Abstract
En el presente documento se proporcionan métodos y sistemas para detectar una secuencia de ácido nucleico diana. El método comprende poner en contacto un oligonucleótido que comprende la secuencia de ácido nucleico diana con un complejo de transposón; insertar uno o más promotores de T7 ARN en el oligonucleótido utilizando la transposasa; y (c) amplificar la secuencia de ácido nucleico diana. El complejo de transposón puede comprender una transposasa y una secuencia de transposón que comprende uno o más promotores de T7 ARN. La secuencia nucleica diana se puede amplificar generando oligonucleótidos de ARN que comprenden la secuencia de ácido nucleico diana mediante la transcripción de uno o más promotores de T7 ARN insertados. El ácido nucleico diana amplificado puede detectarse utilizando un sistema de detección basado en CRISPR Cas13.
