Abstract
La presente invención se refiere a kits y métodos de ensayos para la detección temprana de patógenos, la identificación precisa del agente etiológico, y vigilancia de enfermedad mejorada; más específicamente, la presente invención describe un inmunoensayo que conduce a la detección rápida y simultánea de anticuerpos para un amplio rango de patógenos infecciosos en fluidos biológicos de pacientes infectados; este inmunoensayo involucra el acoplamiento covalente y orientado de proteínas de fusión que comprenden una enzima de AGT y un antígeno viral en un soporte sólido identificable (por ejemplo, microesferas fluorescentes), dicho soporte que está recubierto previamente con un sustrato de AGT; este acoplamiento esta mediado por la reacción irreversible de la enzima de AGT sobre su sustrato; las microesferas acopladas con antígeno así obtenidas muestran mayor captura de anticuerpos específicos en comparación con las microesferas acopladas con antígeno producidas por procedimientos de acoplamiento de amina estándar; los métodos de la invención poseen la capacidad para multiplexear, minimizar la cantidad de muestra biológica, y tienen sensibilidad y especificidad mejoradas hacia los anticuerpos objetivo en comparación con ELISA clásico o ensayo de radio-inmunoprecipitación.
