치료용 항체 시장은 1986년에 승인된 이후 급속히 성장해오고 있다. 기존 단클론 항체가 비용이 많이 들고 힘든 포유류 발현 시스템을 필요로 하고 조직 침투 문제를 겪는 문제로 인해, 이의 대안으로 다양한 항체 단편 형식이 연구되고 있다.
이러한 항체 유도체 중에서 scFvs(single-chain variable fragments)는 1988년에 여러 scFv 기반 치료제가 FDA의 승인을 받아 임상 가능성을 입증받았다. scFv는 하나의 중쇄 및 경쇄 가변 도메인(VH 및 VL) 면역글로불린(Igs)은 폴리펩티드 링커에 의해 연결되는 구조이다. scFv는 원치 않는 결정화 단편(Fc) 매개 면역 반응을 일으키지 않고, 전장 항체보다 종양 침투 및 분포가 더 좋은 이점이 있지만, scFv의 혈청은 일반적으로 크기가 작고(약 25kDa) Fc 영역이 없기 때문에 반감기가 매우 짧은 단점이 있다.
이에 그 유용성에도 불구하고 임상적 적용이 혈액암이나 황반변성과 같은 일부 경우에 국한되고 있는 실정으로, 이를 개선할 수 있는 새로운 scFv 기반 치료제 개발이 필요하다.
본 발명은 신규 구조의 단일쇄 가변 단편의 변이 단백질을 제공하는 것을 목적으로 한다.
본 발명은 단일쇄 가변 단편의 변이 단백질을 포함하는 암의 예방 또는 치료용 약학 조성물을 제공하는 것을 목적으로 한다.
1. 항체의 중쇄 가변 영역(VH)과 경쇄 가변 영역(VL)이 링커로 연결된 단일쇄 가변 단편(scFv)에 알부민 결합 모이어티가 융합된, 단일쇄 가변 단편 변이 단백질.

알부민 결합 모이어티가 링커 내에 결합되는 경우 중쇄 가변 영역의 C-말단과 경쇄 가변 영역의 N-말단으로부터 각각 2 aa 이상, 3 aa 이상, 4 aa 이상, 5 aa 이상, 6 aa 이상, 7 aa 이상, 8 aa 이상, 9 aa 이상 또는 10 aa 이상 이격된 위치에 융합되는 것이 바람직하다.
예컨대 10 내지 15 aa 길이를 갖는 링커의 4 내지 8 번째 아미노산과 이의 인접한 아미노산 사이에 융합되는 것일 수 있다. 또는 16 내지 20 aa 길이를 갖는 링커의 7 내지 14 번째 아미노산과 이의 인접한 아미노산 사이에 융합되는 것일 수 있다.
구체적으로는 서열번호 10 또는 11의 링커의 5 내지 8번째 아미노산과 이의 인접한 아미노산 사이에 융합되거나, 서열번호 12의 링커의 11 내지 16번째 아미노산과 이의 인접한 아미노산 사이에 융합되는 것일 수 있다.
또는 서열번호 10 또는 11의 링커의 6번째와 7번째 아미노산 사이, 또는 서열번호 12의 링커의 13번째와 14번째 아미노산 사이에 융합되는 것일 수 있으나 이에 제한되는 것은 아니다.
알부민 결합 모이어티가 중쇄 가변 영역의 C-말단과 경쇄 가변 영역의 N-말단으로부터 적어도 3 aa 이격되어 융합되지 않는 경우 인접한 상보성 결정 영역(CDR)과의 간섭이 일어나 항체에 대한 결합력이 떨어질 수 있다.
알부민 결합 모이어티가 링커 내에, 또는 단일쇄 가변 단편의 C-말단에 융합되는 경우에는 알부민 결합 모이어티는 중쇄 가변 영역 및 경쇄 가변 영역의 상보성 결정 영역(CDR)이 위치하는 방향과 반대 방향에 위치하게 된다. 여기서 반대 방향이라는 것은 예컨대 상보성 결정 영역(CDR)이 단일쇄 가변 단편 변이 단백질의 아래쪽에 위치하면 알부민 결합 모이어티는 위쪽에 위치한다는 것이다. 또한 반대 방향이라는 것은 예컨대 상보성 결정 영역(CDR)이 단일쇄 가변 단편 변이 단백질의 좌측에 위치하면 알부민 결합 모이어티는 우측에 위치한다는 것이다.
알부민은 인간 혈청 알부민(HSA)일 수 있다.
본 발명 변이 단백질은 상기 알부민 결합 모이어티를 통해 생체 내 투여시 혈청 알부민과 결합하여 변이 단백질의 대사 및 분해를 지연시키고, 암세포에 대한 투과성을 높일 수 있다.
알부민 결합 모이어티가 단일쇄 가변 단편의 링커 내에 융합되면 단일쇄 가변 단편의 N-말단 또는 C-말단에 생리활성물질을 추가 결합할 수 있다. 또한 알부민 결합 모이어티가 단일쇄 가변 단편의 C-말단이외의 위치에 융합되는 경우 단일쇄 가변 단편의 C-말단에는 생리활성물질을 결합시킬 수 있다. 이 경우 본 발명의 단일쇄 가변 단편 변이 단백질은 생리활성물질을 타겟에 전달하는 전달체로 기능한다.
생리활성물질은 체내에 투여되었을 때 소정의 활성을 나타낼 수 있는 모든 물질을 의미한다. 생리활성물질은 예컨대 사이토카인, 인슐린, 인슐린 유사 성장인자(IGF)-1, IGF-2, 상피 성장인자(EGF), 변환성장인자(TGF), 인간 성장 호르몬 및 혈관 내피 성장인자(VEGF)로 이루어진 군에서 선택되는 어느 하나일 수 있다.
사이토카인은 예컨대, 인터루킨(IL-1 내지 IL-17), 종양 괴사 인자(TNF), 인터페론(IFN), 에리스로포이에틴(EPO), 트롬보포이에틴(TPO) 등일 수 있다.
알부민 결합 모이어티는 단일쇄 가변 단편의 링커 아미노산 서열의 중앙에 융합되는 것일 수 있다. 또는 중앙으로부터 1 내지 5 aa 또는 1 내지 10 aa 떨어진 아미노산과 이와 인접한 아미노산 사이에 융합되는 것일 수 있다.
알부민 결합 모이어티는 예컨대 알부민 단백질 도메인, 어피바디 또는 펩타이드일 수 있다. 이는 항체(IgG)와 비교하여 작은 크기이며 scFv에 융합되어 단일한 폴리펩타이드 체인을 구성한다. 이는 구조에 이황화 결합을 포함하지 않는 것일 수 있다.
알부민 결합 모이어티는 특정 단백질의 일부분일 수도 있고, 인공적으로 설계된 도메인일 수 있다.
예컨대 이는 그람 양성 박테리아에 의해 발현되는 다양한 표면 단백질에서 발견되는 작은 3중 나선 단백질 도메인일 수 있다. 구체적으로는 streptococcal protein G 또는 Finegoldia magna의 protein PAB에서 유래하거나 이로부터 알부민 결합 친화도를 더욱 높이도록 변형된 것일 수 있다.
예컨대 이는 바다상어의 카르틸라지 올리고머 매트릭스 단백질에서 유래한 VNAR(V domain of cartilage oligomeric matrix protein, chondrocyte-derived)이거나 이로부터 알부민 결합 친화도를 더욱 높이도록 변형된 것일 수 있다.
구체적으로 알부민 결합 모이어티는 서열번호 1 내지 3의 아미노산 서열을 갖는 것일 수 있다.
본 발명의 변이 단백질은 해당 단백질을 코딩하는 서열을 발현하는 미생물 내에서 제조되는 것일 수 있다.
미생물은 당 분야에 공지된 미생물이 제한없이 사용될 수 있다. 예를 들면 대장균(Escherichia coli)일 수 있으나, 이에 제한되는 것은 아니다.
미생물 시스템으로 변이 단백질을 제조하는 경우에는 단백질이 PTM(post-translational modification)을 포함하지 않고 이황화 결합이 없거나 적어야 발현이 용이하다. lgG 등 항체 형태에는 PTM와 이황화 결합을 다수 포함하며, 화합물을 이용하는 경우에는 추가적인 결합과 정제과정이 필요하다. 본 발명의 변이 단백질은 융합된 하나의 폴리펩타이드 형태이며, PTM이 없고 이황화 결합이 적은 특징으로 미생물 시스템 발현을 용이하게 할 수 있다.
전술한 scFv의 링커 서열은, 제조 과정에 있어 유전자 클로닝에서 알부민 결합 모이어티의 삽입을 위해, 제한효소가 인식할 수 있는 부위로 일부분이 대체되거나 이들을 더 포함할 수 있다. 예컨대 알부민 결합 모이어티의 양측에 위치한 링커 서열에서 제한효소(Spe I)의 인식부위인 TS 서열로 일분이 대체되거나 이를 더 포함할 수 있다. TS 서열의 잔기는 상기 링커의 G, S 서열의 잔기와 유사한 이점이 있다.
본 발명은 이상의 단백질을 포함하는 암의 예방 또는 치료용 약학 조성물을 제공한다. 이상의 단백질에 관하여 설명된 사항은 모두 본원의 약학 조성물의 유효성분으로서의 단백질에 그대로 적용된다.

본 발명의 약학 조성물은 약학적으로 허용 가능한 담체를 포함할 수 있다. 본 발명에서 용어 "약학적으로 허용 가능한 담체"란 생물체를 상당히 자극하지 않고 투여 성분의 생물학적 활성 및 특성을 저해하지 않는 담체 또는 희석제를 말한다. 본 발명에서의 약학적으로 허용 가능한 담체는 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로즈 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 또는 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액 및 정균제 등 다른 통상의 첨가제를 첨가하여, 조직 또는 장기에 주입하기에 적합한 주사제의 형태로 제형화할 수 있다. 또한, 등장성 멸균 용액, 또는 경우에 따라 멸균수나 생리 식염수를 첨가하여 주사 가능한 용액이 될 수 있는 건조 제제(특히 동결 건조제제)로 제형화할 수도 있다. 또한, 표적 기관에 특이적으로 작용할 수 있도록 표적 기관 특이적 항체 또는 기타 리간드를 상기 담체와 결합시켜 사용할 수 있다.

본 발명에서 용어, "유효량"은 목적하는 치료되어야 할 특정 질환의 발병 또는 진행을 지연하거나, 전적으로 증진시키는데 필요한 양을 의미한다.
본 발명에서 조성물은 약학적 유효량으로 투여될 수 있다. 상기 약학 조성물의 적합한 총 1일 사용량은 올바른 의학적 판단범위 내에서 처치의에 의해 결정될 수 있다는 것은 당업자에게 자명한 일이다.
본 발명의 목적상, 특정 환자에 대한 구체적인 약학적 유효량은 달성하고자 하는 반응의 종류와 정도, 경우에 따라 다른 제제가 사용되는지의 여부를 비롯한 구체적인 조성물, 환자의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여시간, 투여 경로 및 조성물의 분비율, 치료 기간, 구체적 조성물과 함께 사용되거나 동시 사용되는 약물을 비롯한 다양한 인자와 의약 분야에 잘 알려진 유사 인자에 따라 다르게 적용하는 것이 바람직하다.
본 발명에 있어서, 상기 약학 조성물은 필요에 따라 약물의 제조, 사용 및 판매를 관장하는 정부 기관에 의해 지시된 형태로 포장과 연계된 지시서가 수반될 수 있으며, 상기 지시서는 조성물의 형태 또는 인간이나 동물 투여에 관하여 사익 기관의 승인을 나타내고 있고, 예를 들어, 약물의 처방에 대하여 미국 식품의약국에서 승인된 표지일 수 있다.
본 발명은 이상의 단백질을 투여하는 단계를 포함하는 암의 치료 방법을 제공한다. 이상의 단백질에 관하여 설명된 사항은 모두 본원의 암의 치료 방법의 유효성분으로서의 단백질에 그대로 적용된다.
본 발명의 방법은 암에 걸린 개체에 상기 단백질을 투여하는 단계를 포함한다.
암에 걸린 개체는 암에 걸린 동물, 구체적으로 암에 걸린 포유류일 수 있고, 보다 구체적으로는 암에 걸린 인간일 수 있다.
단백질은 치료상 유효량으로 투여될 수 있다.
본 발명에서 용어 "투여"는 어떠한 적절한 방법으로 환자에게 본 발명의 조성물을 도입하는 것을 의미하며, 본 발명의 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 경구 또는 비경구의 다양한 경로를 통해 투여될 수 있다. 복강내 투여, 정맥내 투여, 근육내 투여, 피하 투여, 피내 투여, 경구 투여, 국소 투여, 비내 투여, 폐내 투여 또는 직장내 투여될 수 있으나, 이에 제한되지 않는다.
이하, 본 발명을 실시예를 들어 보다 상세하게 설명한다.
실시예
scFv 및 알부민 결합 도메인의 종류 및 알부민 결합 도메인 위치를 달리하여 실시예 1-9 및 비교예 1-4을 설계하고 구조 모델을 확인하였다. scFv 단백질의 아미노산 서열은 링커에 의해 연결된 N-말단 VH 및 C-말단 VL 영역으로 구성된다. N-말단에 NcoI 효소의 제한 부위로 'MG' 서열이 삽입되었고, C-말단에는 단백질 정제 태그로 헥사히스티딘 태그(His-tag) 서열이 삽입되었다. 알부민 결합 도메인의 삽입을 위해 아미노산 잔기 "TS"를 암호화하는 SpeI 제한 부위를 사용했다.

도 1 내지 14에서 4D5 scFv(서열번호 4, 도 1), 실시예 1-9 및 비교예 1-4의 AlphaFold2를 사용하여 계산된 구조를 나타내었다. 도면에서 각 중쇄 및 경쇄 가변 영역 내 밝은 3개 구간은 CDR을 나타낸다. 실시예 모두 단백질 구조가 링커 내부에 융합되어 중쇄 가변 영역 및 경쇄 가변 영역의 구조를 해치지 않고 정상적으로 폴딩되는 것을 확인할 수 있었다. 도 1은 ABD가 없는 본래 4D5scFv의 구조이며, ColabFold 웹 서버를 사용하여 AlphaFold2에 의해 구현되었다. 이와 비교하여 실시예 1 내지 5가 4D5Ab의 원래 구조를 상당 부분 유지하여 항 HER2 결합 친화력이 유지될 것임을 확인할 수 있었다.
추가적으로 4D5-S-ABD 및 4D5-L-ABD에 대해서는 pTM 점수와 pLDDT로 분석했다. 이러한 익숙하지 않은 형태의 4D5-ABD 변형의 구조적 무결성을 확인하기 위해 AlphaFold2를 사용하여 구조를 계산적으로 예측하고 pTM 점수 및 pLDDT로 분석했다(도 15, 표 3). 그 후 4D5-ABD 변이체의 모델 구조를 4D5scFv 및 ABD의 모델 구조와 정렬하였다(도 16, 표 4). 모든 정렬에 대한 TM 점수는 0.94점 이상으로 매우 높았다. 즉, 구성된 4D5-ABD 변이체에서 4D5scFv 및 ABD의 구조가 많이 유지되어 기능도 유지될 것임을 의미한다. 또한 예측된 모델 구조는 삽입된 ABD의 위치가 예상대로 4D5scFv의 CDR에서 떨어져 있음을 보여주었다. 이 사실은 4D5-ABD 및 HER2 복합체(도 17)의 시각화에서도 볼 수 있으며, 여기서 ABD는 HER2 결합 부위에서 명확하게 분리된다. 따라서, 4D5scFv의 항원 결합 친화도는 ABD의 존재 또는 ABD에 대한 알부민 결합에 의해 크게 방해받지 않을 것이라고 예측할 수 있다. 결론적으로 각 구성 요소의 높은 구조적 일관성과 위치 독립성은 4D5scFv와 ABD의 기능을 상당 부분 유지하게 된다.

실험예
1. 단백질의 발현 및 정제
1.1. 플라스미드
상기 설계로 구축된 변이 단백질 중 실시예 1 내지 5에 대하여 박테리아 숙주에서 세포질 발현을 통해 단백질로 생산하였다. 구체적으로 상기 아미노산 서열을 암호화하는 유전자는 ExpOptimizer(NovoPro Bioscience, 중국 상하이)를 사용하여 최적화 하였으며, 마크로젠(서울, 대한민국)에서 합성 및 서브클론하여 pBAD 벡터 내 NcoI/KpnI 제한 부위에 삽입하여 pBAD_4D5scFv 플라스미드를 생성했다.
4D5scFv 및 실시예 1 내지 5의 플라스미드를 대장균 TOP10 숙주 세포로 형질 전환했다. 형질 전환된 세포는 암피실린 100μg/mL을 함유한 표준 2xYT 배지에서 37°C에서 16시간 동안 배양하면서 200rpm으로 흔들었다. 세포를 동일한 신선한 배지에 접종하고 600nm(OD600)에서 광학 밀도가 0.5 ~ 0.6이 될 때까지 배양했다. 단백질 발현은 최종 농도 0.2%(w/v)의 L-(+)-아라비노스에 의해 유도되었다. 온도를 23°C로 낮춘 후 배양액을 24시간 동안 배양하고, 성장한 세포를 8000rpm, 4°C에서 원심분리하여 수확한 후 세포 펠릿을 -80°C에서 보관했다. 각 변종에 대한 유도 전(BI) 및 유도 후(AI) 샘플을 수집하고 13,000rpm에서 1분간 원심분리한 다음, 2M 요소가 포함된 PBS(pH 7.4)에 다시 현탁하여 SDS-PAGE 분석을 수행했다.
His 태그가 부착된 상기 실시예들을 Ni-NTA를 사용한 금속 친화성 크로마토그래피로 정제했다. 세포 펠릿을 용해 완충액(10mM 이미다졸, 50mM NaH2PO4, 300mM NaCl, pH 8.0)에 1mg/mL 리소자임 및 5mg/mL DNase로 용해하고 최소 5분 동안 얼음 위에서 배양했다. 용액을 1초 펄스, 2초 휴식, 28% 앰프(500W, 20kHz)로 총 15분 동안 초음파 처리하고 5분 휴식 후 이 과정을 반복했다. 이후 혼합물을 4°C에서 20분간 10,000×g으로 원심분리하고 상층액을 4°C에서 30분간 Ni-NTA 아가로스 수지와 함께 배양했다. 배양된 수지를 필터가 있는 폴리프로필렌 컬럼에 로드한 후 세척 버퍼(20mM 이미다졸, 50mM NaH2PO4, 300mM NaCl, pH 8.0)로 세척하고 용출 버퍼(250mM 이미다졸, 50mM NaH2PO4, 300mM NaCl, pH 8.0)로 용출했다. 제조업체의 프로토콜에 따라 PD-10 탈염 컬럼을 사용하여 버퍼를 PBS(pH 7.4)로 교환했다. 정제된 단백질을 4°C에서 보관하고 SDS-PAGE로 분석했다.
1.2. SDS-PAGE 분석
재부유된 BI 및 AI 샘플과 정제된 단백질을 포함하여 준비된 단백질을 SDS-PAGE(Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis)를 사용하여 분석했다. 모든 샘플은 환원을 위해 100mM 디티오트레이톨(DTT)이 포함된 2× 샘플 로딩 염료(0.2% 브로모페놀 블루, 4% SDS, 20% 글리세롤, 100mM Tris-HCl, pH 6.8)와 혼합되었다. 환원되지 않는 샘플을 준비하기 위해 DTT는 제외되었다. 샘플을 100°C에서 10분간 끓인 후 12% SDS-PAGE 젤에 넣었다. 전기영동 후, 겔을 쿠마시 브릴리언트 블루(0.25% 쿠마시 블루, 50% 에탄올, 10% 아세트산)로 염색하고 데스티네이트(50% 에탄올, 10% 아세트산)했다. 겔은 ChemiDoc XRS+ 시스템(Bio-Rad, 미국 캘리포니아주 헤라클레스)을 사용하여 이미지화 및 시각화했다.
도 18a에 발현 중에 수집한 세포 용해물의 SDS-PAGE 분석 결과를 나타내었다. 각각 27, 32, 33kDa의 분자량을 가진 AI 샘플이 로드된 레인에서 밴드가 검출되었으며, 이는 4D5scFv, 4D5-S-ABD 및 4D5-L-ABD의 분자량에 해당한다. 정제는 아가로스 수지의 Ni-NTA와 각 단백질의 C-말단에 있는 His-tag 친화성 태그 간의 상호작용을 이용하여 수행했다. 정제된 단백질의 SDS-PAGE 분석 결과, 발현 겔과 동일한 위치의 밴드가 검출되었다(도 18b).
또한 실시예 1 및 2를 통해 확인한 결과 변이 단백질 환원 및 비환원 조건 모두에서 단량체 단백질 밴드만 나타났는데(도 19), 이는 실질적인 분자 간 이황화 결합이 형성되지 않았음을 의미한다.
1.3. MALDI-TOF 분석
MALDI-TOF(Matrix-Assisted Laser Desorption/Ionization Time-of-Flight) 분석을 수행했다. 정제된 샘플을 PBS(pH 7.4)에서 Zeba Spin Desalting Column (7K MWCO)을 사용하여 탈염하고 0.1% TFA 용액으로 준비했다. 샘플을 TA30(30% 아세토니트릴, 0.1% TFA) 용액에 포화시킨 SA 매트릭스와 1:1(v/v)의 비율로 혼합했다. 에탄올 용액에 포화시킨 SA 매트릭스를 강철 타겟 플레이트에 사전 증착한 다음, 샘플 혼합 용액을 층 위에 증착하여 Autoflex speed(미국 매사추세츠주 빌레리카, 브루커)를 사용하여 분자량 분석을 수행했다. 보정 표준품으로 Protein Standard II를 사용했다. 이 세 가지 단백질은 MALDI-TOF 분석을 통해 추가로 확인되었다(도 20).
도 20에서, 온전한 4D5scFv, 4D5-S-ABD, 4D5-L-ABD의 질량 스펙트럼에서 26,645, 32,013, 32,816 m/z에서 피크가 검출되었다. 1% 미만의 편차로, 이는 각각 26,801, 32,155, 32,759 m/z인 4D5scFv 변이체의 예상 값과 잘 일치했다. 정제된 4D5scFv, 4D5-L-ABD 및 4D5-S-ABD의 생산 수율은 각각 22.0 ± 0.5, 18.8 ± 1.3 및 21.5 ± 0.8 mg/L였다. 전반적으로 이러한 결과는 4D5scFv 변종 샘플이 성공적으로 발현 및 정제되었음을 보여준다.
1.4. SEC 분석
추가적으로 실시예 1 및 2에 대해서 정제된 샘플은 고속 단백질 액체 크로마토그래피 SEC(Size Exclusion Chromatography)로 특성화했다. PBS(pH 7.4)에서 준비된 등몰량(5 μM)의 샘플을 Superdex 75 Increase 10/300 GL 컬럼(Cytiva, 스웨덴 웁살라)에 주입하고 0.2mL/min의 유속으로 용출시켰다. 표준 단백질로는 Gel Filtration Calibration Kit HMW(Cytiva, 스웨덴 웁살라)의 오발부민, 코날부민 및 알돌라제를 사용했다. 분석은 NGC Quest 10 크로마토그래피 시스템(Bio-Rad, 미국 캘리포니아주 헤라클레스)을 사용하여 수행했다.
도 21의 SEC 분석 결과를 보면, 4D5scFv는 단량체와 이량체의 혼합물에서 안정적이며, 비환원 SDS-PAGE에 따라 이황화 결합이 형성되지 않는 것으로 나타났다. 반대로 4D5-ABD 변이체는 이합체를 형성하지 않고 단량체로만 존재했는데, 이는 ABD의 융합으로 인해 생성된 입체 장애 때문일 수 있다.
2. 결합 친화성 분석
(1) HER2에 대한 결합 친화성 분석
HSA의 유무에 따라 항 HER2 ELISA(Enzyme-Linked Immunosorbent Assay)를 통해 정제된 4D5scFv 변이체의 항 HER2 표적 효율을 분석했다. 구체적인 방법으로, 코팅 완충액(PBS, pH 7.4)에 재조합 HER2 항원(500 pg/μL, 100 μL/웰)을 4°C에서 하룻밤 배양하여 면역 플레이트에 코팅했다. 플레이트는 200μL/웰에서 PBST(0.05% Tween 20)로 흔들어 세 번 세척했다. 이후 차단 완충액(5% 탈지유를 PBST에 넣어 200 μL/웰에서 실온에서 배양하여 플레이트를 차단한 후 4회 세척했다. 정제된 단백질을 실온에서 1:2 몰 비율로 HSA를 첨가하거나 첨가하지 않은 PBS(pH 7.4)에서 배양했다. 배양된 단백질을 차단 완충액에서 준비하여 50 nM 스톡을 만들고, 이를 연속적으로 3배 희석했다. 준비된 샘플(100 μL/웰, n = 2)을 플레이트에 로드하고 실온에서 배양한 후 결합되지 않은 단백질을 4회 세척했다. 플레이트는 차단 완충액에 1:1500으로 희석한 토끼 항-His-태그 항체 100μL/웰로 배양되었다. 배양 후 웰을 4회 세척했다. 마지막으로, 차단 완충액에 1:3000으로 희석한 100 μL/웰의 HRP-결합 항-토끼 IgG 항체와 함께 배양했다. 웰을 4회 세척했다. 100 μL/웰의 TMB를 첨가하여 검출을 수행한 후, 반응은 2 M HCl로 켄칭했다. 결합된 단백질은 450nm에서 흡광도를 측정하여 정량화했다.
도 22 및 하기 표 4에서 실험 결과를 나타내었다. 모든 단백질은 유사한 농도 의존적 곡선을 보였으며, 이는 ABD 삽입이 4D5scFv의 구조에 상당한 왜곡을 일으키지 않았으며, 결과적으로 4D5-ABD 변이체의 항 HER2 결합 친화력을 유지함을 시사한다.
구체적으로 Albumin이 ABD에 붙었을 때와 안 붙었을 때의 결합력을 비교하기 위해 항체와 HSA를 미리 incubation한 실험군(HSA+)과 하지 않은 실험군(HSA -)을 비교하였다. ABD가 없는 4D5scFv의 HER2 결합은 크게 변하지 않아, 4D5scFv의 항 HER2 결합 친화력이 HSA의 존재에 의해 방해받거나 차단되지 않음을 알 수 있다. 4D5-ABD 변이체에 HSA가 결합하면 항 HER2 결합 친화도가 약간 감소할 수 있는데, 이는 부피가 큰 HSA가 4D5scFv와 결합할 때 HER2에 대한 입체 장애가 약하기 때문인 것으로 보인다. HSA가 있을 때와 없을 때 모두 ABD를 링커 중간에 위치 시킨 4D5-L-ABD와 4D5-S-ABD가 상대적으로 강한 결합력을 보였으며, 그 중에서도 링커 길이가 가장 긴 4D5-L-ABD가 가장 강한 결합력을 보였다. AlphaFold2로 예측한 구조를 토대로, ABD가 CDR 근처에 위치하여 낮은 binding affinity를 가질 것으로 예상되었던 LinkC와 TermN이, 예상 외로 ABD가 CDR과 떨어져 있는 LinkN 변이체보다 높은 binding affinity를 가지는 것으로 나타났다. 이는 LinkC와 TermC의 경우 ABD C-term에 최소한의 linker residue가 삽입된 데 반해, LinkN은 N-term에 추가적인 linker residue가 없기 때문으로 추측된다. 이는 각 변이체를 만들기 위한 cloning 과정에서 발생한 차이이다. 이는 ABD와 항체 사이에 linker가 적거나 없을 경우 입체 장애가 발생하며, 결합력을 위해서는 항체와 ABD 사이에 최소한의 linker가 필요하다는 것을 의미한다. 따라서 링커 영역 중간에 ABD를 삽입하여 항체와 ABD가 가장 입체 장애의 영향을 덜 받은 4D5-S-ABD와 4D5-L-ABD가 가장 높은 결합력을 가지는 것으로 보인다.

(2) HSA 결합 친화력 분석
ELISA를 통해 정제된 4D5-ABD 변이체의 항 HSA 효율도 조사했다. 구체적인 방법으로, 코팅 완충액에 있는 HSA(500 pg/μL)를 플레이트에 코팅했다. 정제된 단백질을 차단 완충액에서 20 nM으로 제조하고 연속적으로 3배 희석했다. 이 외 다른 조건과 절차는 위에서 설명한 대로 수행되었다.
HSA가 존재할 때 4D5scFv 변이체의 결합 친화력도 조사하기 위해, 모든 변이체를 1:2 몰 비율로 HSA로 사전 배양한 후 ELISA를 실시했다. 도 23 및 하기 표 5에서 항-HSA 결합 친화력 결과를 나타냈다.
4D5-L-ABD와 4D5-S-ABD가 가장 강한 결합력을 가지는 것으로 나타났다. ABD의 결합력은 그 감소세가 적어 항체의 경우보다 링커 길이에 의한 영향을 덜 받는 것으로 보이나, 경향은 항체 결합력과 동일하였다.

2.9. 약동학 분석
약동학 분석을 위해 마우스를 사용해 4D5scFv 변이체의 혈청 반감기를 조사했다. 실험은 광주과학기술원(GIST)의 동물실험윤리위원회의 가이드라인에 따라 수행되었다. 구체적으로, 200 μL PBS(pH 7.4)에서 정제된 4D5scFv 및 4D5-ABD 변이체(100 μg/mL)를 9주령 암컷 BALB/c 마우스(n = 4)의 꼬리 정맥에 주사했다. 후안와 채혈을 통해 주사 후 3분, 1, 2, 4, 8, 24, 48시간에 혈액 샘플(<100 μL)을 채취했다. 혈청은 4°C에서 10분간 10,000 × g으로 원심분리하여 혈액에서 분리한 후 추후 사용할 때까지 -20°C에서 보관했다. 각 시점의 혈청 단백질 농도는 위에서 설명한 대로 ELISA로 측정했다. 농도는 표준 보정 곡선을 보간하여 계산되었다.
4D5scFv 및 4D5-ABD 변이체의 혈청 반감기를 BALB/c 마우스에 정맥 주사한 후 측정했다(n = 4). 혈청에 남아있는 단백질을 정량하기 위해 ELISA를 수행했다(도 23). 4D5scFv는 혈청 반감기가 18분으로 혈액에서 빠르게 제거되었다. 반면, 4D5-S-ABD 및 4D5-L-ABD는 34시간의 현저하게 연장된 반감기를 보였으며, 이는 4D5scFv에 비해 114배 증가한 수치이다. 또한 두 ABD 융합 변이체 모두 AUC 값이 약 70배 증가했으며, 4D5scFv, 4D5-S-ABD 및 4D5-L-ABD의 경우 각각 42, 2887 및 3008로 계산되었다. 4D5-ABD 변종의 경우 검출 한계에 도달하지 않았다는 점을 고려하면 실제 AUC 증가는 훨씬 더 높을 것으로 예상된다.

4D5-ABD 변이체에서 반감기가 크게 증가한 것은 알부민과 결합한 4D5-ABD 분자가 FcRn 매개 재활용을 거쳤기 때문이라고 추론할 수 있다. 이 결과는 또한 내부에 삽입된 ABD가 혈청 알부민에 대한 결합 친화력을 유지한다는 것을 보여주며, 이는 구조 계산 및 ELISA 결과와 일치하는 것이다. 그리고 4D5-ABD 변이체 간에는 차이가 관찰되지 않았으며, 이 또한 ELISA 결과와 일치한다. 또한 이는 4D5-S-ABD 변이체의 15개 아미노산 잔기의 링커 길이가 알부민에 결합하기 위해 ABD 모이어티를 자유롭게 이동시키기에 충분하다는 것을 시사한다.

- 청구항 1에 있어서, 상기 알부민 결합 모이어티는 상기 링커 내에 융합되고, 상기 단일쇄 가변 단편의 C-말단에 생리활성물질이 융합되어 있는, 단일쇄 가변 단편 변이 단백질.
