本発明は、作用物質としてメラトニン、イチョウおよびビオチンの組合せ物を含有する組成物に関する。この組成物は、殊に毛髪における局所的使用のための調製物の製造に適している。
毛髪の成長にプラスに影響を及ぼすためのメラトニンを使用することは、公知である。メラトニンは、毛髪の成長を制御する、毛髪の減退の増殖細胞との選択的な相互作用のために、毛髪の成長に対して刺激作用を有する。この刺激作用は、メラトニンの種々の濃度を使用する場合に観察されうる。
米国特許第５９５２３７３号明細書には、作用物質として１つ以上のフラボニドを含有する組成物が施こされる、皮膚の治療方法が記載されている。毛髪の成長の改善のために、メラトニンを作用物質として使用することは、何も指摘されていない。
米国特許出願ＵＳ２００２／００６１８７０には、聴力の損失または耳鳴りを軽減させるための組成物が開示されている。錠剤の形で投与される前記組成物は、数多くの作用物質を含有し、この作用物質の中には、場合によってはメラトニン、ビオチンまたはイチョウも含まれる。毛髪の成長を改善するための使用または局所的投与については、指摘されていない。
意外なことに、本発明を生じる試験の範囲内で、イチョウおよびビオチンの添加は、メラトニンとの同時の使用の際にメラトニンの作用を増強させ、頭皮表面上でのメラトニンの保持率を高めることが確認された。
従って、本発明の対象は、作用物質として（ａ）メラトニンまたはその誘導体、（ｂ）抽出物としてのイチョウおよび／またはその１つ以上の内容物質および（ｃ）ビオチンを含有する組成物である。この組成物は、有利に殊に頭皮上への施与のために製薬学的使用または／および化粧術的使用に適している局所的調製物である。
本発明による組成物は、作用物質の組合せ物を含有する。この組合せ物の第１の成分は、メラトニンまたはメラトニン誘導体である。メラトニン誘導体は、有利に５−メトキシトリプタミン、５−メトキシトリプトファン、５−メトキシトリプトフォール、５−メトキシインドール−３−酢酸および６−ヒドロキシ−メラトニンから選択される。また、この物質と共に、メラトニンの生理的に認容性の塩、エステルおよび複合体化合物が使用されてもよい。
この組成物の第２の成分は、イチョウである。イチョウは、種々の物質交換機能に対してプラスの影響を及ぼす。更に、イチョウの好ましい性質は、ラジカル捕捉能力を強化させることである。好ましくは、イチョウは、抽出物として、殊に乾燥抽出物として使用されるかまたは／およびその１つ以上の内容物質が使用される。特に本発明による組成物の使用に適当なのは、イチョウの葉からの乾燥抽出物である。
更に、本発明による組成物の成分は、ビオチンである。ビオチンは、例えばトランスカルボキシル化反応の場合に脂肪酸物質代謝およびアミノ酸物質代謝におけるコエンザイムとして作用を発揮する。
組成物中の作用物質の濃度は、使用に応じて広い範囲内で変動可能である。好ましくは、作用物質の濃度は、それぞれ独立に組成物の全質量に対して０．０００１％（質量）〜１％（質量）の範囲内にある。メラトニンまたはメラトニン誘導体の濃度は、有利に０．００１％（質量）〜０．０１％（質量）である。イチョウの濃度は、有利に０．０１％（質量）〜０．１％（質量）である。ビオチンの濃度は、有利に０．００２％（質量）〜０．０５％（質量）である。
１回の適用当たり、有利には、メラトニンまたはメラトニン誘導体約０．００１ｍｇ〜約１０ｍｇ、有利に０．０１ｍｇ〜約１ｍｇが投与される。
本発明による組成物の３つの作用物質成分は、毛髪の成長を促進し、老化保護を調整し、毛髪の濃さを改善するかまたは／および休止期率を減少させるという作用プロフィールを発揮する。毛髪の減退に対するメラトニンの作用は、イチョウとビオチンとの組み合わせによって通常強化される。従って、１回の適用当たりのメラトニンの極めて低い濃度、例えば０．００１〜１０ｍｇ、有利に約０．０１〜１ｍｇは、申し分ない作用をもって使用されてもよい。
メラトニン、イチョウおよびビオチンの組合せ物は、適当な担持剤系中に添加されてよい。好ましくは、作用物質は、溶解してかまたは／および分散して液状、半固体または固体の担持剤系中に含有されている。適当な担持剤系の例は、液体、例えば水または緩衝水溶液、生理的に認容性の有機溶剤、例えばエタノールまたはこれらの組合せ物、水中油型乳濁液、油中水型乳濁液、脂肪、ポリエチレングリコール、プロピレングリコール、グリセリン、乳化剤またはこれらの組合せ物ならびに製薬学的調製物および化粧術的調製物中に使用される担持剤または助剤である。
水性担持剤系、即ち５０％（質量）またはそれ以上、殊に６０％（質量）またはそれ以上の含水量を有する担持剤系の使用は、好ましい。水性担持剤系のｐＨ値は、有利にｐＨ３〜ｐＨ６の範囲内、特に有利にｐＨ３．５〜ｐＨ４の範囲内で調節される。
この担持剤系は、有利に毛髪の減退に対して意図的に作用物質の放出を可能にする。それによって、毛髪の減退における作用物質の位置決めは、最適化される。更に、作用物質メラトニン、イチョウおよびビオチンの制御された吸収は、担持剤系によって可能になることができる。
更に、適当な担持剤系中のメラトニンをイチョウおよびビオチンと一緒に投与することは、メラトニン吸収の抑制または遅延を生じることができる。それによって、標準的なヒトの血漿濃度は、影響を及ぼされない。その上、作用物質のよりいっそう長く維持される作用を達成することができる。
担持剤系中で調製することにより、化粧術において、殊に化粧用溶液の形で使用される安定した調製物を得ることができる。
特殊な調製物系、例えばリポソーム、ナノソーム（nanosome）または固体の包接担体、例えばアガロースを使用する場合には、作用物質、殊に全ての作用物質の少なくとも１つの制御された放出を可能にする組成物を得ることができる。
本発明による組成物は、殊に例えば毛髪への製薬学的使用または／および化粧術的使用に適している。この組成物は、溶液、懸濁液、乳濁液、マイクロエマルジョン、ナノ系、クリーム剤、ゲル剤、ローション、スプレー剤、フォーム剤または軟膏として存在することができるかまたは局所的使用に適した全ての別の形で存在することができる。この組成物は、通常、チューブ、瓶、スプレー用瓶、プラスター、海綿および織物担持剤またはプラスチック担持剤から選択された包装物系または使用系中ならびに毛髪への適用に適した別の系中で使用される。特に好ましい包装物系は、適用のために単位用量が準備される１回用量−アンプル剤である。アンプル剤は、種々の材料、例えばガラスまたはプラスチックから完成されていてよい。特に好ましいのは、プラスチックアンプル剤である。それというのも、プラスチックアンプル剤は、安全で良好な取り扱いを保証するからである。
本発明による組成物は、作用物質とともに１つ以上の化粧術的助剤もしくは添加剤または／および製薬学的助剤もしくは添加剤、例えば濃稠化剤、鉱物質、油、ビタミン、例えば殊にレチン酸の形でのビタミンＡ、または芳香物質を含有することができる。
本発明による組合せ調製物は、殊に夕方に使用するのが望ましく、作用を殊に夜間に発揮する。この種の使用の場合、前記組成物の作用は、特に強力である。本発明による組成物は、殊に毛髪の減退に刺激を与えるためおよび毛髪の成長にプラスの影響を与えるために適している。
治療は、殊に毛髪への使用に適した局所的投与形で行なわれ、この投与形は、それぞれの使用目的に適した相応する、作用物質の濃度を有する。
本発明による組成物は、毛髪の成長の促進、殊に男性の場合または女性の場合の脱毛症予防または／および治療に使用されることができる。特に好ましい適用症は、男性のタイプのアンドロゲン性脱毛症、女性のタイプのアンドロゲン性脱毛症、男性のタイプの広汎性脱毛症および女性のタイプの広汎性脱毛症である。
更に、本発明を次の実施例につき詳説する。
実施例：作用物質メラトニン、イチョウおよびビオチンを有する本発明による調製物の組成物
この組成物は、イチョウ乾燥抽出物０．０５質量％、ビオチン０．０１質量％、メラトニン０．００３３質量％ならびに他の添加剤、水およびエタノールを含有する。この組成物のｐＨ値は、３．５〜４である。

Claims (19)

  1. 組成物において、作用物質として
    （ａ）メラトニンまたはその誘導体、
    （ｂ）イチョウおよび
    （ｃ）ビオチンを含有することを特徴とする、組成物。
  2. ５−メトキシトリプタミン、５−メトキシトリプトファン、５−メトキシトリプトフォール、５−メトキシインドール−３−酢酸および６−ヒドロキシ−メラトニンならびにその生理的に認容性の塩、エステルおよび複合体化合物から選択されたメラトニンまたはメラトニン誘導体を含有する、請求項１記載の組成物。
  3. イチョウを乾燥抽出物および／またはその１つ以上の内容物質として含有する、請求項１または２記載の組成物。
  4. 作用物質の濃度がそれぞれ組成物の全質量に対して０．０００１％（質量）〜１％（質量）の範囲内にある、請求項１から３までのいずれか１項に記載の組成物。
  5. 作用物質が溶解してかまたは／および分散して液状、半固体または固体の担持剤系中に存在する、請求項１から４までのいずれか１項に記載の組成物。
  6. 作用物質の少なくとも１つの制御された放出を可能にしている、請求項１から５までのいずれか１項に記載の組成物。
  7. 製薬学的使用または／および化粧術的使用のための請求項１から６までのいずれか１項に記載の組成物。
  8. 局所的調製物としての請求項１から７までのいずれか１項に記載の組成物。
  9. 毛髪中で使用するための請求項７または８記載の組成物。
  10. 作用物質の少なくとも１つの経皮的吸収が制御可能である、請求項８または９記載の組成物。
  11. 溶液、懸濁液、乳濁液、マイクロエマルジョン、ナノ系、クリーム剤、ゲル剤、ローション、スプレー剤、フォーム剤または軟膏としての請求項１から１０までのいずれか１項に記載の組成物。
  12. プラスチックまたはガラス、チューブ、瓶、スプレー用瓶、プラスター、海綿および織物担体またはプラスチック担体からなる１回用量−アンプル剤から選択された包装物系または使用系中での請求項１から１１までのいずれか１項に記載の組成物。
  13. イチョウおよびビオチンの添加によるメラトニンの毛髪濾胞の刺激作用を強化するための請求項１から１２までのいずれか１項に記載の組成物。
  14. 毛髪の成長を促進させるための請求項１から１３までのいずれか１項に記載の組成物の使用。
  15. 毛髪の減退を予防または／および治療する薬剤を製造するための請求項１４記載の使用。
  16. 男性の場合の毛髪の減退を治療するための請求項１５記載の使用。
  17. 女性の場合の毛髪の減退を治療するための請求項１５記載の使用。
  18. １回の適用当たりメラトニン０．００１ｍｇ〜１０ｍｇが投与される、請求項１４から１７までのいずれか１項に記載の使用。
  19. １回の適用当たりメラトニン０．０１ｍｇ〜１ｍｇが投与される、請求項１８記載の使用。
