Description

에스트로겐/프로게스틴 경피용량 단위, 시스템 및 방법

제1도는 두 개의 분리된 약제 저장층, 즉 에스트로겐을 함유하는 층(점을 사용하여 약제의 존재를 나타낸 층) 및 프로게스틴을 함유하는 층(납작한 타원형으로 약제의 존재를 나타낸 층)을 포함한 6개의 층을 가진 본 발명의 투여단위의 횡단면도이며,
제3도는 5개의 층, 즉(1) 방출층, (2)레보로르게스트렐/증진제층, (3)오파놀(Oppanol) B80, (4)아크릴 접착체중의 에스트라디올, 및 (5) 지지층을 가진 본 발명의 투여단위의 횡단면도이고,
제3도는 젊은 여성 사체의 피부를 통한 각각의 피부투과 속도에 대하여 프로게스틴 및 에스트로겐 함유 층을 분리하는 층의 두께가 달라지는 효과를 나타낸 그래프이다.

Claims (38)

  1. a) 경피 전달되는 에스트로겐 및 프로게스틴을 사실상 통과시키지 않는 지지층, b) 상기 지지층에 접착되어 있으며, 경피 흡수될 수 있고 약제학적으로 허용되는 유호 투여량의 하나 이상의 유효한 에스트로겐이 용해 및/또는 마이크로분산되어 있는 중합체(여기서, 중합체는 생물학적으로 허용되며 상기 하나 이상의 에스트로겐에 적합한 환경을 제공하고 상기 하나 이상의 에스트로겐이 경피 흡수될 수 있게 전달한다) 층 및 c) 경피 흡수될 수 있고 약제학적으로 허용되며 노르겐스트렐, 레보르게스트렐 및 이들의 생물학적 혼화성 유도체로 이루어진 그룹 중에서 선택된 유효 투여량의 하나 이상의 유효한 프로게스틴이 용해 및/또는 마이크로분산되어 있으며, 생물학적으로 허용되고 상기 하나 이상의 프로게스틴이 용해 및/또는 마이크로본산되어 있으며, 생물학적으로 허용되고 상기 하나 이상의 유효한 프로게스틴이 용해 및/또는 마이크로분산되어 있으며, 생물학적으로 허용되고 상기 하나 이상의 프로게스틴에 적합한 환경을 제공하며 상기 하나 이상의 프로게스틴 및 상기 하나 이상의 에스트로겐 (여기서, 이들 호르몬을 제공하여 임신을 조절하거나 에스트로겐 대체 요법이 수행되도록 한다)이 경피 흡수될 수 있게 전달하고, 유효량의 경피 피부 흡수 강화제를 갖는, 상기 중합체 층을 포함하는 경피 에스트로겐/프로게스틴 투여단위.
  2. 제1항에 있어서, 상기 하나 이상의 에스트로겐으로서 에티닐 에스트라디올 또는 17-베타-에스트리디올 또는 이들의 혼합물을 갖는 경피 투여 단위.
  3. 제1항에 있어서, 하나 이상의 프로게스틴으로서 노르게스트렐 또는 레보노르게 스트렐 또는 이들의 혼합물을 갖는 경피 투여 단위.
  4. 제1항에 있어서, 에스트로겐으로서 에티닐 에스트라디올을 가지며 프로게스틴으로서 노르게스트렐 또는 레보노르게스트렐을 갖는 경피 투여 단위.
  5. 제1항에 있어서, 에스트로겐으로서 17-베타-에스트라디올을 가지며 프로게스틴으로서 노르게스트렐 또는 레보노르게스트렐을 갖는 투여 단위.
  6. 제1항에 있어서, 접착제 층 또는 중합체 층 또는 두층 모두가 폴리아크릴 접착제 중합체로부터 제조된 경피 투여 단위.
  7. 제6항에 있어서, 폴리아크릴 접착제 중합체가 하기 일반식을 갖는 경피 투여 단위.
    상기식에서, X는 접착제 층 또는 중합체 층 또는 두층 모두의 정해진 특성을 제공하기에 충분한 반복단위의 수를 나타내며, R은 수소 또는 저급알킬 중에서 선택된다.
  8. 제1항에 있어서, 지지층이 미세다공성이며 호흡이 가능한 경피 투여 단위.
  9. 제1항에 있어서, 접착제 층 또는 중합체 층 또는 두층 모두가 실리콘 접착제 중합체 또는 플리이소부틸렌 접착제 중합체로부터 제조된 경피 투여 단위.
  10. 제1항에 있어서, 프로게스틴에 대한 강화 인수가 1.2, 1.3, 1.5 또는 2.0 이상인 경피 투여 단위.
  11. 제1항에 있어서, 피부 침투 흡수도 정화제가 n-데실 알콜을 포함하는 경피 투여 단위.
  12. 제1항에 있어서, 약 1주일 동안 적어도 최소한의 1일 투여량의 에스트로겐 및 프로게스틴을 효과적으로 제공하며, 프로게스틴에 대해 약 1.5이상의 강화 인수를 제공하는 일정량의 피부 침투 흡수도 강화제를 갖는 경피 투여 단위.
  13. 제1항에 있어서, 하나 이상의 에스트로겐이 존재하는 중합체 층 및 하나 이상의 프로게스틴이 존재하는 접착체 층이 하나 이상의 에스트로겐이 목적하는 대로 경피 흡수되게 전달되는 생물학적 접착제 또는 중합체 분리층(여기서, 분리층은 에스트로겐, 프로게스틴 및 강화제를 함유하지 않거나 사실상 함유하지 않는 접착제 또는 중합체를 사용하여 제조된다)에 의해 분리되지만 각자 이들과 밀착되어 있는 경피 투여 단위.
  14. 제13항에 있어서, 하나 이상의 에스트로겐으로서 에티닐 에스트라이올 또는 17-베타-에스트라디올 또는 이들의 혼합물을 가지며 하나 이상의 프로게스틴으로서노르게스트렐 또는 레보노르게스트렐을 갖는 경피투여 단위.
  15. 제1항에 있어서, 분리층이 치수적으로 안정한 분리층을 제공하며 하나 이상의 에스트로겐 전달 속도를 현저하게 감소시키기에 충분한 고점도 또는 분자량을 갖는 생물학적으로 허용되는 접착제 또는 중합체로부터 제조된 경피 투여 단위.
  16. 제15항에 있어서, 분리층이 플라이소부틸렌 접착제로부터 제조된 경피 투여 단위.
  17. 제13항에 있어서, 에스트로겐 호르몬에 대한 경피 흡수된 프로게스틴의 약 1/1내지 약 13/1의 범위인 경피투여 단위.

  23. 제22항에 분리층이 약 75내지 약 125μ의 두께를 가지며 폴리이소부틸렌 약 800,000이상의 상대 분자질량 Mv(평균점도)를 갖는 경피 투여 단위.
  24. 임신 조절을 필요로 하는 대상자의 피부에서 제1항의 투여 단위 하나 이상을 사용하여 연속적인 월경주기에 있어서 약 3주간의 적당한 월경주기 기간 동안 유효한 1일 투여량의 에스트로겐 및 프로게스틴을 제공함으로써 임신을 조절하는 방법.
  25. 제24항에 있어서, 에스트로겐이 에티닐 에스트라이올 또는 17-베타 에스트라디올 또는 이들의 혼합물이고 프로게스틴이 노르게스트렐 또는 레보노르게스트렐 또는 이들의 혼합물인 방법.

  34. 제30항에 있어서, 제21항의 경피 투여 단위를 사용하는 임신 조절 시스템.
  35. 제30항에 있어서, 제22항의 경피 투여 단위를 사용하는 임신 조절 시스템.
  36. a) 경피 전달되는 에스트로겐 및 프로게스틴을 사실상 통과시키지 않는 지지층, b) 상기 지지층에 접착되어 있으며, 경피 흡수될 수 있고 약제학적으로 허용되는 유효 투여량의 하나 이상의 유효한 에스트로겐이 용해 및/또는 마이크로분산되어 있는 중합제(여기서, 중합체는 생물학적으로 허용되며 상기 하나 이상의 에스트로겐에 적합한 환경을 제공하고 상기 하나 이상의 에스트로겐이 경피 흡수될 수 있게 전달한다)층, c) 경피 흡수될 수 있고 약제학적으로 허용되며 유효 투여량의 하나 이상의 유효한 프로게스틴이 용해 및/또는 마이크로분산되어 있으며, 생물학적으로 허용되고 상기 하나 이상의 프로게스틴에 적합한 환경을 제공하며 상기 하나 이상의 프로게스틴 및 상기 하나 이상의 에스트로겐 이 경피 흡수될 수 있게 전달하고, 유효량의 경피 피부흡수 강화제를 갖는 상기 중합체 층 및 d) 중합체층과 접착제 층의 사이에 위치하여 이들과 밀착되어 있으며 하나 이상의 에스트로겐(여기서, 호르몬은 중합체 및 접착제 층 내에서 인정하며 동시에 흡수되어 적어도 최소한의 유효한 1일 투여량의 상기 호르몬을 제공하여 임신을 조절하거니 에스트로겐 대체 요법이 수행되도록 한다)이 통과하여 목적하는 만큼 경피 흡수되게 하고 에스트로겐, 프로게스틴 및 강화제를 함유하지 않거나 사실상 함유하지 않는 접착제 또는 중합체를 사용하여 제조한 생물학적으로 허용되는 상기 접착제 또는 중합체로 이루어진 분리층을 포함함을 특징으로 하며, a) 프로게스틴으로서 레보노르게스트렐 또는 또다른 프로게스틴으로서 생물학적으로 동등한 양이 경피 흡수되느 레보노르게스트렐을 사용하고 에스트로겐으로서 17-베타-에스트리디올 또는 또아른 에스트로겐으로서, 생물학적으로 동등한 양이 경피 흡수되는 17-베타-에스트라디올을 사용시 프로게스틴/에스트로겐 경피 흡수 속도비가 약 75/1 내지 약1.27/1인 제1투여 단위, b)프로게스틴으로서 레보노르게스트렌 또는 또 다른 프로게스틴으로서 생물학적으로 동등한 양이(경피 흡수되는 레보노르게스트렐을 사용하고 에스트로겐으로서 17-베타-에스트라디올 또는 또다른 에스트로겐으로서 생물학적으로 동등한 양이 경????수되는 17-베타-에스트라디올을 사용시 프로게스틴/에스트로겐 경피 흡수 속도비가 약 1.25내지 약2.5/1 제2투여 단위, c) 프로게스틴으로서 레보노르게스트렌 또는 또다른 프로게스틴으로서 생물학적으로 동등한 양이 경피 흡수되는 레보르게스트렐을 사용하고 에스트로겐으로서 17-베타-에스트라디올 또는 또 다른 에스트로겐으로서 생물학적으로 동등한 양이 경피 흡수되는 17-베타-에스트라디올을 사용시 프로게스틴/ 에스트로겐 경피 흡수 속도비가 약2/1 내지 약5/1인 제3투여 단위를 추가 특징으로 하는, 각각 약 1주 동안 적어도 최소한의 유효한 1일 투여량의 에스트로겐 및 프로게스틴을 제공하는 경피 흡수 투여 단위 3개를 포함하며, 이들 각각을 약 1주씩, 즉, 제1투여 단위는 월경 주기의 약 5일 째에, 제2투여 단위 및 제3투여 단위는 각각 이보다 약 7일 후 및 약 14일 후에 사용하여 임신을 조절하는 임신 조절 시스템.

  38. 에스트로겐 대체 요법을 필요로하는 대상자의 피부에 유효 투여량의 대체 요법용 에스트로겐 하나 이상 및 프로게스틴 하나 이상을 각각 제공하는 제1항의 투여 단위를 연속 사용함을 특징으로 하는 에스트로겐 대체요법.
