제1도는 Ei-ON19 및 Ei-ON20이 프라이머(primer)이고 주형이 세포라인 U-937로부터 분리된 RNA인 경우의 프라이머 연장(extension) 실험의 결과를 도시한 것이다.
제2도는 반점-블롯 하이브리드화의 결과를 도시한 것이다.
제3a도는 HLZ cDNA(클론 HL14-1)의 구조를 도시한 것이고,
제3b도는 HLZ cDNA의 뉴클레오타이드 서열을 측정하는
제5도는 공개된 HLZ 아미노산 서열(윗줄)과 클론 HL14-1의 cDNA로부터 유도된 아미노산 서열(아랫줄)과의 비교도이다.
제6도는 다복제 하이브리드 벡터 pEAS 102를 도시한 것이다.
Claims (12)
- a) HLZ를 코드화하는 mRNA를 인체 세포라인, 바람직하게는 U-937 세포라인으로부터 분리하여, 이의 진정한 DNA 서열을 합성하고,b) 상기 DNA 서열을 플라스미드에 삽입하여 숙주 세포에서 복제 및 발현 가능한 하이브리드 플라스미드를 생성시킨후,c) 상기 하이브리드 플라스미드를 사용하여 숙주세포, 바람직하게는 원시핵 세포, 진핵세포 또는 포유동물 세포를 형질전환시켜, 상기 단백질이 숙주세포에 의해 발현되도록 한 다음,d) 숙주세포에 의해 분리된 상기 단백질을 분리하여 정제함을 특징으로 하여, 인체 라이소자임(HLZ)을 제조하는 방법.
- 제1항에 있어서, 상기 DNA 서열이 하기 서열(A) 또는 (B) 이고, 이 서열 다음에 정지코돈이 있는 방법.
- 제2항에 있어서 상기 DNA 서열을 플라스미드 pUC9의 Pst I-절단부위사이에 삽입하는 방법.
- 제1항에 있어서 형질전환에 사용되는 하이브리드 플라스미드가 플라스미드 pHL2, pHL8, pHL14-1, pHL21, pHL23, pHL14-23 또는 pHL35인 방법.
- 제4항에 있어서 HLZ를 코드화하는 DNA 서열을 엄격한 조건하에서 플라스미드 pHL14-1 또는 pHL14-23의 삽입물을 사용하여 하이브리드화시키고, 이 DAN 서열이 85% 이상, 바람직하게는 90% 이상 동종성을 나타내는 방법.
- 제1내지 5항중 어느 한 항에 있어서, HLZ를 코드화하는 상기 하이브리드 플라스미드의 DNA 영역을 프로모터, 리이더 서열 또는 터미네이터에 커플링시키고, 해독개시 시그널을 HLZ에 대한 코드화 영역과 프로모터 사이에 위치시키는 방법.
- 제1 내지 6항중 어느 한 항에 있어서 형질전환 및 발현에 사용되는 숙주 세포가 이. 콜라이(E. coli), 사카로마이세스 세레비지애(Saccharomyces cerevisiae)또는 포유동물 세포인 방법.
- 제1내지 7항중 어느 한 항에 있어서, 발현 벡터가 성숙 HLZ를 코드화하거나, 제2항에 기술한 뉴클레오타이드 서열을 함유하는 방법.
- 제8항에 있어서 발현벡터 바람직하게는 pWS290D 또는 pWS296D가 ADHⅠ프로모터 α-인자 리이더 HLZ 유전자 및 ADHⅡ 터미네이터로 이루어지거나 성분 α-인자 프로모터, α-인자 리이더, 유전자 HLZ 및 α-인자 타미네이터로 이루어지고, 상기 발현 벡터가, 분비가능한 성숙 HLZ를 만드는 방법.
- HLZ에 동종인 혼합 서열의 올리고뉴클레오타이드 푸울2개를 고체상 포스포트리-에스테르 방법으로 합성하여, 올리고뉴클레오타이드의 DNA 서열이 아미노산 서열 Ala Asn Trp Met Cys Leu 또는 Tyr Trp Cys Asn Asp Gly에 상응함을 특징으로 하여 DNA 올리고뉴클레오타이드를 제조하는 방법.
- 제7항에 기술된 숙주세포를, 플라스미드 129/29를 사용하여 형질전환시켜, 형질전환된 숙주가 복제 및 발현 가능한 벡터에서 바람직하게는 제4, 5또는 9항에 기술한 벡터에서 제2항에 기술된 뉴클레오타이드 서열을 함유함을 특징으로 하는 형질전환 된 숙주를 제조하는 방법.
- 제1내지 11항중 어느 한 항에 있어서, 재조합 HLZ 단백질이 진정한 HLZ와 동일하거나, 그의 생물학적 활성 변형체를 구성하거나 또는 하기의 아미노산 서열(Ⅰ) 또는 (Ⅱ)를 함유함을 특징으로 하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
