkloridet fordampet, residuet ble oppløst i 50 ml av en én-normal løsning av HCl, og løsningen ble ekstrahert med ethylacetat. Det vandige lag ble gjort alkalisk med mettet kaliumcarbonat og ble ekstrahert med ether. Destillasjon av det tørkede etherekstrakt gav 5,4 g 3-[4-[4-(2-pyrimidinyl)-1-piperazin-yl] -bu tyl] -aminocarbonylamino- 3-methylbutanoat som en sirup.
2-[ 4-[ 4-( 2- pyrimidinyl)- 1- piperazinyl]- butylimino]- tetra-hydro- 4, 4- dimethyl- 6H- l, 3- oxazin- 6- on
En løsning av 1 g kalium tert.-butoxyd i 5 ml ethanol ble tilsatt til en løsning av 5,4 g ethyl-3-[4-[4-(2-pyrimidinyl )-1-piperazinyl]-butyl]-aminocarbonylamino-3-methylbutanoat i 10 ml ethanol, og den omrørte blanding ble kokt under tilbakeløpskjøling i 30 min. Ethanolen ble fordampet, residuet ble behandlet med 50 ml vann, og den dannede olje ble ekstrahert med methylenklorid. Fordampning av de kombinerte, tørkede methylenkloridekstrakter gav en sirup som delvis krystalliserte. Sirupen ble behandlet med ether, og krystallene ble fjernet ved filtrering (2 g). En løsning av krystallene i vandig methanol ble behandlet med carbon, ble filtrert og fikk omkrystallisere under dannelse av 1 g 2-[4-[4-(2-pyrimidinyl)-1-piperazinyl]-butylimino]-tetra-hydro-4,4-dimethyl-6H-l,3-oxazin-6-on, sm.p. 124-6°C.
Eksempel III
In vitro- bestemmelse av anxiolytiske egenskaper via 5- HT1A-binding
Radioligandbindingsstudier av 5-HT1A gjenkjennelsessteder ble utført på følgende måte. Pannebark fra normoten-sive Sprague-Dawley-hanrotter ble dissekert, fryst i flytende nitrogen og lagret ved +20°C inntil de skulle anvendes. Vev fra 4-8 rotter ble oppsamlet og homogenisert i 70 vol tris-HCl-buffer (50 mM, pH 7,7) under anvendelse av en kinematica Polytron (innstilt til 2/3 av maksimal hastig-het, 20 sekunder). Homogenatet ble sentrifugert (36500 x g i 10 min.), pelleten ble rehomogenisert i samme volum av buffer, og prosessen ble gjentatt ytterligere to ganger. Mellom andre og tredje sentrifugering ble vevhomogenatet inkubert ved 37°C i 10 min. Den sluttelige pellet ble suspendert i samme volum av tris-buffer inneholdende 10 M pargylin, 5,7 mM CaCl2 og 0,1 % askorbinsyre. Denne suspensjon ble inkubert i 10 min. ved 37°C og deretter lagret på is inntil den ble anvendt ved bindingsbestemmelsen.
0,7 ml vevhomogenat, 0,1 ml radioaktiv ligand og 0,1 ml av den egnede konsentrasjon av testforbindelse, sammen med buffer til et sluttvolum på 1 ml, ble inkubert ved 37°C i 15 min. Inkuberingene ble avsluttet ved hurtig filtrering gjennom Whatman GF/B-filtre, etterfulgt av tre 5 ml vaskinger med iskald tris-HCl-buffer (50 mM, pH 7,0). Radioaktivitet ble målt etter ekstråksjon i Aquasol-Z (NEN) til en effektivitet på 45-50 %. Radioliganden anvendt for å merke 5-HT^A gjenkjennelsessteder og dets konsentrasjon, var [<3>H]-8-hydroxy-2-(di-n-propylamino)-tetralin, [<3>H]-8-OH-DPAT, 1 mM.
Ved hovedsakelig å følge den ovenfor angitte prose-dyre, ble forbindelsen 2-[4-[4-(2-pyrimidinyl)-1-pipera-zinyl ] -butylimino ]-tetrahydro-4 ,4-dimethyl-6H-l,3-oxazin-6-on (forbindelse A) testet. Resultatene er uttrykt som pICj-Q (log^Q-konsentrasjon av testforbindelse som inhiberer spesifikk binding med 50%).

karakterisert ved at en alkylester av 3-[(N-alkyl)-ureido]-propionsyre med formel:
hvori n, R^ og R2 er som ovenfor angitt, R er en lavere alkylgruppe, og Y er en egnet forlatende gruppe, omsettes med l-(2-pyrimidinyl)-piperazin under dannelse av en alkylester av 3-[N-[uu-[4-(2-pyrimidinyl) ]-1-piperazinyl]-alkyl]-ureido]-propionsyre, angitte alkylester av (2-pyrimidinyl)-1-piperazinyl]-alkyl]-ureido]-propionsyre sykliseres, og det ønskede 4-(2-pyrimidinyl)-1-piperazinyl heterosyklisk carbonylderivat isoleres derfra.
