Abstract
MUTAGÊNESE DIRECIONADA USANDO EDITORES DE BASES. A presente invenção refere-se a métodos para descobrir traços e gerar sistemas celulares com fenótipos aprimorados. Em particular, a presente invenção fornece métodos para o desenvolvimento de plantas com fenótipos agronomicamente otimizados usando mutagênese direcionada com poucos ou nenhum efeito fora-do-alvo. Mutagênese direcionada é alcançada pela introdução de um complexo editor de bases ou de um complexo STEME que compreende uma série de RNAs guia que têm como alvo uma sequência de ácidos nucleicos de interesse. A presente invenção refere-se também a sistemas celulares obtidos por meio dos métodos descritos no presente documento e ao uso de um complexo editor de bases ou o complexo STEME que compreende uma matriz de RNAs guia para gerar um sistema celular que tem um fenótipo agronomicamente importante e para a identificação de um fenótipo agronomicamente importante.
