Description

필로카핀(Pilocarpine)이 배합된 타액분비 촉진 츄잉껌
본 발명은 감미료, 향료등이 배합된 통상의 껌베이스에 대하여 0.13∼0.20중량%의 필로카핀(pllocar·pine)을 배합하여 된 것을 특징으로 한 필로카핀이 배합된 츄잉껌에 관한 것이다.
본 발명의 목적은 나이가 많아지면서 입안에 침이 적게 나오는 사람이 껌을 씹으면 껌에 배합된 필로카핀이 흡수되면서 침샘의 부교감신경을 자극하여 입안에 침이 많이 나오게 하는 새로운 껌을 제공하려는데 있다.
일반적으로 츄잉껌은 씹는 자체를 목적으로 하고 있으나, 껌에 여러가지 목적에 따른 기능을 부여하여 입에 냄새를 제거하게 된 것, 졸음을 각성하게 된 것, 향기를 내게 된 것등 다양한 종류의 껌이 있으며, 이러한 껌베이스에는 각각 기 기능을 발휘하는 조성물이 배합되어 있다.
본 발명은 종래의 여러 종류의 껌과는 달리 껌을 씹으면 껌베이스에 배합된 감미료와 향료에 의하여 입안이 상쾌하고 맛이 있을 뿐 아니라 입안에 침이 많이 나와 구강위생을 건강하게 유지하고 언어발생을 원활하게 할 수 있게 한 것이다.
즉 젊은이들과는 달리 60세 이상의 노년에 이르면 생리적으로 입안의 침이 젊은 이의 약 8분 1 정도로 적게 나온다. 이렇게 입안에 침이 적게 나오게 되면 상대적으로 세균 번식이 높게 되어 가령 젊은이의 구강에 7만 마리/㎠ 정도의 세균이 번식되고 있는 반면 8분의 1 정도 침이 나오는 경우에는 약 3O만 마리/㎠ 정도로 세균의 번식율이 대단히 높다. 이는 구강위생이 좋지 않고 여러가지 질병에 감염될 위험이 큰 것을 의미한다.
또 입안에 침이 부족하면 말하기 불편하고 정확하게 발음하기도 어려워 알아들을 수 없게 말하는 경우도 허다하다. 또 침이 적게 나오면 많이 나오는 것에 비하여 소화기관의 활동이 약하고 소화불량의 원인이 되기도 한다.
입에 침이 적게 나오는 경우는 나이든 사람 뿐 아니라 긴장하거나 질병에 걸려 있을 때도 그러하다.
인간이 노화에 따라 청년기에 비하여 침(타액)의 분비가 현격하게 감소되는 것은 일반적인 상피현상이나 타액선 선세포의 퇴축과 기능저하를 유발하는 선천적인 외배염성 발육부전증이나 40대 이후에 호발되는 쇠그린증후군등에서 더욱 현저한 타액의 분비감소가 야기되므로 인하여 일반적으로 인간의 타액 1ml에 약 1억개 이상의 미생물이 포함되어 있고 이중 3분의 1 이상이 병원성 미생물임을 감안할 때 타액의 세균에 대한 항균 및 정균작용은 매우 필요한 것이며 이같은 타액의 기능이 저하되거나 상실될 때 인체의 모든 장기에 감염증을 발생시킬 수 있는 것으로, 타액의 정상적인 분비는 일상의 음식물 소화현상과 구강에서의 제반 운동활동 가운데에서의 윤활현상을 보장하는 것 이외에도 전신건강과 직접적인 관계를 가지고 있는 것이다.
현재까지 타액 분비 촉진제로는 부교감신경흥분제로 다양한 투약이 시도되어 왔으나 필로카핀이 타액 분비 촉진에 탁월한 효과가 있음이 증명된 이래 주로 경구투여로 1회에 20mg씩 1일 2회씩 투여하여 왔다.(Drug Information Handbook p p.926, LAXY COMP Inc.1993. U.S A.)
그러나 필로카핀이 가지고 있는 중추신경과 심근에의 흥분효과(참고문헌:① Fox, P.C., Van DerVen, P.F., Baum, Boj and Mandel, I. D.:Pilocarpine for the treatment of xerostomia associated with salivary gland dysfunction,Oral Surg.61(3) : 243-248, 1986.② Deborah Greenspan & Troy E. No. 2 : 186-190, 1993 )로 노령층에 빈발하는 고혈압 또는 파킨슨병 등의 환자나 그 가능성이 높은 유사증상이 있는 환자들에게 장기간 투여할 수 없어 그 투약은 크게 제한적으로 이루어져 왔다.
본 발명은 치과에서 임상적으로 사용하고 있는 필로카핀을 껌베이스의 씹는 기능적 작용과 결합시키어 특별하게 의도함이 없이 껌을 씹으면 동시에 침이 많이 나올 수 있게 하므로써 침에 의한 세균 번식을 억제하여 효과적으로 구강위생을 높힐 수 있고 소화에 촉진작용을 하며 언어발성을 원활하게 할 수 있게 한 것이다.
본 발명을 상세히 설명하면 다음과 같다.
한 개의 껌(일회용)은 스틸렌-부다디엔 공입합체 또는 치콜, 라덱스 고무의 주재로 된 통상의 껌베이스(gum base)에 수소화 에스텔 고무점성제, 파라핀 왁스, 스테아틴산, 당알코올인 키스톨등을 배합하여 형성된 2.5∼3g의 베이스 조성물에 4∼5mg의 필로카핀을 배합하여 조제하였다. 이는 통상 경구투여 용량 20mg의 5분의 1에 해당되는 용량을 제제에 함유케 하였을 뿐 아니라 평균 20분간 껌 저작시 약 50%의 필로카핀만이 구강에서 타액에 용해되어 직접 점막을 통하고 타액선 배출구를 통하여 타액선에 작용하게 되어 실제로 인체에 작용되는 전체의 필로카핀양은 통상 20mg의 8분의 1에 해당되는 미량이 되어, 발생될 수도 있는 필로카핀에 의한 부작용을 최소한으로 할 수 있다.
필로카핀의 껌을 이용한 투여는 상피의(한선,땀샘)와 누선의 기능을 향상시켜 건성피부와 건조한 눈각막을 습윤케 하여 인체의 다른 장기에서도 건강에 도움을 주는 기능을 유도하게 할 수 있는 것이다. 특히 구강에서는 껌을 씹는 도중에 구강의 점막을 통하여 타액선에 도달된 필로카핀은 타액의 분비를 촉진시켜 구강의 점막을 습윤시켜 구강점막을 보존하고, 원활한 저작과 발음운동을 할 수 있게 하며 감염된 구강의 세균막(plaque)과 타액, 치석에 있는 병원균을 희석, 자정, 정균, 항균시키는 기능을 하여 병원성 미생물로부터 인체를 방어하게 하는 것이다.
그러므로 필로카핀 껌은 구강이 가지고 있는 제반 기능 즉, (1) 언어, 소화의 기능을 건강하게 유지하는데 기여하고, (2) 구강에 상존하는 병원성 미생물의 감염으로부터 인체를 방어하는데 큰 역할을 하게 된다.
본 발명에서 배합되는 필로카핀(C11H16O2N2)은 세리과(科) 필로카푸스(pilocarpus) 속(屬) 식물 잎에 함유되어 있는 알칼토이드(alkaloid)의 침상 결정체로서 아미노산인 히스티딘(Histidine)으로부터 유도할 수있다.
필로카핀은 아도로핀의 중독을 해독하는데도 사용되고 땀을 나게하는 발한제(發汗劑)로도 사용되고 있는 물질이다.
본 발명의 껌을 씹으면 껌베이스에 함유된 감미료와 향기등에 의하여 입안이 시원하고 상쾌하며 또한 껌베이스에 배합된 필로카핀이 입안의 체온과 침에 의하여 용출되어 구강세포에 흡수되고 따라서 입안의 침샘의 부교감신경을 자극시켜 부지중에 입안에 침이 많이 나오게 된다.
입안에 침이 많게 되면 입안의 세균 번식을 억제할 수 있어 구강위생에 유효할 뿐 아니라 언어 발성이 정확하게 되고 음식에 대한 소화력이 증진되는 등의 효과가 있다.

상기 실시예 1,2의 비율로 배합하여 조성된 조성물을 각각의 교반기에 의하여 중탕으로 가열하면서 균일하게 교반 배합하고 상온으로 냉각시킨 후 나머지 타액 촉진제로서 필로카핀 0.13∼0.20중량%을 균질로 배합하여 -5℃로 냉각시킨 다음 판상으로 압연하고 소정의 크기로 재단 및 포장하여 본 발명의 필로카핀이 배합된 츄잉껌을 얻는다.

Claims (2)

  1. 치콜, 라텍스 고무의 주재로 된 통상의 껌베이스에 수소화 에스텔 고무점성제, 파라핀 왁스, 스테아린산, 키스톨 당알코올을 배합하여 조성된 2.5∼3g의 베이스 조성물에 4mg∼5mg의 필로카핀을 균질로 배합하여서 된 것을 특징으로 하는 필토카핀이 배합된 츄잉껌.
  2. 스틸렌-부다디엔 공중합물 30중량％, 수소화 에스텔 고무점성제 3중량%, 파라핀 왁스 5중량％, 스테아린산 2중량%, 당 알코올 50∼60중량％를 배합한 껌베이스에 있어서, 껌베이스에 0.13∼0.20중량％의 필로카핀을 배합하는 것을 특징으로 한 필로카핀이 배합된 츄잉껌.
