Abstract
본 발명은 생물제제 생성물로 처리하는 동물에서 감소된 수준의 항원성을 갖는 핵산 생성물 및 폴리펩타이드 단백질 생성물을 제조하는 방법을 제공한다. 체세포는 다능성 줄기 세포로 형질전환되고, 원하는 핵산(들)으로 형질주입되며, 목적하는 핵산 생성물의 높은 수준의 제조자로 알려져 있는 체세포로 재-분화되는 동물로부터 분리된다. 본 발명은 집단을 치료하기 위한 일반적인 세포주, 인종 군을 치료하기 위한 인종 특이적 세포주 또는 개개를 치료하기 위한 환자 특이적 세포주를 얻기 위해서 사용될 수 있다. 또한, 본 발명은 유도된 다능성 줄기 세포가 원래의 이들의 체세포로 재-분화되도록 하여 세포를 기형종을 형성하지 않고 동물을 치료적으로 치료하기 위해 사용할 수 있는 방법을 제공한다.

본 출원은 미국 특허 가출원 제61/429,409호(출원일: 2011년 1월 3일) 및 미국 특허 가출원 제61/431,376호(출원일: 2011년 1월 10일)의 우선권을 주장하며, 이 기초 출원들은 참고로서 이의 전문이 본 명세서에 포함된다.
현재 다수의 질병을 치료하기 위해 치료적으로 사용되는 많은 재조합 폴리펩타이드 및 단백질이 존재한다. 이러한 재조합 폴리펩타이드 및 단백질은 모두 일차 연속 비인간 또는 인간 2배체 세포계를 사용하여 상업적으로 제조된다. 예를 들면, 이들 중 일부는 대장균과 같은 세균을 사용하여 제조되는 반면에 나머지는 동물 유래의 여러 난소 세포주 또는 효모를 사용해서 제조된다. 세균은 특히 글라이코실화 패턴 및 기타 단백질 변형이 생물제제-수용체 결합 친화도, 생물제제 활성, 생물 분류, 또는 생체 또는 수용자 면역 인식을 위한 약물 동력학에 매우 중요한 경우 몇몇 폴리펩타이드 및 단백질의 제조에 사용될 수 없다. 글라이코실화가 결정적 변수인 경우 중국 햄스터 난소 세포가 현재 생체 제조에 있어서 가장 일반적으로 사용되는 세포주 중의 하나이다.
불행하게도, 장기간 또는 만성 치료에서의 재조합 폴리펩타이드 또는 단백질의 사용 의무로 인해서 제품에 대한 환자 개발 중화 항체가 수득되어 환자가 약물에 덜 반응하거나 또는 반응하지 않게 된다. 몇몇 경우에 환자는 류머티스성 관절염, 소아류머티스관절염, 건선, 건선성 관절염, 강직성 척추염, 궤양성 대장염 및 크론 병을 일상적으로 치료하기 위해 사용되는 것으로서, 동일한 부류, 예컨대 다양한 항-TNF 생물제제, 예컨대 에타너셉트(Etanercept)라고도 알려져 있는 엔브렐(Enbrel); 인플릭시맙(Infliximab)이라고도 알려져 있는 레미카이드(Remicaide); 세톨리주맙(Certolizumab) 및 D2E7라고도 알려져 있는 Humira 중 다른 약물로 교체할 수 있다. 그러나 하나의 특별한 항-TNF 생물제제에 대한 중화 항체의 생성으로 환자가 궁극적으로 또 다른 항-TNF 생물제제 제품에 대한 중화 항체를 생성하도록 한다. 몇몇 경우에, 환자에 대한 대안적인 치료법이 존재하지 않아서 이러한 중화 항체의 형성으로 환자에게 치료 선택권이 없어진다. 심지어 적당한 치료 선택권이 있다고 해도, 동일한 부류에서 다른 약물에 대한 중화 항체의 개발에 대한 소인은 결국 이러한 환자에게 치료 선택권이 없게 될 수 있다는 것을 의미한다.
인간 항체에 의해 중화될 수 있는 다른 생물제제 제품은 하기를 포함한다: HRT 및 호르몬 피임제에 의해 유발되는 유방암을 치료하기 위해 잠재적으로 사용할 수 있으며, 골다공증 및 화학요법 유래 골절 치료로 승인된 것으로서 다발성 경화증에 대한 혁신적 치료인 나탈리주맙(Natalizumab) 또는 티사브리(Tysabri) 및 핵 인자 카파-B 리간드의 수용체 활성제(Receptor activator of nuclear factor kappa-B ligand: RANKL)에 대한 완전한 인간 단일클론 항체인 데노수맙(Denosumab). 아바테셉트(Abatecept)는 항-TNF 요법에 대해 치료효과가 없는 류머티스성 관절염 환자에서의 사용이 승인된 CTLA-4 융합 단백질이다. 이의 용도가 아바테셉트 유도 중화 항체에 대한 증거를 갖기에는 너무 새로운 것이지만, 이는 또한 인간 항체 중화 항체 반응을 유도할 수 있는 가능성을 갖는다.
항체 및 융합 단백질계 약물 이외의 다른 폴리펩타이드 및 단백질도 장기간 치료에 의한 면역 반응을 이끄는 것으로 나타났다. 예를 들면, 재조합 인간 에리트로포이에틴이 이의 효험을 감소시키는 중화 항체 형성을 이끌며, 희귀 무형성증 증상을 유발할 수 있다. 또한, 혈우병 환자의 응고 인자 요법에 대한 항체의 발생은 환자 중 25 내지 30%만큼 높다. 이는 응고 인자에 의한 일반적인 문제이다. 암 및 B형 간염에 대한 재조합 인터페론 알파 2a 요법도 또한 치료에 대한 중화 항체의 발생으로 방해를 받는다. 단신의 어린이에 대한 장기간의 성장 호르몬 요법에 대한 무반응도 또한 문제가 많다. 몇몇 인슐린 제품에 대한 중화 항체 형성에 대한 보고들도 있다.
중화 항체를 끌어내는 잠재력이 있는 다른 생물제제 약물에는 전혈, 혈청, 혈장 풀 또는 기타 생물제제 공급의 일차 공급원, 예를 들면, 인간 알부민, 인간 알파1-프로테아제 억제제, 인간 항 혈우병 인자/폰 빌레브란트 인자 복합체, 베이비빅 보툴리눔중독 면역 글로불린 정맥 주사, C1 에스터라제 억제제, 섬유소 밀봉제, 피브리노겐, 면역 글로불린 정맥 주사, 면역 글로불린 피하 주사, 단백질 C 농축물, Rho(D) 면역 글로불린 정맥 주사, 트롬빈, 폰 빌레브란트 인자/응고 인자 VIII 복합체를 포함한다.
재조합 폴리펩타이드 및 단백질은 하기를 포함하는 여러 특성들을 기본으로 하는 중화 항체 발생 및 면역 반응을 이끈다: 생물제제 치료기간, 반복 요법의 간격, 생물제제의 아미노산 조성, 및 생물제제의 변형, 예컨대 글라이코실화, 메틸화, 니트로실화, 시알릴화, 인산화, 황산화, 프레닐화, 셀렌환, 유비퀴틸화, 비타민-의존적 변형, 단백질 결합 관계, 아실화, 당화반응, 3차원 배열 및 다중코일화. 따라서, 생물제제로 치료하는 동물에서 감소된 항원성을 갖는 폴리펩타이드 단백질 제품을 생성하는 방법이 필요하다.

본 발명은 합성적으로 생산된 다능성 줄기 세포(synthetically produced pluripotent stem cell: spPSC) 또는 내생 다능성 줄기 세포(endogenous pluripotent stem cell: ePSC)를 형질주입(transfecting) 또는 형질전환(transforming)시킴으로써 폴리펩타이드 또는 단백질과 같은 생물제제, 핵산, 바이러스 및 백신을 제조하기 위한 방법을 제공함으로써 이러한 필요를 충족시킨다. 이러한 세포들은 처리되고 목적하는 생물제제를 발현시키고 형질주입 또는 형질전환된 spPSC 또는 ePSC에 의한 생물제제 제품의 발현을 유도하는 벡터로 형질전환되는 종으로부터 유래된다.
본 발명은 추가로 성인 세포로부터 spPSC를 생성하거나 또는 동물의 ePSC를 분리하는 단계 및 폴리펩타이드 또는 단백질이 줄기 세포에 의해 발현되는 조건 하에서 상기 폴리펩타이드 또는 단백질에 있어서 코팅하는 핵산으로 상기 spPSC 또는 ePSC를 형질주입 또는 형질전환시키는 단계를 포함하는 재조합 폴리펩타이드 또는 단백질의 제조 방법을 제공한다.
본 발명의 대안적인 실시형태에서, 상기 재조합 폴리펩타이드 또는 단백질을 투여하는 개체의 것으로서 동일한 인종 군의 세포로부터 상기 spPSC가 제조되거나 또는 ePSC가 분리된다. 다른 인종 군은 여러 가지 글라이코실화 패턴 및 하나의 군에서 다른 군으로의 뚜렷한 다형성을 가질 수 있다. 따라서, 본 발명에 따르면, 재조합 폴리펩타이드 및 단백질은 spPSC 또는 ePSC로부터 제조되며, 상기 ePSC 또는 spPSC는 폴리펩타이드 또는 단백질이 투여된 개체와 동일한 인종 군으로부터 분리된 세포로부터 생성된다. 개체에는 spPSC 또는 ePSC로부터 제조된 재조합 폴리펩타이드 또는 단백질을 투여할 것이며, 상기 spPSC는 제조되거나 또는 ePSC는 개체가 속하는 인종 군에 속하는 세포로부터 분리된다.
본 발명은 추가로 개체 동물에 폴리펩타이드 또는 단백질을 투여하는 방법을 제공하며, 상기 방법은 spPSC, 예컨대 유도 다능성 줄기 세포(iPSC)를 상기 동물의 세포로부터 제조하는 단계 또는 ePSC를 분리하는 단계, 및 상기 폴리펩타이드 또는 단백질이 상기 다능성 줄기 세포에 의해 발현되는 조건하에서 상기 폴리펩타이드 또는 단백질에 대해 코딩하는 핵산으로 상기 spPSC 또는 ePSC를 형질주입 또는 형질전환시키는 단계, 상기 폴리펩타이드 또는 단백질을 상기 유도 다능성 줄기 세포로부터 분리하는 단계, 및 상기 분리된 폴리펩타이드 또는 단백질을 상기 개체에 투여하는 단계를 포함한다.
본 발명은 폴리펩타이드 또는 단백질의 맞춤형 제조 방법을 제공하며, 이로 인해서 spPSC 또는 ePSC가 상업적으로 실행가능하고 유용하도록 한다. 본 발명은 장기간 또는 만성 용도에 있어서 폴리펩타이드 또는 단백질에 의해 통상적으로 발생하는 중화 항체 형성의 문제를 극복하기 위해서 환자에 특이적인 폴리펩타이드 또는 단백질을 제조하기 위한 환자 특이적 spPSC 또는 ePSC의 제조 방법에 관한 것이다. 상기 환자는 임의의 동물일 수 있으며, 바람직하게는 포유류이고, 보다 바람직하게는 인간이다. 본 발명은 또한 핵산 또는 바이러스를 제조하는 방법을 제공하며, 상기 방법은 목적하는 핵산 또는 바이러스를 제조하는 조건하에서 벡터로 spPSC 또는 ePSC를 형절주입 또는 형질전환시키는 단계를 포함한다.
또한, 본 발명은 치료 용도의 밀접하게 매칭된 번역 이후 변형된 생물제제의 제조를 위한 환자-특이적 및 기간 또는 세포형 특이적 세포주를 얻기 위한 방법을 제공한다. 환자 특이적 줄기 세포는 SCNT, 유도 재프로그래밍, 단위생식 또는 ANT-OAR 재프로그래밍 방법을 사용하여 유도할 수 있거나, 표적 환자로부터 분리할 수 있다. 상기에 의해 유래되거나 분리된 다능성 줄기 세포는 예를 들면 삽입 또는 에피솜 발현 백터 또는 상동 재조합 방법을 사용하여 원하는 치료를 발현시키기 위한 표준 분자 생물공학 절차를 사용하여 유전적으로 변형시킬 수 있다. 유전적 변형 세포주는 배양으로 늘릴 수 있으며, 제조된 생물제제의 수명을 기본으로 예정될 수 있는 주기적 생물제제 생산 시행을 위해서 기탁될 수 있다(실시예 2). 대안적으로, 유래된 환자 특이적 줄기 세포주는 목적하는 치료 단백질을 가장 높은 수준으로 발현시키는 것이 일반적인 세포 형태로 배양으로 분화시킨 다음에 생물제제 제조에 사용될 수 있다. 분화는 각 생산 시행에 대해서 실행될 수 있거나, 제조된 치료요법의 수명을 기초로 이후 생산 시행을 위해 기탁된 분화된 환자 특이적 세포주 및 대규모로 시행될 수 있다(실시예 3).
또한, 글라이코실화 패턴 및 기타 번역 후 변형은 조직 및 세포 형태에 따라 다르다고 알려져 있으므로, 환자 특이적 줄기 세포주는 생물제제를 내생으로 발현시키는 세포 타입 중에서 또는 기관으로부터 분리된 성인 또는 체세포로부터 제조할 수 있다. 예로서, 다음에 SCNT, PGA, ANT-OAR 또는 재프로그래밍 기술을 적용시켜 생물제제 생산을 위한 다능성 세포주를 유도할 수 있다. 이렇게 해서 유래 또는 분리된 다능성 줄기 세포는 원하는 치료를 발현시키기 위한 표준 분자 생물공학 절차를 사용하여 유전적으로 변형시킬 수 있다. 유전적으로 변형된 세포주는 배양으로 늘릴 수 있으며, 생산된 생물제제의 수명을 기초로 지정될 수 있는 주기적 생물제제 생산 시행을 위해서 기탁될 수 있다(실시예 4 참조). 재프로그래밍된 세포(iPS 세포)의 "메모리" 특성의 추가의 이점을 취하면, 환자 및 조직 또는 세포-형태 특이적 iPS 세포는 내생적 번역 후 변형이 가능한 세포주를 보다 완전하게 생성하기 위해 원래의 이들 세포-형태로 다시 분화되도록 유도될 수 있다. 상기 iPS 세포는 원래의 세포 형태로 재-분화된 후에 또는 원래의 분리된 세포-형태로 재-분화되기 이전에 원하는 치료를 발현시키도록 유전적으로 변형될 수 있다(실시예 5 참조).
예를 들면, 성장 호르몬은 하수체 전엽에서 소마토트로프 셀에 의해 매우 높게 생성되는 것이 일반적이며, 또한 태반(영양 아층) 및 혀 및 음문 또는 항문의 피부 내의 세포에서 높게 발현된다. 인자 VIII 단백질 발현은 신장의 세관 세포에서 높으며, 인간 단백질 아틀라스에 따르면 다 조직 및 세포 형태에 의해 적당하게 발현된다. 항체는 일반적으로 비장 및 기타 림프계 기관의 배중심에서 성숙하는 B 세포에 의해서 생성된다. 항체-의존성 세포-매개 세포 독성(ADCC)이 높은 항체의 치료적 생성은 제조 세포주에 존재하는 IgG1 서브타입 항체의 2등분 위치에 N-아세틸글루코사민(GlcNac)을 둘 수 있는 GDP-D-만노스-4,6-데하이드라타제(GMD)의 수준에 의해서 결정된다(J Biol Chem., Vol. 273, pp. 14582-14587, 1998 및 BMC Biotechnol., 7:84-97 (2007). 높은 ADCC 활성을 갖는 항체의 생성은 CHO 제조 세포주를 사용하는 것이 항상 최적은 아니다(J Biol Chem. 278: 3466-3473, 2003). 본 발명은 제조된 항체, 융합 단백질 및 세포 세포독성 생물제제의 최적의 ADCC 활성을 위한 신규의 포유동물 세포주를 제공한다.
다량의 생물제제 생성과 관련이 있는 것으로 알려진 전사 인자는 환자 특이적 세포주로부터 발현 수준을 최적화하기 위해 원하는 유전자와 함께 형질주입시킬 수 있다. 예를 들면, 다량의 Pit-1 발현으로 성장 호르몬을 차단하거나 또는 방지하면서 세포 형태로 프로락틴 발현을 높일 수 있다(Genes Dev, 3: 946-958 1989).
단일클론 항체 생성은 Sino Biological Inc, Peoples Republic of China, morphogenics(Proc Natl Acad Sci, 103: 3557-3562, 2006)에 의해서 개발된 시스템 또는 기타 표준 생물공학 방법을 사용해서 유전자 코돈을 최적화함으로써 강화시킬 수 있다.
합성적으로 생산된 다능성 줄기 세포의 생성
임의의 형태인 합성적으로 생산된 다능성 줄기 세포를 본 발명의 맞춤형 생물제제를 제조하기 위해 사용할 수 있다. 두 개의 주요한 카테고리는 유도되거나 재프로그래밍된 다기능 줄기 세포(iPSC) 및 핵이식(SCNT), ANT-OAR 및 단위생식에 의해 제조된 줄기 세포이다.
체세포 핵 이식(somatic cell nuclear transfer: SCNT)은 적출된 난자에 성인 체세포의 핵을 주입하고, 준비된 수용기 자궁에 이식하여 정상 출산시켜 완전한 핵 유전적 클론을 수득하는 기술이다. 또한, 다능성 줄기 세포는 SCNT 방법을 사용해서 배양으로 유도하였다(Cell Reprogram. 12:105-113, 2010 및 Genome Res., 19: 2193-2201, 2009).
변형된 핵 이식-난모 세포 지원 재프로그래밍(altered nuclear transfer, oocyte assisted reprogramming: ANT-OAR)은 SCNT와 유사한 기술이지만 기부자 핵을 수용자 난자에 주입하기 이전에 유전적으로 변형시켜 ANT-난모 세포가 완전한 기관으로 분화 및 완전한 기관을 형성하는 것을 막는다(Genome Res. 19: 2193-2201, 2009).
단위 생식(parthenogenesis: PGA)은 또한 무투명대 핵 이식, 단위 생식의 활성화와 같은 기술; 및 SCNT와 같은 클로닝 기술을 사용해서 다능성 줄기 세포를 발생시키기 위해 사용되며, 단위 생식(PGA)은 또한 재프로그래밍된 줄기 세포를 발생시키기 위해서도 사용하였다(Cell Reprogram., 12: 105-113, 2010 및 Nature, 450:497-502 2007).
이러한 다능성 줄기 세포는 어느 정도 오랫동안의 장기간 배양으로 유지하여 재조합 단백질, DNA 및 바이러스용으로의 생물제제 제조에 있어서의 잠재적인 공급원으로 사용할 수 있다.
유도 또는 재프로그래밍된 spPSC

원래의 세포 타입으로의 재-분화되는 재프로그래밍된 세포에 대한 선호의 유사한 관찰은 인간 태아 신경 전구 세포의 비-바이러스성 재프로그래밍을 사용해서 확인하였다(PLoS One., 4: e7076-e7088, 2009). 수득된 재프로그래밍된 세포는 몇몇 다능성 마커, 즉 세 개의 배엽 모두 용의 마커를 발현하며, 배양으로 배양체, 및 그러나 유전자 칩 분석(GeneChip analysis)을 이용하여 기형종을 형성할 수 있으며, 저자들은 재프로그래밍된 신경 전구 세포가 신경 줄기 세포 유전자의 몇몇 발현을 보유한다는 것을 입증했다. 치료를 위해 표적화된 특수한 장기로부터의 체세포의 분리, 이러한 체세포의 재프로그래밍, spPSC의 내인성 "메모리"를 유지하도록 하는 배양에서의 짧은 기간의 팽창, 이후의 치료적 응용이 따르는 원래 세포 타입으로 재-분화시키는 것으로서 다능성 줄기 세포 방법을 사용할 수 있어 암 또는 기형종 형성의 위험을 줄여 환자를 치료할 수 있다.
재프로그래밍된 체세포는 1 내지 12 계대, 가장 바람직하게는 4 계대에서 사용할 수 있다. 재프로그래밍된 체세포는 표준 세포 생물학 및 분화 기술에 따라서 목적하는 재생성용 세포 타입으로 분화될 수 있다. 수득된 치료적 세포는 카테터, 예컨대 NOGAStar 또는 MyoStar 주입 카테터 또는 기타 승인된 카테터 주입 장치를 포함할 수 있는 표준 주입 기술을 사용해서 정맥 주사, 동맥 주사, 근육 주사 또는 기타 주입 방법으로 투여할 수 있다. 대안적으로, 치료적 세포는 최소 침습 또는 보다 침습적인 수술 방법에 의해서 표적 조직에 투여될 수 있다. 치료적 세포는 0 내지 15%, 보다 바람직하게는 5%의 자기유래의 인간 혈청 알부민을 함유하는 완충 조성물로 투여할 수 있다. 중추신경계 질환을 치료를 위해 치료적 세포는 자기 유래의 뇌척수액을 사용해서 완충되는 것이 바람직할 것이다. 치료적 세포는 또한 스캐폴드, 예컨대 콜라겐, 피브리노겐 또는 기타 세포외 매트릭스 또는 세포외 매트릭스들의 조합을 사용해서, 또는 스레드, 예컨대 알지네이트 또는 기타 표준 방법을 사용해서 만든 것을 사용해서 투여하여 제 위치에 세포를 유지하고, 재생 수선을 요구하는 장기와의 접촉을 유지할 수 있다.
바람직하게 최소 500개의 치료적 세포가 투여될 것이며, 통상적인 치료 요법에서는 15 밀리언 내지 500 밀리언 개의 세포를 이용할 것이다. 보다 바람직하게, 15 밀리언 내지 100 밀리언의 세포를 최적의 이점을 위해 투여할 것이다.
신경 질환, 예컨대 알츠하이머 질환, 파킨슨병, 뇌졸증, 헌팅턴병, 다발성 경화증, 마비 및 기타 중추 신경계(CNS) 질환을 위한 세포 치료에 있어서의 예로서는 공개된 고정 내시경을 사용해서 비점막을 분리하는 것이 바람직할 것이다(J Spinal Cord Med. 29:191-203, 2006). 신경 줄기 및 전구 세포 또는 후각 초성 세포는 재생 의약 분야에서 실행하고 있는 것으로서 잘 알려져 있는 방법에 따라서 비점막으로부터 분리된다. 대안적으로, 신경능 줄기 세포는 인간 모낭으로부터 분리될 수 있다(Folia Biol . 56:149-157, 2010). 가장 바람직하게는, 신경 줄기 세포는 규정 인자의 첨가에 의해 재프로그래밍되며, 계대 4에 있어서 팽창 및 계대배양되고, 규정된 인자의 첨가에 의한 특수한 중추 신경계 세포로의 재-분화가 이루어지며, 이후에 재생 요법을 위해 환자로 투여된다.
본 발명은 재생 의학에 있어서의 체세포로 안전하게 재프로그래밍되고, 재-분화되는 방법을 제공한다. 유용한 체세포는 완전하게 분화된 체세포, 전구 세포 또는 보다 원시적인 줄기 세포를 포함할 수 있다. 재생 요법을 요하는 장기에 따라서, 보다 원시적인 줄기 세포가 장기 천자, 생체 검사, 찰과 또는 수술적 접근에 의해서 보다 또는 덜 접근가능할 수 있다. 보다 원시적인 줄기 세포의 사용은 이러한 줄기 세포로의 접근이 가능한 경우에 바람직하다. 완전하게 분화된 체세포보다는 더 바람직하지만 전구 세포를 사용하는 것은 덜 바람직하다.
척추 손상 치료에 있어서, 비점막은 견고한 내시경에 의해서 제거되며, 신경 줄기 세포는 Reynolds 및 Weiss(Science, 255:1707-1710, 1992)에 의해서 1992년에 최초로 기술된 신경구 분석(neurosphere assay)을 이용해 분리하며, 표피 성장 인자(EGF) 및 염기성 섬유모세포 생장 인자(bFGF)는 비점막 배양 수일 동안 생존하는 세포에 첨가해서 신경구 성장을 자극하고, 신경 줄기 세포는 배양 7 내지 10일 내에 회수할 수 있다. 수득된 신경 줄기 세포는 5 내지 7일 동안 히그로마이신 선별로 OCT4 및 NANOG의 운반을 위한 에피솜 벡터를 첨가해서 재프로그래밍한다. 세포는 4 계대까지 팽창 및 계대배양하여 집적화가 이루어지지 않는 콜로니를 수득한 다음에 자가 뇌척수액 또는 규정된 인자를 사용해서 비점막 및 줄기-유사 신경 전구 세포로 분화한다. 손상된 척수는 표준 정중선 절개 및 뒤쪽 정중선 척수절개술을 사용하는 수술 중에 노출되며, 반흔 조직은 허용가능한 만큼 제거되고, 치료적 세포는 자가 뇌척수액 중에서 완충되며, 생물작용 스캐폴드 상에 접종된 후에 손상된 척수로 직접 적용된다.

재조합 단백질 생성을 위해 환자 특이적으로 합성적으로 생산된 다능성 줄기 세포의 유전적 변형

항체 제조 B 세포는 치료적 항체 생물제제를 제조하기 위해 환자 특이적 제조 세포주를 생성할 목적으로 말초혈, 골수 및 기타 손쉽게 접근가능한 조혈 세포 공급원으로부터 분리한다. B 세포는 CD19 발현을 기본으로 하는 이용가능한 키트를 사용해서 분리한다(StemCell Technologies). 희석 제한 또는 세포 선별 방법을 적용해서 가장 높은 수준의 면역글로불린(Ig)을 제조하는 세포를 선별할 수 있다. 짧은 팽창 후에 클론 고 Ig 제조 세포는 표준 재프로그래밍 기술을 사용해서 다능성 또는 전구체 상태로 재프로그래밍한다. 수득된 환자 특이적 줄기 세포는 표준 생물학적 기술 및 방법을 사용해서 목적하는 항체 유전자 구조물로 형질도입한다. 수득된 발현 항체 치료제제는 항체 치료제제용 물질 조성물의 소유자에 의해서 내밀히 유지되거나 또는 공개적으로 이용가능한 당업의 생물공학적 방법의 상태에 따라 처리 및 정제된다. 목적하는 정제 항체 생성물을 수득하기 위한 방법에는 이온 교환 크로마토그래피가 포함된다.

고 활성 ADCC 항체의 생성을 위한 환자 특이적 세포주의 생성
면역글로불린의 N-아세틸글루코사민(GlcNac) 번역 후 변형은 항체-의존적 세포-변형 독성(ADCC)에 중요하며, 비-퓨코실화 GlcNac 잔기는 Fc 감마 수용체에 있어서 매우 높은 친화도를 갖는다[Mori K., et al., "Non-fucosylated therapeutic antibodies: the next generation of therapeutic antibodies", Cytotechnology., 55:109-114(2007)].
따라서, 높은 수준의 ADCC를 갖는 항체 치료제제를 목적하는 경우에 적당한 수준에서 GlcNac를 이동시키고, 비-퓨코실화된 GlcNac를 남길 수 있는 환자 특이적 세포주가 바람직하다. 암종 세포는 GMD를 높은 수준으로 발현시키는 것으로 알려져 있으므로, 암 줄기 세포 및 다능성 세포는 유사한 유전자 표시를 가지므로, 다능성 세포는 또한 번역 후 GlcNac 부착에 원인이 될 수 있는 효소인 GMD를 높은 수준으로 발현시키는 것으로 기대할 수도 있다. 정상 조직 중 결장 및 췌장은 GMD를 가장 높은 수준으로 발현한다. 항체를 퓨코실화시키는 효소에 원인이 될 수 있는 FUT8의 부족은 리툭시맙(Rituximab) 또는 허셉틴(Herceptin)과 같은 효험에 있어서 ADCC 활성에 의존하는 치료적 항체 생성용의 환자 특이적 줄기 세포주로 바람직할 수 있다. 예를 들면, 쥐 하이브리도마 YB2/0 세포에서 생성되는 단클론 항체는 CHO 세포를 사용해서 생성되는 동일한 단클론 항체보다 50배 높은 ADCC 활성을 갖는다. B 세포 림프종뿐만 아니라 지방조직-유래 줄기 세포 및 생식 세포주는 FUT8의 평균 수준보다 더 높게 발현하며, 반면에 조혈 줄기 세포(hematopoietic stem cell: HSC), 미성숙 B 세포, 정상 골격 근육은 평균 FUT8보다 낮게 발현한다.

- 제1항에 있어서, 상기 수용성 수용체는 TNFα, TNFβ 및 Blys로 이루어진 군으로부터 선택되는 표적에 결합하는 것인, 재조합 폴리펩타이드 또는 단백질의 제조방법.
- 제1항에 있어서, 형질주입된 spPSC를 불멸화시키는(immortalizing) 단계를 추가로 포함하는, 재조합 폴리펩타이드 또는 단백질의 제조방법.
