Abstract
본 발명은 혈뇌장벽 또는 뇌혈관장벽을 통과하는 폴리펩타이드 및 상기 폴리펩타이드의 물질 전달 용도에 관한 것으로 상기 폴리펩타이드는 단백질과 같이 자연적으로는 혈뇌장벽을 통과할 수 없는 물질을 뇌 조직 내로 전달할 수 있다.
Description
본 발명은 혈뇌장벽 또는 뇌혈관장벽을 통과하는 폴리펩타이드 및 상기 폴리펩타이드의 물질 전달 용도에 관한 것이다.
혈뇌장벽 또는 뇌혈관 장벽 (Blood-Brain Barrier, BBB)은 뇌척수액과 혈액을 분리시키는 장벽이다. 높은 선택적 투과성을 갖고 있어 몸의 주요 조절중추를 세균 등과 같이 혈액으로부터 운반될 수 있는 병원체 및 혈액 내의 잠재적인 위험물질로부터 격리시키는 역할을 한다.
혈뇌장벽은 포도당, 필수아미노산, 전해질 등은 내피세포를 통해 수동적으로 통과시키지만 혈중의 대사산물, 독소, 약물은 뇌세포로 들어오지 못하게 막는 역할을 한다. 그러나 물, 공기, 이산화탄소와 같은 지용성 물질은 자유롭게 장벽을 통과할 수 있으므로 알코올, 니코틴, 마취제 등은 뇌에 영향을 미치게 된다.
혈뇌장벽의 이러한 선택적 투과성은 알츠하이머 질환, 파킨슨 질환, 세균성 및 바이러스성 감염 및 암과 같은 뇌 및 중추신경계 (central nervous system, CNS) 질환의 약리학적 치료에 주요 장애가 된다. 뇌 및 중추신경계 (CNS) 질환 및 장애를 치료하기 위한 많은 치료제는 친수성이어서 혈뇌장벽을 직접 통과하지 못하기 때문이다.
상기와 같은 상황에서 본 발명자들은 신규한 혈뇌장벽 투과성 펩타이드를 발굴하기 위해 연구하였고, 5종의 신규 펩타이드와 GST의 융합 단백질이 혈뇌장벽을 통과하는 것을 생체 내 실험 결과로 확인하였다.
따라서, 본 발명의 목적은 신규한 혈뇌장벽 투과성 펩타이드 및 상기 펩타이드의 목적물질 뇌전달 용도를 제공하는 것이다.
상기 목적을 달성하기 위하여 본 발명의 일 양상은 서열번호 3 또는 서열번호 5의 아미노산 서열을 포함하는 혈뇌장벽 투과성 펩타이드를 제공한다.
본 발명의 일 구체예에 따르면, 서열번호 3의 아미노산 서열을 포함하는 혈뇌장벽 투과성 펩타이드는 서열번호 1 내지 3으로 이루어진 군에서 선택될 수 있고, 서열번호 5의 아미노산 서열을 포함하는 혈뇌장벽 투과성 펩타이드는 서열번호 4 또는 서열번호 5의 아미노산 서열로 이루어질 수 있다.
본 발명에서, 서열번호 3의 아미노산 서열을 포함하는 혈뇌장벽 투과성 펩타이드는 서열번호 1 내지 3으로 이루어진 군에서 선택되는 아미노산 서열과 70% 이상, 바람직하게는 80% 이상, 더욱 바람직하게는 90% 이상, 더더욱 바람직하게는 95% 이상, 가장 바람직하게는 98% 이상의 서열 상동성을 가지는 아미노산 서열을 포함할 수 있다.
본 발명에서, 서열번호 5의 아미노산 서열을 포함하는 혈뇌장벽 투과성 펩타이드는 서열번호 5의 아미노산 서열과 70% 이상, 바람직하게는 80% 이상, 더욱 바람직하게는 90% 이상, 더더욱 바람직하게는 95% 이상, 가장 바람직하게는 98% 이상의 서열 상동성을 가지는 아미노산 서열을 포함할 수 있다.
한편, 상기 혈뇌장벽 투과성 펩타이드는 다른 세포막 투과성 펩타이드와 융합되어 사용될 수 있으며, 세포막 투과성 펩타이드의 예에는 페네트라틴 (Penetratin), TAT 펩티드, Pep-1펩티드, S413-PV, 메가이닌 2 (Magainin 2) 또는 부포린 2 (Buforin 2)가 있다.
본 발명자들은 발굴한 혈뇌장벽 투과성 펩타이드와 GST의 융합 단백질이 마우스의 혈뇌장벽을 통과하는 것을 구체적인 실험으로 확인하였다 (도 1 내지 5).
따라서, 본 발명의 다른 양상은 혈뇌장벽 투과성 펩타이드; 및 상기 혈뇌장벽 투과성 펩타이드의 일 말단에 결합된 목적 물질;을 포함하는 목적 물질 뇌전달용 시스템을 제공한다.
상기 목적 물질은 화합물, 단백질, 당단백질, 펩타이드, 뉴클레오티드, 핵산, 탄수화물, 지질, 당지질, 나노입자, 리포좀, 약물, 지질 기반 제제 (lipid-based formulation), 바이러스, 양자점 (quantum dots) 및 형광색소 (fluorochrome)로 이루어진 군으로부터 선택될 수 있다.
본 발명에서, 상기 혈뇌장벽 투과성 단백질과 목적 물질은 당 업계에 공지된 단백질과 목적 물질을 결합시키는 방법으로 연결될 수 있으며, 예를 들어 공유결합, 화학결합, 펩타이드 결합 등으로 연결될 수 있다.
상기 기술하였듯이 본 발명자들은 혈뇌장벽 투과성 펩타이드와 GST의 융합 단백질이 혈뇌장벽을 통과하는 것을 확인한 바, 목적 물질로 단백질(목적 단백질)을 사용할 때는 혈뇌장벽 투과성 단백질과 목적 단백질의 융합 단백질 형태로 뇌조직에 도입할 수 있다.
한편, 목적 물질로 뇌 또는 중추 신경계 질환의 치료제나 진단제를 사용한다면 목적 물질 뇌전달용 시스템은 뇌 또는 중추 신경계 질환의 치료 또는 예방용 약학 조성물, 뇌 또는 중추 신경계 질환의 진단용 조성물로 활용될 수 있다.
예를 들어, 본 발명은 알파-시누클레인 (alpha-synuclein), 아밀로이드 베타 (amyloid beta), 타우 (tau), SOD1 (Superoxide dismutase1), TDP-43 등 퇴행성 뇌질환에서 응집이 증가하는 다양한 타겟물질에 선택적으로 결합하여 응집을 억제하는 물질과 혈뇌장벽 투과성 펩타이드를 연결시킨 후, 이를 체내에 주입하면 뇌실질 내의 상기 타겟물질의 분포를 PET를 통해 조기에 조영하거나 응집을 억제할 수 있다.
상기 뇌 또는 중추 신경계 질환에는 일반적으로 말하는 퇴행성 신경 질환이 포함되며, 알츠하이머성 치매, 파킨슨 병, 전두측두치매, 루이치매, 및 헌팅톤병 등이 포함된다.
상기 뇌 또는 중추 신경계 질환의 진단용 조성물은 뇌 또는 중추 신경계 질환의 위험도 예측 또는 진단용 키트 형태로 구현되어 제공될 수 있다. 이 키트에는 혈뇌장벽 투과성 펩타이드를 검출할 수 있는 제제를 포함할 뿐만 아니라, 분석 방법에 적합한 1종 이상의 다른 구성성분 조성물, 용액, 또는 장치가 포함될 수 있다.
상기 뇌 또는 중추 신경계 질환의 치료 또는 예방용 약학 조성물은 목적하는 방법에 따라 경구투여하거나 비경구투여(예를 들어, 정맥 내, 피하, 복강 내 또는 국소에 적용)할 수 있으며, 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 제조된다.
경구투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제, 트로키제 등이 포함되며, 이러한 고형 제제는 유효성분에 적어도 하나 이상의 부형제 예를 들면, 전분, 탄산칼슘, 수크로스(sucrose) 또는 락토오스(lactose) 또는 젤라틴 등을 섞어 제조된다. 또한, 단순한 부형제 외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용된다. 경구 투여를 위한 액상 제제로는 현탁제, 내용 액제, 유제 또는 시럽제 등이 해당되는데, 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁용제, 유제, 동결건조제제, 좌제 등이 포함된다. 비수성용제, 현탁용제로는 프로필렌글리콜, 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세롤, 젤라틴 등이 사용될 수 있다.
본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에 있어서 '약학적으로 유효한 양'은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 예를 들어, 본 발명의 약학적 조성물은 0.1 ㎎/㎏ 내지 200 ㎎/㎏, 바람직하게는 1 ㎎/㎏ 내지 50 ㎎/㎏ 용량으로 1일 1회, 또는 1일 3회로 분할하여 투여할 수 있다. 상기 투여량은 어떠한 방법으로도 본 발명의 범위를 한정하는 것이 아니다. 그러나 투여 경로, 비만의 중증도, 성별, 체중, 연령 등에 따라서 증감될 수 있으므로 상기 투여량이 어떠한 방법으로도 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 또 다른 양상은 상기 혈뇌장벽 투과성 펩타이드를 코딩하는 폴리뉴클레오티드를 제공한다.
본 명세서에 사용된 용어, '폴리뉴클레오티드(polynucleotide)'는 단일가닥 또는 이중가닥 형태로 존재하는 디옥시리보뉴클레오티드 (deoxyribonucleotide) 또는 리보뉴클레오티드(ribonucleotide)의 중합체이다. RNA 게놈 서열, DNA(gDNA 및 cDNA) 및 이로부터 전사되는 RNA 서열을 포괄하며, 특별하게 다른 언급이 없는 한 천연의 폴리뉴클레오티드의 유사체를 포함한다.
본 발명에서, 상기 폴리뉴클레오티드는 세포막 투과성 펩타이드를 코딩하는 뉴클레오티드 서열뿐만 아니라, 그 서열에 상보적인(complementary) 서열도 포함한다. 상기 상보적인 서열은 완벽하게 상보적인 서열뿐만 아니라, 실질적으로 상보적인 서열도 포함하다. 또한, 상기 폴리뉴클레오티드 서열은 변형될 수 있으며, 변형은 뉴클레오티드의 추가, 결실 또는 비보존적 치환 또는 보존적 치환을 포함한다.
본 발명의 또 다른 양상은 상기 혈뇌장벽 투과성 단백질을 코딩하는 폴리뉴클레오티드 서열을 포함하는 재조합 벡터를 제공한다.
본 명세서에 사용된 용어, '벡터(vector)'는 숙주세포에서 목적 유전자를 발현시키기 위한 수단을 의미한다. 예를 들어, 플라스미드 벡터, 코즈미드 벡터 및 박테리오파아지 벡터, 아데노바이러스 벡터, 레트로바이러스 벡터 및 아데노-연관 바이러스 벡터와 같은 바이러스 벡터를 포함할 수 있으나, 이에 한정하지는 않는다. 상기 재조합 벡터로 사용될 수 있는 벡터는 당 업계에서 종종 사용되는 플라스미드 (예를 들면, pSC101, pGV1106, pACYC177, ColE1, pKT230, pME290, pBR322, pUC8/9, pUC6, pBD9, pHC79, pIJ61, pLAFR1, pHV14, pGEX 시리즈, pET 시리즈 및 pUC19 등), 파지 (예를 들면, λgt4λB, λ-Charon, λΔz1 및 M13 등) 또는 바이러스(예를 들면, CMV, SV40 등)를 조작하여 제작될 수 있다.
상기 재조합 벡터는 상기 폴리뉴클레오티드 서열과 폴리뉴클레오티드 서열에 작동적으로 연결된(operatively linked) 프로모터를 포함할 수 있다.
본 명세서에 사용된 용어, '작동적으로 연결된'은 뉴클레오티드 발현 조절 서열(예를 들어, 프로모터 서열)과 다른 뉴클레오티드 서열 사이의 기능적인 결합을 의미하며, 이에 의해 상기 조절 서열은 상기 다른 뉴클레오티드 서열의 전사 및/또는 해독을 조절하게 된다.
본 발명의 일 구체예에 따르면, 목적 물질로 단백질을 사용하는 경우 벡터에 혈뇌장벽 투과성 단백질과 목적 단백질을 코딩하는 폴리뉴클레오티드 서열을 같이 도입하여 융합 단백질 형태로 발현시킬 수 있다.
따라서, 상기 재조합 벡터는 융합 단백질의 정제를 용이하게 하는 태그(tag) 서열, 예를 들어, 연속된 히스티딘 코돈, 말토우즈 바인딩 단백질 코돈 및 Myc 코돈 등을 포함할 수 있다. 또한, 상기 재조합 벡터는 융합 단백질의 가용성(solubility)을 증가시키기 위한 융합파트너(partner) 등을 추가로 포함할 수 있다. 또한 융합 단백질 발현 시 불필요한 부분을 제거하기 위하여 효소에 의해 특이적으로 절단되는 서열, 발현 조절 서열 및 세포 내 전달을 확인하기 위한 마커(marker) 또는 리포터 유전자 서열을 포함할 수 있다.
본 발명의 혈뇌장벽 투과성 폴리펩타이드를 이용하면 자연적으로는 혈뇌장벽을 통과할 수 없는 물질을 뇌 조직 내로 효과적으로 전달할 수 있다.

도 4는 서열번호 4의 폴리펩타이드에 형광물질을 표지하여 마우스에 투여한 후 뇌 조직에서 폴리펩타이드의 시간별 혈뇌장벽 투과도를 확인한 결과이다 (적색: 혈관, 녹색: 폴리펩타이드).
도 5는 서열번호 5의 폴리펩타이드에 형광물질을 표지하여 마우스에 투여한 후 뇌 조직에서 폴리펩타이드의 시간별 혈뇌장벽 투과도를 확인한 결과이다 (적색: 혈관, 녹색: 폴리펩타이드).
이하 하나 이상의 구체예를 실시예를 통하여 보다 상세하게 설명한다. 그러나 이들 실시예는 하나 이상의 구체예를 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1: 펩타이드 준비
펩타이드를 다음과 같이 준비하였으며, 펩타이드 서열은 표 1에 기재하였다.

투과성 펩타이드가 약물 전달체로 활용될 수 있는지 평가하기 위해 자연적으로는 혈뇌장벽을 투과할 수 없는 GST를 서열번호 1 또는 2에 해당하는 펩타이드의 N-말단에 접합시켰다.
구체적으로 pGEX 벡터 시스템에 서열번호 1 또는 2의 펩타이드 서열과 GST를 인코딩하는 뉴클레오티드 서열을 삽입하여 재조합 벡터를 만들고, 이 재조합 벡터를 대장균 균주 BL21(DE3)에 형질전환으로 도입하였다. 상기 균주를 LB 배지에서 26℃로 3시간 동안 배양한 후 1 mM의 IPTG (isopropyl-β-D-thiogalactopyranoside)를 처리하고, 다시 18℃에서 12시간 배양하였다.
이후 배양액을 회수하여 원심분리로 세포를 회수하고, 세포에 초음파를 처리 (sonication)하였다. 친화성 크로마토그래피로 GST+펩타이드의 융합 단백질을 얻었다. 수득한 융합 단백질은 PBS(pH 7.4) 완충액을 이용해 2~3일간 염 농도를 맞추고 순도를 높였다.
서열번호 1, 2를 제외한 서열번호 3, 4 및 5의 펩타이드는 합성하였고, 순도는 95 % 이상인 것을 확인 후 이후 실험에 활용하였다.
실시예 2: 펩타이드의 BBB 투과능 평가
본 발명의 펩타이드가 생체 시스템에서 혈뇌장벽을 투과할 수 있는지 다음과 같이 확인하였다.
생체 내에서 펩타이드를 추적하기 위해 실시예 1에서 제작한 GST+펩타이드의 융합 단백질 또는 서열번호 3 내지 5의 펩타이드에 형광신호를 나타내는 Dylight 488을 결합시켰다. 형광물질이 결합된 각 융합 단백질 또는 펩타이드 시료를 인체와 유사한 대사작용을 나타내는 것으로 알려진 마우스 (계통명: C57BL/6N, 8주령)에 주입하고 뇌 조직 및 혈관부위에서 시료의 존재 여부를 관찰하였다
마우스는 움직임을 제한하고 이에 따른 부적절한 결과 해석을 방지하고자 호흡마취 (isoflurane)을 진행하였다. 뇌 조직과 혈관을 구분하기 위하여 항-CD31과 FSD fluor 647 화합물을 통해 혈관을 염색하였으며, 조직 이미징을 위해 두개골 위의 피부 조직을 절개하고, 절개 부위에 두개골 창 (cranial window)를 부착하여 고정된 이미지를 얻었다. 이후 시료 주입 직전 (0 hr)의 조직 이미지를 형광현미경 (IVM-CM Confocal & Two-photon convertible microscopy)으로 촬영하고, 시료 (10 mg/kg)를 정맥주사로 주입하여 시간에 따른 형광신호 양상을 관찰하였다.
그 결과, 펩타이드 1 및 2는 주입 후 0.16시간 (10분) 시점부터 뇌 조직 내 혈관에서 형광신호를 관찰할 수 있었고, 펩타이드 1은 주입 후 1시간, 펩타이드 2는 주입 후 4시간 시점에 뇌 조직 내 형광신호가 가장 높은 것을 알 수 있었다 (도 1 및 도 2).
펩타이드 3 내지 5는 모두 주입 후 0.5시간 (30분) 시점부터 뇌 조직 내 혈관에서 형광신호를 관찰할 수 있었으며, 주입 후 1시간 시점에 형광신호가 가장 높았다 (도 1 내지 5).
도 1 내지 5의 결과를 통해 본 발명의 펩타이드가 혈뇌장벽을 투과하고, GST와 같은 큰 물질을 효과적으로 뇌 조직 내로 전달할 수 있음을 확인하였다.
Claims (10)
- 서열번호 3 또는 서열번호 5의 아미노산 서열을 포함하는 혈뇌장벽 투과성 펩타이드.
- 제1항에 있어서, 서열번호 3의 아미노산 서열을 포함하는 혈뇌장벽 투과성 펩타이드는 서열번호 1 내지 3으로 이루어진 군에서 선택되는, 혈뇌장벽 투과성 펩타이드.
- 제1항에 있어서, 서열번호 5의 아미노산 서열을 포함하는 혈뇌장벽 투과성 펩타이드는 서열번호 4 또는 서열번호 5의 아미노산 서열로 이루어지는, 혈뇌장벽 투과성 펩타이드.
- 제1항 내지 제3항 중 어느 한 항에 따른 혈뇌장벽 투과성 펩타이드; 및
상기 혈뇌장벽 투과성 펩타이드의 일 말단에 결합된 목적 물질;을 포함하는 목적 물질 뇌전달용 시스템. - 제4항에 있어서, 상기 목적 물질은 화합물, 단백질, 당단백질, 펩타이드, 뉴클레오티드, 핵산, 탄수화물, 지질, 당지질, 나노입자, 리포좀, 약물, 지질 기반 제제 (lipid-based formulation), 바이러스, 양자점 (quantum dots) 및 형광색소 (fluorochrome)로 이루어진 군으로부터 선택되는, 목적 물질 뇌전달용 시스템.
- 제4항에 있어서, 상기 혈뇌장벽 투과성 펩타이드의와 목적 물질은 공유결합으로 연결되는, 목적 물질 뇌전달용 시스템.
- 제1항 내지 제3항 중 어느 한 항에 따른 혈뇌장벽 투과성 펩타이드를 코딩하는 폴리뉴클레오티드.
- 제7항의 폴리뉴클레오티드를 포함하는 재조합 벡터.
- 제4항 내지 제6항 중 어느 한 항에 따른 목적 물질 뇌전달용 시스템을 유효성분으로 포함하는, 뇌 또는 중추 신경계 질환의 치료 또는 예방용 약학 조성물.
- 제4항 내지 제6항 중 어느 한 항에 따른 목적 물질 뇌전달용 시스템을 유효성분으로 포함하는, 뇌 또는 중추 신경계 질환의 진단용 조성물.
