Abstract

요오드화 GHRH 표지자를 사용하는 독특한 결합 검정법을 사용하여 성장 호르몬 방출 호르몬(GHRH)수용체 결합을 특성화했다. 광친화성 GHRH 표시자는 수용체의 광라벨링 및 특성화를 위해 구성했다.
게다가, 고 친화성 비오틴화 GHRH 동족체를 구성했다. GHRH-R/GHRH 복합체의 용도 및 온화한 세정액, 이어서 친화 크로마토그래피를 사용하는 특이적으로 결합된 GHRH 의 추출은 실질적으로 정제된 GHRH-R 분리물을 산출했다. GHRH- R 분리물의 전기영동 처리는 수용체의 서열화를 수행하기에 충분한 순도의 GHRH-R 을 생성한다.

제2도는 포화 결합 분석을 위해 사용된 그래프이다.
제3도는 수용체의 광친화성 가교를 나타내는 SDS겔의 방사선 사진이다.
제4도는 10nM GHRH 에 의한 경쟁을 나타내며 또한 탈글리코 실화를 보이는 CHAPS 추출 ANB-NOS 가교를 보이는 제3도에서와 같은 SDS 겔의 방사선 사진이다.

Claims (21)

  1. 서열화하기에 충분한 순도의 성장 호르몬 방출 호르몬 수용체.
  2. 성장 호르몬 방출 호르몬 수용체 분리물.
  3. 제2항에 있어서, 성장 호르몬 방출 호르몬 수용체를 서열화 하기에 충분한 순도인 분리물.
  4. 복합 성장 호르몬 방출 수용체를 친화 크로마토그래피 지지체 상에 고정시킨후 상기 지지체상의 상기 복합체를 해리시켜 성장 호르몬 방출 호르몬 수용체 분리물을 생성하는 단계들로 구성되는 성장 호르몬 방출 호르몬 수용체 분리를 생성방법.
  5. 제4항에 있어서, 상기 성장 호르몬 방출 홀몬 수용체는 성장 호르몬 방출 호르몬 또는 성장 호르몬 방출호르몬 동족체화 복합되며, 부가적으로 상기 복합체를 상기 지지체에 고정하기 전에, 상기 복합체를 온화한 세정액에 용해시키고, pH를 낮추므로써 상기 지지체상에 고정된 복합체를 해리시키는 단계들을 포함하는 방법.
  6. 제5항에 있어서, 상기 수용체는 성장 호르몬 방출 호르몬 동족체에 결합되고, 상기 동족체는 친화 크로마토그래피 지지체에 결합할 수 있는 관능을 갖는 방법.
  7. 제6항에 있어서, 상기 동족체는 비오친화되고 상기 크로마토 그래피 지지체는 상기 비오틴화 동족체에 결합할 수 있는 스트렙타비딘 관능가를 함유하는 방법.
  8. 성장 호르몬 방출 호르몬 수용체에 결합할 수 있고 친화 크로마토그래피 지지체에 결합할 수 있는 성장호르몬 방출 호르몬 동족체.
  9. 제8항에 있어서, [His1, Nie27, 비오티닐-Lys41]-GHRH-(1-4)-NH2인 동족체.
  10. UV 방사선 부재하에 성장 호르몬 방출 호르몬 수용에 결합할 수 있고 UV 방사선 존재하에 성장 호르몬 방출 호르몬 수용체에 가교 할 수 있는 성장 호르몬 방출 호르몬 동족체.
  11. 제10항에 있어서, GHRHa-SANPAH로서, 이때 GHRHa가 [His1, NIe27]-GHRH-(1-32)-NH2이고, SANPAH가 설포숙신이미딜 6-(4,-아지도-2,니트로페닐아미노)헥사노에이트인 동족체.
  12. 제10항에 있어서,GHRHa가 [His1,NIe27]-GHRH-(1-32)-NH2이고 ANB-NOS 가 N-S-아지도-2-니트로벤조일옥시숙신이미드인 동족체.
  13. N-5-아지도-2-니트로벤조일옥시숙신이미드와 GHRH 동족체 [His1, NIe27]-GHRH-(1-32)-NH2의 커플링 단계로 구성되는 광-친화성 표지가 생성방법.
  14. 설포숙신이미딜6-(4´-아지도-2´-니트로페닐 암모니오)헥사노 에이트를 GHRH동족체[His1,NIe27]-GHRH-(1-32)-NH2에 커플링시키는 광-친화성 표지가 생성 방법.
  15. 세공-형성 항생제의 존재하에 조 뇌하수체 전엽 조직과 GHRH의 접촉 단계로 구성되는 조 뇌하수체 전엽조직에 대한 성장 호르몬 방출 호르몬의 결합 방법.
  16. 제5항에 있어서, 상기 항생제가 약 0.05 ㎎/㎖로 존재하는 아라메티신인 방법.

  18. 제17항에 있어서, 상기 요오드화 성장 호르몬 방출 호르몬 또는 성장 호르몬 방출 호르몬 동족체를 HPLC를 거쳐 상기 요오드화 후에 정제하는 방법.
  19. 제4항의 방법에 의해 생성된 성장 호르몬 방출 호르몬 수용체 분리물.
  20. 제7항의 방법에 의해 생산된 성장 호르몬 방출 호르몬 수용체 분리물.
