本発明は、単離されたクロストリジウムのプロペプチドおよび神経毒、そのワクチンまたは抗体、被験体を免疫して処置する方法、クロストリジウムのプロペプチドおよび神経毒をコードする単離された核酸分子、発現の方法、キメラタンパク質、ならびに処置方法に関する。
クロストリジウム神経毒は、シナプス小胞エキソサイトーシスのニューロン機構を標的にする構造的に類似したタンパク質のファミリーである。クロストリジウム(Clostridium)属の嫌気性菌により産生される、ボツリヌス(botulinum)神経毒(「BoNT」、7つの免疫学的に別個のサブタイプA〜G)および破傷風(Tetanus)神経毒(「TeNT」)は、重量あたりで公知の最も有毒な物質であり、静脈内または筋肉内経路により投与される場合、0.5〜2.5ng/kgの範囲のLD50を有する(National Institute of Occupational Safety and Health,「Registry of Toxic Effects of Chemical Substances(R-TECS),」Cincinnati, Ohio: National Institute of Occupational Safety and Health (1996))。BoNTは、コリン作動性神経をそれらの神経筋接合部においてターゲットし、アセチルコリン放出を阻害し、末梢神経筋遮断を引き起こす(Simpson,「Identification of the Major Steps in Botulinum Toxin Action,」Annu. Rev. Pharmacol. Toxicol. 44:167-193 (2004))。BoNT血清型A、BおよびEは特に、それらが噴霧され、吸入により送達され得るため、軍隊および一般市民にとって最も多大なる脅威を示すと考えられる(Arnon et al.,「Botulinum Toxin as a Biological Weapon: Medical and Public Health Management,」JAMA 285:1059-1070 (2001))。

以下の実施例は、本発明の態様を例証するために提供されるが、決してその範囲を限定することを意図するものではない。
すべての中間精製段階由来の試料、および純粋な組換えタンパク質を、日常的に分離し、Laemmli手順(その全体が参照により本明細書に組み入れられる、Laemmli,「Cleavage of Structural Proteins During the Assembly of the Head of Bacteriophage T4,」Nature 227:680-685 (1970))に従って8%分離ポリアクリルアミドゲル上で可視化した。タンパク質バンドを、Bio-Safe Coomassie G-250 Stain(Bio-Rad, カタログ#161-0786)により可視化した。
ウェスタンブロット分析のための試料を、8%SDS-ポリアクリルアミドゲル上で分離した。分離後、タンパク質を、20%メタノールを追加した1xTris/グリシン緩衝液(Bio-Rad, カタログ#161-0734)において、100ボルト、4℃で2時間、Hybond-Cニトロセルロース膜(Amersham Biosciences, カタログ#RPN303C)へ転写した。転写後、膜を蒸留水でリンスし、タンパク質バンドを1%酢酸中の0.2% Ponceau Sで1分間、染色することにより可視化した。膜由来の色素を、Tris緩衝食塩水/0.1% Tween-20緩衝液、pH7.5中で洗い流し、続いてブロッキング試薬(Tris緩衝食塩水/0.1% Tween-20緩衝液、pH7.5中の5%無脂肪粉乳)中で膜を、4℃で16時間、インキュベーションした。免疫検出のために、膜を1:7,000希釈の一次抗体/免疫血清と、Tris緩衝食塩水/0.1% Tween-20緩衝液、pH7.5中の0.5%無脂肪乳において、室温で2時間インキュベートした。膜を洗浄し(6x5 min)、1:10,000希釈の二次抗体と室温で25分間、インキュベートした。一連のさらなる洗浄(6x5 min)後、免疫反応性バンドをECL(増強化学ルミネセンス(enhanced chemiluminescence)) Plus Western Blotting Reagent(Amersham Biosciences, カタログ#RPN2124)を用いて、製造会社の使用説明書に従って可視化した。Hyperfilm ECL(Amersham Biosciences, カタログ#RPN1674K)を、化学ルミネセンスバンドを可視化するのに十分な曝露時間でのオートラジオグラフィーに用いた。タンパク質を、陽性対照および分子量タンパク質標準との比較により同定した。
精製組換えタンパク質画分のタンパク質濃度を、標準として用いたウシ血清アルブミンと共に、BCA Proteinアッセイ試薬(Pierce, カタログ#23225)を用いて測定した。
様々な組換えタンパク質の毒性を、マウス横隔神経-片側横隔膜調製物においてバイオアッセイした(その全体が参照により本明細書に組み入れられる、Simpson et al.,「Isolation and Characterization of a Novel Human Monoclonal Antibody that Neutralizes Tetanus Toxin,」J. Pharmacol. Exp. Ther. 254:98-103 (1990))。組織を切除し、生理的緩衝液に懸濁し、95% O2、5% CO2で通気し、35℃で保持した。生理的溶液は、以下の組成物を有する:137mM NaCl、5mM KCl、1.8mM CaCl2、1mM MgSO4、24mM NaHCO3、1mM NaH2PO4、11mM D-グルコース、および0.01%ゼラチン。横隔神経を連続的に刺激し(1.0 Hz; 0.1〜0.3 msec持続時間)、筋収縮を記録する。毒素誘発性麻痺を、神経性刺激への筋収縮応答における50%低下として測定した。
細胞を、Transwell(登録商標)多孔性底部インサート(Corning-Costar)において0.4μm細孔サイズを有するポリカーボネート膜上で増殖させた(図10)(その全体が参照により本明細書に組み入れられる、Zweibaum et al.,「Use of Cultured Cell Lines in Studies of Intestinal Cell Differentiation and Function,」Schulz et al.,により編集されたHandbook of Physiology, Section 6:「The Gastrointestinal System,」American Physiological Society, Bethesda, Vol. IV, 223-255において; Dharmsathaphorn et al.,「A Human Colonic Tumor Cell Line that Maintains Vectorial Electrolyte Transport,」Am. J. Physiol. 246:G204-G208 (1984);およびDharmsathaphorn et al.,「Established Intestinal Cell Lines as Model Systems for Electrolyte Transport Studies,」Methods Enzymol. 192:354-389 (1990)) 。各インサート内での細胞増殖領域は、1cm2と等しい。細胞を蒔く前に、インサート膜を10μg/cm2ラット尾コラーゲンI型でコーティングした。コラーゲン原液(6.7mg/ml)を、滅菌1%酢酸に調製し、4℃で保存した。コラーゲン原液を、氷冷60%エタノール中に必要に応じて希釈し、10μgの希釈したコラーゲンを含む結果として生じた溶液の150μlを各ウェル(cm2)へ添加する。
対象となるタンパク質の毒性を、マウスにおいてバイオアッセイする。タンパク質を、1mg/mlウシ血清アルブミンを含むリン酸緩衝食塩水中で希釈し、動物へ腹腔内に(i.p.)に注射する。タンパク質を、動物(平均体重〜25g)あたり1〜100ngの濃度の溶液の100μlのアリコートで投与する。毒性誘導体への曝露にも関わらず生きている任意の動物を、いずれの非特異的毒性も検出するように、合計2週間モニターする。
操作したタンパク質を、BoNT AおよびBoNT Eについての基質の供給源として、マウス脳シナプトソームならびに組換えSNAP-25のいずれかを用いてエンドプロテアーゼ活性についてアッセイする。天然または還元型タンパク質を、50mM HEPES、pH7.1、20μM ZnCl2、および1%N-オクチル-β-D-グルコピラノシドを含む反応緩衝液においてシナプトソーム膜の10〜50μgとインキュベートする。還元型タンパク質を、リン酸緩衝食塩水におけるDTT(20mM;1hr;室温)とのインキュベーションにより調製する。切断反応は、操作されたタンパク質の基質への添加(200nM最終濃度)により開始され、反応を、37℃で3時間進行するようにさせる。エンドプロテアーゼ活性を、ウェスタンブロット分析および基質の免疫検出のための抗C末端SNAP-25抗体(StressGen)を用いてアッセイする。SNAP-25の可視化のために、試料を16.5% Tris-トリシン(tricine)ゲル上で分離する。分離後、タンパク質を、Tris-グリシントランスファー緩衝液において50ボルトで1時間、ニトロセルロース膜(Micron Separations)へ転写する。ブロットされた膜を、蒸留水中でリンスし、1%酢酸中の0.2% Ponceau Sで1min、染色する。蒸留水での短時間リンスの後、分子量マーカーおよび転写されたタンパク質を可視化する。膜を、リン酸緩衝食塩水-Tween(pH7.5;0.1% Tween 20)中で脱染し、その後、リン酸緩衝食塩水-Tweenにおける5%無脂肪粉乳で、室温で1hr、ブロッキングする。その後、膜を、0.5%乳において、抗SNAP-25ポリクローナル抗体の1:5,000希釈とインキュベートする。二次抗体を、1:20,000希釈で用いる。膜を再び洗浄し(5X)、増強化学ルミネセンス(SuperSignal(登録商標)West Pico, Pierce)を用いて製造会社の使用説明書に従って可視化する。膜を、化学ルミネセンスバンドを可視化するのに十分な時間、フィルム(Hyperfilm ECL, Amersham Biosciences)へ感光させる。ペプチドを、既知の標準との比較により同定する。BoNT B基質切断アッセイを、公表されたプロトコール(その全体が参照により本明細書に組み入れられる、Caccin et al.,「VAMP/Synaptobrevin Cleavage by Tetanus and Botulinum Neurotoxins is Strongly Enhanced by Acidic Liposomes,」FEBS Lett. 542:132-136 (2003))に従って行う。
BoNT A誘導体を操作するために用いる手順を、以下に詳細に概説する。すべてのBoNT誘導体を操作するための同様のストラテジーを実行することができる。
1つの100μl PCR反応設定あたり、25μgの混合ゲノムDNAまたは5μgの純粋ボツリヌス菌(Clostridium botulinum)A型ゲノムDNAを用いた。反応条件は、Platinum(登録商標)Pfxポリメラーゼ(Invitrogen, カタログ#11708-021)と共に供給された製造会社のプロトコールに従って設計した。すべてのオリゴヌクレオチドおよびリンカーは、Genebank(アクセッション番号:M30196)から得られたボツリヌス(botulinum)神経毒A型cDNAの配列により設計した。アニーリング温度は、PCRに用いる各セットのオリゴヌクレオチドの構造から推論した。

pFBSecBoNTAを、以下のプロトコールにより得た:オリゴヌクレオチド:

PCR産物を、前消化かつ脱リン酸されたベクターpLitBoNTAへサブクローニングし、結果として、プラスミドpLitSecBoNTAを生じた。BssHIIおよびNotIで消化されてpLitSecBoNTAから単離されたDNA断片を、前消化かつ脱リン酸されたベクターpFastBac(商標)1(Invitrogen, カタログ#10360-014)へサブクローニングし、結果として、8764b.p.プラスミドpFBSecBoNTAを生じた。
DNA鋳型は、ボツリヌス菌A型培養物から単離された純粋なゲノムDNAとして、または土壌の嫌気性細菌から単離された混合ゲノムDNAとしてのいずれかで得られた。BoNT A DNAを、高忠実性Platinum Pfxポリメラーゼ(Invitrogen, Carlsbad, CA)を用いるPCRより増幅した。BoNT A毒性誘導体(td)の完全長コード配列は、5つのPCR断片および2つのリン酸化リンカーの改変ベクターpLitmus38i(New England Biolabs, Beverly, MA)への連続的サブクローニング後に得られ、結果として、プラスミドpLitBoNTAを生じた。このストラテジーは、PCR反応中の不忠実性を最小限にするため、ならびに発現された毒素産物のその後の操作のための標的突然変異およびエンドヌクレアーゼ制限部位の導入を可能にするために用いられた。
発現プラスミドを、大腸菌Rosetta-gami B(DE3)コンピテント細胞(Novagen, カタログ#71136-3)へ製造会社のプロトコールに従って熱ショック方法によりトランスフェクションさせた。細菌培養物を、50mg/lカルベニシリン、15mg/lカナマイシン、12.5mg/lテトラサイクリン、および34mg/lクロラムフェニコールを含むLB培地で増殖させた。細菌培地へのL-アラビノース(0.01%最終濃度)の添加無しおよび有りでの、細胞あたりのプラスミドコピー数に影響を及ぼす様々な条件を試験した。タンパク質発現に用いられるすべての細菌培養物を、OD@600nm 〜0.3-0.4に達するまで37℃で増殖させた。発現の誘導の前に、細菌培養物を、発現された産物の収率および品質への温度の影響を試験するために分割した。誘導されると(OD@600nm 〜0.5-0.7)、培養物を、37℃、25℃および12℃において増殖させた。誘導に用いられた増殖培地における最終IPTG濃度は、0.5mMであった。経時的研究について、誘導後1時間目、3時間目、6時間目、9時間目および12時間目における培養物の試料を収集し、分析した。最適な条件下で、大腸菌培養物を、OD 〜0.4@600nmに達するまで37℃でL-アラビノースの存在下で一晩、インキュベートした。細菌懸濁液の温度を、その後、1時間かけて12℃まで低下させ、IPTGを、0.5mMの最終濃度まで添加した。誘導後、培養増殖を、振盪インキュベーターにおいて12℃でもう6時間、継続するようにさせておいた。細菌ペレットをその後、Sorwall GS3ローターにおける遠心分離(7000rpm、25min、4℃)により収集し、組換えタンパク質単離のために処理した。氷上に保たれた細胞を、BugBuster溶解試薬(Novagen, カタログ#70584-4)に、容量比細胞ペースト:BugBuster可溶化試薬1:5で再懸濁した。混合物超音波処理の代わりに用いられる、核酸分解試薬ベンゾナーゼ(Novagen, カタログ#70746-3)、1000U/ml最終濃度、組換えリゾチーム(Novagen, カタログ#71110-4)、50U/ml最終濃度、およびプロテアーゼ阻害剤のカクテル「Complete」(Roche(Switzerland), カタログ#1697498)、1タブレット/50ml最終濃度を、再懸濁の過程でペーストへ同時に添加した。再懸濁から約30分後、非粘稠性可溶化液をSorwall SS34ローターにおける遠心分離(17000rpm、25min、4℃)により透明にし、さらなるタンパク質精製のために処理した。
Bac-to-Bac(登録商標)バキュロウイルス発現系(Invitrogen, カタログ#10359-016)が、組換えバキュロウイルスの作製のために用いられた。昆虫細胞培養についてのプロトコールは、キットと共に供給されたマニュアルから採られた。組換えドナープラスミドを、Max Efficiency DH10Bac(商標)コンピテント細胞(Invitrogen, カタログ#10361-012)へ形質転換した。組換えバクミドを含むコロニーは、lacZαの破壊により同定され、色素生産性基質Bluo-gal(Invitrogen, カタログ#15519-028)を含むプレート上で増殖するが、青色の発色の欠如により選択された。高分子量DNAを、DNAプラスミド精製系(Qiagen, カタログ#12245)における選択されたコロニーから単離した。対象となるDNAのバキュロウイルスゲノムへの転位は、結果として、転位が起こった試料における1170b.p. DNAバンドの増幅を生じる、オリゴヌクレオチドCBA14およびCBA17を用いての高分子量DNAにおけるPCRにより確認された。バクミドは、無血清培地適応Sf9昆虫細胞(Invitrogen, カタログ#11496-015)をトランスフェクションして、バキュロウイルスを作製するために用いられた。トランスフェクションは以下のプロトコールにより行われた:2mlの補充されていないGraceの昆虫細胞培地(Invitrogen, カタログ#11595-030)において1個の35mmのウェルあたり9x105細胞を蒔いた。>97%の生存度を有する対数期の中間部における3〜4日経った懸濁培養物由来の細胞が、実験に用いられた。細胞を、前もって、27℃で少なくとも1時間、プラスチックに付着させ、製造会社により供給されたプロトコールに従って、バクミドをCellfectin(登録商標)トランスフェクション試薬(Invitrogen, カタログ#10362-010)と混合した後に形成された親油性複合体をトランスフェクションした。トランスフェクションから72時間後、組換えバキュロウイルスを含む上清を採取し、低速度遠心分離(Sorwall GS 3 Rotor、2000rpm、20min、4℃)により細胞から分離した。上清は、一次バキュロウイルス系統を示す。このバキュロウイルス系統の増幅およびウイルスプラークアッセイは、製造会社により供給されたプロトコールに従って行われた。最適MOIの同定および組換えタンパク質発現の経時的研究に関する実験もまた、製造会社の推奨に従って行われた。
が、PCR産物のすべての構築物へのクローニングにより導入された。図5は、バキュロウイルス系における発現に向けられたBoNT A誘導体の概略図を提供する。図解した組換えタンパク質に示されたシグナルペプチドは、細胞内輸送中にセクレターゼプロセシングにより除去される(その全体が参照により本明細書に組み入れられる、von Heijne,「Signals for Protein Targeting Into and Across Membranes,」Subcell. Biochem. 22:1-19 (1994))。ベクターpFastBac(商標)1における遺伝子の発現は、オートグラファ・カリフォルニカ(Autographa californica)多核多角体病ウイルス(AcMNPV)ポリヘドリン(PH)プロモーターにより制御される。組換えドナープラスミドをDH10Bac(登録商標)大腸菌コンピテント細胞へ形質転換した。一旦pFastBac(商標)に基づいた発現プラスミドが細胞質に存在すれば、pFastBac(商標)に基づいたベクターにおける発現カセットに隣接するmini-Tn7エレメントと、すでに細胞に存在するバキュロウイルスシャトルベクター(バクミド)上のmini-attTn7標的部位のアーム間で転位が起こる。この事象は、組換えバクミドを生じる。転位は、同じくコンピテント細胞に存在するヘルパープラスミドにより供給される追加のタンパク質を必要とする。組換えバクミドクローンの選択は、視覚的に(サイズおよび色により)行われた。単離されたDNAの分子性質は、特異的オリゴヌクレオチドプライマーを用いるPCRにより確認された。
図7は、例としてtd誘導体を用いる、組換えエンテロキナーゼ(rEK)でのプロペプチド構築物の切断についての用量滴定曲線を示す。単鎖(SC)タンパク質のプロセシングについて、異なる量がBoNT A tdへ加えられている。20℃で8時間、0.5Uの酵素を用いることが、1μgのsc BoNT A tdを完全に消化することが見出された。
完全長BoNT A誘導体の適量を精製するための方法は、例としてBoNT A tdを用いて開発した。組換えタンパク質はNi-NTA樹脂に結合することが見出されたが(図5)、親和性は、1段階精製スキームを確立するには十分ではなかった。タンパク質は、5mMイミダゾールにおいてNi-NTA樹脂に結合したが、40mMイミダゾールによりアフィニティーカラムから溶出された。イミダゾールのこの濃度において、溶出液に存在する他のタンパク質があり、それゆえに、追加の段階が、混入物質から組換えタンパク質を分離するために必要とされる。
2つの型の実験を、組換え毒素が天然毒素の生物活性を保持したかどうかを評価するために行った。これらは、生物学的試験に十分な量で産生したBoNT A td誘導体を用いて行われた。第一の試験において、組換えBoNT A tdを、静脈内経路によりマウスへ投与し(マウスあたり〜1ng)、死までの時間をモニターした。死は、注射から約12分後に観察された。筋肉の脱力または麻痺の先行症状は明らかではなかった。第二試験において、組換えBoNT A tdを、マウス横隔神経-片側横隔膜調製物へ添加し、神経中枢の刺激(0.2Hz)により誘起されるアセチルコリン放出を阻害するその能力を、筋収縮をモニターすることにより評価した。〜1x10-11Mの濃度において、組換えBoNT A tdは、167±17min(n=4)で神経筋遮断を生じた。遮断がボツリヌス毒素型作用に起因し得ることを保証するために、そのポリペプチドを、天然BoNT A重鎖のカルボキシ末端側の半分(すなわち、受容体結合ドメイン)に対して産生されたウサギ抗血清とプレインキュベートする最終実験を行った。これらの実験(n=3)において、組織を約400分間、モニターした場合でさえも、神経筋遮断はなかった。「BoTox」としてAllergan Inc.により市販されている薬学的調製物は、約1x10-11M(1mlあたり60ユニット)の濃度で100分で神経筋遮断を生じる。Rummel(前記)により作製されたBoNT A、BおよびG組換え産物は、同様の時間枠で神経筋遮断をもたらすのに60〜1000倍多いBoNTを必要とする。
クロストリジウム毒素についてのDNA配列およびタンパク質配列は、GeneBankからアクセスされる。完全長毒素をコードする構築物は、いくつかの研究所から入手できる。これらの公知の配列および構築物は、計画された遺伝子操作のための効率的な出発点を提供する。
天然毒素にそれらの構造および生理学的活性に関して最も良く似ている毒性誘導体(「ad」)を作製するために、単一アミノ酸点突然変異が、毒素のメタロプロテアーゼ触媒ドメインの活性部位へ導入される。この分子におけるたいていの毒素特徴は、天然毒素におけるのと同じままであるが、シナプスエキソサイトーシス機構においてその基質を切断することができないため、それは毒性を欠いている。このように作製した毒性誘導体は、それらの天然毒素との構造的類似性、ならびに天然毒素の吸収および輸送経路に沿った多様な部位において免疫応答を生じるそれらの能力のため、ワクチンとして開発中の他のBoNT調製物より優れている。これらの誘導体は、天然毒素と同じ結合部位および輸送経路において競合する可能性が高いため、それらはまた、BoNT中毒に対する解毒剤として抗体調製物より優れている可能性がある。
PCR増幅のためのすべてのBoNT血清型についてのDNA鋳型は、純粋なクロストリジウム培養物(血清型特異的)か、または出発物質としての混合ゲノムDNAが調製され得る土壌由来の嫌気性培養物かのいずれかから得られ得る。高忠実性Platinum(登録商標)Pfxポリメラーゼが、増幅エラーを最小限にするためにすべてのPCR反応に用いられる。BoNT BおよびE血清型を次の実施例に記載する。
BoNT A tdの完全長コード配列を得るために用いられたものと類似した1組のオリゴヌクレオチドが、Gene bankから入手できる配列(アクセッション番号BoNT BについてM81186およびBoNT EについてX62683)を用いてBoNT BおよびEについて設計され得る。配列は、大腸菌、バキュロウイルスおよびピチア・パストリス発現系においてタンパク質発現に影響を及ぼし得る望ましくないDNA制御エレメントおよび他の特徴について入念に評価され、そのようなエレメントは、沈黙部位特異的突然変異誘発により除去される。毒素のLC-HC接合部における低特異性タンパク分解部位を除去するように、およびLC-HC接合部においてエンテロキナーゼ切断部位を導入するように、ターゲットされた追加の突然変異が導入される。図1〜3に示した、毒素配列アラインメントおよびドメイン構造に基づいて、組換え毒素の生物学的性質に影響を及ぼすことなく改変され得る遺伝子領域が同定され、BoNT A tdについて実行した設計スキームと同様に、NheI、XbaI、KpnIおよびXhoI制限部位が導入される。そのような突然変異が、沈黙突然変異誘発を通して生じることが不可能である場合には、制限部位は、タンパク質配列の構造的可動部へ中性アミノ酸挿入を介して導入される。生じたいずれの重複性制限部位も、沈黙部位特異的突然変異誘発により除去される。発現系において遺伝子転写の中途での終結を引き起こし得る、またはタンパク質発現に干渉し得るBoNT DNA配列もまた改変される。ピチア・パストリスにおける発現(その全体が参照により本明細書に組み入れられる、Henikoff et al.,「Sequences Responsible for Transcription Termination on a Gene Segment in Saccharomyces Cerevisiae,」Mol. Cell Biol. 4:1515-1520 (1984); Irniger et al.,「Different Classes of Polyadenylation Sites in the Yeast Saccharomyces Cerevisiae,」Mol. Cell Biol. 11:3060-3069 (1991); Scorer et al.,「The Intracellular Production and Secretion of HIV-1 Envelope Protein in the Methylotrophic Yeast Pichia Pastoris,」Gene 136:111-119 (1993))は、ATリッチな鋳型からの転写の中途での終結を抑制することができる1組のPCRオリゴヌクレオチドプライマーを設計することによりそのような配列の排除を必要とする。これらの手順は、次の発現研究に用いられる、BoNT B tdおよびBoNT E tdの改変型コード配列を含む構築物を生じる。
完全長の機能的に活性のある毒素の発現および精製は、代わりの構築物設計および発現系を用いて実験室において困難であることが証明されている。理想的な構築物および発現系は、好ましくは、それらは宿主にとって非典型的なコード配列を含むため、クロストリジウム毒素を分離しない;たいていの真核生物において同様である、制御エキソサイトーシスのための装置を破壊し得るサイトゾルへの活性毒素の侵入により毒されない;および発現された毒素の正常な翻訳後修飾、特に、ジスルフィド架橋の形成を可能にする。
大腸菌において完全長BoNT Aを産生しようとする試みは、結果として、BoNT Aプロペプチドについて予想されたより有意に小さい、主要なC末端が切り詰められたプロペプチドをもたらした(図4)。将来、この産物のC末端組成物を、ミクロシーケンシングにより分析し、大腸菌系に特異的な推定上のタンパク分解性切断部位を同定し、この効果を抑制するように設計されるアミノ酸置換を有するpETcoco発現構築物を再設計するつもりである。タンパク分解部位が、過剰量で添加した場合、C末端BoNT A受容体結合ドメイン内を切断することが実証されているトリプシンにより認識されるものと同様であることは可能性がある(その全体が参照により本明細書に組み入れられる、Chaddock et al.,「Expression and Purification of Catalytically Active, Non-Toxic Endopeptidase Derivatives of Clostridium Botulinum Toxin Type A,」Protein Expr. Purif. 25:219-228 (2002)) 。再設計した構築物の発現は、以下に記載するような、進化したRosetta-gami B(DE3)大腸菌株を用いる。
組換えタンパク質を大腸菌周辺質への分泌のためにターゲットすることは、安定性および翻訳後ジスルフィド結合形成を向上させることができる。BoNT A td配列のコード部分を、標的タンパク質の搬出および周辺質折り畳みに必要とされるシグナルを含むpET39b(+)ベクター(Novagen, Cat.#70090-3)へサブクローニングする。この系は、208アミノ酸DsbA・Tag(商標)と融合したペプチド配列のクローニングおよび発現のために設計される。DsbAは、ジスルフィド結合の形成および異性化を触媒する周辺質酵素である(その全体が参照により本明細書に組み入れられる、Rietsch et al.,「An In vivo Pathway for Disulfide Bond Isomerization in Escherichia coli,」Proc. Natl. Acad. Sci. USA 93:13048-13053 (1996); Sone et al.,「Differential In vivo Roles Played by DsbA and DsbC in the Formation of Protein Disulfide Bonds,」J. Biol. Microbiol. 51:179-202 (1997))。以下に記載したBoNT Aの分解は、大腸菌が、細胞質に蓄積するタンパク質についてジスルフィド架橋を正しく形成する能力がないことの結果として起こる可能性がある。DsbAベクターは、非還元周辺質環境において組換えBoNTの可溶性および正しい折り畳みを増強し得る(その全体が参照により本明細書に組み入れられる、Collins-Racie et al.,「Production of Recombinant Bovine Enterokinase Catalytic Subunit in Escherichia coli Using the Novel Secretory Fusion Partner DsbA,」Biotechnology 13:982-987 (1995))。周辺質へターゲットされる組換えタンパク質の収量は通常は低いが、大腸菌における周辺質発現は、継続的な考慮に値する。
多コピーピチア・パストリス(Pichia pastoris)発現キット(Invitrogen, カタログ#K1750-01)は、組換えタンパク質を得るために用いられる。対象となる遺伝子を有し、かつピチアゲノムへの取り込みのためにターゲットされたベクターpPIC9Kのバックボーン上の組換えプラスミドは、直線化のために制限エンドヌクレアーゼSalIにより消化され、供給された製造会社のプロトコールに従って、ザイモリアーゼを用いるスフェロプラスト方法によりピチア株GS115およびKM71に形質転換される。ヒスチジン欠損培地上のジェネテシン(Geneticin)の存在下における形質転換体選択の一次および二次ラウンドは、同じプロトコールに従って行われる。タンパク質発現は、メタノール(0.5%最終濃度)の培地への添加により誘導される。破壊された細胞および培地を、SDS-PAGEおよびウェスタンブロッティングにより分析する。
改変型N末端6-Hisタグ付きBoNT A tdのコード部分を、発現プラスミドにおいてS. セレビシエ(S. cerevisiae)由来のα-因子分泌シグナルを供給するベクターpPIC9Kへサブクローニングした。これは、結果として、発現されたタンパク質の培地への分泌を生じるはずである。構築物を制限エンドヌクレアーゼSalIにより直線化し、ピチア・パストリスのKM71およびGS115株へのスフェロプラスト方法によりトランスフェクションした。形質転換体の一次選択は、ヒスチジン欠損培地において増殖する細胞の能力、野生型細胞に欠けている形質、を試験することにより行われた。選択の第二ラウンドは、対象となる遺伝子のコピー数と増殖培地におけるジェネテシンの濃度の増加の間の相関により対象となる遺伝子の複数の挿入断片を有するクローンの同定を可能にする抗生物質ジェネテシンの存在下で行われた。同定されたクローンの、対象となるタンパク質を発現させる能力を、メタノール含有培地において細胞を増殖させることにより試験する。
BoNT A tdのN末端におけるヒスチジンアフィニティータグの長さを増加させ、もう2個の構築物−8-Hisおよび12-Hisタグ付き、を組換えBoNT産物におけるNi-NTAアガロースに対するより高い親和性を与えるそれらの能力について試験した。このアプローチは、Ni-NTAアフィニティー精製媒体に対して類似した親和性の減少を示した他の組換えタンパク質について成功裡に用いられている(その全体が参照により本明細書に組み入れられる、Ichtchenko et al.,「Alpha-Latrotoxin Action Probed with Recombinant Toxin: Receptors Recruit Alpha-Latrotoxin but do not Transduce an Exocytotic Signal,」EMBO J. 17:6188-6199 (1998); Rudenko et al.,「Structure of the LDL Receptor Extracellular Domain at Endosomal pH,」Science 298:2353-2358 (2002))。組換えBoNT A tdの精製の改善が達成され得る場合には、イオン交換クロマトグラフィーの少なくとも2段階が、現行の精製スキームから省かれ得る。

BoNTに冒されたニューロンの細胞質を効果的にターゲットすることができるBoNT解毒剤を産生するように設計された遺伝子操作プラットフォームが開発されている。このプラットフォームに準じて設計された解毒剤は、毒性クロストリジウム神経毒への曝露後長期間、被験体へ効果的に投与される可能性があり、緊急設定において大集団へ解毒剤を投与することの実際的ロジスティックスを改善する。単離されたBoNT A軽鎖の遺伝子構築物を用いて、タンパク質モチーフを、神経毒に冒されたニューロンのサイトゾルからの排除のために毒性野生型クロストリジウム神経毒(例えば、ボツリヌス菌)軽鎖を結合、不活性化または別なふうに標識するように導入する。最適化解毒剤活性を有するキメラ軽鎖は、クロストリジウム神経毒に冒されたニューロンのサイトゾルへ解毒活性を送達することができる完全長クロストリジウム神経毒キメラを作製するためにクロストリジウム神経毒重鎖の誘導体化構築物と、その後、再結合され得る。発現系は、開発され、天然クロストリジウム神経毒輸送に関与する構造的特徴および翻訳後修飾が保存されていることを保証するために試験され得る(上記のように)。この種の作製されたクロストリジウム神経毒解毒剤は、経口または吸入経路により投与した場合、神経毒に冒されたニューロンを効果的にターゲットすることができ、クロストリジウム神経毒中毒の症状をすでに経験している患者(例えば、人工呼吸器による患者)を救うために用いられ得る。
メタロプロテアーゼ不活性化突然変異E224>Aを有するBoNT A cDNAの突然変異型軽鎖をコードするプラスミド(pLitBoNTALCME224A)を、製造会社により供給されたプロトコールに従ってGeneTailor(商標)Site-Directed Mutagenesis System (Invitrogen, カタログ#12397-014)およびPlatinum(登録商標)Pfx DNA Polymerase(Invitrogen, カタログ#11708-021)を用いて、リン酸化オリゴヌクレオチド
