Abstract

La presente invención proporciona un proceso de método de purificación posterior para la producción de polipéptidos de unión a antígeno biespecíficos. El método comprende al menos las etapas de (i) proporcionar una resina de separación que comprende una parte de matriz de polímero y una parte de ligando, en donde la parte de matriz comprende polimetracrilato y tiene un tamaño de partícula de aproximadamente 30 a 60pm, en donde la parte de ligando comprende proteína L recombinante, y en donde la proteína L de la parte de ligando se une covalentemente a las partículas de la parte de matriz, (ii) poner en contacto un fluido del proceso que comprende el polipéptido de unión a antígeno biespecífico con la resina de separación, (iii) capturar el polipéptido de unión a antígeno biespecífico por la parte de ligando de la resina de separación, en donde el polipéptido de unión a antígeno biespecífico se une reversiblemente a la parte de ligando de la resina de separación, y en donde el resto del fluido del proceso no se une a la parte de ligando de la resina de separación, (iv) lavar el polipéptido de unión a antígeno biespecífico unido con un tampón de lavado que no eluye el polipéptido de unión a antígeno biespecífico de la porción de ligando, y (v) eluir el polipéptido de unión a antígeno biespecífico de la parte de ligando con un tampón de elución a un pH bajo.
