Abstract
Se proporciona un método de edición de genoma dirigida de un animal usando la integración homóloga específica del sitio. Una composición que comprende un oligonucleótido de una sola hebra o molécula de ácido nucleico de doble hebra que comprende una molécula de ácido nucleico de interés y secuencias que flanquean un sitio de segmentación de locus objetivo se inyecta en el cigoto de un animal junto con una nucleasa y una molécula de ácido nucleico de guía que dirige la nucleasa a un locus objetivo. La composición se inyecta en el cigoto después de la fertilización y antes de la formación de un núcleo. La molécula de ácido nucleico de interés se recombina en el genoma con alta eficiencia. El proceso permite la integración de moléculas de ácido nucleico en el genoma de animales en el cual los pronúcleos no pueden ser visualmente observados durante la inyección.
