Abstract
La invención se refiere a un método para la determinación de la constante de aglutinación de un compuesto de interés con respecto a las proteínas que comprende las siguientes etapas: a) agregar el compuesto de afinidad elevada a un sistema de dos cámaras, en donde las dos cámaras están separadas por una membrana semipermeable, que es permeable para el compuesto de interés, y determinar la cantidad del compuesto de afinidad elevada de interés en una de las cámaras después que el equilibrio de la distribución ha sido alcanzado, b) agregar un compuesto de deposición a una de las cámaras por lo cual el compuesto de deposición puede no permearse hacia la membrana, y determinar el coeficiente de distribución del compuesto de interés con respecto al compuesto de deposición después que el equilibrio de la distribución ha sido alcanzado, c) agregar una proteína no específica a la otra cámara, por lo cual la proteína no específica no puede permearse hacia la membrana, y determinar el coeficiente de distribución del compuesto de interés con respecto a la proteína no específica en la presencia de un compuesto de deposición después que el equilibrio de la distribución ha sido alcanzado, y d) determinar la constante de aglutinación del compuesto de prueba con el coeficiente de distribución de las etapas b) y c).
