Description

프로스타글란딘 E 유도체의 안정한 제제의 제조방법
본 발명은 프로스타글란딘 E 유도체, 즉 프로스타글란딘 E 류 및 이들의 15R 체의 안정한 제제의 제조 방법에 관한 것이다. 프로스타글란딘 E 류 및 이들의 15R 체(이하에서는 ＂PGE 류＂로 약칭함)는 일반적으로 미량으로서 위산분비억제, 평활근 수축, 혈장판응집억제 등과 같은 약리작용을 나타낸다.
그러나 PGE는 극히 불안정한 화합물로서 취급이 불편하므로, 실제의 임상에 적용하기에는 부적합하다. 따라서 현재까지 각종 안정화법이 검토되어오고 있으며, 그 예로는 PGE류를 중쇄 지방산 트리글리세리드에 용해 또는 혼합하는 안정화법(일본 특개소57-20304), 알킬렌글리콜류 또는 그의 모노에스테르류에 용해 시키는 안정화법(일본 특개소53-139716), 탄소수 4-10인 제3알코올에 용해시키는 방버(일본 특개소50-145515), 식물성 오일 및 (또는) 산에스테르에 용해시키는 방법(일본특개소50-105815)등, 특정 용매에 용해시키거나 분산 매내에 분산시킴으로써, 안정화시키는 방법이 주로 알려져 있다.
이와 같이, 용액화 또는 현탁액화하여 안정화시키는 경우, 상기 약액을 그대로 환자에게 투여하는 일은 적으며 일반적으로 복용 및 휴대가 간편하도록 임의의 방법으로 제제화한 후, 임상 적용되는 경우가 많다. 상기 경우에 복용 및 휴대성 등을 고려하여 액체를 충전할 수 있는 제제로서 가장 일반적인 연질 캡슐제로 제조하는 방법이 많이 이용되나, 용매나 분산매를 선택 사용하여 안정화시킨 PGE류가 연질 캡슐제로 제조할 경우 그의 안정성이 손상되는 문제점이 있다.
상기 원인은 해명되지 못하고 있으며, 따라서 현재 PGE류의 연질 캡슐제의 안정성을 확보하는데 필요하다고 인정되는 각종 수단을 강구하고 있는 실정이다.
예를 들면, 완전방습포장을 하는 견지에서, 포장에 다대한 비용을 들이거나, 제품의 수송 및 보관도 보냉한 상태로 행하거나, 나아가 환자에게 인도한 후에도 냉장고내 보관을 엄중히 지시하는 등, 제조자 및 소비자 모두에게 큰 부담이 되는 수단을 시도해왔다. 이와 같은 노력에도 불구하고 충분한 효과는 얻어지지 못하고 있다.
본 발명자들은 이와 같은 사실들을 감안하여, 안정화법에 대하여 여러가지 검토해온 결과, PGE류의 약액을 연질 캡슐이 아니 경질 캡슐에 충전할 경우함유된 PGE류의 안정성이 현저하게 향상되고, 상온에서의 취급 및 보관이 가능하게 됨을 알아내어 본 발명을 완성하였다.
즉 본 발명은 프로스타글란딘 E 류 및/또는 이들의 15R 체로부터 선택되는 1 또는 2이상의 프로스타글란딘류 함유액을 경질캡슐에 충전함을 특징으로 하는 프로스타글라딘 E 유도체의 안정한 제제의 제조방법에 관한 것이다.
본 발명에서 사용하는 경질캡슐은 일본 약국방 규격에 적합한 것이면 모두가 사용하능하다. 또한 필요에 따라 장용성 경질 캡슐(예, 일본특개소49-42819호 명세서에 기재되어 있음)등도 사용할 수 있다.
경질 캡슐은 주약제를 충전한 후, 캡과 보디의 접합 부분을 젤라틴 등을 사용하여 접착시켜 내용액의 누출을 방지할 수 있으므로 바람직하다.
본 발명의 대상이 되는 PGE 유도체로는 프로스타글란딘 E1(이하 ＂PGE1＂으로 약칭함), 프로스타글란딘 E2(이하 ＂PGE2＂로 약칭함), 13,14-디히드로-PGE1, 13,14-디히드로-PGE2, 15-메틸-PGE1, 15-메틸-PGE2, 15-메틸-13,14-디히드로-PGE1, 15-메틸-13,14-디히드로-PGE2, 16-메틸-PGE1, 16-메틸-PGE2, 16-메틸-13,14-디히드로-PGE1, 16-메틸-13,14-디히드로-PGE2, 17-메틸-PGE1, 17-메틸-PGE2, 15,16-디멜틸-PGE1, 15,16-디멜틸-PGE2, 16-페닐-ω-트리놀-PGE1, 16-페닐-ω-트리놀-PGE2, 16-페닐-ω-트리놀-13,14-디히드로-PGE1, 16-페닐-ω-트리놀-13,14-디히드로-PGE2, 16-시클로헥실-ω-트리놀-PGE1, 16-시클로헥실-ω-트리놀-PGE2, 16-시클로헥실-ω-트리놀-13,14-디히드로-PGE1, 16-시클로헥실-ω-트리놀-13,14-디히드로-PGE2, 및 이들의 에스테르체 및 이들의 15R체 등이 있다.
또한 본 발명에서 사용하는 용매, 분사매로는 상기 용매 또는 분산매내에 PGE 류를 용해 또는 균일하게 분산 시키고 안정화시킬 수 있는 것으로서, 동시에 캡슐 제피와 상호작용을 일으켜 제피의 물성을 변화시키는 것이 아니면 무엇이든지 가능하다. 대표적이 예로는 중쇄 지방산 트리글리세리드, 글리세릴 트리아세테이트 또는 땅콩오일, 참기름 등의 식물성 오일을 들 수 있다.
하기 실험에서는 본 발명의 유용성을 입증한다.
[실험예]
PGE 류 10mg을 트리카프릴린 또는 글리세릴 트리 아세테이트에 용해시켜 전량이 100g이 되도록 한다. 이어서 이를 100mg씩 분취하여 연질 캡슐 및 경질캡슐로 제조한다. 상기에서 수득된 PGE 류 제제의 안정성을 표 1에 나타낸다. 상기 연질 캡슐제의 제피는 젤라틴 63%, 글리세린 29.61%,정제수 7%, 메틸파라벤 0.23%, 프로필파라벤 0.06%로 이루어진다. 또한 경질캡슐로는 일본 약국방 4호 캡슐을 사용한다.
40℃에서 3개월 동안 방치한 후, 각 캡슐내의 주약제의 잔존율을 하기 방법으로 산출한다.
캡슐 10개의 내용액을 회수한 후, 이를 얇은 막 크로마토그래피법으로 분리한다. 상기 얇은 막 크로마트크리피법은 클로로포름/테트라히드로푸란/진한 빙초산 용액(10:2:1)을 전개 용매로, 및 얇은 막 크로마토그래피용 플레이트(Kiesegel F254/영국 메르크사의 상표)를 사용하여 수행하고, 인몰리브덴산의 에탄올 용액을 분무한 후, 각 점적의 흡광도를 덴시토 미터로 측정하여 각 점적의 피크면적을 구하고, 그 면적비로부터 PGE 류의 함유량을 산출한다.

하기 실시예에서는 본 발명을 상세히 설명하며, 본 발명은 하기 실시예에 한정되는 것은 아니다.
[실시예 1]
PGE 10mg을 트리카프릴린에 용해시켜 전량이 100g이 되도록 한다. 이어서 이를 150mg씩 분취하여 일본 약국방 4호 경질캡슐에 충전하고, 캡슐의 캡과 보디의 접합부를 젤라틴을 사용하여 접착시킨다.
[실시예 2-11]
하기 표 2에 나타낸 주약제 용매 또는 분사매, 경질캡슐을 사용하여, 실시예 1과 동일한 방법으로 인정한 PGE 류 제제를 제조한다.

  1. 프로스타글란딘 E 류 및 이들의 15R 체로 구성된 프로스타글란딘 E 유도체로부터 선택된 1종 이상의 프로스타글란딘류를 함유하는 용액 또는 현탁액을 경질 캡슐에 충전함을 특징으로 하는, 프로스타글란딘 E 유도체의 안정한 제제의 제조방법.
