Description
Fremgangsmåte for fremstilling av terapeutisk virksomme tetrahydroisokinolin-derivater.
Foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av tetrahydroisokinolin-derivater med den generelle formel:
i hvilken R, betegner lavere alkyl, lavere alkoxy, nitro eller halogen, R2 lavere alkyl eller lavere alkenyl og R:1 lavere alkoxy, lavere aralkoxy eller hydroxy, hvor to substituenter R,, i nabostilling også kan danne en lavere aikylendioxygruppe, og m og n har verdien 0, 1, 2 eller 3, deres ethere og estere, og salter av disse forbindelser.
Fremgangsmåten karakteriseres ved at man reduserer en tetrahydroisokinolinforbindelse med den generelle formel: i hvilken substituentene R,-R:) så vel som m og n har ovenstående betydning, eller et salt av denne og, hvis ønsket, forethrer eller forestrer, og overfører de således erholdte baser til et salt.
De i ovenstående formler forekom-mende alkylrester R,, R2 og R;! er f. eks.
lavere alkylrester, slik som methyl-, ethyl-, isopropyl-, butyl- eller heptylresten. Eks-empler på alkoxy- og lavere aralkoxygrup-per er lavere alkoxygrupper, slik som me-thoxy, ethoxy, propoxy, butoxy, henholds-vis benzyloxy, fenethoxy. Under betegnel-sen alkylendioxy faller f. eks. methylen-dioxy- og ethylendioxygruppene. Substituentene R, og R.t, i tilfelle halogen, kan være fluor, klor, brom eller jod. Vinyl, allyl, methallyl eller dimethallyl er eks-empler på alkenyl-substituenter av resten
R2. Salter av forbindelsene i formelen II er
f. eks. addisjonssalter med syrer, slik som mineralsyrer, dvs. hydrohalidene (som hy-droklorider, hydrobromider), sulfatene, fos-fatene osv.
Ethere av forbindelsene i den generelle formel I er f. eks. alkylether (som methyl, ethyl-, butylether), alkenylether (som allylether) eller også aralkyl- (som benzyl-, fenethylether) eller arylether (som fenylether).
Blant estere av forbindelsene med den generelle formel I forstås f. eks. de pro-dukter som acylrest har en alkanoylrest (f. eks. acetyl-, propionyl-, butyryl-, pival-oylresten), aralkanoylrest (f. eks. fenace-tylrest) eller aroylrest (f. eks. benzoyl-eller en substituert benzoylrest, som f. eks. en p-nitrobenzoylrest, veratroylrest eller trimethoxybenzoylrest).
Reduksjonen som skal utføres i hen-hold til oppfinnelsen av oxo- til hydroxy-gruppen kan utføres efter på i og for seg kjente metoder.
Hensiktsmessig utføres reduksjonen f. eks. ved hjelp av alkalimetall-metallhy-drider, slik som lithiumaluminiumhydrid, eller særlig natriumborhydrid, kaliumbor-hydrid. En foretrukket utførelsesform be-står i at man reduserer med natriumborhydrid, fortrinnsvis i et egnet oppløsnings-middel som er bestandig overfor reduk-sjonsmidlet, slik som et lavere alkanol, f. eks. methanol eller ethanol, eller i di-methylformamid. Reduksjonen foregår al-lerede i kulden. Denne reduksjonsmåte kan også utføres når nitrogrupper eller, alkenyl-rester er tilstede, hvilke ved ovenstående reduksjonsmidler ikke angripes. Efter ut-ført reduksjon kan eventuelt tilstedevæ-rende aralkyloxyrester R;!, derav særlig benzyloxyresten, spaltes ved hydrogeno-lyse, hvorved den frie hydroxygruppe dannes. Denne debenzylering utføres fordel-aktig katalytisk, f. eks. i nærvær av en edelmetallkatalysator, slik som palladiumkull.
I et ytterligere fremgangsmåte-trinn kan forbindelsene i formel I forethres eller forestres. Forestringen kan utføres ved be-handling med vanlige acyleringsmidler, f. eks. carboxylsyre, carboxylsyreanhydrider eller -halogenider. Til foretring tjener f. eks. diazoalkaner, alkylhalogenider, alke-nylhalogenider, aralkylhalogenider eller arylhalogenider.
Fremgangsmåteproduktene er basiske, oftest krystalliserte forbindelser, som med de anvendelige anorganiske eller organiske syrer f. eks. med halogenhydrogen-syre, som saltsyre, bromhydrogensyre eller med svovelsyre, fosforsyre, vinsyre, sitron-=yre osv. danner mest krystalliserte, i vann oppløselige salter.
De dannede tetrahydroisokinolin-derivater med den generelle formel I, har to asymmetriske carbonatomer. Således er dannelsen av to stereoisomere racemater mulig, hvilke, hvis de forekommer ved si-den av hverandre i form av en blanding, kan skilles efter på i og for seg kjente metoder, f. eks. kromatografi eller fraksjonert krystallisasjon. Racematene på sin side kan, hvis ønsket, oppskilles under anven-delse av på i og for seg kjente metoder, f. eks. ved fraksjonert krystallisasjon av sal-tene med optisk aktive syrer, som D-vinsyre, di-benzoyl-D-vinsyre, D-kamfersul-fonsyre, i deres optiske antipoder. Ved reduksjon av optisk aktivt utgangsmateriale med formelen II får man direkte optisk aktive sluttforbindelser.
Fremgangsmåteproduktene er i besid-delse av verdifulle farmakologiske, særlig analgetiske, spasmolytiske og hostelind-rende egenskaper.
I sammenligningsforsøk, hvilke ble gjennomført på mus efter metode av Gross, ble konstatert at de efter fremgangsmåten ifølge oppfinnelsen oppnåelige fenyl-hydroxyethyl-forbindelser er de analoge fenethylforbindelsene overlegne, så vel med hensyn til den absolutte analgetiske virk-ning som også den therapeutiske indeks.
Eksempel I.
30 g l-p-klorfenacyl-2-methyl-6,7-di-methoxy-l,2,3,4-tetrahydroisokinolin opp-løses i 1000 ml methanol. Til denne opp-løsning tilsetter man under iskjølning og under stadig omrøring porsjonsvis ialt 6 g natriumborhydrid. Efter at hydrogenut-viklingen er opphørt lar man det henstå i to timer ved rumtemperatur og fjerner til slutt oppløsningsmidlet i vannstråleva-kuum. Man tilsetter vann til resten og opp-tar de organiske andeler i 200 ml ether. Man dekanterer den etheriske oppløsning, lar den henstå i 15 timer i kulden og av-filtrerer derpå av de utfelte krystaller. For rensning omkrystalliseres de fra ed-dikester/petrolether. Man erholder på denne måte l-hydroxy-lp-klorfenyl-2-[2-methyl-6,7-dimethoxy-l,2,3,4-tetrahydroisokinolinyl-(l)]-ethan i form av en av de to stereoisomerer med smeltepunkt 120
—121° C. Til utvinning av den annen stereoisomer konsentrerer man det etheriske
filtrat, oppløser resten i aceton og tilsetter en oppløsning av 9 g vannfri oxalsyre i aceton. Efter tilsetning av ether krystalli-serer den annen stereoisomer i form av det sure oxalat, hvilket smelter efter omløs-ning fra methanol/ether ved 123—125° C. Den frie base som erholdes efter oppløs-ning av det sure oxalat med fortynnet natronlut og efter vanlig opparbeidelse, er en f arveløs olje.
Også efter reduksjon av fenacylfor-bindelsen med lithiumaluminiumhydrid i kokende tetrahydrofuran erholdes de ovenfor beskrevne stereoisomerer.
Eksempel II.
l-p-klorfenacyl-2-methyl-6,7-dime-thoxy-l,2,3,4-tetrahydroisokinolin-hydroklorid (smp. 152°) reduseres efter angivelsene i eksempel I i methanol med natriumborhydrid, og reduksjonsblandingen opparbeides tilsvarende ovenstående an-givelser, hvorved man erholder det sure oxalat av det annet stereoisomer som er beskrevet i eksempel I, med smeltepunkt 123—125°.
Eksempel III.
13 g l-p-nitrofenacyl-2-methyl-6,7-dimethoxy-l,2,3,4-tetrahydroisokinolin
(smp. 120°) oppløses i 300 ml. dimethyl-formamid. Til denne oppløsning tilsetter man porsjonsvis 2 g natriumborhydrid under stadig omrøring. Man lar det stå i 15 timer ved romtemperatur, konsentrerer derpå oppløsningen i vakuum og tilsetter resten forsiktig med fortynnet saltsyre. Efter vasking med ether stilles den saltsure opp-løsning med natronlut alkalisk, for å opp-ta de frigjorte basiske andeler i ether. Efter 15 timers henstand utkrystalliserer fra den etheriske oppløsning 1-hydroxy-l-p-ni-trofenyl-2-[2-methyI-6,7-dimethoxy-l,2,3,
4-tetrahydroisokinolinyl-(l)]-ethan i form av en av de to steroisomerer, hvilket efter omløsning fra isopropylether smelter ved 150—151°. Det annet stereoisomere kan utvinnes fra den etheriske moderlut efter konsentrasjon, oppløsning av resten i aceton og tilsetning med oxalsyre i form av det sure oxalat, hvilket efter omløsningen fra methanol/ether smelter ved 121—122°. Det fra det sure oxalat over den frie base fremstilte hydrobromid smelter ved 126—128°.
Eksempel IV.
Efter angivelsene i eksempel I reduseres l-p-klorfenacyl-2-methyl-6-benzyloxy-7-methoxy-l,2,3,4-tetrahydroisokino-Iin (smp. 93°) til 1-hydroxy-l-p-klorfenyl-2-[2-methyl-6-benzyloxy-7-methoxy-l,2,3,4-tetrahydroisokinolinyl-(l)]-ethan. Smeltepunktet for den ene isomer ved 130°. Smeltepunktet for den annen isomer som hydroklorid ved 186°.
I et ytterligere reduksjonstrinn av-spaltes benzylgruppen hydrogenolytisk som følger: 700 mg av isomeren med smeltepunkt 130° oppløses i 50 ml methanol og hydreres over 100 mg 5 pst.'s palladiumkull. Efter opptagelse av den nødvendige mengde hydrogen avfiltreres fra katalysatoren, filtratet konsentreres noe, tilsettes alkoholisk saltsyre og derpå ether inntil blakk - het. Derved utskiller hydrokloridet seg fra l-hydroxy-l-p-klorfenyl-2-[2-methyl-6-hydroxy-7-methoxy-l,2,3,4-tetrahydroisokinolinyl-(l)]-ethan, hvilket avfiltreres og tørres. Smp. 178—179°. U.V. absorp-sjonsmaksima i finsprit ved 284 mjx (e = 3790); i 0,01 alkoholisk natronlut ved 247 —299 rti(x e = 9400 og 4850).
Eksempel V.
2,5 g av de efter eksempel I erholdte isomerer med smeltepunkt 120—121° opp-løses i 10 ml pyridin. Efter tilsetning av 10 ml acetanhydrid lar man blandingen henstå i en time ved vannbadtemperatur. Derpå konsentrerer man i vakuum, oppløser resten i benzol, konsentrerer den benzol-iske oppløsning igjen og tilsetter resten ether. Den etheriske oppløsning vaskes med vann, tørkes og konsentreres. Resten oppløses i aceton og tilsettes en oppløsning av vannfri oxalsyre i aceton, hvorved det sure oxalat av l-acetoxy-l-p-klorfenyl-2-[2-methyl-6,7-dimethoxy-l,2,3,4-tetrahydroisokinolin-(l)]-ethan ved tilsetning av ether bringes til utskillelse. Det dannes nåler med smeltepunkt 80—82°.
På lignende måte erholdes fra isomerer med smeltepunkt 120—121° med pro-pionylklorid i pyridin det amorfe 1-propio-nyloxy-lp-klorfenyl-2-[2-methyl-6,7-di-methoxy-l,2,3,4-tetrahydroisokinolinyl-(1)]-ethan, hvis hydroklorid oppviser smeltepunktet 225—226°. Analogt hertil fremstilles med benzoylklorid-l-benzoyloxy-1-klorfenyl-2-[2-methyl-6,7-dimethoxy-l,2, 3,4- tetrahydroisokinolinyl- (1) ] -ethan, hvis sure oxalat smelter ved 136—137°. Smeltepunktet for hydrokloridet 185—186. l-[|3-(4-klorfenyl)-propionyloxy]-l-p-klorfenyl-2-[2-methyl-6,7-dimethoxy-l,2,3,4-tetrahydroisokinolinyl-(l)]-ethan-hydroklorid: smeltepunkt 175—177°. l-(pivaloyloxy)-l-p-klorfenyl-2-[2-methyl-6,7-dimethoxy-l,2,3,4-tetrahydroisokinolinyl- (1) ] -ethan-hydroklorid:
smeltepunkt 198—201°.
Efter angivelsene i eksempel I kan fra de tilsvarende fenacylforbindelser de følg-ende reduksjonsprodukter fremstilles: 1-hydroxy-l-p-klorfenyl-2-[2-allyl-6,7-di-methoxy-1,2,3,4-tetrahydr oisokinolinyl-(l)]-ethan med smeltepunkt 122°. (Smeltepunktet for utgangsmaterialet: 72°). l-hydroxy-l-fenyI-2-[2-methyl-6,7-dimethoxy-l,2,3,4-tetrahydroisokinolinyl-(l)]-ethan med smeltepunkt 128°. (Smeltepunktet for utgangsmaterialet: 80—81°). 1 -hydroxy-1 - (3,4-diklor f enyl) -2- [2-methyl-6,7-dimethoxy-l,2,3,4-tetrahydroisokinolinyl- (1) ] -ethan. Smeltepunktet for den ene isomer: 136°. Smeltepunktet for den annen isomer som sur oxalat: 118 —120°. (Smeltepunkt for utgangsmaterialet: 149°). l-hydroxy-l-p-methoxyfenyl-2-[2-methyl-6,7-dimethoxy-l,2,3,4-tetrahydroisokinolinyl-(l)]-ethan med smeltepunkt 130°. (Smeltepunktet for utgangsmaterialet: 92°). l-hydroxy-l-p-tolyl-2-[2-methyl-6,7-dimethoxy-l,2,3,4-tetrahydroisokinolinyl-(l)]-ethan med smeltepunkt 127°.
(Smeltepunktet for utgangsmaterialet: 116°). l-hydroxy-l-p-klorfenyl-2-[2-methyl- 6,7 - methy lendioxy- 8- me thoxy-1,2,3, 4-tetrahydroisokinolinyl- (1) ] -ethan-hydroklorid med smeltepunkt 192—194°. (Smeltepunkt for utgangsmaterialet: 113°). 1 -hydroxy-1 - (2-ethyl-4,5 -dimethoxy-fenyl)-2-[2-methyl-6,7-dimethoxy-l,2,3,
4-tetrahydroisokinolinyl-(l)]-ethan i form
av en oljeaktig isomerblanding. (Smeltepunktet for utgangsmaterialet: 110—111°).
Claims (2)
1. Fremgangsmåte til fremstilling av terapeutisk virksomme tetrahydroisokinolin-derivater med den generelle formel
En forsøksrapport av 3. mars 1965 var
hvor R, er lavere alkyl, lavere alkoxy, nitro eller halogen, R2 betegner lavere alkyl eller lavere alkenyl og R;1 lavere alkoxy, lavere aralkoxy eller hydroxy, hvor to substituenter R3 i nabostilling også kan danne en lavere alkylendioxygruppe, og m og n betyr verdien 0,1,2 eller 3, deres ethere og estere, og av salter av disse forbindelser, karakterisert ved at man reduserer en tetrahydroisokinolinforbindelse med den generelle formel
hvor R2, R3, m og n har den ovenfor nevn-te betydning, eller et salt av den samme og hvis ønsket forethrer eller forestrer reaksjonsproduktet og overfører en således erholdt base eventuelt til et salt.
2. Fremgangsmåte ifølge påstand 1, karakterisert ved at man reduserer l-p-.klorfenacyl-2-methyl-6,7-dimeth-oxy-l,2,3,4-tetrahydroisokinolin eller 1-p-mtrofenacyl-2-methyl-6,7-dimethoxy-1,2,3,4-tetrahydr oisokinolin.
