Abstract

La presente invención se refiere a un proceso cromatográfico para la separación de componentes de proteína de una solución que contiene proteína, la solución comprende un péptido de insulina y uno o más iones metálicos di- o polivalentes, el péptido de insulina es capaz de auto-asociación y/o cambios estructurales en presencia de iones metálicos di- o polivalentes, el proceso comprende las etapas de: a) aplicar la solución que contiene proteína a una columna de un material en fase sólida cromatográfica, en donde la carga del péptido de insulina es de al menos 6.0 g por litro de volumen de la columna (g/LCV), y b) eluir el péptido de insulina a partir del material en fase sólida por medio de un eluyente que tiene un pH como máximo de 8.5; y recoger un fondo del péptido de insulina que corresponda a por lo menos 75% en peso del péptido de insulina aplicado a la columna en la etapa (a).
