멜라토닌 일일 투여 유닛{Daily melatonin dosing units}
본 발명은, 전신적인 효과는 나타내지 않으면서 국소 부위에만 효과를 나타내는 양의 멜라토닌 또는 멜라토닌 유도체의 1회 투여량을 포함하는, 국소 적용을 위한 약제 또는 화장료용 1회용 용기에 관한 것이다.
멜라토닌-함유제는 다양한 의학 및 화장료용 목적으로 사용될 수 있다. 예를 들면, DE 101 10 418.9는, 피부 및 모발에 대한 국소 적용을 위한 멜라토닌-함유 조성물을 개시하고 있다.
본 발명은 첨부된 도면 1 과 2, 그리고 실시예를 통하여 더 설명될 것이다.

본 발명의 주제는, 전신적인 효과는 나타내지 않으면서 국소 부위에만 효과를 나타내는 양의 멜라토닌 또는 멜라토닌 유도체의 1회 투여량을 함유하는, 국소 적용을 위한 약제 또는 화장료용 1회용 용기에 관한 것이다.
본 발명에 따른 1회용 용기는, 정상적인 멜라토닌 혈장 레벨에 해로운 변화를 유발하지 않으면서 국소 부위에만 유효한 멜라토닌 또는 멜라토닌 유도체의 1회 투여량의 적용을 가능하게 한다.
본 출원의 본문에서 사용되는 "멜라토닌 또는 멜라토닌 유도체"라는 표현은, 멜라토닌 및/또는 멜라토닌 유도체 및 염, 에스테르 또는 그 착물을 포함한다. 바람직한 멜라토닌 유도체는, 예를 들면, 5-메톡시트립타민, 5-메톡시트립토판, 5-메톡시트립토폴, 5-메톡시인돌-3-아세트산 및 6-히드록시멜라토닌을 포함한다.
본 발명에 따른 1회 투여량은, 혈장 레벨의 심각한 변화 없이 치료적 효과 및/또는 긍정적인 미용 효과를 가능하게 하는 투여량을 나타낸다. 본 발명에 따르면, 멜라토닌 약 0.001 내지 약 1.0 mg의 1회 투여량이 바람직하며, 멜라토닌 약 0.01 내지 약 0.2 mg의 1회 투여량이 더욱 바람직하다. 약제의 의도하는 용도 또는 도포 부위에 따라서, 1회 투여량은 하루에 1 회 또는 수 회 도포될 수 있다. 바람직한 구현예에서, 1회 투여량은, 일일 투여량에 해당하며, 더욱 바람직하게는, 저녁에 사용되는 일일 투여량에 해당한다.
의약 또는 화장료는 바람직하게는 액상 제형으로 존재한다. 약제의 국소적 적용을 위한 적절한 제형의 예는, 용제, 현탁제, 에멀션, 마이크로에멀션, 나노시스템, 크림, 겔, 로션, 스프레이, 폼(foam) 또는 연고, 그리고 국소적으로 적용될 수 있는 기타 제형이다. 특히 본 약제는, 화장료 용액의 형태로 바람직하게 제공된다.
그 의도하는 용도에 따라서, 본 발명에 따른 1 회용 용기 내에 함유된 의약 또는 화장료는, 여러 가지 농도의 멜라토닌 또는 멜라토닌 유도체를 함유할 수 있다. 본 약제는, 바람직하게는, 약 0.001 내지 약 0.01 중량%, 더욱 바람직하게는 약 0.003 내지 0.004 중량%의 농도의 멜라토닌 또는 멜라토닌 유도체를 포함한다. 특히 바람직한 농도는 약 0.0033 중량%이다.
한 구현예에서, 치료제 또는 화장료는 멜라토닌 또는 멜라토닌 유도체를 단일 유효 성분으로서 포함한다. 그러나 본 약제는, 또한 비타민 A, 비타민 A 산 또는 기타 비타민 A 유도체, 비오틴 및/또는 징코 빌로바와 같은, 한 가지 이상의 다른 활성 성분도 포함할 수 있다.

본 발명에 따른 조성물은 모발 성장을 촉진하는 데에, 특히 남성형 또는 여성형 탈모증을 예방 및/또는 치료하는 데에 사용될 수 있다. 특히 바람직하게는, 남성형 안드로겐성 탈모증, 여성형 안드로겐성 탈모증, 남성형 확산성 탈모증, 및 여성형 확산성 탈모증이다.
의도하는 적용분야 또는 용도에 따라서, 본 발명에 따른 1회용 용기는 여러 가지 부피의 의약 또는 화장료를 포함할 수 있다. 바람직한 구현예에서, 상기 1회용 용기는 약 2.5 내지 약 3.5 ml, 바람직하게는 약 2.9 내지 약 3.2 ml의 약제를 포함한다. 특히 바람직한 용량은 약 3.0ml이다.
치료제 또는 화장료는 종래에 알려진 적절한 방법을 이용하여 용기 내로 충전할 수 있다. 약제는 바람직하게는, GMP 조건 하에서 용기에 충전한다. 이러한 방법으로써, 약제를 확실히 멸균상태로 할 수 있고, 약제가 도포될 때 균 감염에 관한 문제가 발생하지 않는다.
본 발명에 따른 1회용 용기는 원칙적으로 어떠한 원하는 재료로써도 만들 수 있으며, 구체적으로, 식품 및 약품의 포장용으로 승인된 재료를 이용한다. 바람직한 재료의 예는, 플라스틱 및 플라스틱 혼합물이다. 특히 적절한 플라스틱의 예는, 폴리에틸렌, 염화 폴리비닐, 폴리스티렌, 폴리프로필렌, 폴리시클로올레핀 또는 이들의 혼합물 또는 공중합체이며, 특히 바람직한 것은 폴리에틸렌 또는 폴리시클로올레핀이다.
원하는 외관 모양에 따라서, 1회용 용기는 투명, 불투명, 무색 또는 유색일 수 있다. 예를 들면, 원하는 염료를 플라스틱에 첨가하여, 유색 용기를 얻을 수 있다. 만약 약제가 빛에 민감한 물질을 포함하고 있다면, 불투명 용기를 사용하는 것이 바람직하다. 왜냐하면 불투명 용기는 부가적으로 빛으로부터의 보호를 제공하기 때문이다.
본 발명에 따른 1 회용 용기는, 원칙적으로 어떠한 원하는 형상이라도 가질 수 있다. 바람직한 용기의 예는 도 1 및 2에 도시되어 있다. 적절한 1회용 용기는 바람직하게는, 본 약제가 포함되어 있는 부분, 및 소정의 파쇄 라인을 통하여 저부에 연결된 폐기가능한 마개 부분을 포함한다.
용기의 간편하고 안전한 조작은, 바람직하게는, 돌려서 여는 폐기가능한 마개 부분을 상부에 포함함으로써 보장된다. 따라서 용기는 용이하게 열릴 수 있다. 또한 열린 후에, 압력을 가하는 경우만 약제가 나올 수 있다. 이러한 압력은 예를 들면, 도포하는 동안 손으로 가할 수 있다. 이러한 특성으로 인하여, 사용자는 약제를 정확하게 투여할 수 있고, 표적 부위에 위치시킬 수 있다. 만약 약제를 두피 또는 모발에 도포하고자 한다면, 열린 용기를 빗처럼 모발 사이로 직접 움직여서 도포한다. 이를 통하여 매우 수월한 작업이 가능하고, 용기를 안전하게 취급할 수 있으며 약제의 투여량을 정확하게 맞출 수 있다.
의도하는 용도에 따라서, 1회용 용기는, 그 표면에 인쇄되거나 요철로 된 문자를 포함하며, 그리고/또는 라벨이 제공된다.
더욱이, 둘 이상의 1회용 용기는 분리가능한 방법으로 서로 연결될 수 있다. 예를 들면, 바람직하게는 5 또는 10 개의 용기를 서로 연결하여 배열을 형성한다. 용기 또는 용기의 배열은 또한 포장할 수 있는데, 바람직하게는 차광성 포장, 특히 바람직하게는 예를 들어 알루미늄 백에 포장할 수 있다.
본 출원에서 사용된 "알루미늄 백"이라는 표현은, 알루미늄으로 만들어진 백, 및 또한 예를 들면 외측 또는 내측에 알루미늄을 피복한 백을 포함한다. 예를 들면, 하나 이상의 플라스틱층 및/또는 종이층으로 만들어진 백을 알루미늄으로 피복할 수 있다. 바람직하게는, 백은, 예를 들면, 폴리에틸렌, 폴리에스테르 및/또는 종이를 부가적으로 포함하는 알루미늄-함유 박층 호일을 포함한다.
포장, 특히 차광성 포장은, 약제의 효능을 손상시키지 않고 상당히 긴 시간 동안 저장을 가능하게 한다. 더욱이, 그러한 포장은 가스 손실에 대한 보호를 제공할 수 있다.
의도하는 용도에 따라서, 하나 이상의 1회용 용기가 포장된 알루미늄 백은, 그 위에 인쇄를 할 수 있고 그리고/또는 라벨을 붙일 수 있다.
본 발명의 또 다른 구현예에서는, 1회용 용기의 배열을 복수 개 포함하는 포장 단위를 제공한다.
실시예 1 : 활성 물질로 멜라토닌, 징코 빌로바 및 비오틴을 갖는 본 발명의 조성물의 구성
본 조성물은 징코 빌로바 건조 추출물 0.05 중량%, 비오틴 0.01 중량%, 멜라토닌 0.0033 중량% 및 부가적인 추출물, 물 및 에탄올을 포함한다. 본 조성물의 pH는 3.5 내지 4이다.
실시예 2 : 임상 시험
실시예 1에 기재된 조성물을, 위약 대조 및 크로스오버 디자인으로써, 무작위 이중맹검법으로 8 명의 건강한 여성 환자에게 테스트하였다. 일일 투여량은 0.1 mg 멜라토닌 (조성물 3 ml)으로 하였으며, 이것을 14일 동안 잠들기 전에 매일 두피에 도포하였다.
소변 및 혈액 샘플을 환자로부터 채취하여, 멜라토닌 및 대사산물 6-히드록시멜라토닌 설페이트에 대해 분석하였다.
멜라토닌 또는 위약으로 치료한 환자의 멜라토닌 혈장 프로파일은, 내생성 멜라토닌에 대한 문헌에 기록된 농도/시간 프로파일에 상응하는 것으로 밝혀졌다. 이러한 사실은, 14일 동안의 멜라토닌-함유 조성물의 국소 적용이 내생성 멜라토닌 분비를 변화시키지 않았음을 증명한다.
14 일 동안의 조성물의 투여는, 멜라토닌의 혈장 농도 또는 6-히드록시멜라토닌 설페이트의 뇨 중 농도를, 생리학적 농도 이상으로 증가시키지 않았다. 멜라토닌-함유 조성물의 국소 적용은 부작용도 없었다. 또한 반응 시간 또는 피질 자극 파라미터에 대한 어떠한 영향도 발견되지 않았다.
모든 것을 고려할 때, 본 조성물의 투여가 전신적인 효과를 유발하지 않는다는 결론을 얻을 수 밖에 없었다.

Claims (29)

  1. 전신적인 효과는 나타내지 않으면서 국소 부위에만 효과를 나타내는 양의 멜라토닌 또는 멜라토닌 유도체의 1회 투여량을 포함하는, 국소 적용을 위한 의약 또는 화장료용 1 회용 용기.
  2. 제 1 항에 있어서, 상기 1회 투여량은, 0.001 내지 1.0 mg의 멜라토닌 또는 멜라토닌 유도체인 것을 특징으로 하는 용기.
  3. 제 1 항 또는 제 2 항에 있어서, 상기 1회 투여량은 0.01 내지 0.2 mg의 멜라토닌 또는 멜라토닌 유도체인 것을 특징으로 하는 용기.
  4. 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 상기 1회 투여량은 일일 투여량에 해당하는 것을 특징으로 하는 용기.
  5. 제 1 항 내지 제 4 항 중 어느 한 항에 있어서, 상기 약제는 액상 제형으로 존재하는 것을 특징으로 하는 용기.
  6. 제 5 항에 있어서, 상기 액상 제형은 0.001 내지 0.01 중량%의 멜라토닌 농도를 갖는 것을 특징으로 하는 용기.
  7. 제 6 항에 있어서, 상기 액상 제형은 0.003 내지 0.004 중량%의 멜라토닌 농도를 갖는 것을 특징으로 하는 용기.
  8. 제 1 항 내지 제 7 항 중 어느 한 항에 있어서, 상기 약제는 단일 활성 물질로서, 멜라토닌 또는 멜라토닌 유도체를 포함하는 것을 특징으로 하는 용기.
  9. 제 1 항 내지 제 7 항 중 어느 한 항에 있어서, 상기 약제는 활성 물질로서, 멜라토닌 또는 멜라토닌 유도체와, 비오틴 및/또는 징코 빌로바의 조합물을 포함하는 것을 특징으로 하는 용기.
  10. 제 1 항 내지 제 7 항 중 어느 한 항에 있어서, 상기 약제는 활성 물질로서, 멜라토닌 또는 멜라토닌 유도체와, 비타민 A, 비타민 A 산, 또는 기타의 비타민 A 유도체의 조합물을 포함하는 것을 특징으로 하는 용기.
  11. 제 1 항 내지 제 10 항 중 어느 한 항에 있어서, 2.5 내지 3.5 ml의 약제를 포함하는 것을 특징으로 하는 용기.
  12. 제 11 항에 있어서, 2.9 내지 3.2 ml의 약제를 포함하는 것을 특징으로 하는 용기.
  13. 제 1 항 내지 제 12 항 중 어느 한 항에 있어서, 상기 약제는, GMP 조건 하에서 용기에 충전되는 것을 특징으로 하는 용기.
  14. 제 1 항 내지 제 13 항 중 어느 한 항에 있어서, 플라스틱으로 만들어진 것을 특징으로 하는 용기.
  15. 제 14 항에 있어서, 폴리에틸렌, 폴리프로필렌, 염화폴리비닐, 폴리스티렌, 또는 이들의 혼합물로 만들어진 것을 특징으로 하는 용기.
  16. 제 1 항 내지 제 15 항 중 어느 한 항에 있어서, 무색인 것을 특징으로 하는 용기.
  17. 제 1 항 내지 제 15 항 중 어느 한 항에 있어서, 유색인 것을 특징으로 하는 용기.
  18. 제 1 항 내지 제 17 항 중 어느 한 항에 있어서, 투명인 것을 특징으로 하는 용기.
  19. 제 1 항 내지 제 17 항 중 어느 한 항에 있어서, 불투명인 것을 특징으로 하는 용기.
  20. 제 1 항 내지 제 19 항 중 어느 한 항에 있어서, 상부에, 돌려서 여는 1회용 마개 부분을 구비하는 것을 특징으로 하는 용기.
  21. 제 1 항 내지 제 20 항 중 어느 한 항에 있어서, 용기를 연 후 압력을 가할 때만 약제가 나오는 것을 특징으로 하는 용기.
  22. 제 1 항 내지 제 21 항 중 어느 한 항에 있어서, 그 위에 인쇄되거나 요철로 된 문자를 가지며, 그리고/또는 라벨이 제공된 것을 특징으로 하는 용기.
  23. 분리가능한 방식으로 서로 연결된, 제 1 항 내지 제 22 항 중 어느 한 항에 따른 용기의 복수 개의 배열.
  24. 제 23 항에 있어서, 5 개의 용기가 분리가능한 방식으로 서로 연결된 것을 특징으로 하는 배열.
  25. 제 23 항에 있어서, 10 개의 용기가 분리가능한 방식으로 서로 연결된 것을 특징으로 하는 배열.
  26. 제 23 항 내지 제 25 항 중 어느 한 항에 있어서, 알루미늄 백 내에 포장되는 것을 특징으로 하는 배열.
  27. 제 26 항에 있어서, 상기 백이 알루미늄-함유 박층 호일로 이루어지고, 폴리에틸렌, 폴리에스테르 및/또는 종이를 부가적으로 포함하는 것을 특징으로 하는 배열.
