Abstract
Description
The present invention relates to an antibacterial composition comprising an extract of Antarctic Meckel's smelt ( Champsocephalus gunnari ) or an extract of Antarctic marbled female fish ( Notothenia rossii ) as an active ingredient.
Microorganisms exist all around us. In particular, telephones, mobile phones, flooring materials, refrigerators, air conditioners, humidifiers, toys, wallpaper, and various gas kits that we can easily access in our daily lives are prone to propagation of various germs and molds, causing odors, and even cause human body odor. Harmful bacteria proliferate and cause various diseases, and serious social problems such as food poisoning by pathogenic Escherichia coli O-157 and legionella bacteria that cause pneumonia in public baths occur. Therefore, a highly functional antibacterial material is required to prevent this, and many studies on these antibacterial materials have been conducted to date.
Direct or indirect damage caused by pathogenic microorganisms causes many economic, environmental, and medical problems. There are many problems throughout society, such as losses due to spoilage during food distribution in the food industry, harm to the human body and environmental pollution due to the use of excessive chemical pesticides on crops in the agricultural industry, and the emergence of antibiotic-resistant strains due to misuse and abuse of antibiotics. are causing them
In addition, the awareness of the importance of the microbial contamination problem of cosmetics is increasing due to the hygienic aspect of the skin area as well as the deterioration of the product value due to the deterioration of the product itself. Cosmetics use a lot of nutrients derived from natural products and raw materials that are easily decomposed by microorganisms, so they are easily contaminated by microorganisms such as mold, yeast, and bacteria during distribution or use by consumers. In order to suppress, preservatives are added, and the minimum amount should be used to show the maximum antibacterial activity. Therefore, it is necessary to consider the combination of nutrients in cosmetic products, pH, emulsified form, mixed use of preservatives, and their stability, as well as antibacterial activity and skin irritation of all raw ingredients in the product beforehand to determine each product Selecting an appropriate preservative system for each type is very important. And because of the various side effects of synthetic preservatives, the development of natural preservatives has emerged as a major concern in the cosmetic industry.

Therefore, the composition of the present invention containing the Antarctic Meckel's smelt extract or the Antarctic marble-patterned female chum extract as an active ingredient can be usefully used as an antibacterial material.
In the present invention, 'antibacterial composition' may mean the same as antibiotics, which is a generic term for antimicrobial agents, and may mean the same as antibacterial agents, bactericides, preservatives, preservatives, or disinfectants, and may cause food poisoning or purulent It refers to a substance capable of inhibiting or suppressing the growth and life function of pathogenic microorganisms such as Staphylococcus aureus , which is the cause of infection.
Staphylococcus aureus ' is a gram-positive facultative anaerobic bacterium, which is a pathogen commonly present on the skin and nasal surfaces of healthy people or livestock, and causes food poisoning by producing heat-resistant exotoxins. It is also known to secrete phagocyte-killing toxins (leucocidin), hemolysin, coagulation enzymes, etc., to break free from the resistance of infected host cells and cause pyogenic infections. Staphylococcus aureus is commonly found on the skin and nasal passages of more than 50% of healthy people and animals, and can cause a variety of diseases ranging from mild skin infections such as acne and impetigo to life-threatening diseases such as pneumonia, meningitis, and sepsis. . It is also one of the five most common causes of nosocomial infections, often leading to postoperative wound infections. About 500,000 patients in US hospitals are known to contract staph infections each year. Staphylococcus aureus can secrete various toxins depending on the strain, which are classified into three types: pyrogenic toxin superantigens, exfoliative toxins, and other toxins. The strains that produce the 7 enterotoxins that cause food poisoning belong to superantigens of febrile toxins. Enterotoxin produced by Staphylococcus aureus causes gastroenteritis, which causes symptoms such as nausea, vomiting, diarrhea, and abdominal pain, and the symptoms are relieved and healed naturally within 8-24 hours. Food poisoning symptoms are caused by enterotoxins, not by the bacteria themselves, and because they are heat resistant, they are not destroyed even when heated or cooked, so it is best not to consume foods contaminated with Staphylococcus aureus.

First, in order to confirm the antibacterial activity, as a qualitative analysis, the inhibition of microbial growth was determined by dropping the Antarctic fish extract of the present invention on a solid culture medium. In detail, 10 8 cells of the two bacterial species were applied to LB solid medium, and then 10 ul of each of the Antarctic fish extracts of the present invention prepared in <Example 1> were treated for each concentration. The experiment was repeated twice for the same concentration. The concentration of the treated extract was 0.5, 1, 2, and 5 ug/ul, so that the amount of the extract finally used was 5, 10, 20, and 50 ug. The extract was dissolved in 50% DMSO, and 0.1 and 0.2 ug/ul of Kanamycin were used as positive controls. Each solid medium was incubated at 37 ° C. for 16 to 24 hours, and the degree of growth of the applied microorganisms was visually confirmed.
