Abstract
Método para determinar la constante de unión de un compuesto de alta afinidad de interés a proteínas, comprendiendo las etapas siguientes: a) adición de compuesto de alta afinidad a un sistema de dos cámaras, en el que las dos cámaras están separadas por una membrana semipermeable, que es permeable al compuesto de interés, y determinación de la cantidad del compuesto de alta afinidad de interés en una de las cámaras tras alcanzarse el equilibrio de distribución, b) adición de un agente acomplejante como compuesto sumidero a una de las cámaras, en el que el compuesto sumidero es una sustancia que reduce significativamente la afinidad del compuesto de interés para una proteína no específica; dicha proteína no específica se une a un amplio abanico de compuestos y el compuesto sumidero no permea la membrana, y determinación del coeficiente de distribución del compuesto de interés respecto al compuesto sumidero tras alcanzarse el equilibrio de distribución, c) adición de una proteína del plasma como proteína no específica a la otra cámara, en la que la proteína del plasma no puede permear la membrana, y determinación del coeficiente de distribución del compuesto de interés respecto a la proteína del plasma en presencia de un compuesto sumidero tras alcanzarse el equilibrio de distribución, y d) determinación de la constante de unión del compuesto de alta afinidad de interés con el coeficiente de distribución de las etapas b) y c), y en la que el compuesto de alta afinidad de interés se refiere a un compuesto que bajo condiciones fisiológicas y en presencia de un exceso de proteína del plasma, por lo menos el 95% de la cantidad total del mismo se encuentra unida a dicha proteína del plasma.
