Abstract
본 발명은 IgE-의존성 히스타민 방출인자와 결합하는 항체 및 이의 용도에 관한 것으로, 본 발명에 따른 항-HRF 항체는 HRF에 특이적으로 높은 결합력을 가지고 결합함으로써, HRF의 활성을 억제하는 것을 확인한 바, 알레르기 질환, 염증성 질환, 자가면역질환 또는 암, 고혈압, 말라리아, 골다공증과 같은 HRF 관련 질환의 예방 및 치료제 개발에 유용하게 이용할 수 있다.
Description
본 발명은 IgE-의존성 히스타민 방출인자와 결합하는 항체 및 이의 용도에 관한 것이다.
히스타민방출인자 (histamine-releasing factor, HRF, Translationally Controlled Tumor Protein, TCTP, 이하 HRF로 칭함)는 호염구 (basophil)를 활성화시키고 히스타민의 방출을 유도하여 후기 알러지 반응을 유발한다고 알려져 있으며, 호염구 외에도 후기 알러지 반응에 관여하는 염증세포를 활성화시켜 다양한 사이토카인을 유리시키기 때문에 후기 알러지 반응에 중요한 인자로 인식되고 있다 (MacDonald et al., Science, 269, 688-690, 1995).
HRF는 특정 IgE가 존재할 경우 호염구에서 히스타민을 분비하게 하는 것으로 알려졌으나, 이후 IgE와 이의 수용체인 FcεR 존재 유무에 상관없이 HRF가 염증세포에서 히스타민, IL-4 및 IL-13 등의 분비를 조절하는 것이 관찰됨으로써 Bheekha-Escura 등은 HRF가 IgE가 아닌 특이적 세포막 수용체에 결합하여 작용할 것이라는 가능성이 제시되었다.
본 발명자들은 선행연구를 통해 이량체를 형성할 수 있는 HRF가 세포에서 히스타민 및 IL-8 분비함을 규명하여, HRF의 이량체형이 알레르기를 유발하는 물질임을 최초로 증명하였고 (대한민국 등록특허 제100780255호, 유럽 등록 특허 제1683866호, 일본 등록특허 제4564926호, 미국 등록특허 제7772368호), HRF가 친수성 단백질임에도 불구하고, 세포막을 건너갈 수 있다는 사실과, 세포내 존재 하는 HRF는 (Na,K)ATPase 알파서브유닛 중 대형 세포질 루프(large cytoplasmic loop) CD3 (cytoplasmic domain 3)에 결합한다는 사실을 최초로 규명하였고, 세포내 HRF을 과발현하는 형질전환 마우스에서 고혈압이 유발됨을 입증하였다 (대한민국 등록특허 제10-0457350호, 유럽 등록 특허 제1167526호, 일본 등록 특허 제4295449호, 미국 등록 특허 제6710165호). 또한, HRF에서 막투과 단백질 도메인 기능을 갖는 펩타이드를 발견하였다 (대한민국 등록특허 제10-0859972호). 또한, 이량체 HRF를 타겟으로 하여 이를 제어함으로써 항알레르기 효능을 갖는 펩타이드 약물(dTBP2)을 자체적으로 개발하였으며, 알레르기 질환과 류마티스성 관절염에서 억제 효능을 입증하였다 (대한민국 등록특허 제10-1830838호). 또한, HRF 구조 부위로서 세포막에 존재하는 HRF의 수용체에 결합하는 유연성 루프(flexible loop) 도메인 (대한민국 등록특허 제101804291호)또는 헬릭스 2(helix 2) 도메인 (대한민국 등록특허 제101843051)및 C-말단 (대한민국 등록특허 제101804285호)을 규명하였으며, 이들에 대한 결합 물질이 IL-8의 분비능을 억제시키는 것을 확인하였다. 또한, 항알레르기제로서 디히드로코스터스 락톤 (Phytomedicine 2018), 카다모닌(cardamonin)이 이량체 HRF를 억제하여 항염증 효과를 발휘함을 규명하였고 (FRONTIERS IN PHARMACOLOGY, 2021, v.12, 765521), 단량체와 이량체 HRF가 류마티스 관절염의 질환 활성에 밀접한 관련이 있다는 것을 확인하였고, HRF가 류마티스 관절염의 진단 및 치료를 위한 새로운 바이오마커이자 치료 대상임이 확인하였다(EXPERIMENTAL AND MOLECULAR MEDICINE, 2021, 67-80). 이에 더하여, 이량체 HRF의 저해제인 dTBP2은 비만 세포의 탈과립을 직접적으로 억제하여 전신 아나필락시스 반응을 약화시킴을 규명하였고 (FRONTIERS IN PHARMACOLOGY, 2021, v.12, 764321), HRF가 제2형 당뇨병을 포함한 비만 및 비만 관련 대사 장애의 치료 표적이 될 수 있음을 규명하였으며 (International Journal of Obesity, 2021, v.45 no.7, 1576-1587), 이량체 HRF가 기도 상피세포의 활성화를 통해 기도염을 악화시킴을 규명하였다 (Biomedicine and Pharmacotherapy, 2021, v.144, 112316).
이에 본 발명자들은 상기한 바와 같이 다양한 질병과 연관이 있는 HRF에 대하여 특이적으로 결합하는 항체를 개발하고자 노력하였으며, 특히 단량체 및/또는 이량체 HRF에 결합하는 항체를 개발하였다. 본 발명에 따른 항-HRF 항체는 HRF에 특이적으로 높은 결합력을 가지고 결합함으로써, HRF의 사이토카인 유사 활성을 억제하는 것을 확인하였으며, 알레르기 질환, 만성 염증성 질환, 자가면역질환, 또는 암, 고혈압, 말라리아, 골다공증과 같은 HRF 관련 질환의 예방 및 치료제 개발에 유용하게 이용할 수 있음을 발견하고 본 발명을 완성하였다.
본 발명의 일 목적은 단량체 및/또는 이량체 HRF에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 제공하는 것이다.
본 발명의 다른 일 목적은 상기 항체 또는 이의 항원 결합 단편을 암호화하는 폴리뉴클레오티드를 제공하는 것이다.
본 발명의 다른 일 목적은 상기 폴리뉴클레오티드를 포함하는 발현 벡터를 제공하는 것이다.
본 발명의 다른 일 목적은 상기 발현 벡터가 도입된 형질전환체를 제공하는 것이다.

이하, 본 발명을 상세하게 설명한다.
본 발명은 단량체 및/또는 이량체 HRF에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편을 제공한다.
상기 HRF는 히스타민 유리활성이 있는 IgE 의존적 히스타민 방출 인자로, 번역 제어 종양 단백질 (translationally controlled tumor protein, TCTP)로 알려진, 모든 세포질에 존재하는 172개의 아미노산으로 구성되어 있는 공지의 단백질이다. 상기 HRF는 IgE-감작된(sensitized) 호염구(basophil)를 자극하여 히스타민 및 인터루킨-4 (IL-4)의 유리를 촉진시킴으로써 알레르기성 비염, 천식 및 아토피성 피부염과 같은 후기반응 알레르기 질환을 일으킨다고 보고되어 있다.
상기 HRF는 이량체형(dimer)이 활성형이고 상기 HRF의 구조 중에서 유연성 루프 (flexible loop: FL) 도메인 또는 헬릭스 2 (helix 2: H2) 도메인이 HRF에 대한 수용체와 결합하는 부위이며, 상기 HRF에 특이적으로 결합하는 항체는 전장 및 N-말단이 결실된 것으로 구성되는 군으로부터 선택되는 어느 하나에 결합하는 것이 바람직하다. 본 발명의 일 실시태양에서 이량체 HRF는 N-말단의 아미노산 10개가 결실된 것이다. 본 발명의 일 실시태양에서 단량체 또는 이량체 HRF는 각각 서열번호 82 또는 서열번호 83의 아미노산 서열로 구성되는 것일 수 있다.
구체적으로 본 발명의 단량체 및/또는 이량체 HRF에 특이적으로 결합하는 항체 또는 이의 항원 결합 단편은 하기 (1) 내지 (8)으로 이루어진 군에서 선택될 수 있다.

본 발명에서 항체(Antibody)는 단량체 및/또는 이량체 HRF 단백질과 같은 항원에 특이적으로 결합하고 인식하는 폴리펩티드를 의미한다. 본 발명은 단량체 및/또는 이량체 HRF 단백질에 결합하는 완전한 항체 형태뿐만 아니라, 상기 항체 분자의 항원 결합 단편도 포함한다.
본 발명에서 항체는 단일클론항체, 다클론항체 및 다중특이성 항체 (예를 들어, 이중특이성 항체)를 모두 포함할 수 있다. 항체는 2개의 중쇄 (heavy chain)와 2개의 경쇄 (light chain)로 구성되며, 대상 항원의 종류에 따라 아미노산 서열이 달라지는 가변 영역 (variable region)과 서열이 달라지지 않는 불변 영역 (constant region)을 갖는다.
상기 항체 또는 이의 항원 결합 단편은 전장항체 (Full-lenght antibody) 또는 항체 단편일 수 있다. 바람직하게는, 본 발명에서 상기 항체는 단일클론항체일 수 있다.
본 발명의 상기 항체는 인간 항체, 인간화 항체, 또는 키메라 항체일 수 있다.
본원에서 사용된 용어 "인간 항체(human antibody)" 또는 "인간화 항체(humanized antibody)"는 인간 또는 인 간 세포에 의해 생성된 항체, 또는 인간 항체 레퍼토리(repertoires) 또는 다른 인간 항체 코딩 서열을 이용하 는 비인간 근원으로부터 유래된 항체의 아미노산 서열에 상응하는 아미노산 서열을 보유한다.
본원에서 사용된 용어 "키메라(chimeric) 항체"는 중쇄 및/또는 경쇄의 일부가 특정 근원(source) 또는 종 (species)으로부터 유래되고, 중쇄 및/또는 경쇄의 나머지가 상이한 근원 또는 종에서 유래한 항체를 의미한다.
상기 항원 결합 단편은 scFv (single-chain variable fragment), Fab, Fab', F(ab')2, Fd 및 Fv 일 수 있다. 일 실시태양에서 상기 항원 결합 단편은 scFv-Fc (Fragment, crystallizable) 형태의 항원 결합 단편이다.
상기 scFv는 항체의 경쇄 및 중쇄의 가변도메인을 15개 내외의 아미노산이 연결된 펩타이드 사슬로 이루어진 링커 (linker)로 연결한 단백질을 지칭한다. 경 쇄가변도메인 (VL)-링커-중쇄가변도메인 (VH), 또는 중쇄가변도메인 (VH)-링커-경쇄가변도메인 (VL)의 순서 모두 가능하며, 원 항체와 동일 혹은 유사한 항원 특이성을 가진다.
상기 단량체 및/또는 이량체 HRF에 대한 항체의 항원 결합 단편, 예컨대, 항 HRF scFv에 있어서, 상기 중쇄 가변 영역과 경쇄 가변 영역은 링커, 예컨대, 펩타이드 링커를 통하거나 통하지 않고 연결될 수 있다. 상기 펩타이드 링커는 1 내지 100개 또는 2 내지 50개의 임의의 아미노산으로 이루어진 폴리펩타이드일 수 있으며, 그 포함된 아미노산 종류는 제한이 없다. 상기 펩타이드 링커는, 예컨대, Gly, Asn 및/또는 Ser 잔기를 포함할 수 있으며, Thr 및/또는 Ala과 같은 중성 아미노산들도 포함될 수 있다. 펩타이드 링커에 적합한 아미노산 서열은 당 업계에 공지되어 있다. 한편, 상기 링커는 상기 이중 특이 항체의 기능에 영향을 미치지 않는 한도 내에서, 그 길이를 다양하게 결정할 수 있다. 예컨대, 상기 펩타이드 링커는 Gly, Asn, Ser, Thr 및 Ala로 이루어진 군에서 선택된 1종 이상을 총 1 내지 100개, 2 내지 50개, 또는 5 내지 25개를 포함하여 이루어진 것일 수 있다. 일 실시태양에서, 상기 펩타이드 링커는 18-mer 링커인, GGSSRSSSSGGGGSGGGG (서열번호 81)이다.
본 발명의 상기 항체 또는 이의 항원 결합 단편은, HRF 단백질을 특이적으로 인식할 수 있는 범위 내에서 첨부한 서열목록에 기재된 아미노산 서열의 변이체를 포함할 수 있다. 예를 들면, 항체의 결합 친화도 및/또는 기타 생물학적 특성을 개선시키기 위하여 항체의 아미노산 서열에 변화를 줄 수 있다. 이러한 변형은, 예를 들어 항체의 아미노산 서열 잔기의 결실, 삽입 및/또는 치환을 포함한다.

또한, 발명은 상기 항체 또는 이의 항원 결합 단편을 암호화하는 폴리뉴클레오티드를 제공한다.
본 발명의 상기 항체 또는 이의 항원 결합 단편을 암호화하는 폴리뉴클레오티드는 상기한 폴리뉴클레오티드 염기 서열에 대하여 실질적인 동일성을 나타내는 폴리뉴클레오티드 염기 서열도 포함하는 것으로 해석된다. 상기의 실질적인 동일성은, 상기한 본 발명의 염기 서열과 임의의 다른 서열을 최대한 대응되도록 얼라인하고, 당업계에서 통상적으로 이용되는 알고리즘을 이용하여 얼라인된 서열을 분석한 경우에, 최소 80%의 상동성, 보다 바람직하게는 최소 90%의 상동성, 가장 바람직하게는 최소 95%의 상동성을 나타내는 염기 서열을 의미한다. 서열비교를 위한 얼라인먼트 방법은 당업계에 공지되어 있다. 얼라인먼트에 대한 다양한 방법 및 알고리즘은 문헌 [Smith and Waterman, Adv. 상기 HRF 표면 항원에 특이적인 항체는 바람직하게는 파아지-디스플레이 기술(Smith, Science, 228, 1315-1317,1985; 및 Hoogenboom & Chames, Immunol Today, 21, 371-378, 2000)을 응용하여 선별할 수 있다. 다음과 같이 파아지-디스플레이 기술을 응용하여, 필라멘트(filamentous) 파아지(M13, Fd 및 F1) 표피 단백질을 발현하는 유전자(gene III)에 목적하는 항체를 발현하는 유전자를 융합시켜, 상기 융합된 항체가 박테리오 파아지 입자의 표면에 노출된 항체-파아지 형태의 바이러스 입자를 생성시키고, 상기 노출된 항체의 높은 특이도 및 친화도, 파아지의 높은 감염성을 이용하여 바이오패닝 (biopanning) 기법을 응용하여 파아지 라이브러리로부터 원하는 항체를 선별할 수 있다 (Burton & Barbas, Adv. 본 발명에서 용어 "도입"은 상기 항체 또는 이의 항원 결합 단편를 암호화하는 폴리뉴클레오티드를 포함하는 벡터를 숙주세포에 전달하는 방법을 의미한다. 이와 같은 도입은 칼슘 포스페이트-DNA 공침전법, DEAE-덱스트란-매개 트랜스펙션법, 폴리브렌-매개 형질감염법, 전기충격법, 세주사법, 리포좀 융합법, 리포펙타민 및 원형질체 융합법 등의 당 분야에 공지된 여러 방법에 의해 수행될 수 있다.
또한, 본 발명은 상기 항체 또는 이의 항원 결합 단편을 포함하는 HRF 관련 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
상기 HRF 관련 질환은 염증성 질환, 자가면역질환, 암, 고혈압, 말라리아 및 골다공증으로 구성된 군에서 선택될 수 있다.
상기 염증성 질환 또는 자가면역질환은 천식, 기관지염, 만성폐쇄성폐질환, 기관지확장증, 비염, 아토피 피부염, 두드러기, 건초열, 결막염, 아나필락시스의 알레르기 질환, 기관지염, 폐렴, 관절염. 신장염, 건선, 피부염, 크론병, 장염, 치은염, 동맥경화증, 관상동맥염, 간염, 베체트병, 방광암, 전립선염, 신우신염, 사구체신염, 골수염, 갑상선염, 포도막염, 복강염, 수막염, 폐섬유화증 및 류마티스 관절염으로 구성되는 군에서 선택될 수 있다.

IL-8은 만성기관지염 등 만성 염증성 기관지 질환(Richman-Eisenstat et al., Am J Physiol, 264, L413- 418, 1993), 폐렴 등 염증성 폐질환(Erger and Casale, Eur Respir J, 11, 299-305, 1998; Pease & Sabroe, Am J Respir Med, 1, 19-25, 2002), 관절염 또는 신장염(Harada et al., J Leukoc Biol, 56, 559-564, 1994), 건선(Schulz et al., J Immunol, 151, 4399-4406, 1993; Bruch-Gerharz et al., J Exp Med, 184, 2007-2012, 1996), 피부염(Sticherling et al., Arch Dermatol Res, 284, 82-85, 1992), 크론병(Izutani et al., Inflamm Bowel Dis, 1, 37-47, 1995), 염증성 장질환(Mitsuyama et al., Clin Exp Immunol, 96, 432-436, 1994), 치은 염(Haake & Huang, Clinical Periodontology, 9th Edition. Philadelphia: W.B.Saunders Co. 2002. page 162), 동맥경화증, 관상동맥질환 등 심혈관계 질환(Apostolakis et al., Cardiovasc Res, 84, 353-360, 2009; Boekholdt et al., Arterioscler Thromb Vasc Biol, 24, 1503-1508, 2004), 만성간질환(Zimmermann et al., PLoS ONE, 6, e21381, 2011), 베체트병(Behcet's disease, Katsantonis et al., Dermatology, 201, 37-39, 2000), 방광암, 전립선염, 신우신염 또는 골수염(Shahzad et al., Int arch med, 3, 11, 2010), 갑상선질환 (Kobawala et al., J Thyroid Res, 8, 270149, 2011), 포도막염(uveitis, Klok et al., Br J Ophthalmol, 82, 871-874, 1998), 사구체 신염, 복강염, 수막염 및 폐섬유화증(Harada et al., Mol Med Today, 2, 482-489, 1996)의 다양한 염증성 질환에 관여한다고 알려져 있다. 따라서, 폐질환, 류머티스성 관절염, 염증성 장질환, 건선, 수장족저농포증(palmoplantar pustulosis) 등 만성 염증성 피부질환, 안구 염증 등 염증성 질환에서 IL-8을 저해하는 것이 치료 전략으로 제시되었다(Mukaida, Am J Physiol Lung Cell Mol Physiol, 284, L566-L577, 2003; Skov et al., J Imm unol, 181, 669-679, 2008; Harada et al., J Leukoc Biol, 56, 559-564, 1994). IL-8의 차단항체(blocking antibody)나 IL-8 수용체를 코딩하는 유전자를 저해하는 치료법은 염증 치료 효과가 있으며(Harada et al., Mol Med Today, 2, 482-489, 1996), 일례로 만성 염증성 피부질환 환자에서 IL-8에 대한 항체를 투여하여 염증이 감소되었다(Skov et al., J Immunol, 181, 669-679, 2008). HRF에 의해 분비가 증가하는 GM-CSF도 다양한 염증성 질환과 관련성이 있어(Hamilton, Trends Immunol, 23, 403-408, 2002), GM-CSF 또한 류머티스성 관절염 등의 염증성 질환의 타겟으로 제시되고 있다(Cornish et al., Nat Rev Rheumatol, 5, 554-559, 2009). 따라서, HRF 활성 억제제가 IL-8 분비 억제에 관여하는 것을 확인함으로써, 본 발명의 HRF 활성 억제제가 상기 질환을 예방 및 치료할 수 있는 이론이 확실한 것으로 나타났다. 따라서 본 발명의 HRF 항체는 염증성 질환의 예방 및 치료용 약학적 조성물로 이용될 수 있다.
또한, HRF에 의해 증가되는 IL-8은 천식 또는 기관지염 (Chanez et al., Int Arch Allergy Immunol, 111, 83-88, 1996), 만성폐쇄성폐질환 (Nocker et al., Int Arch Allergy Immunol, 109, 183-191, 1996), 기관지 확장증 (bronchiectasis, Simpson et al., Thorax, 62, 211-218, 2007), 비염 (Benson et al., Pediatr Allergy Immunol, 10, 178-185, 1999; Kuna et al., J Allergy Clin Immun, 97, 104-112, 1996), 아토피성 피부염 (Kimata & Lindley, Arch Dis Child 70,119-122, 1994), 두드러기 (urticaria, Choi et al., J Clin Immunol, 28, 244-249, 2008), 건초열 (hay fever, Ciprandi et al., Otolaryngol Head Neck Surg, 133, 429-435, 2005), 결막염 (Miyoshi et al., Cornea, 20, 743-747, 2001) 아나필락시스 (anaphylaxis) 등의 여러 알레르기 질환에 관여한다고 알려져 있다. 따라서 본 발명의 HRF 항체는 알레르기 질환의 예방 및 치료용 약학적 조성물로 이용될 수 있다.
또한, HRF는 1980년대까지 종양에 관련된 단백질 (tumor-specific protein)로 알려져 왔으며, 그 합성은 종양의 증식 단계 (proliferative stage)와 관련이 있을 것으로 생각되어 왔다. 마우스 적백혈병 (erythroleukemia) 세포에서는 21kDa의 종양 단백질 p21으로 밝혀졌고 (Chitpatima et al,1988), 에를리히 복수 암(Ehrlich ascites tumor)에서 세포성장과 관련이 있는 단백질 p23이 HRF와 동일한 것으로 밝혀졌다 (Bohm et al, 1989). 따라서 본 발명의 HRF 항체는 암의 예방 및 치료용 약학적 조성물로 이용될 수 있다.
또한, HRF는 나트륨/칼륨 APT아제 펌프 작용을 저해하여 혈관 평활근 근육 및 심장 근육의 반응도와 수축도, 그리고 뇌의 신경전달물질 분비에 영향을 미쳐 고혈압을 유발한다고 알려져 있다 (대한민국 공개특허 10-2004-0111051). 따라서 본 발명의 HRF 항체는 고혈압의 예방 및 치료용 약학적 조성물로 이용될 수 있다.
또한, 1998년 항말라리아제인 아르테미시닌 (Artemisinin)이 말라리아 단백질인 HRF에 결합하여 작용하는 것이 밝혀진 바 있다 (Bhisutthibhan et al., J Biol Chem, 273, 16192-16198, 1998). IL-8은 말라리아 환자에서도 분비되며(Friedland et al., Trans R Soc Trop Med Hyg, 87, 54-55, 1993), 말라리아의 HRF가 IL-8 분비를 촉진시킨다고 보고되었다 (MacDonald et al., Proc Natl Acad Sci U S A, 98, 10829-32, 2001). 따라서 본 발명의 HRF 항체는 방 및 치료용 약학적 조성물로 이용될 수 있다.
또한, HRF가 뼈를 없애는 파골세포의 분화 시 유도되며, HRF가 과발현된 마우스에서 골량이 감소하고 파골세포가 증가함이 알려져있다 (SW Choi et al., FEBS letters, 588(21), 4026-31, 2004). 따라서 본 발명의 HRF 항체는 골다공증 예방 및 치료용 약학적 조성물로 이용될 수 있다.
건강한 성인 기증자의 단핵백혈구에 호흡기 바이러스인 인플루엔자 (influenza) 또는 호흡기세포융합바이러스 (respiratory syncytial virus)를 시험관 내에서 노출 시 HRF 생성을 유도하며, 생성된 HRF는 호염기구 탈과립 (basophil degranulation)을 유발하여 바이러스 유발성 기관지 경련을 일으킬 수 있다 (Chonmaitree et al., J Infect Dis, 164(3), 592-4, 1991). 따라서 본 발명의 HRF 항체는 바이러스로 인한 호흡기 질환 예방 및 치료용 약학적 조성물로 이용될 수 있다.
상기 약학적 조성물은 하나 이상의 약제학적으로 허용가능한 담체, 희석제를 더 포함할 수 있다. 일례로, 상기 약학적 조성물은 약학적으로 허용 가능한 담체를 포함할 수 있고, 인체 또는 수의용으로 제형화되어 다양한 경로로 투여될 수 있다. 투여 경로는 경구, 복강, 정맥, 근육, 피하, 피내 등의 경로로 투여될 수 있다. 바람직하게는, 주사제로 제형화하여 투여하는 것이다. 주사제는 생리식염액, 링겔액 등의 수성용제, 식물유, 고급 지방산 에스텔 (예, 올레인산에칠 등), 알코올류 (예, 에탄올, 벤질알코욜, 프로필렌 글리콘, 글리세린 등) 등의 비수성 용제 등을 이용하여 제조할 수 있고, 변질 방지를 위한 안정화제 (예, 아스코르빈산, 아황산수소나트륨, 피로아황산나트륨, BHA, 토코페롤, EDTA 등), 유화제, pH 조절을 위한 완충제, 미생물 발육을 저지하기 위한 보존제 (예, 질산페닐수은, 치메로살, 염화벤잘코늄, 페놀, 크레솔, 벤질알코올 등) 등의 약제학적 담체를 포함할 수 있다. 상기 약학적 조성물은 약제학적 유효량으로 투여할 수 있다. 이때, 용어 "약제학적 유효량"이란 예방 또는 치료 효과를 나타낼 수 있을 정도의 충분한 양과 부작용이나 심각한 또는 과도한 면역반응을 일으키지 않을 정도의 양을 의미하며, 정확한 투여 농도는 환자의 연령, 체중, 건강, 성별, 환자의 약물에 대한 민감도, 투여경로, 투여 방법 등 의학 분야에 잘 알려진 요소에 따라 당업자에 의해 용이하게 결정될 수 있으며, 1회 내 지 수회 투여가 가능하다. 일반적으로는 체중 1 kg 당 0.1 mg 내지 100 mg, 바람직하게는 0.5 mg 내지 10 mg을 2주 또는 4주 간격으로 투여할 수 있다. 그러나 투여 경로, 질환의 중증도, 성별, 체중, 연령 등에 따라서 증감될 수 있으므로 상기 투여량이 어떠한 방법으로도 본 발명의 범위를 한정하는 것은 아니다.
또한, 본 발명은 상기 항체 또는 이의 항원 결합 단편을 포함하는 HRF 관련 질환의 진단용 조성물을 제공한다.

또한, 본 발명은 상기 항체 또는 이의 항원 결합 단편을 이용하여 HRF 관련 질환이 의심되는 개체의 분리된 생물학적 시료의 HRF 단백질을 항원-항체 반응을 통하여 검출하는 단계를 포함하는 HRF 관련 질환을 위한 정보 제공 방법을 제공한다.
상기 생물학적 시료는 진단 대상 환자로부터 얻어진 세포, 조직, 체액 (예컨대, 혈액, 혈청, 림프액, 등) 등으로 이루어진 군에서 선택된 것일 수 있으며, 생체로부터 분리된 것일 수 있다. 상기 개체는 인간, 원숭이 등을 포함하는 영장류, 마우스, 래트 등을 포함하는 설치류 등을 포함하는 포유류에서 선택된 것일 수 있다.
상기 항원-항체 반응은 당업계에 공지된 다양한 방법을 통하여 수행할 수 있다. 예컨대, 통상적인 효소 반응, 형광, 발광 및/또는 방사선 검출을 통하여 하여 측정될 수 있으며, 구체적으로, 면역크로 마토그래피 (Immunochromatography), 면역조직화학염색 (Immunohistochemistry), 효소결합 면역흡착 분석 (enzyme linked immunosorbent assay: ELISA), 방사선 면역측정법 (radioimmunoassay: RIA), 효소 면역분석 (enzyme immunoassay: EIA), 형광면역분석 (Floresence immunoassay: FIA), 발광면역분석 (luminescence immunoassay: LIA), 웨스턴블라팅 (Western blotting), 마이크로어레이, 표면 플라즈몬 공명법 (surface plasmon resonance: SPR) 등으로 이루어진 군으로부터 선택된 방법에 의하여 측정될 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 구체적인 실시예 및 실험예에서, 단량체 및/또는 이량체인 HRF 단백질을 합성하여 분리 및 정제하였다 (도 1a 내지 도 1e 참조). 인간항체인 scFv 파지 라이브러리를 이용하여 상기 단량체 및/또는 이량체인 HRF에 특이적으로 결합하는 파지를 획득하였으며 (도 2a 참조), HRF 이량체에 대하여 높은 결합력을 가지는 38개의 클론 및 HRF 단량체에 대하여 높은 결합력을 가지는 33개의 클론을 선별하였다 (도 2b 및 도 2c 참조). 또한, 상기에서 선별된 클론의 scFv 항체의 아미노산 서열을 확인하였고 (도 2d 및 도 2e 참조), 상기 항체를 scFv-Fc 형태로 전환하여 제작하였다 (도 3a 내지 도 3b). 상기 제작된 항-HRF scFv-Fc 항체 중에서, 항원 결합력이 우수한 항체를 확인한 결과, 사람 HRF 이량체에 대한 결합력은 JEW-D195 ≥ JEW-M449 > JEW-D121 > JEW-D4 > JEW-M353 > JEW-M357 > JEW-D199 > JEW-M491 순으로 높았고, 마우스/랫트 HRF 이량체에 대한 결합력은 JEW-M449 ≥ JEW-D195 > JEW-D121 > JEW-D4 > JEW-M353 > JEW-M357 > JEW-D199 > JEW-M491 순으로 높음을 확인하였다 (도 4a 및 4b 참조). 또한, 인체 기관지 상피세포주인 BEAS-2B 세포에 HRF 이량체를 처리하여 BEAS-2B 세포의 활성화를 유도하고, 본 발명의 항-HRF 단일클론 항체를 처리한 후, 염증성 매개물질인 IL-8이 억제됨을 확인하였다 (도 5 참조). 또한, 천식 및 비염 동물 모델에 본 발명의 항-HRF scFv-Fc 항체를 투여하여 총 백혈구 세포수가 감소하는 것을 확인하였고 (도 7 참조), IL-4, IL-5 및 IL-13 수준이 감소함을 확인하였으며 (도 8a 내지 도 8c 참조), 혈장 내 오발부민 특이적인 IgE 생산량도 감소함을 확인하였으며 (도 9 참조), 염증세포 침윤과 점액질 분비가 감소함을 확인하였다 (도 10 참조). 따라서 본 발명의 항-HRF 항체는 염증을 억제하는 효과가 있음을 알 수 있으며, HRF 관련 질환의 치료제로 유용하게 이용될 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명한다.
이들 실시예는 단지 본 발명을 예시하기 위한 것이므로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지는 않는다.
<실시예 1> HRF의 발현, 분리 및 정제
HRF 단량체 또는 이량체를 제작하기 위한 실험을 수행하였다.
구체적으로, 도 1a에 나타낸 바와 같이, HRF 이량체를 생산하기 위해 N-말단의 아미노산 10개가 결실되도록 발현 벡터를 제작하였다. 이후, HRF 유전자가 삽입된 pRSET A (Invitrogen) 발현 벡터를 E. coli BL21(DE3)pLysS (Promega)에 형질전환 (transformation)시키고 배양하여 재조합 단백질의 합성을 유도하였다 (도 1b 및 도 1c). 배양액은 His · Bind resin (Novagen)을 이용하여 Ni2+과 결합한 His-tagged HRF 단백질을 용출 완충액 (elution buffer)을 이용하여 용출시켜 수거하였다. His-tag column으로 분리 및 정제한 재조합 HRF의 정제도를 높이기 위해 2차 단백질 정제를 실시하였다. 2차 단백질 정제는 음이온 교환 칼럼 (anion exchange column)인 Mono Q HR 5/5 column (Amersham Pharmacia Biotech)을 사용하여 재정제하였다. 보다 자세하게, Mono Q HR 5/5 column은 3차 증류수를 약 5분간 흘려주어 세척한 후 20 mM Tris-HCl, 50 mM NaCl, 1 mM EDTA (pH 7.4)을 약 40분간 흘려보내 안정화시켰다. 앞에서 1차 정제한 단백질을 칼럼 (column)에 로딩 (loading)하고 다시 0~400 mM의 NaCl을 흘려보내 농도구배별로 1ml/min씩 샘플 (sample)을 받았다. 이 분획들을 SDS-PAGE 한 후, 쿠마시 브릴리언트 블루 (coomassie brilliant blue) 염색을 하여 HRF가 용출되는 분획만 선별하였다 (도 1d 및 도 1e). 이를 vivaspin (vivascience)을 사용하여 버퍼 조성을 PBS로 치환하면서 농축하였다.
<실시예 2> 패닝(panning)에 의한 특이적인 항체들의 선별
인간항체인 단쇄가변분절 (single-chain variable fragment; scFv) 파지 라이브러리를 이용하여 패닝 실험을 진행하여 HRF 항원에 특이적으로 결합하는 파지를 증가시켰다.
구체적으로, 5 μg의 HRF 항원을 2 ml의 PBS에 가한 다음 면역 시험관 (immunotube)에 넣고 4℃에서 하룻밤 방치시켜 시험관 표면에 항원을 흡착시켰다. 다음날 PBS-T(phosphate buffered saline-Tween20)로 3회 세척하고 PBS로 희석한 3% 스킴 밀크 (skim milk)를 사용하여 상온에서 2시간 동안 차단 (blocking)하였다. 이후, PBS-T로 3회 세척하였다. 항원에 결합하는 파지를 선별하기 위해 1012 PFU의 항체 파지 라이브러리가 포함된 용액 3 ml를 가하여 2시간 동안 반응시켰고, 이후 시험관 안의 용액을 버리고 PBS-T로 10회 세척하였다. 항원에 결합한 scFv 파지 항체를 용리하여 수거하기 위해 0.1 M glycine/HCl (pH 2) 1 ml을 넣고 상온에서 10분간 방치하여 파지를 용리시켰다. 이후, 2M Tris-base 50 μl를 넣어 중화시켰다. 이와 같은 패닝 (panning) 과정을 3회 반복함으로써 항원에 특이적으로 결합하는 파지를 획득 및 증가시켰다 (도 2a).
<실시예 3> 양성 클론(clone)의 선별 및 확인
HRF 항원에 높은 결합력을 가지는 항-HRF 단일클론 항체를 선별하기 위하여, 96-well plate의 well에 각각 200 μl의 SB-carbenicillin 배지를 넣고, scFv 유전자가 포함된 ER2537 E. coli 단일 콜로니를 멸균된 이쑤시개를 이용하여 각각의 well에 접종하고 배양 (37 ℃, 4 시간) 후 VCSM13 헬퍼 파지를 넣어 감염시켰다. 헬퍼 파지 감염 후 대장균을 원심 분리하여 감염되지 않은 헬퍼 파지를 제거한 후 침전된 대장균을 250 μl의 배지에 현탁하였다. 이후 37℃에서 16시간 배양하였다. 배양액을 원심 분리하여 상층에 부유한 파지 용액을 회수하여 ELISA를 시행하였다.
항원에 결합하는 파지 클론을 확인하기 위해 96-well plate에 항원 100 ng을 하룻밤 코팅하였다. PBS-T로 항원을 코팅한 plate는 PBS로 3회 세척하였다. 이후 3% 스킴밀크로 상온에서 플레이트를 1시간 동안 블라킹 (blocking)시켰다. 이후 플레이트를 PBS-T로 3회 세척한 다음 파지 용액을 가하여 상온에서 1시간 동안 반응시켰다. 플레이트를 PBS-T로 세척하고, HRF가 결합되어 있는 항-M13-HRF를 첨가하여 실온에서 1시간 동안 반응시켰다. 플레이트를 PBS-T로 3회 세척하여 결합되지 않은 항체를 제거하였다. 그리고 TMB 기질을 첨가하고 10분 동안 반응시켜 발색시킨 후에, 2 N 염산 용액을 첨가하여 발색 반응을 종료시켰다. 흡광도 평판 판독기를 이용하여 450 nm의 흡광도를 측정하여 평가하였다.
그 결과, 도 2b에 나타난 바와 같이, 288개의 클로니에서 항원인 HRF 이량체에 대하여 양성반응을 보이는 38개의 클로니를 선별하였다. 또한, 도 2c에 나타난 바와 같이, 항원인 HRF 단량체에 대하여 양성반응을 가지는 33개의 클로니를 선별하였다.
<실시예 4> 항-HRF 단클론 항체의 아미노산 서열 확인
실시예 3에서 선별된 양성 파지 클론의 scFv 유전자 서열은 ABI 프리즘 3730XL 유전자 분석기 (Applied Biosystems)로 분석하여 항체의 아미노산 서열을 확인하였다.
그 결과, 도 2d 및 도 2e에 나타난 바와 같이, 서로 다른 아미노산을 가지고 있는 8종의 항-HRF scFv 항체를 획득하였다.
<실시예 5> 항-HRF 단일클론 항체의 제작 및 정제
실시예 4를 통해 확인한 항-HRF 단클론 항체를 scFv-Fc 형태로 전환하기 위해, scFv 유전자를 scFv-Fc 발현 벡터에 삽입하였고, 이후 HEK293F 세포에 형질전환하여 항체 발현을 유도하였다. 이후, 세포 밖으로 분비되는 항체를 얻기 위해 배양액을 원심분리하여 상층액을 회수하였다. 획득한 배양액은 여과 과정을 거친 후 친화성 크로마토그래피인 protein G column을 이용하여 항체를 정제하였다. 제작 및 정제한 항체를 확인하기 위하여, SDS-PAGE 분석을 진행하였다. 보다 자세하게는, 정제된 항체 4 μg에 각각 환원 샘플버퍼 (reducing sample buffer)와 비환원 샘플버퍼 (Non-reducing sample buffer)를 첨가한 후에, 10% 아크릴아미드 겔 (acrylamid gel)에 로딩하였다. 이후 전기영동을 실시한 다음, 이후 쿠마시 블루를 이용하여 단백질을 염색하였다.
그 결과, 도 3a 및 도 3b에 나타난 바와 같이, 순도 95% 이상의 항-HRF 단일클론 항체가 제작 및 정제되었음을 확인하였다.
<실험예 1> 항-HRF 단일클론 항체의 항원 결합력
실시예 5에서 제작 및 정제된 항-HRF scFv-Fc 항체 중에서 항원 결합력이 우수한 단일클론 항체를 선별하기 위하여 ELISA binding 실험을 실시하였다.
구체적으로, 코팅 버퍼 (coating buffer, 50 mM의 sodium carbonate, pH 9.6)에 HRF 항원을 96-well plate에 1 μg/ml의 농도가 되도록 분주한 다음, 항원을 4℃에서 하룻밤 코팅하였다. 그리고 PBS-T를 이용하여 3회 세척하여 결합되지 않은 항원을 제거하였다, 이후 3% skim milk로 plate를 실온에서 1시간 동안 blocking 하였고, PBS-T로 3회 이용하여 다시 세척하였다. 그리고 항-HRF 단일클론 항체를 희석하여 넣고 1시간 동안 실온에서 반응시켰다. 항원-항체 결합 1시간 후에 PBS-T를 이용하여 3회 세척하여 결합되지 않은 항체를 제거해 준 후에, HRF가 결합되어 있는 검출용 항체 (HRF conjugated anti-human IgG)를 첨가하고 실온에서 1시간 동안 반응시켰다. 이후, 플레이트를 PBS-T로 5회 세척하여 결합되지 않은 검출용 항체를 제거하였다. 그리고 TMB 기질을 첨가하고 10분 동안 반응시켜 발색을 유도시킨 다음, 2 N의 염산 용액을 첨가하여 발색 반응을 종료시켰다. 흡광도 평판 판독기를 이용하여 450 nm의 흡광도를 측정하여, 항원 결합력이 우수한 항체를 확인하였다.
그 결과, 도 4a에 나타낸 바와 같이, 사람 HRF 이량체에 대한 결합력은 JEW-D195 ≥ JEW-M449 > JEW-D121 > JEW-D4 > JEW-M353 > JEW-M357 > JEW-D199 > JEW-M491 순으로 높음을 확인하였다. 또한, 도 4b에 나타난 바와 같이, 마우스/랫트 HRF 이량체에 대한 결합력은 JEW-M449 ≥ JEW-D195 > JEW-D121 > JEW-D4 > JEW-M353 > JEW-M357 > JEW-D199 > JEW-M491 순으로 높음을 확인하였다.
<실험예 2> 기관지 상피세포의 IL-8 분비 억제 효능 확인
항원 결합력인 우수한 단일클론 항체의 in vitro 효능을 확인하기 위해, 인체 기관지 상피세포주인 BEAS-2B 세포를 이용하였다. BEAS-2B 세포의 활성화를 유도하기 위하여 HRF 이량체를 처리하였고, 일정 시간이 경과한 뒤 세포 상등액에 포함된 염증성 매개물질인 IL-8의 증가된 생성량을 확인하여 제작한 항-HRF 단일클론 항체의 효능을 검정하였다.
보다 자세하게는, BEAS-2B 세포는 37℃, 5% 이산화탄소 존재하에 DMEM 배지에서 배양하였으며, 세포가 약 80% 성장했을 때 48-well plate로 옮겨 24시간 동안 성장시켰다. 세포가 80~90% 성장했을 때 DMEM 배지로 2회 세척하였고, 각각의 단일클론 항체 (30 μg/ml)는 HRF (2 μg/ml)와 미리 혼합한 다음, 60분 후 이 혼합물을 세포에 처리하여 18시간 동안 배양하였다. 시판용 ELISA kit (Biolegend)를 이용하여 세포 상등액 내에 존재하는 IL-8의 생성량을 정량하였다.
그 결과, 도 5에 나타낸 바와 같이, BEAS-2B 세포에 HRF만 처리한 경우 세포 활성화를 유도하여 세포 상등액에 포함된 염증성 매개물질인 IL-8의 증가된 생성량을 확인하였다. 반면에 항-HRF 단일클론 항체 (JEW-M449, JEW-D121, JEW-D4, JEW-D195)를 처리한 경우 IL-8 생성량이 억제되는 것을 확인하였다.
<실험예 3> 천식 모델에서 항-HRF 단클론 항체의 항염증 효능
질병 모델에서 항-HRF scFv-Fc 항체의 효능을 확인하기 위해 오발부민(chicken egg ovalbumin)으로 천식 및 비염 동물 모델을 도 6에 나타낸 바와 같이 유도하였다.
구체적으로, 실험동물은 7~8주령의 암컷 BALB/c 마우스을 이용하였으며, 알러지성 천식의 유도하기 위해 오발부민 50 ㎍ (Sigma)과 1 mg Alum을 PBS 0.2 ml에 혼합하여 마우스의 복강 내 주사하였다. 1차 전신감작 (sensitization) 후 14일째에 동일한 방법으로 2차 전신감작을 통해 면역 부스팅 (boosting)을 유도하였다. 1차 전신감작 후 28, 30, 32, 34일째에 오발부민(200 ㎍)을 녹인 PBS 20 ㎕를 마우스의 비강 내 투여하여 흡입감작시켰다. 항-HRF scFv-Fc 항체인 JEW-M449의 약효를 평가하기 위해 항체 투여양을 100, 200 ㎍/mouse로 설정하였다. 도 6에 나타낸 바와 같이, 항체 투여 시기는 오발부민 흡입감작 15분 전에 복강 내 투여하였다. 이틀에 한 번 총 4회 투여하였고, 마지막 항체 투여 48시간 후 실험동물을 희생하였다.
오발부민의 반복적인 감작에 의한 알러지성 천식 동물 모델의 기관지폐포세척액에서는 호산구를 포함한 염증세포와 염증성 사이토카인이 증가한다. 따라서, 본 발명자들은 항-HRF 단클론 항체의 항염증성 효능을 확인하기 위해 기관지폐포세척액을 채취하였다. 구체적으로, 각 군별로 마지막 오발부민 흡입감작 48시간 후 마우스의 복강 내 졸레틸과 럼푼을 썩어 주사하여 마취시켰다. 마취 후 개흉한 다음 기도 부위를 절개하여 기도 내로 20 게이지의 혈관 내 튜브카테터를 삽입하였다. PBS 0.8 ml를 주입하고 흡입하기를 3회 반복하여 세척액을 회수하였다. 회수한 기관지폐포세척액은 4℃, 2,000 rpm에서 15분간 원심분리하고, 상층액은 -70℃에 보관하였다. 세포침전물은 PBS 200 ㎕로 재분산하여 염증성 세포수, 즉 총 백혈구 세포수를 측정하였다. Cytospin을 사용하여 재분산한 세포침전물을 슬라이드 글라스에 도말한 후 Diff-Quick(Baxter Healthcare)을 사용하여 염색하였다. 이후 현미경 하에서 총 백혈구 세포수를 측정하여 비교하였다.
그 결과, 도 7에 나타낸 바와 같이, 총 백혈구 세포수는 정상대조군 (Control)에 비교하여 알러지 유발군(OVA)에서 증가하였고, 알러지 유발군에 비해 항체 치료군(Anti-HRF)은 용량 의존적으로 총 백혈구 세포수가 감소하는 것을 확인하였다.
다음으로, 기관지폐포세척액을 원심 분리한 상층액에서 알러지 염증반응의 지표로, Th2의 면역 반응도를 보기 위해 IL-4, IL-5 및 IL-13 수준을 시판용 ELISA kit (Biolegend)를 이용하여 측정하였다.
그 결과, 도 8a 내지 8c에 나타낸 바와 같이, IL-4, IL-5 및 IL-13 수준은 정상대조군에 비교하여 알러지 유발군에서 증가하였고, 알러지 유발군과 비교하여 항체 치료군에서 용량 의존적으로 감소하였다.
각 군별로 혈장 내 오발부민 특이적인 IgE 생성량을 확인하기 위해 마우스의 혈액 채취하였다. 구체적으로, 마지막 오발부민 흡입감작 48시간 후 마취 상태인 마우스의 심장에서 ~0.5 ml의 혈액을 채취한 다음, 4℃, 1,000 x g에서 15분간 원심분리하여 혈장을 얻었다. 혈장 내 오발부민 특이적인 IgE 생산량은 시판용 ELISA kit (Biolegend)을 이용하여 측정하였다.
그 결과, 도 9에 나타낸 바와 같이, 혈장 내 오발부민 특이적인 IgE는 정상대조군에서는 검출되지 않았고, 알러지 유발군에서 증가하는 것을 확인하였다. 또한, 오발부민 특이적인 IgE 생성량은 알러지 유발군과 비교하여 항체 치료군에서 용량 의존적으로 감소하는 것을 확인하였다.
각 군별로 기관지폐포세척액 채취한 다음, 폐 조직 일부를 10% 포르말린 용액에 고정하였다. 고정된 조직은 탈수과정을 거쳐 파라핀 블록을 제작하였고, 5 ㎛ 두께로 절편을 만들어 hematoxylin & eosin (H&E) 염색과 periodic acid-Schiff (PAS) 염색을 실시하였다. 염색한 폐 조직 절편은 광학현미경을 이용하여 병리조직학적 검사를 하였다.
염증세포 침윤 정도를 관찰하기 위해 폐 조직을 H&E 염색한 결과와 점액질 분비 정도를 측정하기 위해 PAS 염색한 결과를 도 10에 제시하였다. 정상대조군에서는 이상 소견이 관찰되지 않은 반면, 알러지 유발군에서는 기도 협착과 기도벽의 두께가 증가되고 다량의 염증세포가 침윤된 것이 확인되었고, PAS 양성세포의 양이 증가는 것으로 관찰되었다. 반면, 항체 치료군의 폐 조직 염색 결과, 알러지 유발군에 비해 염증세포 침윤과 PAS 양성세포 양이 용량 의존적으로 감소하는 것으로 관찰되었다.

- 제9항에 있어서, 상기 염증성 질환 또는 자가면역질환은 천식, 기관지염, 만성폐쇄성폐질환, 기관지확장증, 비염, 아토피 피부염, 두드러기, 건초열, 결막염, 아나필락시스의 알레르기 질환, 기관지염, 폐렴, 관절염. 신장염, 건선, 피부염, 크론병, 장염, 치은염, 동맥경화증, 관상동맥염, 간염, 베체트병, 방광암, 전립선염, 신우신염, 사구체신염, 골수염, 갑상선염, 포도막염, 복강염, 수막염, 폐섬유화증 및 류마티스 관절염으로 구성되는 군에서 선택되는 것을 특징으로 하는, HRF 관련 질환의 예방 또는 치료용 약학적 조성물.
- 제1항 내지 제3항 중 어느 한 항의 항체 또는 이의 항원 결합 단편을 포함하는 것을 특징으로 하는, HRF 관련 질환의 진단용 조성물.
- 제11항의 조성물을 포함하는 것을 특징으로 하는, HRF 관련 질환의 진단용 키트.
- 제1항 내지 제3항 중 어느 한 항의 항체 또는 이의 항원 결합 단편을 포함하는 조성물을 포함하는 것을 특징으로 하는, HRF 검출용 키트.
- 제1항 내지 제3항 중 어느 한 항의 항체 또는 이의 항원 결합 단편을 이용하여 HRF 관련 질환이 의심되는 개체의 분리된 생물학적 시료의 HRF 단백질을 항원-항체 반응을 통하여 검출하는 단계를 포함하는 것을 특징으로 하는, HRF 관련 질환의 진단 방법.
