배양 후 세포를 트립신-EDTA(trypsin-ethylenediamine tetra acetic acid)로 떼어내어 세포수를 측정한 다음 원심분리 하여 세포를 회수하였다. 세포 내 멜라닌의 정량은 로탄(Lotan: Cancer Res., 40: 3345-3350, 1980)의 방법을 변형하여 실시하였으며, 셀 펠릿을 PBS(phosphate buffered saline)로 1회 세척한 후 균질화 버퍼액(50mM 소듐 포스페이트, pH 6.8, 1% Triton X-100, 2mM PMSF) 1㎖를 첨가하여 5분간 와류시켜 세포를 파쇄 하였다. 상기 파쇄된 세포를 원심분리(3,000rpm, 10분)하여 얻은 세포 여액에 1N NaOH(10% DMSO)를 첨가하여 추출된 멜라닌을 용해한 후, 상기 마이크로 플레이트 판독기로 405㎚에서 멜라닌의 흡광도를 측정하여 멜라닌을 정량하였다. B16F10 멜라닌 형성세포의 멜라닌 생성 억제율은 하기 식에 의거하여 계산하였으며, 그 결과를 하기 표 4에 나타내었다. 이때 멜라닌 생성 저해 효과가 있는 것으로 알려진 물질인 알부틴(arbutin)을 비교군으로 사용하였다.After cultivation, the cells were separated with trypsin-ethylenediamine tetra acetic acid (EDTA), the number of cells was measured, and then the cells were recovered by centrifugation. 상기 표 4에서 확인되는 바와 같이, 본 발명 실시예의 혼합추출물은 제조예의 각각의 추출물에 비하여 우수한 멜라닌 생성 억제율을 나타내었다. 실시예 2 내지 13은 실시예 1에 비하여 더 우수한 효과를 나타내었으며, 실시예 11은 가장 우수한 효과를 나타내었다.As can be seen in Table 4, the mixed extract of Examples of the present invention exhibited superior melanin production inhibition compared to each of the extracts of Preparation Example.
