혈청알부민의 정제방법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 환원조건하의 사람 혈청 알부민의 SDS-PAGE(은-염색)를 도시한다. 탈지된 HSA:레인1(100ng); 5(500ng); 9(10μg). 정제된 HSA:레인 2,3(100ng); 6,7(500ng); 10,11(10μg). 분자량마커:레인 4,8.
제2도는 사람혈청 알부민의 등전점 전기영동(CBB R-250 염색)을 도시한다. I:레인 1,2,3의 샘플에 대한 적용점. II:레인 4,5,6의 샘플에 대한 적용점. Ⅲ : 레인 7,8,9의 샘플에 대한 적용점, 탈지된 HSA(10mg) :레인 1,4,7. 정제된 HSA(10μg): 나머지 레인.
제3도는 사람혈청 알부민의 Mono QR상에서의 고성능 이온교환 크로마토그래피를 도시한다. 상단크로마토그램:정제된 HSA;하단크로마토그램:탈지된HSA.

Claims (10)

  1. 정제된 재조합 혈청 알부민의 제조방법에 있어서, 혈청알부민이 발현되는, 혈청알부민 코드화 구조유전자를 포함하는 발현구조로 형질전환된 미생물을 배지중에서 성장시키는 단계; 청징된 발효 브로스 또는 세포 용해물로서 생성배지를 단리하는 단계; 생성배지를 알칼리성으로 만들어 알칼리배지를 침전물로부터 분리하는 단계; 상기 알칼리 배지의 pH를 농축된 혈청 알부민을 제공하기 위하여 생리적부근의 pH로 변화시키고 상기 농축된 배지를 친유성 고정상으로 크로마토그래피하여 흡착제거제로서 수용성 지질 이온을 수성상에 첨가함으로써 알부민을 용출하는 단계; 및 실질적으로 오염물이 없는 정제된 혈청 알부민을 상기 미생물로 부터 단리하는 단계로 이루어지는 방법.
  2. 제1항에 있어서, 상기 방법이 추가로 상기 크로마토그래피단계에 앞서 상기 알칼리 배지를 산성화하고 그 결과의 산성화된 배지를 음이온 교환수지와 항온 반응시키는 단계를 포함하는 것을 특징으로하는 방법.
  3. 제2항에 있어서, 상기 산성화된 배지가 4.0 내지 5.5의 범위의 pH인 것을 특징으로하는 방법.
  4. 제2항 또는 3항에 있어서, 상기 항온반응후 상기 산성화된 배지를 투석하는 추가의 단계를 포함하는 것을 특징으로하는 방법.
  5. 제1항 내지 4항중 어느하나에 있어서, 상기 알칼리 배지가 약7.5 내지 9.0의 범위의 pH인 것을 특징으로 하는 방법.
  6. 제1항 내지 5항중 어느하나에 있어서, 상기 흡착제거제가 약 6내지 12 탄소원자의 알칼리를 가지는 것을 특징으로하는 방법.
  7. 제6항에 있어서, 상기 흡착제거제가 약6내지 12탄소원자의 카르복실산염인 것을 특징으로하는 방법.
  8. 제1항 내지 7항중 어느하나에 있어서, 상기 정제된 혈청 알부민을 투석하는 추가의 단계를 포함하는 것을 특징으로하는 방법.
  9. 제1항 내지 8항중 어느하나에 있어서, 상기 미생물이 효모인 것을 특징으로하는 방법.
  10. 제1항 내지 9항중 어느하나에 있어서, 상기 혈청 알부민이 사람 혈청 알부민인 것을 특징으로하는 방법.
