De uitvinding heeft betrekking op een N-(2-gesubstitueerde alkyl)-N ’-£( imidazol-4-yl)alkyl]guanidine.
Impromidine , ofwel N-/2-6-niethy Timidazol^-ylrEthyl-thio)ethyl/-XT-£-(iinidazol-4-yl) propyl/guanidine is bekend als 5 een specifieke en de meest potente histamine H^-agonist.
Afhankelijk van het gebruikte testsysteem gedraagt het zich hetzij als partiele hetzij als volledige agonist met een potentie van 5-800 maal die van histamine (Proc.VIIIth Internat.Symp.Med.
Chem.Uppsala , blz. 202-203 (1985) Eds R. Dahlbom and J.L.G.
10 Nilsson). Vanwege zijn effekt op de afgifte van histamine uit mestcellen zou men enig gebruik van impromidine kunnen maken bij de behandeling van allergische aandoeningen. Een groot nadeel voor het klinisch gebruik van impromidine is echter zijn betrekkelijk grote potentie ten aanzien van de maagsapsecretie 15 en zijn effekt op vasoconstrictie en vasodilatie.
Er werd nu een reeks nieuwe aan impromidine verwante verbindingen gevonden die een sterke histamine H?-agonistische werking op het rechter atrium van het guinese biggetje vertonen naast een betrekkelijk geringe werking 20 op de maagzuursecretie van het guinese biggetje en een potent histamine -antagonisme, als beproefd aan zowel het ileum van het guinese biggetje als de trachea van het guinese biggetje.
Vanwege deze combinatie van histamine -antagonisme en I^-agonisme in één verbinding zijn deze verbindingen van klinische 25 betekenis voor bijvoorbeeld de behandeling van congéstieve hartgebreken en sommige allergische aandoeningen.
Deze nieuwe verbindingen zijn N-(<ü-gesubstitu-eerde alkyl)-N’-Ztimidazol-4-yl)alkyl/guanidinen met de formule 1, waarin: 30 85 C15 8 5 * - 2 - m 1, 2 of 3 is; η 2 of 3 is; X a) S, 0 of CH2 is, waarbij Y een met R gesubstitueerde difenylmethyl-groep of (10,11-dihydro)-5H-dibenzo-/a, d7“cyclohepten-5-ylgroep is, of b) N is , waarbij *1 Y, een met R gesubstitueerde fenylgroep 10 .1 is en Y eveneens een met R gesubstitueerde fenylgroep is, waarbij Y en Y^ niet gelijk gesubstitueerd behoeven te zijn of Y een met R gesubstitueerde benzvlgroep is, of 15 v ..
c; =CH- is, waarbij Y een met R gesubstitueerde difenyl-methylyleengroep of (10,11-dihydro)-5H-dibenzo-/a,d7~cyclohepten-5-ylideengroep is, 20 R H, alkyl, alkoxy, halogeen en/of trihalo-• geenmethyl is, met dien verstande , dat van alle betrokken fenylringen er een of meer vervangen kunnen zijn door een met R gesubstitueerde heterocyclische aromatische ring en hun zuuradditiezouten.
Eventueel aanwezige, met R gesubstitueerde heterocyclische aromatische ringen zijn bijvoorbeeld: 2-, 3- en 4-pyridiny], 4-imidazolyl, 4-thiazolyl,2-guanidino-4-thiazolyl, 2- en 3-30 furanyl, 2-dimethylaminomethy1-5-furanyl,enz, 3801585 -2a- 0 «.
De resultaten van farmacologische proeven met deze nieuwe verbindingen zijn samengevat in tabel A (H^-werking) en tabel B (^“werking )· De waarden voor de H1-werking worden gegeven als het gemiddelde van twee experimenten in quadruplo, 5 terwijl de waarden voor de ^-werking zijn verkregen uit tenminste twee experimenten in duplicaat.
In tabel A zijn de beproefde verbindingen gedefinieerd met eai formule , waarbij de betekenis van en n in de formule in de tabel is weergegeven. De verbindingen 10 zijn voorts van een volgnummer voorzien, dat overeenkomt met de in tabel 3 gebruikte volgnummers. Opgemerkt wordt, dat de laatste drie verbindingen van beide tabellen niet onder de uitvinding vallen maar ter vergelijking worden vermeld.

Discussie Η .j-werking
Er treedt geen verandering in -werking op als men het 5-methylimidazoolgedeelte van impromidine verandert in een fenylgroep (verbinding XII). Invoering van nog een extra fenylgroep in verbinding XEI resulteert echter in een toeneming van pA£ van 5,5 naar 6,5 (verbinding I) bij het ileum van het guinese biggetje . Verkorting van de trimethyleehketen in verbinding I tot een ethyleenketen (verbinding Π) heeft geen effekt op de 10 H.j-werking. Hetzelfde ziet men bij vergelijking van verbinding IV met verbinding V en verbinding VU net verbinding IX.
Vervanging van het zwavelatoom in verbinding I door zuurstof (verbindirg III) leidt tot een 10 maal zo grote potentie vergeleken met de uitgangsstof ten aanzien van het 15 ileum van het guinese biggetje en zelfs tot een 25 maal zo grote potentie ten aanzien van de trachea.
Invoering van een p-methylgroep (verbindingen IV en V) of een p-fluorgroep (verbinding VI) heeft weinig of geen effekt op het H.-antagonisme.

Ook verbindingen VII,VIII,IX en X zijn bijna even potent als -antagonist als verbinding I. Verbinding XI schijnt zelfs een nog wat potentere ^-antagonist te zijn dan het zuurstofanalogon verbindingn.
25 H0-werking
Vervanging van de 5-methylimidazoolgroep van impromidine door een fenylgroep (verbinding XII.) gaat gepaard met een 4- voudige afneming in ^-werking op het atrium van het 3Q guinese biggetje en een 8-voudige afneming van de werking op de maagfundus. Invoering van een extra fenylgroep in verbinding XII (verbinding I) leidt tot een 2- a 3-voudige vermindering in ^-agonisme op het atrium en een zelfs 32-voudige afneming 8S 0 1 5 8 5 - 6 - in Η-,-agonistne op de maag fund us.
Am
Verkorting van de trimethy leen keten van verbinding I tot een ethyleenketen (verbinding II) resulteert in een opmerkelijke afneming in H9~werking, zowel op het atrium als op de 5 raaagfiindus. Analoge resultaten zijn verkregen bij verbinding IV ten opzichte van verbinding V en verbinding VTTT ten opzichte van verbinding XX.
Vervanging van het zwavelatoom in verbinding X door een zuurstofatoom (verbinding III)resulteert in een 20-voudige 10 afneming in H0-agonisme op het atrium. Op de maagftndus is de verandering in pD9 minder uitgesproken, maar in dit proefsysteem gaat deze verandering in structuur gepaard met een opmerkelijke afneming van de intrinsieke aktiviteit.
Ook de invoering van een p-methylgroep (verbin-15 ding IV en V) of een p-fluorgroep (verbinding VI) resulteert in een sterke afneming in H9-werking,
Verbinding VEC bleek als H9-antagonist even potent te zijn als de p-methyl- en p-fluoranalogen.
Als men het zwavelatoom in verbindingen I en II 20 weglaat, verkrijgt men de difenylpropylanaloga , verbindingen VIH en E. Deze verbindingen vertonen een opmerkelijk grote potentie op het rechter atrium van het guinese biggetje. Verbinding IX bleek op het atrium bijna even potent te zijn als impromidine.
Invoering van een dubbele binding resulteert 25 in een 4-voudige afneming in H2-wer7{CinS °P ^et atrium (verbinding X) . Het aminoanaloog , verbinding XI, bleek ongeveer 2 maal zo potent als histamine te zijn op het atrium en ongeveer 4% van de werking van impromidine te hebben in dit proefsysteem. Op de maagbodem heeft deze verbinding ongeveer 1% van de werking 30 van impromidine en 10 maal de werking van histamine.

GevoIgtrekkingen
Het zuurstofanaloog , verbinding m, an het amino-analoog , verbinding XI, bleken de meest potente -antagonisten uit deze reeks impromidineanalogen te zijn.
Verbindingen I, IX en X. bleken de meest potente verbindingen uit deze reeks op het rechter atrium van het guinese biggetje te zijn.
Verbindingen Ι,ΙΧ, X ai XI verdienen vanwege hun combinatie van eigenschappen de meeste voorkeur.

De farmacologische proeven werden als volgt uitgevoerd:
Trachea guinees biggetje (H^)
Mannelijke guinese biggetjes (350-500 g) werden gedood door een tik op de kop en de trachea werd verwijderd.

Uit de trachea werden enkele segmenten gesneden, met 0,4 g verzwaard en geplaatst in een orgaanbad , (35°C), dat 120 mM NaCl, 6 mM KC1 , 1 mM MgSO^, 2,5 mM CaCl2, 1 mM NaH?P04, 2,5 mM NaHC03 en 6 mM glucose bevatte. Men begaste het orgaanbad met zuurstof, die 5% C09 bevatte. pD^,intrinsieke werking en antagonistische on werking van de te beproeven verbindingen werden bepaald uit iso- toon opgenomen cumulatieve dosisresponskrommen.
De H.j-specificiteit van het orgaan is vastgesteld door blokkering van de door histamine opgewekte contracties van de trachea met mepyramine. Bovendien bewezen zowel de -3 23 specifieke histamine ^-agonist dimaprit (tot 10 M) en de specifieke histamine ^-antagonist cimetidine (tot 10 Sl) geen effekt te hebben op de rusttoestand van het orgaan en ook niet op door histamine opgewekte contracties.

De histamine H7-werking van de zuursecretie van de geïsoleerde mmgfundus van het guinese biggetje is bepaald volgens Impicciatore et al. Eur.J.Pharmacol. ,48_,249-254(1975) .
13 Synthese • De onderhavige verbindingen worden bereid volgens reaktieschema A of B . In deze reaktieschema's komt overeen met de desbetreffende groep Y-X-(CH?) - in de formule 1.
De primaire aminen, die bij stap i en stap 5 20 worden gebruikt, kunnen worden bereid volgens in de literatuur beschreven methoden of zijn in de handel verkrijgbaar (3,3-difenylpropylamine).
De reaktie van de primaire aminen met benzoyl-isothiocyanaat tot de benzoylthioureumderivaten ( staD 1) 25 verliep met hoge opbrengsten (66-88 Z). De hydrolyse van deze benzoylthioureumderivaten (stap 2) verliep ook met hoge opbrengsten (80-93%). De isothioureums, die resulteren uit de reaktie van de thioureums met methyljodide ( stap3) werden niet geïsoleerd, 30 maar men liet ze rechtstreeks na verdamping van de overmaat methyl-jodide met 4-(3-aminopropyl)imidazool of 4-(2-aninoethyl)imidazool reageren (stap4). Deze laatste reaktie gas—slechte op- 88 CM 3 S 5 # *· - 9 -
De verbindingen met een basische groep in het "I^-gedeelte” ( verbindingen VEE en XI) werden gesynthetiseerd 3 via hun overeenkomstige cyaanguanidine. Men liet de primaire aminen eerst met dimethylcyaaniminodithiocarbonaat reageren (stap 5). Deze reaktie verliep zeer goed en resulteerde in hoge opbrengsten aan isothioureums (ongeveer 70%) . Men liet de N-cyaanisothioureums ma:4 (et-aainopropyl)imidazool tot de cyaan-^ guanidinen reageren ( stap 6). Deze reaktie gaf slechte opbrengsten (10-20%).
De hydrolyse van deze cyaanguanidinen tot de eindprodukten (verbindingen VII en XI gaf een bijna 15 kwantitatieve opbrengst. Deze produkten werden gezuiverd als hun tripikraten teneinde het bij deze hydrolyse gevormde ammonium-chloride te verwijderen.

Algemene werkwijze voor de synthese van 2-
Zr<^-fenyl-4-methvl/benzvlthio7ethvlamine (An) en 2-[&- fenvl-4- 30 ---:-:-*--- fluorbenzylthio_/ethylamine.HCl (A^).
Verbindingen A^ en zijn bereid in analogie 86 0 1 5 8 5 *' «* - 10 - met de voor de bereiding van verbinding genoemde methode.
Men verwarmde een oplossing van 0,2 mol van de gewenste gesubstitueerde benzhydrol, 0,2 mol cysteamine en 28 ml boriumtri-fluorideetheraat in 200 ml azijnzuur 1 uur aan een terugvloei-5 koeler. Na afkoeling dampte men de oplossing in en herkristalli-seerde het residu uit propanol-2/ether.
Resultaten: A^ :de vrije base werd onder verlaagde druk gedestilleerd, kookpunt _ .:115-120°C.

Men voegde een oplossing van ongeveer 20 g van de vrije base van het overeenkomstige amine (A.-AJ in 100 ml 5 ° CHCl^ langzaam toe aan een oplossing van een equimolaire hoeveelheid benzoylisothiocyanaat in 100 ml CHCl^. De resulterende oplossing werd 15 minuten aan een terugvloeikoeler verwarmd en vervolgens onder verlaagde druk tot ongeveer 50 ml geconcentreerd. Toevoeging van diethylether veroorzaakte kristallisatie. Het ^ precipitaat werd afgefiltreerd, met ether gewassen en gedroogd.

N-cyaan-S-methyl-N'-{2-/2-methyl-a-(2-pyridyl)benzylthio? ethyl}isothioureum (Cfl) . - —- Een oplossing van 20 g 2-methylfenyl-2-pyridylmethanol .methanol en 11,4 g cysteamine in 300 ml 48% HBr werd 5 uur aan een terugvloeikoeler verwarmd en vervolgens ingedampt. Het 10 residu werd in water opgelost, met KOH op pH 11 gebracht, waarna de waterfase met ΟΗΟΙβ werd geextraheerd. De CHCl^-laag werd op MgSO^ gedroogd en ingedampt. Het residu werd in ether opgelost en langzaam toegevoegd aan een oplossing van 15 g N-cyaandimethyl-iminodithiocarbonaat. Na 3 uur roeren werd het precipitaat afge-filtreerd,met ether gewassen en gedroogd.
Opbrengst: 60%, smpt.: 96-99°C.

Men roerde een oplossing van 10 g van een thioureum (C^-Cg) en 1,2 equivalent methyljodide 18 uur bij kamertemperatuur in 200 ml methanol. Na afdamping van het oplosmiddel voegde men een oplossing van 2,5 g 4-(3-aminopropyl)imidazool of 4-(2-aminoethyl)imidazool in 200 ml ethanol aan het residu toe.

Men verwarmde het resulterende mengsel 70 uur aan een terug-, vloeikoeler en zuiverde het produkt door kolomchromatografie (silicagel 0,063-0,200 mm).

Algemene werkwijze voor de synthese van de guanidinederivaten van voorbeelden VII en XI.
Men verwarmde een oplossing van 12 g van het betrokken S-methylisothioureumderivaat (C, of Cg) en 2,5 g 5 4-(3-aminopropyl)imidazool in 300 ml ethanol 70 uur aan een terugvloeikoeler. Na afdamping van het oplosmiddel werd het residu over een silicakolom geleid en geelueerd met een 50% mengsel van ethanol en chloroform. De frakties , die het zuivere nitril bevatten , werden opgevangen, het oplosmiddel werd afgedampt 10 en het residu in 2 N HC1 opgelost. Na 3 uur verwarmen aan een terugvloeikoeler werd het raaktiemengsel ingedampt en werd het residu opgelost in methanol en toegevoegd aan een oplossing van pikrinezuur in methanol. De geprecipiteerde olie werd grondig met methanol gewassen en in vacuum gedroogd, waarop zij 15 verstarde.

10 Yt een met R gesubstitueerde fenylgroep is en Y eveneens een met R gesubstitueerde fenylgroep is, waarbij Y en niet gelijk gesubstitueerd behoeven te zijn of Y een met R gesubstitueerde 15 benzylgroep is of • c) =CH- is, waarbij Y een met R gesubstitueerde difenylmethyleen groep of (10,11-dihydro)-5H-dibenzo-/a,d_/-cyclo- 20 hepten-5-ylideengroep is, R H, alkyl, alkoxy, halogeen en/of trihalogeen- methyl is, met dien verstande, dat van alle betrokken fenylringen er een of meer vervangen kunnen zijn door een met R gesubstitueerde hetero- 25 cyclische aromatische ring een zuuradditiezout van een dergelijke verbinding is.
