Description
Oppfinnelsen angår en fremgangsmåte for fremstilling av en vegetabilsk proteinekstrakt av den typen som er kjent under betegnelsen isolat (minst 85 - 90% protein) eller konsentrat (70% protein).
For fremstilling av en ekstrakt, rik på vegetabilsk protein, (isolat eller konsentrat) blir korn eller andre vegetabilske råstoffer knust til mel, hvis kornet på forhånd er behandlet med fettoppløsningsmiddel som f. eks. heksan, får man da et avfettet mel.
For fremstilling av et isolat blir avfettet mel eller fett-holdig mel ført i kontakt med enten et alkalisk medium (pH-verdi lik 7-14) eller en sur væske (pH-verdi lik 1-3,5). Den uoppløselige resten skilles fra, vaskes og vaskevannet blandes med den oppløselige fraksjon. Fraksjonen blir nøytralisert til isoelektrisk pH for proteinet som under disse betingelser ut-felles med godt utbytte og god spesifisitet: for soya vil man f. eks. ved behandling i alkalisk miljø ved en pH-verdi lik 8 og utfelling ved en pH-verdi lik 4,5, kunne overføre 80-90%
av det oppløselige protein til isolatet som får et proteininn-hold på 85-90% av tørrstoffet i det minste. Resten av tørr-stoffet består av oppløselige karbohydrater, salter og organiske stoffer bundet til proteinene. Blant disse utgjør fytinsyre (myoinositol-heksafosforsyre) 1,5-2% av tørrstoffet.
For fremstilling av et konsentrat består en fremgangsmåte i å vaske avfettet mel med vann eller en vandig oppløsning ved den isoelektriske pH-verdi og skille den proteinrike rest fra den overstående væske. Restens tørrstoff består av 70% protein og 20% uoppløelige polysakkarider samt salter og organiske stoffer hvoriblant 1,2-1,7% fytinsyre.
Proteinfraksjonen inneholder videre i begge tilfelle biologisk aktive proteiner, bl.a. trypsininhibitorer, i ikke uvesentlige mengder.
Imidlertid har nærvær av disse to stoffer, fytinsyre og trypsininhibitor negativ virkning på næringsverdien av proteinekstrakten. Trypsininhibitoren kan forårsake pankreashypertrofi og kan også være årsak til redusert veksthastighet hos unge rotter. Fytinsyren på sin side kan danne komplekser med for-skjellige essensielle metallkationer som kalsium, jern, mag-nesium, sink og nedsetter deres tilgjengelighet og opptak i mage-tarmkanalen.
Oppfinnelsen reduserer disse ulemper fordi den på særlig enkel og elegant måte muliggjør fremstilling av vegetabilske proteinekstrakter som samtidig er fattige på fytinsyre og tryptininhi-bitor og på oligosakkarider.
Foreliggende oppfinnelse angår i henhold til dette en fremgangs-imåte for fremstilling av en vegetabilsk proteinekstrakt omfattende, ved hjelp av et vandig miljø, å vaske et mel, et ca. 70%-ig proteinkonsentrat eller et 85-90%-ig proteinisolat, fortrinnsvis oppnådd fra belgfrø eller oljefrø, og deretter å separere vaskevannet og gjenvinne den uoppløselige fraksjon som ut-gjør den vegetabilske proteinekstrakten, karakterisert ved at det anvendes et vandig miljø hvor pH-verdien ligger innenfor området 5 <<> pH i 5,7.
Foreliggende fremgangsmåte skiller seg således ut fra kjente metoder ved at arbeids-pH-verdien ikke er den isoelektriske pH-verdi, men en pH-verdi som ligger vesentlig høyere, noe som fører til en svak reduksjon av det kvantitative utbyttet, d.v.s. utvunnet mengde proteiner, men som imidlertid blir mer enn komp-ensert ved forbedringen av proteinkvaliteten.
Med isoelektrisk pH-verdi mener man den isoelektriske pH-verdi for det aktuelle protein og da det her dreier seg om en bland-ing av proteiner vil den isoelektriske pH-verdi i praksis be-tegne den midlere isoelektriske pH-verdi som fremkommer ved minimal oppløselighet. Denne pH-verdi ligger ved ca. 4,5 for de fleste rå planteproteiner (f. eks. soya og bomull), d.v.s.. proteiner som ikke har gjennomgått noen spesiell behandling bortsett fra bløtning i vann. Det aktuelle pH-område strekker seg således fra 5,0 - 5,7, fortrinnsvis 5,3 - 5,5.
Man har generelt konstatert at det kvantitative utbytte reduseres med 10 - 20%, men at innholdet av fytinsyre og trypsininhibitor reduseres med en faktor 2-4, avhengig av den valgte pH-verdi. Innholdet av oligosakkarider ble også nedsatt.
Det skal nevnes her at proteinekstrakter med lavt innhold av
i det minste én av disse uønskede stoffer allerede er fremstilt, men da på basis av enzymatiske behandlinger eller innviklede tekniske prosesser som ultrafiltrering eller ved inkubering av utgangsstoffet over lengre perioder ved temperaturer over romtemperatur. Dette i motsetning til den foreliggende prosess-ens enkelhet.
Som kjent kan utgangsstoffene for fremstilling av proteinekstrakter med godt utbytte være planter som soya, solsikke, mais, hestebønner, limabønner, ricinusfrø, bomullsfrø, lupin, sesam-frø, jordnøtt, kubønner (Vigna sinensie), gråerter etc. og generelt belgplanter og oljefrøplanter. Når det gjelder de, meltyper og fortrinnsvis de fettfrie mel som benyttes som ut-gangsstof fer , er disse omtalt tidligere i innledningen i lik-het med konsentrater eller isolater som skal gjennomgå vaske-behandling. De sistnevnte er følgelig tradisjonelt fremstilt ved en isoelektrisk pH-verdi.
pH-verdien som skal ligge mellom 5,0 og 5,7, fortrinnsvis
mellom 5,3 og 5,5, kan innstilles ved hjelp av ethvert egnet reagens, f. eks. saltsyre, fosforsyre, acetatpuffere eller med natriumkarbonat, kaliumkarbonat eller karbonat-puffere, alt etter ønske. pH-verdien er den bestemmende faktor som mulig-gjør fremstilling av en ønsket proteinekstrakt med hensyn på kvalitet og kvantitet. Når det f. eks. gjelder soya, vil innholdet av fytinsyre og trypsininhibitor ved pH-verdier under 5,0 ikke lenger synke særlig og ved pH-verdier over 5,7, vil de kvantitative tap bli for store og innholdet av trypsininhibitor blir igjen større.
Utførelse av oppfinnelsens fremgangsmåte krever ingen spesiell apparatur eller reagens og heller ingen forandring av eksister-ende produksjonslinjer.
Separasjonene av oppløselige og uoppløselige fraksjoner kan skje på kjent måte ved filtrering, dekantering, sentrifugering eller på annen måte. Naturligvis kan de dannede uoppløselige fraksjoner vaskes med vann eller fortrinnsvis med en vandig oppløsning med en pH-verdi mellom 5,0 og 5,7.
Proteinekstraktene kan brukes som sådan eller etter nøytrali-sering, eventuelt etter standardisering av sammensetningen. Ekstraktene kan også tørkes (f orstøvning, frysetørring etc). Proteinekstraktene finner anvendelse som næringsmiddel, særlig for mennesker og spesielt som diettbestanddel. Næringsverdien er vesentlig forbedret.
De følgende eksempler illustrerer oppfinnelsen. I eksemplene er mengdeforhold på vektbasis og tallverdier ifølge oppfinnelsen er notert med en "<*>" i sammenligningstabeller.
Eksempel 1
Soyakonsentrat
På basis av soyamel, befridd for kapsler og fett, fremstilles et konsentrat ved å suspendere melet i vann i en mengde på
100 g mel i 1900 g destillert vann. Suspensjonen røres ved romtemperatur i 1 time og man deler blandingen i 6 like fraksjoner på hver 300 g, og innstiller pH-verdien i fraksjonene med saltsyre til pH 4,5, 5,0, 5,3, 5,5, 5,7 og 6,0. Fraksjonene sentrifugeres med 3000 g i 15 minutter og bunnfallet frysetørkes. Mengde og sammensetning av disse tørre fraksjoner gjengis i tabellen nedenfor.
Mengde utgangsstoff: 15 g pr. prøve
Det er klart at konsentrater fremstilt ved pH-verdier mellom 5,0 og 5,7 er bedre enn de som er fremstilt ved pH 4,5 uten at tapet blir for stort som ved pH 6,0.
Eksempel 2
Soyaisolat ( ved vasking) og soyakonsentrat
Man vasker 10 g av nedenstående produkter med vann ved pH 5,5 i en mengde på 25-100 g vann pr. 5 g produkt. Den dannede suspensjon sentrifugeres ved 3000 g i 15 minutter, de uopp-løselige stoffer oppsamles og tørkes.
Eksempel 3
Konsentrat av helsoya
Man fremstiller et konsentrat av helsoya ved å suspendere 10g skallfri og malte soyabønner i 190g vann og innstiller pH til 5,5. Den uoppløselige fraksjon fraskilles ved sentrifugering ved 2000g i 10 minutter, det oppflytende fettet tilsettes bunnfallet fra sentrifugeringen. Man får etter tørking et konsentrat av helsoya (6g) som inneholder 51% proteiner av totalt tørrstoff, dvs. 68% av fettfritt tørrstoff og 0,31% fytinsyre.

1. Fremgangsmåte for fremstilling av en vegetabilsk proteinekstrakt omfattende, ved hjelp av et vandig miljø, å vaske et mel, et ca. 70%-ig <p>roteinkonsentrat eller et 85-90%-ig proteinisolat, fortrinnsvis oppnådd fra belgfrø eller oljefrø, og deretter å separere vaskevannet og gjenvinne den uoppløselige fraksjon som utgjør den vegetabilske proteinekstrakten, karakterisert ved at det anvendes et vandig miljø hvor pH-verdien ligger innenfor området 5 ^ pH - 5,7.
2. Fremgangsmåte ifølge krav 1 , karakterisert ved at det anvendes et vandig miljø med en pH-verdi mellom 5,3 og 5,5.
