Claims (30)
- 아미노그룹이 보호된 Z 아미노산을 에스테르화시켜, 공유결합에 의한 불용성수지 중합체로 만든 후, Z아미노산잔기물로 부터 아미노 보호그룹을 제거하고, 아미노 그룹이 보호된 Y아미노산과 반응시켜 Y아미노산을 Z-수지에 커플링시키고, Y아미노산 잔기로부터 알파-아미노보호그룹을 제거하고, 아미노그룹이 보호된 X 아미노산과 반응시켜 X 아미노산을 Y-Z-수지에 커플링시키고, X 아미노산 잔기로 부터 아미노 보호그룹을 제거하고, 아미노그룹이 보호된 W아미노산과 반응시켜 W아미노산을 X-Y-Z-수지에 커플링시키고, W아미노산 잔기로부터 아미노 보호그룹을 제거하고, 아미노 그룹이 보호된 N-R 치환된 V아미노산과 반응시켜 N-R 치환된 V아미노산을 W-X-Y-Z-수지에 커플링시키고, 산(R=OH), 암모니아(R1=NH2) 또는 적절한 아민을 사용하여 펩타이드로 부터 수지를 분리시킨 후, 모든 보호그룹들을 제거함을 특징으로 하여, 다음 일반식(I)의 펩타이드 및 이의 약제학적으로 무독한 산-또는 염기-부가염을 제조하는 방법.R-V-W-X-Y-Z-R1(I)상기식에서, R은 H,NH2, 아실-NH, CH3NH 또는 피로(pyro)-GLU-NH이고,V는이고, W는 PRO, 데하이드로 PRO 또는[여기에서, A는 각기 저급알킬이고, B는 각기 H(A가 C4-C6저급 알킬인 경우) 또는 C1-C3저급알킬이거나, A 및 B는 함께 -(CH2)4- 또는 -(CH2)5-를 형성한다]이고,X는 D-ASP,ASP,D-GLU 또는 GLU이고, Y는 GLY,VAL,LEU,노르(nor)-LEU,PHE,ILE,LYS,GLN,GLU,ALA,D-VAL,D-LEU,D-노르 LEY,D-PHE,D-ILE,D-LYS,D-GLN,D-GLU 또는 D-ALA이고,(여기에서 D는 H이거나, 페놀 프로톤의 산성도를 증가시키거나 거의 감소시키지 않는 하나 또는 두개의 치환체이고, E는 H 또는 C1-C3저급알킬이다)이고, R1은 OH,NHR",(여기에서 R" 및 R"'는 각기 H 또는 저급알킬이다)이고, 단, R이 H가 아닌 경우 V는 D- 또는 L-이성체일 수 있고, V,X, 및 Y중 하나만이 D 아미노산이다.
- 제1항에 있어서, 다음 일반식(I)의 펩타이드 및 이의 약제학적으로 무독한 산-또는 염기-부가염의 제조방법.R-V-W-X-Y-Z-R1(I)상기식에서, R-V는 아실-ARG 또는 아실-D-ARG이고, W는 PRO 또는 AIB이고 X는 D-ASP,ASP,D-GLU 또는 GLU이고, Y는 GLY,VAL,LEU,노르-LEU,PHE,ILE,LYS,GLN,GLU,ALA,D-VAL,D-LEU,D-노르 LEY,D-PHE,D-ILE,D-LYS,D-GLN,D-GLU 또는 D-ALA이고,(여기에서 D는 H이거나, 페놀 프로톤의 산성도를 증가시키거나 거의 감소시키지 않는 하나 또는 두개의 치환체이고, E는 H 또는 C1-C3저급알킬이다)이고, R1은 OH,NHR",(여기에서 R" 및 R"'는 각기 H 또는 저급알킬이다)이고, 단, V,X, 및 Y중 하나만이 D 아미노산이다.
