Abstract

La presente invención se refiere a métodos para diseñar dominio VH para mejorar su solubilidad y estabilidad; la invención provee la incorporación de sustituciones de aminoácido definidas basadas en la información estructural tridimensional en los segmentos V de un locus de cadena pesada expresando el locus en un mamífero no humano y seleccionando dominios VH solubles; pueden introducirse otras mutaciones de estabilización o solubilización como resultado de maduración por afinidad durante la maduración de células B in vivo.
