Abstract

Una proteína de fusión de polipéptido de cadena simple, que comprende: una proteasa no citotóxica, cuya proteasa es capaz de escindir una proteína del aparato de fusión exocítico de un aferente sensorial nociceptivo (por ejemplo, cadena L de neurotoxina clostridial o proteasa de IgA); una Porción de Selección de galanina que es capaz de unirse a un Sitio de Unión en el aferente sensorial nociceptivo, cuyo Sitio de Unión es capaz de experimentar endocitosis para ser incorporado en un endosoma dentro del aferente sensorial nociceptivo (por ejemplo, el receptor GALR1, GALR2, o GALR3); un sitio de escisión de proteasa en cuyo sitio la proteína de fusión se escinde por una proteasa, en donde el sitio de escisión de proteasa se ubica entre la proteasa no citotóxica y la Porción de Selección de galanina; un dominio de translocación que es capaz de transiocar la proteasa desde dentro de un endosoma, a través de la membrana endosomal y en el citosol del aferente sensorial nociceptivo (por ejemplo, el dominio HN de neurotoxina clostridial) ; un primer espaciador ubicado entre la proteasa no citotóxica y el sitio de escisión de proteasa, en donde el primer espaciador comprende una secuencia de aminoácidos de 4 a 25 residuos de aminoácidos; y un segundo espaciador ubicado entre la Porción de Selección de galanina y el dominio de translocación, en donde el segundo espaciador comprende una secuencia de aminoácidos de 4 a 35 residuos de aminoácidos. Las secuencias de ácido nucleico que codifican las proteínas de fusión de polipéptido, métodos para preparar las mismas y usos de los mismos también se describen (por ejemplo, para tratar, prevenir o aliviar el dolor).
