레인 M: 100 bp DNA 마커
레인 1: DNA 앱타머로부터 PCR 기법을 통하여 증폭된 dsDNA 결과
레인 2: PCR 기법으로 확보한 dsDNA를 열-냉각기법과 스트랩타비딘을 이용하여 제작한 ssDNA 결과
도 2는 SPINK1 단백질과 특이적으로 결합하는 DNA 앱타머 제작을 위하여 실시한 각 SELEX 라운드에서 회수된 GST 융합 SPINK1 단백질과 특이적으로 결합하는 DNA 앱타머 또는 SPINK1 단백질과 특이적으로 결합하는 DNA 앱타머 후보군들의 ssDNA 농도를 Nanodrop spectrophotometer를 이용하여 정량적으로 나타낸 결과이다.
도 3A, 3B는 SPINK1 단백질과 특이적으로 결합하는 DNA 앱타머 제작을 위한 SELEX 과정에서 선별된 6개의 SPINK1 단백질과 특이적으로 결합하는 DNA 앱타머 후보군 SPINK1-1 내지 SPINK1-6의 2차 구조를 나타낸 도면이다.
도 4는 카르복실기를 가지고 있는 CM5 센서 칩 표면에 SPINK1을 고정하는 단계를 나타낸 도면이다.
도 5는 SPINK1 결합 DNA 앱타머를 이용하여 ELISA 기법을 기반으로 SPINK1을 탐지하는 방법에 대한 모식도로서, 패널 [1]은 SPINK1 탐지를 위해 SPINK1 결합 DNA 앱타머를 이용한 ELISA 법의 모식도를 나타내며, 패널 [2]는 기존 항체를 이용하여 SPINK1을 탐지하는 ELISA 법의 모식도를 나타낸 것이다.
도 6은 SPINK1 단백질과 특이적으로 결합하는 앱타머를 이용한 간암 표적인자인 SPINK1 검출 키트의 모식도를 나타낸 도면이다. 간암으로 의심되는 환자의 시료를 SPINK1 단백질과 특이적으로 결합하는 앱타머가 고정되어 있는 진단 키트에 처리하였을 때, 샘플 내 SPINK1 단백질과 SPINK1 단백질 결합 앱타머의 결합에 의해 SPINK1을 진단하는 시스템을 모식도로 나타낸 도이다.

Claims (10)
- 서열번호 1 내지 서열번호 6 중에서 선택되는 어느 하나의 염기서열을 갖는 SPINK1 단백질에 특이적으로 결합하는 DNA 앱타머.
- 제 1항에 있어서,
상기 SPINK1 단백질은 서열번호 11로 표시되는 아미노산으로 이루어진 것을 특징으로 하는 SPINK1 단백질에 특이적으로 결합하는 DNA 앱타머. - 제1항의 DNA 앱타머를 포함하는 SPINK1 단백질 검출용 키트.
- 제1항의 DNA 앱타머를 포함하는 SPINK1 단백질 검출용 마이크로어레이.
- 제1항의 DNA 앱타머를 포함하는 SPINK1 단백질 발현의 증가를 나타내는 암의 진단 또는 예후 판단용 조성물.
- 제5항에 있어서,
상기 암은 간암인 것을 특징으로 하는 조성물. - 제1항의 DNA 앱타머와의 결합반응을 수행하는 단계를 포함하는 것을 특징으로 하는 SPINK1단백질의 검출 방법.
- 피험자로부터 얻은 생물학적 시료를 대상으로 제1항의 DNA 앱타머와의 결합반응을 수행하는 단계를 포함하는, SPINK1 단백질 발현의 증가를 나타내는 암의 진단 또는 예후 판단을 위한 정보 제공 방법.
- 제8항에 있어서,
상기 암은 간암인 것을 특징으로 하는 방법. - (a) 임의의 염기서열을 갖는 주형 DNA로부터 PCR 반응을 이용하여 DNA 앱타머 풀(pool)을 제작하는 단계;
(b) 가열-냉각 기법을 이용하여 상기 DNA 앱타머 풀(pool)의 이중가닥(double-stranded) DNA를 단일가닥(single-stranded) DNA로 만드는 단계;
(c) 상기 (b)단계에서 제조된 단일가닥 DNA 앱타머들을 GST 융합 SPINK1 단백질과 반응시킨 후 반응액을 기활성화된 니트로셀룰로오스 멤브레인 필터에 투과시켜, GST 융합 SPINK1 단백질과 결합하는 단일가닥 DNA 앱타머를 선별하는 단계;
(d) 상기 (c)단계에서 선별된 DNA 앱타머에 대해 니트로셀룰로오스 멤브레인 필터를 이용한 네가티브(negative) SELEX를 수행하여 SPINK1 단백질에 대해 비특이적인 DNA 앱타머를 제거하는 단계; 및
(e) 선별된 단일가닥 DNA 앱타머의 SPINK1 단백질에 대한 친화성을 측정하여 SPINK1 단백질과 가장 특이적으로 결합하는 최적의 DNA 앱타머를 결정하는 단계를 포함하는, SPINK1 단백질에 특이적으로 결합하는 DNA 앱타머의 제조방법.
